Early prediction is still insufficient in clinical practice. Although most pre- eclamptic patients have pathological uterine perfusion in the second trimester, perfusion disturbance has a positive predictive accuracy (PPA) only of approximately 30%. Methods Non-invasive continuous blood pressure recordings were taken simultaneously via a finger cuff for 30 min. Time series of systolic as well as diastolic beat-to-beat pressure values were extracted to analyse heart rate and blood pressure variability and baroreflex sensitivity in 102 second- trimester pregnancies, to assess predictability for pre-eclampsia (n = 16). All women underwent Doppler investigations of the uterine arteries. Results We identified a combination of three variability and baroreflex parameters to best predict pre-eclampsia several weeks before clinical manifestation. The discriminant function of these three parameters classified patients with later pre-eclampsia with a sensitivity of 87.5%, a specificity of 83.7%, and a PPA of 50.0%. Combined with Doppler investigations of uterine arteries, PPA increased to 71.4%. The model was fit to time-accuracy data of memory-updating tasks from four experiments using nonlinear mixed effect (NLME) models as a framework. The model gave a good account of the data from a numerical and a spatial task version. The performance pattern in a combination of numerical and spatial updating could be explained by variations in the interference parameter: assuming less feature overlap between contents from different domains than between contents from the same domain, the model can account for double dissociations of content domains in dual-task experiments. Experiment 3 extended this idea to similarity within the verbal domain. The decline of memory accuracy with increasing memory load was steeper with phonologically similar than with dissimilar material, although processing speed was faster for the similar material. The model captured the similarity effects with a higher estimated interference parameter for the similar than for the dissimilar condition. The results are difficult to explain with alternative models, in particular models incorporating time-based decay and models assuming limited resource pools. In general, genetic associations between resistances to multiple natural enemies tended to be positive regardless of the breeding design, type of attacker, and type of host plant. Positive genetic correlations between resistances were stronger when both attackers were pathogens or generalist herbivores and when resistance to different enemies was tested independently, suggesting that generalists may be affected by the same plant resistance traits and that interactions among natural enemies are common. T1 - A piezoelectric sensor with propidium as a recognition element for cholinesterases N2 - A piezoelectric biosensor has been developed on the basis of the reversible acetylcholinesterase (AChE) inhibitor propidium. The propidium cation was bound to a 11-mercaptoundecanoic acid monolayer on gold-coated quartz crystals. The immobilization was done via activation of carboxyl groups by 1,3-dicyclohexylcarbodiimide (DCC). Different types of cholinesterases (acetyl- and butyryl-ChE) from different origins were tested for their binding ability towards the immobilized propidium. Binding Studies were performed in a flow system, Furthermore, catalytically active and organophosphate-inhibited enzyme were compared re-aiding their binding capability. The binding constants were derived by using an one to one binding model and a refined model also including rebinding effects. It was shown that organophosphorylation of the active site hardly influences the affinity of AChE towards propidium. However, there is no standard protocol for describing such simulation models, which can make them difficult to understand and to duplicate. This paper presents a proposed standard protocol, ODD, for describing IBMs and ABMs, developed and tested by 28 modellers who cover a wide range of fields within ecology. This protocol consists of three blocks (Overview, Design concepts, and Details), which are subdivided into seven elements: Purpose, State variables and scales, Process overview and scheduling, Design concepts, Initialization, Input, and Submodels. We explain which aspects of a model should be described in each element, and we present an example to illustrate the protocol in use. In addition, 19 examples are available in an Online Appendix. We consider ODD as a first step for establishing a more detailed common format of the description of IBMs and ABMs. Once initiated, the protocol will hopefully evolve as it becomes used by a sufficiently large proportion of modellers. In this communication we present evidence that DPE2 acts on the recently identified cytosolic water-soluble heteroglycans (SHG) as does the cytosolic phosphorylase (EC 2.4.1.1) isoform. By using in vitro two-step C-14 labeling assays we demonstrate that the two transferases can utilize the same acceptor sites of the SHG. Cytosolic heteroglycans from a DPE2-deficient Arabidopsis mutant were characterized. Compared with the wild type the glucose content of the heteroglycans was increased. Most of the additional glucosyl residues were found in the outer chains of SHG that are released by an endo- alpha-arabinanase (EC 3.2.1.99). In 2001 and 2002, a survey with special emphasis oil virus isolation in migrating waders and some other birds were conducted. In one of the most important inland resting sites for migratory waterfowl, tracheal and cloacal swabs were collected from 465 waders representing 19 different species, and 165 other birds that were not captured on purpose. A total of 42 avian viruses were isolated, 34 of these were identified as paramyxoviruses (PMVs). The majority of isolates came from waders and wild ducks, and were characterized as PMV-1. In contrast, PMV-4 was found in wild ducks only, PMV-6 was mainly detected in wader species. Four avian influenza viruses (ATVs), belonging to H4 and H3 haemagglutinin subtype, were isolated from wild duck species. Furthermore, four reo-like viruses were isolated from one particular wader species for the first time. The majority of virus positive birds were < 1 year old and did not show any clinical symptoms. There was no evidence for the presence of West Nile virus in these birds. The virtual machines with appropriate resource allocation, software installation, and system configurations are used to build lightweight security laboratories on a hosting computer. Reliability and availability of laboratory platforms are covered by the virtual machine management framework. This management framework provides necessary monitoring and administration services to detect and recover critical failures of virtual machines at run time. Considering the risk that virtual machines can be misused for compromising production networks, we present security management solutions to prevent misuse of laboratory resources by security isolation at the system and network levels. This work is an attempt to bridge the gap between e-learning/tele-teaching and practical IT security education. We investigated differences in activity patterns in relation to daytime, season, sun radiation, moonlight, age and sex. Bushbuck were found to show peak activities around sunrise and at dawn. No difference in the mean activity rates was found between the dry and wet season. Daytime activity was not predicted by differences in sun radiation, nor was nighttime activity predicted by the presence or absence of moonlight. We found the activity of adult territorial males to be strongly positively correlated with that of females, whereas the activity of young-adult non-territorial males was not significantly correlated with the activity of females. This suggests that young-adult males shift their peak activity to phases when adult territorial males are less active. The chosen model taxon, the genus Campylomormyrus, exhibits a wide diversity of electric organ discharge (EOD) waveform types. Their EOD is age, sex, and species specific and is an important character for discriminating among species that are otherwise cryptic. After having established a complementary set of molecular markers, I examined the radiation of Campylomormyrus by a combined approach of molecular data (sequence data from the mitochondrial cytochrome b and the nuclear S7 ribosomal protein gene, as well as 18 microsatellite loci, especially developed for the genus Campylomormyrus), observation of ontogeny and diversification of EOD waveform, and morphometric analysis of relevant morphological traits. I built up the first convincing phylogenetic hypothesis for the genus Campylomormyrus. Taking advantage of microsatellite data, the identified phylogenetic clades proved to be reproductively isolated biological species. This way I detected at least six species occurring in sympatry near Brazzaville/Kinshasa (Congo Basin). By combining molecular data and EOD analyses, I could show that there are three cryptic species, characterised by their own adult EOD types, hidden under a common juvenile EOD form. In addition, I confirmed that adult male EOD is species-specific and is more different among closely related species than among more distantly related ones. This result and the observation that the EOD changes with maturity suggest its function as a reproductive isolation mechanism. As a result of my morphometric shape analysis, I could assign species types to the identified reproductively isolated groups to produce a sound taxonomy of the group. Besides this, I could also identify morphological traits relevant for the divergences between the identified species. Among them, the variations I found in the shape of the trunk-like snout, suggest the presence of different trophic specializations; therefore, this trait might have been involved in the ecological radiation of the group. In conclusion, I provided a convincing scenario envisioning an adaptive radiation of weakly electric fish triggered by sexual selection via assortative mating due to differences in EOD characteristics, but caused by a divergent selection of morphological traits correlated with the feeding ecology. Ich untersuchte die Radiation der Gattung Campylomormyrus anhand eines kombinierten Ansatzes aus molekularen Daten (Sequenzdaten des mitochondrialen Cytochrom b Gens und des nukleären S7 ribosomalen Protein-Gens, sowie 18 Mikrosatelliten, speziell von mir entwickelt für die Gattung Campylomormyrus), Beobachtungen der Ontogenie und der Diversifikation der Entladungsform, sowie morphometrische Auswertungen der Gestalt relevanter morphologischer Merkmale. Ich erstellte eine erste phylogenetische Hypothese für die Gattung Campylomormyrus. Durch meine Mikrosatellitendaten, die als unabhängiger Beweis dienten, konnte ich zeigen, dass die identifizierten phylogenetischen Gruppen reproduktiv isolierte biologische Arten sind. Auf diese Weise konnte ich mindesten sechs Arten nachweisen, die in Sympatrie nahe Brazzaville/Kinshasa (Kongo-Becken) vorkommen. Durch die Übereinstimmung von molekularen Daten und Entladungsformen konnte ich drei kryptische Arten unterscheiden, die sich hinter einheitlichen juvenilen Entladungsformen verbergen, sich aber zu verschiedenen adulten Formen entwickelten. Des Weiteren konnte ich zeigen, dass die adulten männlichen Entladungsformen artspezifisch sind und, dass der Unterschied in der Entladungsform zwischen nah verwandten Arten deutlicher ausgeprägt ist als zwischen entfernter verwandten Arten. Dieses Ergebnis und die Beobachtung, dass sich die Entladungsform bei der Geschlechtsreife ändert, weisen darauf hin, dass die Entladungsform als reproduktiver Isolationsmechanismus dient. In einer morphometrischen Gestalt-Analyse verglich ich das Typen-Material der beschriebenen Arten mit den zuvor ermittelten reproduktiv isolierten Gruppen, um auf diese Weise deren Art zu bestimmen. Überdies konnte ich maßgebliche morphologische Unterscheidungsmerkmale identifizieren. Diese äußern sich hauptsächlich in der Gestalt der rüsselartigen Schnauze, könnten daher mit einer trophischen Spezialisierung einhergehen und eine ökologische Artbildung ermöglichen. Zusammenfassend entwickelte ich, in Übereinstimmung mit anderen Untersuchungen und theoretischen Überlegungen, eine plausible Hypothese einer adaptiven Radiation der schwach-elektrischen Fische Afrikas, ausgelöst durch sexuelle Selektion. Diese wirkt durch assortative Verpaarung, basierend auf Charakteristika der elektrischen Entladungsform. Verursacht wird der Prozess der adaptiven Radiation jedoch durch divergierende Selektion morphologischer Merkmale, die in Bezug zur Nahrungsökologie stehen. For this purpose, a detailed knowledge of the natural variability of the climate during warm stages is a necessary prerequisite. Beside model simulations and historical documents, this knowledge is mostly derived from analyses of so-called climatic proxy data like tree rings or sediment as well as ice cores. In order to be able to appropriately interpret such sources of palaeoclimatic information, suitable approaches of statistical modelling as well as methods of time series analysis are necessary, which are applicable to short, noisy, and non-stationary uni- and multivariate data sets. Correlations between different climatic proxy data within one or more climatological archives contain significant information about the climatic change on longer time scales. Based on an appropriate statistical decomposition of such multivariate time series, one may estimate dimensions in terms of the number of significant, linear independent components of the considered data set. In the presented work, a corresponding approach is introduced, critically discussed, and extended with respect to the analysis of palaeoclimatic time series. Temporal variations of the resulting measures allow to derive information about climatic changes. For an example of trace element abundances and grain-size distributions obtained near the Cape Roberts (Eastern Antarctica), it is shown that the variability of the dimensions of the investigated data sets clearly correlates with the Oligocene/Miocene transition about 24 million years before present as well as regional deglaciation events. Grain-size distributions in sediments give information about the predominance of different transportation as well as deposition mechanisms. Finite mixture models may be used to approximate the corresponding distribution functions appropriately. In order to give a complete description of the statistical uncertainty of the parameter estimates in such models, the concept of asymptotic uncertainty distributions is introduced. The relationship with the mutual component overlap as well as with the information missing due to grouping and truncation of the measured data is discussed for a particular geological example. An analysis of a sequence of grain-size distributions obtained in Lake Baikal reveals that there are certain problems accompanying the application of finite mixture models, which cause an extended climatological interpretation of the results to fail. As an appropriate alternative, a linear principal component analysis is used to decompose the data set into suitable fractions whose temporal variability correlates well with the variations of the average solar insolation on millenial to multi-millenial time scales. The abundance of coarse-grained material is obviously related to the annual snow cover, whereas a significant fraction of fine-grained sediments is likely transported from the Taklamakan desert via dust storms in the spring season. Hierzu ist eine detaillierte Kenntnis der natürlichen Klimavariabilität während Warmzeiten unerlässlich. Neben Modellsimulationen und historischen Aufzeichnungen spielt hierfür die Analyse von sogenannten Klima-Stellvertreterdaten eine besondere Rolle, die anhand von Archiven wie Baumringen oder Sediment- und Eisbohrkernen erhoben werden. Um solche Quellen paläoklimatischer Informationen vernünftig interpretieren zu können, werden geeignete statistische Modellierungsansätze sowie Methoden der Zeitreihenanalyse benötigt, die insbesondere auf kurze, verrauschte und instationäre uni- und multivariate Datensätze anwendbar sind. Korrelationen zwischen verschiedenen Stellvertreterdaten eines oder mehrerer klimatologischer Archive enthalten wesentliche Informationen über den Klimawandel auf großen Zeitskalen. Auf der Basis einer geeigneten Zerlegung solcher multivariater Zeitreihen lassen sich Dimensionen schätzen als die Zahl der signifikanten, linear unabhängigen Komponenten des Datensatzes. Ein entsprechender Ansatz wird in der vorliegenden Arbeit vorgestellt, kritisch diskutiert und im Hinblick auf die Analyse von paläoklimatischen Zeitreihen weiterentwickelt. Zeitliche Variationen der entsprechenden Maße erlauben Rückschlüsse auf klimatische Veränderungen. Am Beispiel von Elementhäufigkeiten und Korngrößenverteilungen des Cape-Roberts-Gebietes in der Ostantarktis wird gezeigt, dass die Variabilität der Dimension der untersuchten Datensätze klar mit dem Übergang vom Oligozän zum Miozän vor etwa 24 Millionen Jahren sowie regionalen Abschmelzereignissen korreliert. Korngrößenverteilungen in Sedimenten erlauben Rückschlüsse auf die Dominanz verschiedenen Transport- und Ablagerungsmechanismen. Mit Hilfe von Finite-Mixture-Modellen lassen sich gemessene Verteilungsfunktionen geeignet approximieren. Um die statistische Unsicherheit der Parameterschätzung in solchen Modellen umfassend zu beschreiben, wird das Konzept der asymptotischen Unsicherheitsverteilungen eingeführt. Der Zusammenhang mit dem Überlapp der einzelnen Komponenten und aufgrund des Abschneidens und Binnens der gemessenen Daten verloren gehenden Informationen wird anhand eines geologischen Beispiels diskutiert. Die Analyse einer Sequenz von Korngrößenverteilungen aus dem Baikalsee zeigt, dass bei der Anwendung von Finite-Mixture-Modellen bestimmte Probleme auftreten, die eine umfassende klimatische Interpretation der Ergebnisse verhindern. Stattdessen wird eine lineare Hauptkomponentenanalyse verwendet, um den Datensatz in geeignete Fraktionen zu zerlegen, deren zeitliche Variabilität stark mit den Schwankungen der mittleren Sonneneinstrahlung auf der Zeitskala von Jahrtausenden bis Jahrzehntausenden korreliert. The number of hydrophobic chains making up a micellar core is relatively low for the oligomeric surfactants, the spacer length playing an important role. For the dimers, the number decreases from 32 to 21 with increasing spacer length. These numbers decrease further with increasing degree of oligomerization down to values of about 15. As for many conventional ionic surfactants, the micelles of all oligomers studied grow only slightly with the concentration, and they remain in the regime of small micelles up to concentrations of at least 3 wt %. In both cases, the impact of He- 4 cores (alpha particles) that were emitted from actinide-bearing mineral inclusions has caused the formation of radiation damage haloes in the host mineral. These haloes have maximum radii of about 33 mu m (chamosite) and 47 mu m (cordierite). They show notably changed optical properties, i.e., intensified absorption of light as recognised by brown (chamosite) and yellow (cordierite) pleochroism and enhanced or even anomalous interference colours. In spite of the significant disturbance of their short range order, alpha particle haloes are characterised by generally low degrees of structural radiation damage. This is indicated by rather moderate broadening of vibrational bands and, in the case of cordierite, apparently undisturbed electron diffraction patterns in the TEM. Intensive damage, virtually close to an amorphous state, was only found in cordierite up to a few tens of nanometres away from actinide-bearing inclusions. This damage is mainly assigned to recoils of heavy nuclei upon emission of an alpha particle, which have particle trajectory lengths that are three orders of magnitude shorter than those of the alpha particles. These functions are mediated through the binding of serotonin to its receptors, of which 13 subtypes have been characterized in vertebrates. We have isolated a cDNA from the honeybee Apis mellifera (Am5-ht7) sharing high similarity to members of the 5-HT7 receptor family. Expression of the Am5-HT7 receptor in HEK293 cells results in an increase in basal cAMP levels, suggesting that Am5-HT7 is expressed as a constitutively active receptor. Serotonin application to Am5-ht7-transfected cells elevates cyclic adenosine 3',5'-monophosphate (cAMP) levels in a dose-dependent manner (EC50 = 1.1-1.8 nM). The Am5-HT7 receptor is also activated by 5-carboxamidotryptamine, whereas methiothepin acts as an inverse agonist. Receptor expression has been investigated by RT-PCR, in situ hybridization, and western blotting experiments. Receptor mRNA is expressed in the perikarya of various brain neuropils, including intrinsic mushroom body neurons, and in peripheral organs. Sie bilden eine bedeutende Gruppe von Botenstoffen und entfalten ihre Wirkungen über die Bindung an eine bestimmte Klasse von Rezeptorproteinen, die als G-Protein-gekoppelte Rezeptoren bezeichnet werden. Bei Insekten gehören zur Substanzklasse der biogenen Amine die Botenstoffe Dopamin, Tyramin, Octopamin, Serotonin und Histamin. Neben vielen anderen Wirkung ist z.B. gezeigt worden, daß einige dieser biogenen Amine bei der Honigbiene (Apis mellifera) die Geschmacksempfindlichkeit für Zuckerwasser-Reize modulieren können. Ich habe verschiedene Aspekte der aminergen Signaltransduktion an den „Modellorganismen“ Honigbiene und Amerikanische Großschabe (Periplaneta americana) untersucht. Aus der Honigbiene, einem „Modellorganismus“ für das Studium von Lern- und Gedächtnisvorgängen, wurden zwei Dopamin-Rezeptoren, ein Tyramin-Rezeptor, ein Octopamin-Rezeptor und ein Serotonin-Rezeptor charakterisiert. Die Rezeptoren wurden in kultivierten Säugerzellen exprimiert, um ihre pharmakologischen und funktionellen Eigenschaften (Kopplung an intrazelluläre Botenstoffwege) zu analysieren. Weiterhin wurde mit Hilfe verschiedener Techniken (RT-PCR, Northern-Blotting, in situ-Hybridisierung) untersucht, wo und wann während der Entwicklung die entsprechenden Rezeptor-mRNAs im Gehirn der Honigbiene exprimiert werden. Als Modellobjekt zur Untersuchung der zellulären Wirkungen biogener Amine wurden die Speicheldrüsen der Amerikanischen Großschabe genutzt. An isolierten Speicheldrüsen läßt sich sowohl mit Dopamin als auch mit Serotonin Speichelproduktion auslösen, wobei Speichelarten unterschiedlicher Zusammensetzung gebildet werden. Dopamin induziert die Bildung eines völlig proteinfreien, wäßrigen Speichels. Serotonin bewirkt die Sekretion eines proteinhaltigen Speichels. Die Serotonin-induzierte Proteinsekretion wird durch eine Erhöhung der Konzentration des intrazellulären Botenstoffs cAMP vermittelt. Es wurden die pharmakologischen Eigenschaften der Dopamin-Rezeptoren der Schaben-Speicheldrüsen untersucht sowie mit der molekularen Charakterisierung putativer aminerger Rezeptoren der Schabe begonnen. Weiterhin habe ich das ebony-Gen der Schabe charakterisiert. Dieses Gen kodiert für ein Enzym, das wahrscheinlich bei der Schabe (wie bei anderen Insekten) an der Inaktivierung biogener Amine beteiligt ist und im Gehirn und in den Speicheldrüsen der Schabe exprimiert wird. N2 - Biogenic amines are small organic compounds that act as neurotransmitters, neuromodulators and/or neurohormones in vertebrates and in invertebrates. They form an important group of messenger substances and mediate their diverse effects by binding to membrane receptors that primarily belong to the large gene-family of G protein-coupled receptors. In insects, the group of biogenic amine messengers consists of five members: dopamine, tyramine, octopamine, serotonin, and histamine. Besides many other effects, some of these biogenic amines were shown, for example, to modulate gustatory sensitivity to sucrose stimuli in the honeybee (Apis mellifera). I have investigated various aspects of the aminergic signal transduction in the “model organisms” honeybee and American cockroach (Periplaneta americana). So far, I have characterized two dopamine receptors, a tyramine receptor, an octopamine receptor and a serotonin receptor of the honeybee, which is well-known for its learning and memory capacities. The receptors where expressed in cultivated mammalian cells in order to analyze their pharmacological and functional (i.e., second messenger coupling) properties. The spatiotemporal expression patterns of the respective receptor mRNA were investigated in the honeybee brain by using different techniques (RT PCR, Northern blotting, in situ-hybridization). The salivary glands of the American cockroach were used as a model object in order to investigate the cellular effects of biogenic amines. Both dopamine and serotonin trigger salivary secretion in isolated salivary glands. The quality of the secreted saliva is, however, different. Stimulation of the glands by serotonin results in the production of a protein-rich saliva, whereas stimulation by dopamine results in saliva that is protein-free. Serotonin-induced protein secretion is mediated by an increase in the intracellular concentration of cAMP. The pharmacological properties of dopamine receptors associated with cockroach salivary glands were investigated and the molecular characterization of putative aminergic receptors of the cockroach was initiated. Furthermore, I have characterized the ebony gene of the cockroach. This gene encodes an enzyme that is probably involved in the inactivation of biogenic amines in the cockroach (as in other insects). Dopamine and serotonin are present in high concentrations in the bee brain, whereas octopamine and tyramine are less abundant. Octopamine is a key molecule for the control of honeybee behaviour. It generally has an arousing effect and leads to higher sensitivity for sensory inputs, better learning performance and increased foraging behaviour. Tyramine has been suggested to act antagonistically to octopamine, but only few experimental data are available for this amine. Dopamine and serotonin often have antagonistic or inhibitory effects as compared to octopamine. Biogenic amines bind to membrane receptors that primarily belong to the large gene-family of GTP-binding (G) protein coupled receptors. Receptor activation leads to transient changes in concentrations of intracellular second messengers such as cAMP, IP3 and/or Ca2+. Although several biogenic amine receptors from the honeybee have been cloned and characterised more recently, many genes still remain to be identified. The availability of the completely sequenced genome of Apis mellifera will contribute substantially to closing this gap. In this review, we will discuss the present knowledge on how biogenic amines and their receptor-mediated cellular responses modulate different behaviours of honeybees including learning processes and division of labour. Methods 63 (2005) 10-23] was used to analyze the NIR absorption spectra of RC preparations from Rhodobacter (R.) sphaeroides strain 2R and Blastochloris (B.) viridis strain KH, containing bacteriochlorophyll (BChl) a and b, respectively. Q(y) bands of individual RC porphyrin components (BChls and bacteriopheophytins, BPheo) were identified. The results indicate that the upper exciton level Py+ of the photo-active BChl dimer in RCs of R. sphaeroides has an absorption maximum of 810nm. The blue shift of a complex integral band at approximately 800nm upon oxidation of the RC is caused primarily by bleaching of Py+, rather than by an electrochromic shift of the absorption band(s) of the monomeric BChls. Likewise, the disappearance of a band peaking at 842 nm upon oxidation of RCs from B. viridis indicates that this band has to be assigned to Py+, A blue shift of an absorption band at approximately 830nm upon oxidation of RCs of B. viridis is also essentially caused by the disappearance of Py+, rather than by an electrochromic shift of the absorption bands of monomeric BChls. It aims at understanding the complex relationships between ecological processes and landscape patterns, identifying the underlying mechanisms and deriving valid predictions for scenarios of landscape change and its consequences. Today, various studies cope with these tasks through so called "landscape modelling" approaches. They integrate different aspects of heterogeneous and dynamic landscapes and model different driving forces, often using both statistical and process-oriented techniques. We identify two main approaches to deal with the analysis of pattern-process interactions: the first starts with pattern detection, pattern description and pattern analysis, the second with process description, simulation and pattern generation. Focussing on the interplay between these two approaches, landscape analysis and landscape modelling will improve our understanding of pattern-process interactions. The comparison of simulated and observed pattern is a prerequisite for both approaches. Genistein and daidzein, the major isoflavones found in soy, have received the most attention. However, they undergo extensive metabolism in the intestine and the liver, which might affect their biological properties, e.g. their antioxidant capacities. Furthermore, the biological activities of other naturally occurring isoflavones, for instance, glycitein from soy or biochanin A from red clover, have not yet been studied in detail. The aim of this study was to investigate the antioxidant activities of six naturally occurring isoflavones and their corresponding oxidative and bacterial metabolites. The oxygen radical absorbance capacity assay as well as the in vitro oxidation of low density lipoproteins with the conjugated diene and the thiobarbituric acid reacting substances formation as end points were used. The oxidative metabolites of genistein and daidzein as well as equol exhibited the highest antioxidant activities in all three assays. With few exceptions, they were more effective than the positive controls quercetin and ascorbic acid. Formononetin, the 4'-O-methyl ether of daidzein, showed the lowest antioxidant property. More precisely, the work was focused on two main problems: 1. How to separate both oscillations into an affordable model for understanding the behaviour of the whole system. 2. How to model the system in order to achieve a better understanding of the interaction, as well as to predict future states of the system. We focused our efforts in the Sea Surface Temperature equations, considering that atmospheric effects were secondary to the ocean dynamics. The results found may be summarised as follows: 1. Linear methods are not suitable for characterising the dimensionality of the sea surface temperature in the tropical Pacific Ocean. Therefore they do not help to separate the oscillations by themselves. Instead, nonlinear methods of dimensionality reduction are proven to be better in defining a lower limit for the dimensionality of the system as well as in explaining the statistical results in a more physical way [1]. In particular, Isomap, a nonlinear modification of Multidimensional Scaling methods, provides a physically appealing method of decomposing the data, as it substitutes the euclidean distances in the manifold by an approximation of the geodesic distances. We expect that this method could be successfully applied to other oscillatory extended systems and, in particular, to meteorological systems. 2. A three dimensional dynamical system could be modeled, using a backfitting algorithm, for describing the dynamics of the sea surface temperature in the tropical Pacific Ocean. We observed that, although there were few data points available, we could predict future behaviours of the coupled ENSO-Annual Cycle system with an accuracy of less than six months, although the constructed system presented several drawbacks: few data points to input in the backfitting algorithm, untrained model, lack of forcing with external data and simplification using a close system. Anyway, ensemble prediction techniques showed that the prediction skills of the three dimensional time series were as good as those found in much more complex models. This suggests that the climatological system in the tropics is mainly explained by ocean dynamics, while the atmosphere plays a secondary role in the physics of the process. Relevant predictions for short lead times can be made using a low dimensional system, despite its simplicity. The analysis of the SST data suggests that nonlinear interaction between the oscillations is small, and that noise plays a secondary role in the fundamental dynamics of the oscillations [2]. A global view of the work shows a general procedure to face modeling of climatological systems. First, we should find a suitable method of either linear or nonlinear dimensionality reduction. Then, low dimensional time series could be extracted out of the method applied. Finally, a low dimensional model could be found using a backfitting algorithm in order to predict future states of the system. In diesem Gebiet der Welt finden zwei wichtige Phänomene gleichzeitig statt: der jährliche Zyklus und El Niño. Der jährliche Zyklus kann als Oszillation physikalischer Variablen (z.B. Temperatur, Windgeschwindigkeit, Höhe des Meeresspiegels), welche eine Periode von einem Jahr zeigen, definiert werden. Das bedeutet, dass das Verhalten des Meers und der Atmosphäre alle zwölf Monate ähnlich sind (alle Sommer sind ähnlicher jedes Jahr als Sommer und Winter des selben Jahres). El Niño ist eine irreguläre Oszillation weil sie abwechselnd hohe und tiefe Werte erreicht, aber nicht zu einer festen Zeit, wie der jährliche Zyklus. Stattdessen, kann el Niño in einem Jahr hohe Werte erreichen und dann vier, fünf oder gar sieben Jahre benötigen, um wieder aufzutreten. Das Hauptproblem in unserem Fall ist die Mindestanzahl von Punkten zu finden, die ausreicht, um beide Phänomene zu beschreiben. Man hat gefunden, dass diese Anzahl drei ist. Nach diesem Teil, war das Ziel vorherzusagen, wie die Temperaturen sich in der Zeit entwickeln werden, wenn man die aktuellen und vergangenen Temperaturen kennt. Man hat beobachtet, dass eine genaue Vorhersage bis zu sechs oder weniger Monate gemacht werden kann, und dass die Temperatur für ein Jahr nicht vorhersagbar ist. Ein wichtiges Resultat ist, dass die Vorhersagen auf kurzen Zeitskalen genauso gut sind, wie die Vorhersagen, welche andere Autoren mit deutlich komplizierteren Methoden erhalten haben. Deswegen ist meine Aussage, dass das gesamte System von jährlichem Zyklus und El Niño mittels einfacherer Methoden als der heute angewandten vorhergesagt werden kann. The philosophy of RME is based on Hans Freudenthal’s concept of ‘mathematics as a human activity’. Prof. Hans Freudenthal (1905-1990), a mathematician and educator, believes that ‘ready-made mathematics’ should not be taught in school. By contrast, he urges that students should be offered ‘realistic situations’ so that they can rediscover from informal to formal mathematics. Although mathematics education in Vietnam has some achievements, it still encounters several challenges. Recently, the reform of teaching methods has become an urgent task in Vietnam. It appears that Vietnamese mathematics education lacks necessary theoretical frameworks. At first sight, the philosophy of RME is suitable for the orientation of the teaching method reform in Vietnam. However, the potential of RME for mathematics education as well as the ability of applying RME to teaching mathematics is still questionable in Vietnam. The primary aim of this dissertation is to research into abilities of applying RME to teaching and learning mathematics in Vietnam and to answer the question “how could RME enrich Vietnamese mathematics education?”. This research will emphasize teaching geometry in Vietnamese middle school. More specifically, the dissertation will implement the following research tasks: • Analyzing the characteristics of Vietnamese mathematics education in the ‘reformed’ period (from the early 1980s to the early 2000s) and at present; • Implementing a survey of 152 middle school teachers’ ideas from several Vietnamese provinces and cities about Vietnamese mathematics education; • Analyzing RME, including Freudenthal’s viewpoints for RME and the characteristics of RME; • Discussing how to design RME-based lessons and how to apply these lessons to teaching and learning in Vietnam; • Experimenting RME-based lessons in a Vietnamese middle school; • Analyzing the feedback from the students’ worksheets and the teachers’ reports, including the potentials of RME-based lessons for Vietnamese middle school and the difficulties the teachers and their students encountered with RME-based lessons; • Discussing proposals for applying RME-based lessons to teaching and learning mathematics in Vietnam, including making suggestions for teachers who will apply these lessons to their teaching and designing courses for in-service teachers and teachers-in training. This research reveals that although teachers and students may encounter some obstacles while teaching and learning with RME-based lesson, RME could become a potential approach for mathematics education and could be effectively applied to teaching and learning mathematics in Vietnamese school. Im Gegensatz dazu forderte er, den Schülern an ‚realistischen’ Situationen nicht-formale und formale Mathematik wieder entdecken zu lassen. Obwohl die mathematische Schulbildung in Vietnam in den letzten Jahrzehnten schon einige Fortschritte gemacht hat, steht sie noch vor großen Herausforderungen. Derzeit ist die Reform der Unterrichtsmethoden eine dringliche Aufgabe in Vietnam. Augenscheinlich ermangelt es der Mathematikdidaktik in Vietnam an dem dazu notwendigen theoretischen Rahmen. Die Philosophie von RME eignet sich grundsätzlich als Orientierung für die Reform der Unterrichtsmethoden in Vietnam. Allerdings ist die Potenz von RME für die mathematische Schulbildung in Vietnam und die Möglichkeiten, RME im Mathematikunterricht anzuwenden, noch zu klären. Das Hauptziel dieser Arbeit war zu erforschen, wie RME beim Mathematik-Lernen und -Lehren in Vietnam eingesetzt werden kann und die Frage zu beantworten: Wie kann RME den Mathematikunterricht in Vietnam bereichern? Dazu wurde insbesondere der Geometrieunterricht in der Sekundarstufe I betrachtet. This includes expression levels of thousands or ten-thousands of genes as well as concentration levels of metabolites or proteins. Such Profile data, observed at different times or at different experimental conditions (e.g., heat or dry stress), show how the biological experiment is reflected on the molecular level. This information is helpful to understand the molecular behaviour and to identify molecules or combination of molecules that characterise specific biological condition (e.g., disease). This work shows the potentials of component extraction algorithms to identify the major factors which influenced the observed data. This can be the expected experimental factors such as the time or temperature as well as unexpected factors such as technical artefacts or even unknown biological behaviour. Extracting components means to reduce the very high-dimensional data to a small set of new variables termed components. Each component is a combination of all original variables. The classical approach for that purpose is the principal component analysis (PCA). It is shown that, in contrast to PCA which maximises the variance only, modern approaches such as independent component analysis (ICA) are more suitable for analysing molecular data. The condition of independence between components of ICA fits more naturally our assumption of individual (independent) factors which influence the data. This higher potential of ICA is demonstrated by a crossing experiment of the model plant Arabidopsis thaliana (Thale Cress). The experimental factors could be well identified and, in addition, ICA could even detect a technical artefact. However, in continuously observations such as in time experiments, the data show, in general, a nonlinear distribution. To analyse such nonlinear data, a nonlinear extension of PCA is used. This nonlinear PCA (NLPCA) is based on a neural network algorithm. The algorithm is adapted to be applicable to incomplete molecular data sets. Thus, it provides also the ability to estimate the missing data. The potential of nonlinear PCA to identify nonlinear factors is demonstrated by a cold stress experiment of Arabidopsis thaliana. The results of component analysis can be used to build a molecular network model. Since it includes functional dependencies it is termed functional network. Applied to the cold stress data, it is shown that functional networks are appropriate to visualise biological processes and thereby reveals molecular dynamics. Diese Profildaten verschiedener Zeitpunkte oder unterschiedlicher experimenteller Bedingungen (z.B. unter Stressbedingungen wie Hitze oder Trockenheit) zeigen, wie sich das biologische Experiment auf molekularer Ebene widerspiegelt. Diese Information kann genutzt werden, um molekulare Abläufe besser zu verstehen und um Moleküle oder Molekül-Kombinationen zu bestimmen, die für bestimmte biologische Zustände (z.B.: Krankheit) charakteristisch sind. Die Arbeit zeigt die Möglichkeiten von Komponenten-Extraktions-Algorithmen zur Bestimmung der wesentlichen Faktoren, die einen Einfluss auf die beobachteten Daten ausübten. Das können sowohl die erwarteten experimentellen Faktoren wie Zeit oder Temperatur sein als auch unerwartete Faktoren wie technische Einflüsse oder sogar unerwartete biologische Vorgänge. Unter der Extraktion von Komponenten versteht man die Reduzierung dieser stark hoch-dimensionalen Daten auf wenige neue Variablen, die eine Kombination aus allen ursprünglichen Variablen darstellen und als Komponenten bezeichnet werden. Die Standard-Methode für diesen Zweck ist die Hauptkomponentenanalyse (PCA). Es wird gezeigt, dass - im Vergleich zur nur die Varianz maximierenden PCA - moderne Methoden wie die Unabhängige Komponentenanalyse (ICA) für die Analyse molekularer Datensätze besser geeignet sind. Die Unabhängigkeit von Komponenten in der ICA entspricht viel besser unserer Annahme individueller (unabhängiger) Faktoren, die einen Einfluss auf die Daten ausüben. Dieser Vorteil der ICA wird anhand eines Kreuzungsexperiments mit der Modell-Pflanze Arabidopsis thaliana (Ackerschmalwand) demonstriert. Die experimentellen Faktoren konnten dabei gut identifiziert werden und ICA erkannte sogar zusätzlich einen technischen Störfaktor. Bei kontinuierlichen Beobachtungen wie in Zeitexperimenten zeigen die Daten jedoch häufig eine nichtlineare Verteilung. Für die Analyse dieser nichtlinearen Daten wird eine nichtlinear erweiterte Methode der PCA angewandt. Diese nichtlineare PCA (NLPCA) basiert auf einem neuronalen Netzwerk-Algorithmus. Der Algorithmus wurde für die Anwendung auf unvollständigen molekularen Daten erweitert. Dies ermöglicht es, die fehlenden Werte zu schätzen. Die Fähigkeit der nichtlinearen PCA zur Bestimmung nichtlinearer Faktoren wird anhand eines Kältestress-Experiments mit Arabidopsis thaliana demonstriert. Die Ergebnisse aus der Komponentenanalyse können zur Erstellung molekularer Netzwerk-Modelle genutzt werden. Da sie funktionelle Abhängigkeiten berücksichtigen, werden sie als Funktionale Netzwerke bezeichnet. Soluble dietary fibre may modulate gastrointestinal or adipose tissue hormones regulating food intake. The present study investigated the effects of arabinoxylan consumption on serum glucose, insulin, lipids, leptin, adiponectin and resistin in subjects with impaired glucose tolerance. In a randomized, single-blind, controlled, crossover intervention trial, 11 adults consumed white bread rolls as either placebo or supplemented with 15g arabinoxylan for 6 weeks with a 6-week washout period. Fasting serum glucose, insulin, triglycerides, unesterified fatty acids, apolipoprotein A1 and B, adiponectin, resistin and leptin were assessed before and after intervention. Fasting serum glucose, serum triglycerides and apolipoprotein A-1 were significantly lower during arabinoxylan consumption compared to placebo (p = 0.029, p = 0.047; p = 0.029, respectively). No effects of arabinoxylan were observed for insulin, adiponectin, leptin and resistin as well as for apolipoprotein B, and unesterified fatty acids. In conclusion, the consumption of AX in subjects with impaired glucose tolerance improved fasting serum glucose, and triglycerides. However, this beneficial effect was not accompanied by changes in fasting adipokine concentrations.
