Based on the Invest-and-Accrue Model, we argue that conscientiousness explains to some extent why certain students receive better grades despite similar academic accomplishments (i.e., achieving similar scores in domain-specific standardized achievement tests). Therefore, the present study examines to what extent the relationship between student personality and teacher-assigned grades consists of direct as opposed to indirect associations (via subject-specific standardized test scores). We used a representative sample of 14,710 ninth-grade students to estimate these direct and indirect pathways in mathematics and German. Structural equation models showed that test scores explained between 8 and 11\% of the variance in teacher-assigned grades in mathematics and German. The Big Five personality traits in students additionally explained between 8 and 10\% of the variance in grades. Finally, the personality-grade relationship consisted of direct (0.02 | β| ≤ 0.27) and indirect associations via test scores (0.01 | β| ≤ 0.07). Conscientiousness explained discrepancies between teacher-assigned grades and students' scores in domain-specific standardized tests to a greater extent than any of the other Big Five personality traits. After having recently demonstrated that upregulation of EBI3 by histone acetylation alleviates disease symptoms in a dextran sulfate sodium (DSS)-treated mouse model of chronic colitis, we now aimed to examine a possible further epigenetic regulation of EBI3 by DNA methylation under inflammatory conditions. Treatment with the DNA methyltransferase inhibitor (DNMTi) decitabine (DAC) and TNF alpha led to synergistic upregulation of EBI3 in human colon epithelial cells (HCEC). Use of different signaling pathway inhibitors indicated NF kappa B signaling was necessary and proportional to the synergistic EBI3 induction. MALDI-TOF/MS and HPLC-ESIMS/MS analysis of DAC/TNF alpha-treated HCEC identified IL-12p35 as the most probable binding partner to form a functional protein. EBI3/IL-12p35 heterodimers (IL-35) induce their own gene upregulation, something that was indeed observed in HCEC cultured with media from previously DAC/TNF alpha-treated HCEC. This study protocol describes an approach to investigate the possible impacts and implications of SCAs on different levels of health care provision. It considers the perspectives of the users, nonusers, general practitioners (GPs), and health care experts. Objective: We aim to assess a comprehensive overview of the use of SCAs and address problematic issues, if any. The primary outcomes of this study are empirically informed multi-perspective recommendations for different stakeholders on the ethical, legal, and social implications of SCAs. Methods: Quantitative and qualitative methods will be used in several overlapping and interconnected study phases. In study phase 1, a comprehensive literature review will be conducted to assess the ethical, legal, social, and systemic impacts of SCAs. Study phase 2 comprises a survey that will be analyzed with a logistic regression. It aims to assess the user degree of SCAs in Germany as well as the predictors for SCA usage. Study phase 3 will investigate self-observational diaries and user interviews, which will be analyzed as integrated cases to assess user perspectives, usage pattern, and arising problems. Study phase 4 will comprise GP interviews to assess their experiences, perspectives, self-image, and concepts and will be analyzed with the basic procedure by Kruse. Moreover, interviews with health care experts will be conducted in study phase 3 and will be analyzed by using the reflexive thematical analysis approach of Braun and Clark. Results: Study phase 1 will be completed in November 2021. We expect the results of study phase 2 in December 2021 and February 2022. In study phase 3, interviews are currently being conducted. The final study endpoint will be in February 2023. Due to the lack of alternatives, analytical methods and empirical relations are currently applied to estimate evolving hydraulic and mechanical rock properties associated with chemical reactions. However, the predictive capabilities of analytical approaches remain limited, since they assume idealised microstructures, and thus are not able to reflect property evolution for dynamic processes. Hence, aim of the present thesis is to improve the prediction of permeability and stiffness changes resulting from pore space alterations of reservoir sandstones. A detailed representation of rock microstructure, including the morphology and connectivity of pores, is essential to accurately determine physical rock properties. For that purpose, three-dimensional pore-scale models of typical reservoir sandstones, obtained from highly resolved micro-computed tomography (micro-CT), are used to numerically calculate permeability and stiffness. In order to adequately depict characteristic distributions of secondary minerals, the virtual samples are systematically altered and resulting trends among the geometric, hydraulic, and mechanical rock properties are quantified. It is demonstrated that the geochemical reaction regime controls the location of mineral precipitation within the pore space, and thereby crucially affects the permeability evolution. This emphasises the requirement of determining distinctive porosity-permeability relationships by means of digital pore-scale models. By contrast, a substantial impact of spatial alterations patterns on the stiffness evolution of reservoir sandstones are only observed in case of certain microstructures, such as highly porous granular rocks or sandstones comprising framework-supporting cementations. In order to construct synthetic granular samples a process-based approach is proposed including grain deposition and diagenetic cementation. It is demonstrated that the generated samples reliably represent the microstructural complexity of natural sandstones. Thereby, general limitations of imaging techniques can be overcome and various realisations of granular rocks can be flexibly produced. These can be further altered by virtual experiments, offering a fast and cost-effective way to examine the impact of precipitation, dissolution or fracturing on various petrophysical correlations. The presented research work provides methodological principles to quantify trends in permeability and stiffness resulting from geochemical processes. The calculated physical property relations are directly linked to pore-scale alterations, and thus have a higher accuracy than commonly applied analytical approaches. The results demonstrate that the location of mineral growth strongly affects the permeability decrease with variations by up to four orders in magnitude. Precipitation in regions of high flow velocities is characterised by a predominant clogging of pore throats and a drastic permeability reduction, which can be roughly described by the power law relation with an exponent of 20. A continuous alteration of the pore structure by uniform mineral growth reduces the permeability comparable to the power law with an exponent of four or the Kozeny-Carman relation. Preferential precipitation in regions of low flow velocities predominantly affects smaller throats and pores with a minor impact on the flow regime, where the permeability decrease is considerably below that calculated by the power law with an exponent of two. Despite their complete distinctive impact on hydraulics, the spatial precipitation patterns only slightly affect the increase in elastic rock properties with differences by up to 6.3\% between the investigated scenarios. Hence, an adequate characterisation of the spatial precipitation pattern is crucial to quantify changes in hydraulic rock properties, whereas the present study shows that its impact on elastic rock parameters is limited. The resulting evolution in porosity, permeability and rock stiffness is examined and compared to the respective micro-computer tomographic (micro-CT) scans. The grain contact-preferential scheme implies a progressive clogging of small throats and consequently produces considerably less connected and stiffer samples than the two other schemes. By contrast, uniform quartz overgrowth continuously alters the pore space and leads to the lowest elastic properties. The proposed stress-dependent cementation scheme combines both approaches of contact-cement and quartz overgrowth, resulting in granulometric, hydraulic and elastic properties equivalent to those of the respective micro-CT scans, where bulk moduli slightly deviate by 0.8\%, 4.9\% and 2.5\% for the Fontainebleau, Berea and Bentheim sandstone, respectively. Here, we identified differentially methylated regions (DMRs) between free-ranging cheetahs and conspecifics in zoological gardens and prospect putative links to pathways relevant to immunity, energy balance and homeostasis. Comparing epigenomic DNA methylation profiles obtained from peripheral blood mononuclear cells (PBMCs) from eight free-ranging female cheetahs from Namibia and seven female cheetahs living in zoological gardens within Europe, we identified DMRs of which 22 were hypermethylated and 23 hypomethylated. Hypermethylated regions in cheetahs under human care were located in the promoter region of a gene involved in host-pathogen interactions (KLC1) and in an intron of a transcription factor relevant for the development of pancreatic beta-cells, liver, and kidney (GLIS3). The most canonical mechanism of DNA methylation in promoter regions is assumed to repress gene transcription. Taken together, this could indicate that hypermethylation at the promoter region of KLC1 is involved in the reduced immunity in cheetahs under human care. The preschool age is characterized by high neuroplasticity which effects motor skill learning, physical fitness, and cognitive development. The aim of this study was to assess the relation of physical fitness and attention (including its individual dimensions (quantitative, qualitative)) as one domain of cognitive performance in preschool children. We hypothesized that fitness components which need precise coordination compared to simple fitness components are stronger related to attention. Methods: Physical fitness components like static balance (i.e., single-leg stance), muscle strength (i.e., handgrip strength), muscle power (i.e., standing long jump), and coordination (i.e., hopping on one leg) were assessed in 61 healthy children (mean age 4.5 +/- 0.6 years; girls n = 30). Attention was measured with the "Konzentrations-Handlungsverfahren fur Vorschulkinder" [concentration-action procedure for preschoolers]). Analyses were adjusted for age, body height, and body mass. Results: Results from single linear regression analysis revealed a significant (p < 0.05) association between physical fitness (composite score) and attention (composite score) (standardized ss = 0.40), showing a small to medium effect (F-2 = 0.14). Further, coordination had a significant relation with the composite score and the quantitative dimension of attention (standardized ss = 0.35; p < 0.01; standardized ss = - 0.33; p < 0.05). Coordination explained about 11\% (composite score) and 9\% (quantitative dimension) of the variance in the stepwise multiple regression model. Conclusion: The results indicate that performance in physical fitness, particularly coordination, is related to attention in preschool children. Thus, high performance in complex fitness components (i.e., hopping on one leg) tends to predict attention in preschool children. Here we formally couple these equations for transport-limited gravel-bed river long-profile evolution. The result is a new predictive relationship whose functional form and parameters are grounded in theory and defined through experimental data. From this, we produce a power-law analytical solution and a finite-difference numerical solution to long-profile evolution. Steady-state channel concavity and steepness are diagnostic of external drivers: concavity decreases with increasing uplift rate, and steepness increases with an increasing sediment-to-water supply ratio. Constraining free parameters explains common observations of river form: to match observed channel concavities, gravel-sized sediments must weather and fine - typically rapidly - and valleys typically should widen gradually. To match the empirical square-root width-discharge scaling in equilibrium-width gravel-bed rivers, downstream fining must occur. However, underlying neuronal mechanisms are unclear. The aim of the current pilot study with a 3-armed randomized controlled trial design was to examine the effects of sensorimotor training for nonspecific chronic low back pain on (1) cognitive performance; (2) fMRI activity co-fluctuations (functional connectivity) between pain-related brain regions; and (3) the relationship between functional connectivity and subjective variables (pain and depression). Six hundred and sixty two volunteers with non-specific chronic low back pain were randomly allocated to a unimodal (sensorimotor training), multidisciplinary (sensorimotor training and behavioral therapy) intervention, or to a control group within a multicenter study. A subsample of patients (n = 21) from one study center participated in the pilot study presented here. Measurements were at baseline, during (3 weeks, M2) and after intervention (12 weeks, M4 and 24 weeks, M5). Cognitive performance was measured by the Trail Making Test and functional connectivity by MRI. Pain perception and depression were assessed by the Von Korff questionnaire and the Hospital and Anxiety. Group differences were calculated by univariate and repeated ANOVA measures and Bayesian statistics; correlations by Pearson's r. Change and correlation of functional connection were analyzed within a pooled intervention group (uni-, multidisciplinary group). Results revealed that participants with increased pain intensity at baseline showed higher functional connectivity between pain-related brain areas used as ROIs in this study. Though small sample sizes limit generalization, cognitive performance increased in the multimodal group. Increased functional connectivity was observed in participants with increased pain ratings. Pain ratings and connectivity in pain-related brain regions decreased after the intervention. The results provide preliminary indication that intervention effects can potentially be achieved on the cognitive and neuronal level. This compilation allows scientists to identify data gaps in need of further RPP analyses but can also aid them in finding an RPP set for their study region. We also present a taxonomically harmonised, unified RPP dataset for the Northern Hemisphere and subsets for North America (including Greenland), Europe (including arctic Russia), and China, which we generated from the available studies. The unified dataset gives the mean RPP for 55 harmonised taxa as well as fall speeds, which are necessary to reconstruct vegetation cover from pollen counts and RPP values. We developed the 'Animated Alcohol Assessment Tool' (AAA-Tool), a mobile app providing an interactive version of the World Health Organization's Alcohol Use Disorders Identification Test (AUDIT) that facilitates the description of individual alcohol consumption via culturally informed animation features. This pilot study evaluated the Russia-specific version of the Animated Alcohol Assessment Tool with regard to (1) its usability and acceptability in a primary healthcare setting, (2) the plausibility of its alcohol consumption assessment results and (3) the adequacy of its Russia-specific vessel and beverage selection. Methods: Convenience samples of 55 patients (47\% female) and 15 healthcare practitioners (80\% female) in 2 Russian primary healthcare facilities self-administered the Animated Alcohol Assessment Tool and rated their experience on the Mobile Application Rating Scale - User Version. Usage data was automatically collected during app usage, and additional feedback on regional content was elicited in semi-structured interviews. Results: On average, patients completed the Animated Alcohol Assessment Tool in 6:38 min (SD = 2.49, range = 3.00-17.16). User satisfaction was good, with all subscale Mobile Application Rating Scale - User Version scores averaging >3 out of 5 points. A majority of patients (53\%) and practitioners (93\%) would recommend the tool to 'many people' or 'everyone'. Assessed alcohol consumption was plausible, with a low number (14\%) of logically impossible entries. Most patients reported the Animated Alcohol Assessment Tool to reflect all vessels (78\%) and all beverages (71\%) they typically used. Conclusion: High acceptability ratings by patients and healthcare practitioners, acceptable completion time, plausible alcohol usage assessment results and perceived adequacy of region-specific content underline the Animated Alcohol Assessment Tool's potential to provide a novel approach to alcohol assessment in primary healthcare. Hier will die Arbeit Einblicke geben, wie die Geographielehrkr{\"a}fte diese Transformation mitgetragen und umgesetzt haben. Welche Wesensz{\"u}ge aus der Sozialisierung in der DDR haben sich bei der Gestaltung des Unterrichtes und dessen Ausrichtung auf die neuen Erziehungsziele erhalten? Hierzu wurden Geographielehrkr{\"a}fte befragt, die sowohl in der DDR als auch im geeinten Deutschland unterrichtet haben. Die Fragen bezogen sich in erster Linie auf die Art und Weise des Unterrichtens vor, w{\"a}hrend und nach der Wende und der daraus entstandenen Systemtransformation. Die Befragungen kommen zu dem Ergebnis, dass sich der Geographieunterricht in der DDR thematisch von dem in der BRD nicht sonderlich unterschied. Von daher bedurfte es keiner umfangreichen inhaltlichen Ver{\"a}nderung des Geographieunterrichts. Schon zu DDR-Zeiten wurden durch die Lehrkr{\"a}fte offenbar eigenm{\"a}chtig ideologiefreie physisch-geographische Themen oft ausgedehnt, um die Ideologie des Faches zu reduzieren. So fiel den meisten eine Anpassung ihres Unterrichts an das westdeutsche System relativ leicht. Die humanistisch gepr{\"a}gte Werteerziehung des DDR-Bildungssystems wurde unter Ausklammerung des sozialistischen Aspektes ebenso fortgef{\"u}hrt, da es auch hier viele Parallelen zum westdeutschen System gegeben hat. Deutlich wird eine Charakterisierung des Faches als Naturwissenschaft von Seiten der ostdeutschen Lehrkr{\"a}fte, obwohl das Fach an den Schulen den Gesellschaftswissenschaften zugeordnet wird und auch in der DDR eine starke wirtschaftsgeographische Ausrichtung hatte. We targeted comparable language use in bilingual and monolingual speakers, crucially covering broader repertoires than just formal language. A main database was the open-access RUEG corpus, which covers comparable informal vs. formal and spoken vs. written productions by adolescent and adult bilinguals with heritage-Greek, -Russian, and -Turkish in Germany and the United States and with heritage-German in the United States, and matching data from monolinguals in Germany, the United States, Greece, Russia, and Turkey. Our main results lie in three areas. (1) We found non-canonical patterns not only in bilingual, but also in monolingual speakers, including patterns that have so far been considered absent from native grammars, in domains of morphology, syntax, intonation, and pragmatics. (2) We found a degree of lexical and morphosyntactic inter-speaker variability in monolinguals that was sometimes higher than that of bilinguals, further challenging the model of the streamlined native speaker. (3) In majority language use, non-canonical patterns were dominant in spoken and/or informal registers, and this was true for monolinguals and bilinguals. In some cases, bilingual speakers were leading quantitatively. In heritage settings where the language was not part of formal schooling, we found tendencies of register leveling, presumably due to the fact that speakers had limited access to formal registers of the heritage language. Our findings thus indicate possible quantitative differences and different register distributions rather than distinct grammatical patterns in bilingual and monolingual speakers. This supports the integration of heritage speakers into the native-speaker continuum. Approaching heritage speakers from this perspective helps us to better understand the empirical data and can shed light on language variation and change in native grammars. Based on the assumption of a linear deterioration of postural control across the life span, the training effect can be extrapolated as a juvenescence of 10 years. Here, we focus on the influence of the following specifications of single-case research designs: number of measurement times, the initial frequency of the behavior, intervention effect, and data trend. We conducted a Monte-Carlo study. First, simulated datasets were created with specific design conditions based on reviews of published single-case intervention studies. Following, data were analyzed using piecewise Poisson-regression models, and the influence of specific design specifications on the test power was investigated. Our results indicate that piecewise regressions have a high potential of adequately identifying the effects of interventions for single-case studies. At the same time, test power is strongly related to the specific design specifications of the single-case study: Few measurement times, especially in phase A, and low initial frequencies of the behavior make it impossible to detect even large intervention effects. Research designs with a high number of measurement times show robust power. Results: The GTPase ARFRP1 interacts with the Golgi-associated PDZ and coiled-coil motif-containing protein (GOPC). Both proteins are co localized at the trans-Golgi network and regulate the first and second phase of insulin secretion by controlling the plasma membrane localization of the SNARE protein SNAP25. Downregulation of both GOPC and ARFRP1 in Min6 cells interferes with the plasma membrane localization of SNAP25 and enhances its degradation, thereby impairing glucose-stimulated insulin release from 13-cells. In turn, overexpression of SNAP25 as well as GOPC restores insulin secretion in islets from 13-cell-specific Arfrp1 knockout mice.

We focused on the expression levels of chromatin modifiers and SNPs in regulatory sequences. We identified alternative splicing events in islets of diabetes-susceptible mice amongst others in genes linked to insulin secretion, endocytosis or ubiquitin-mediated proteolysis pathways. The expression pattern of 54 histones and chromatin modifiers, which may modulate splicing, were markedly downregulated in islets of diabetic animals. Furthermore, diabetes-susceptible mice carry SNPs in RNA-binding protein motifs and in splice sites potentially responsible for alternative splicing events. They also exhibit a larger exon skipping rate, e.g., in the diabetes gene Abcc8, which might affect protein function. Expression of the neuronal splicing factor Srrm4 which mediates inclusion of microexons in mRNA transcripts was markedly lower in islets of diabetes-prone compared to diabetes-resistant mice, correlating with a preferential skipping of SRRM4 target exons. The repression of Srrm4 expression is presumably mediated via a higher expression of miR-326-3p and miR-3547-3p in islets of diabetic mice. During the survey, eight physiochemical water quality parameters were recorded in high spatial and temporal resolution with the BIOFISH multisensor system. Multivariate statistical methods were applied to identify and delineate processes influencing the water quality. The BIOFISH dataset revealed that phytoplankton activity has a major impact on the water quality of the Elbe River in the summer months. The results suggest that phytoplankton biomass constitutes a substantial proportion of the suspended particles and that photosynthetic activity of phytoplankton is closely related to significant temporal changes in pH and oxygen saturation. An evaluation of the BIOFISH data based on the combination of statistical analysis with weather and discharge data shows that the hydrological and meteorological history of the sampled water body was the main driver of phytoplankton dynamics. Results: In historical data social status is positively associated with body height and weight while both are also normally distributed, which marks a significant difference to modern references. Conclusion: Social status is positively associated with height, signaling social dominance, making children of affluent classes taller. Children from the historical data do not reach the average height of modern children, even under the best environmental conditions. Previously, the potential acceptance of a congregation by and for homosexual Jews triggered an intense and broad debate within Reform Judaism. The work asks how it was possible to successfully establish a gay-lesbian synagogue at a time when homosexual acts were considered unnatural and contrary to tradition by almost the entire Jewish community. The starting point of the argumentation is, in addition to general changes in American synagogues after World War II, the assumption that Los Angeles was the most suitable place for this foundation. Los Angeles has an impressive queer history and the Jewish community was more open, tolerant and innovative here than its counterpart on the East Coast. The Metropolitan Community Church was also founded in the city, and as the largest religious institution for homosexual Christians, it also served as the birthplace of queer synagogues. Reform Judaism was chosen as the place of institutional integration of the community because a relative openness for such an endeavor was only seen here. Responsa written in response to a potential admission of Beth Chayim Chadashim can be used to understand the arguments and positions of rabbis and psychologists regarding homosexuality and communities for homosexual Jews in the early 1970s. Ultimately, the commitment and dedication of the congregation and its heterosexual supporters convinced the decision-makers in Reform Judaism. The decisive impulse to question the situation of homosexual Jews in Judaism came from Los Angeles. On posts from a different platform, our model performs on par with BERT-based large language models. We achieve this result at a fraction of the training and inference costs due to the model's simple design. Among them, base excision repair (BER) plays a decisive role. BER is the major repair pathway for small oxidative base modifications and apurinic/apyrimidinic (AP) sites. We established a highly sensitive non-radioactive assay to measure BER incision activity in murine liver samples. Incision activity can be assessed towards the three DNA lesions 8-oxo-2'-deoxyguanosine (8-oxodG), 5-hydroxy-2'-deoxyuracil (5-OHdU), and an AP site analogue. We applied the established assay to murine livers of adult and old mice of both sexes. Furthermore, poly(ADP-ribosyl)ation (PARylation) was assessed, which is an important determinant in DDR and BER. Additionally, DNA damage levels were measured to examine the overall damage levels. No impact of ageing on the investigated endpoints in liver tissue were found. However, animal sex seems to be a significant impact factor, as evident by sex-dependent alterations in all endpoints investigated. This may be because the influence of psychosocial factors on exercise induced adaptation regarding CLBP is neglected. Therefore, this study assessed psychosocial characteristics, which moderate and mediate the effects of sensorimotor exercise on LBP. A single-blind 3-arm multicenter randomized controlled trial was conducted for 12-weeks. Three exercise groups, sensorimotor exercise (SMT), sensorimotor and behavioral training (SMT-BT), and regular routines (CG) were randomly assigned to 662 volunteers. Primary outcomes (pain intensity and disability) and psychosocial characteristics were assessed at baseline (M1) and follow-up (3/6/12/24 weeks, M2-M5). Multiple regression models were used to analyze whether psychosocial characteristics are moderators of the relationship between exercise and pain, meaning that psychosocial factors and exercise interact. Causal mediation analysis were conducted to analyze, whether psychosocial characteristics mediate the exercise effect on pain. A total of 453 participants with intermittent pain (mean age = 39.5 ± 12.2 years, f = 62\%) completed the training. It was shown, that depressive symptomatology (at M4, M5), vital exhaustion (at M4), and perceived social support (at M5) are significant moderators of the relationship between exercise and the reduction of pain intensity. Further depressive mood (at M4), social-satisfaction (at M4), and anxiety (at M5 SMT) significantly moderate the exercise effect on pain disability. The amount of moderation was of clinical relevance. In contrast, there were no psychosocial variables which mediated exercise effects on pain. In conclusion it was shown, that psychosocial variables can be moderators in the relationship between sensorimotor exercise induced adaptation on CLBP which may explain conflicting results in the past regarding the merit of exercise interventions in CLBP. Results suggest further an early identification of psychosocial risk factors by diagnostic tools, which may essential support the planning of personalized exercise therapy. Objectives: First, to assess the association between stress and chronic low back pain (CLBP) and its simultaneous appearance with fatigue and depression as a symptom triad. Second, to identify the most predictive stress-related pattern set for CLBP for a 1-year diagnosis. Methods: In a 1-year observational study with four measurement points, a total of 140 volunteers (aged 18-45 years with intermittent pain) were recruited. The primary outcomes were pain [characteristic pain intensity (CPI), subjective pain disability (DISS)], fatigue, and depressive mood. Stress was assessed as chronic stress, perceived stress, effort reward imbalance, life events, and physiological markers [allostatic load index (ALI), hair cortisol concentration (HCC)]. Multiple linear regression models and selection procedures for model shrinkage and variable selection (least absolute shrinkage and selection operator) were applied. Prediction accuracy was calculated by root mean squared error (RMSE) and receiver-operating characteristic curves. Results: There were 110 participants completed the baseline assessments (28.2 7.5 years, 38.1\% female), including HCC, and a further of 46 participants agreed to ALI laboratory measurements. Different stress types were associated with LBP, CLBP, fatigue, and depressive mood and its joint occurrence as a symptom triad at baseline; mainly social-related stress types were of relevance. Work-related stress, such as "excessive demands at work"[b = 0.51 (95\%CI -0.23, 1.25), p = 0.18] played a role for upcoming chronic pain disability. "Social overload" [b = 0.45 (95\%CI -0.06, 0.96), p = 0.080] and "over-commitment at work" [b = 0.28 (95\%CI -0.39, 0.95), p = 0.42] were associated with an upcoming depressive mood within 1-year. Finally, seven psychometric (CPI: RMSE = 12.63; DISS: RMSE = 9.81) and five biomarkers (CPI: RMSE = 12.21; DISS: RMSE = 8.94) could be derived as the most predictive pattern set for a 1-year prediction of CLBP. The biomarker set showed an apparent area under the curve of 0.88 for CPI and 0.99 for DISS. Conclusion: Stress disrupts allostasis and favors the development of chronic pain, fatigue, and depression and the emergence of a "hypocortisolemic symptom triad," whereby the social-related stressors play a significant role. Using a genetic driver system for gene manipulation specifically at the posterior part of the WID (EnGal4) provides a robust internal control that allows for direct comparison of signal intensities of control and manipulated compartments of the same WID. Therefore, it also circumvents the high degree of staining variability usually associated with whole-tissue samples. We apply the novel NetCoupler-algorithm to link a data-driven (dihydro)ceramide network to T2D and CVD risk. Controlling for confounding by other (dihydro)ceramides, ceramides C18:0 and C22:0 and dihydroceramides C20:0 and C22:2 are associated with higher and ceramide C20:0 and dihydroceramide C26:1 with lower T2D risk. Ceramide C16:0 and dihydroceramide C22:2 are associated with higher CVD risk. Genome-wide association studies and Mendelian randomization analyses support a role of ceramide C22:0 in T2D etiology. Our results also suggest that (dh)ceramides partly mediate the putative adverse effect of high red meat consumption and benefits of coffee consumption on T2D risk.
