  abstract  = {Reflexion ist eine Schl{\"u}sselkategorie f{\"u}r die professionelle Entwicklung von Lehrkr{\"a}ften, welche als Ausbildungsziel in den Bildungsstandards f{\"u}r die Lehrkr{\"a}ftebildung verankert ist. Eine Verstetigung universit{\"a}r gepr{\"a}gter Forschung und Modellierung in der praxisnahen Anwendung im schulischen Kontext bietet Potentiale nachhaltiger Professionalisierung. Die St{\"a}rkung reflexionsbezogener Kompetenzen durch Empirie und Anwendung scheint eine phasen{\"u}bergreifende Herausforderung der Lehrkr{\"a}ftebildung zu sein, die es zu bew{\"a}ltigen gilt. Ziele des Tagungsbandes Reflexion in der Lehrkr{\"a}ftebildung sind eine theoretische Sch{\"a}rfung des Konzeptes „Reflexive Professionalisierung" und der Austausch {\"u}ber Fragen der Einbettung wirksamer reflexionsbezogener Lerngelegenheiten in die Lehrkr{\"a}ftebildung.   abstract  = {Reversible assembly of the V0V1 holoenzyme from V-0 and V-1 subcomplexes is a widely used mechanism for regulation of vacuolar-type H+-ATPases (V-ATPases) in animal cells. in the blowfly (Calliphora vicina) salivary gland, V- ATPase is located in the apical membrane of the secretory cells and energizes the secretion of a KCl-rich saliva in response to the hormone serotonin. We have examined whether the CAMP pathway, known to be activated by serotonin, controls V-ATPase assembly and activity. Fluorescence measurements of pH changes at the luminal surface of isolated glands demonstrate that CAMP, Sp-adenosine-3',5'-cyclic monophosphorothioate, or forskolin, similar to serotonin, cause V-ATPase-dependent luminal acidification. In addition, V-ATPase-dependent ATP hydrolysis increases upon treatment with these agents. Immunofluorescence microscopy and pelleting assays have demonstrated further that V, components become translocated from the cytoplasm to the apical membrane and V-ATPase holoenzymes are assembled at the apical membrane during conditions that increase intracellular cAMP. Because these actions occur without a change in cytosolic Ca2+, our findings suggest that the cAMP pathway mediates the reversible assembly and activation of V-ATPase molecules at the apical membrane upon hormonal stimulus},

  abstract  = {When we think of extremophiles, organisms adapted to extreme environments, prokaryotes come to mind first. However, the unicellular red micro-alga Galdieria sulphuraria (Cyanidiales) is a eukaryote that can represent up to 90\% of the biomass in extreme habitats such as hot sulfur springs with pH values of 0-4 and temperatures of up to 56 degreesC. This red alga thrives autotrophically as well as heterotrophically on more than 50 different carbon sources, including a number of rare sugars and sugar alcohols. This biochemical versatility suggests a large repertoire of metabolic enzymes, rivaled by few organisms and a potentially rich source of thermo-stable enzymes for biotechnology. The temperatures under which this organism carries out photosynthesis are at the high end of the range for this process, making G. sulphuraria a valuable model for physical studies on the photosynthetic apparatus. In addition, the gene sequences of this living fossil reveal much about the evolution of modern eukaryotes. Finally, the alga tolerates high concentrations of toxic metal ions such as cadmium, mercury, aluminum, and nickel, suggesting potential application in bioremediation. To begin to explore the unique biology of G. sulphuraria, 5270 expressed sequence tags from two different cDNA libraries have been sequenced and annotated. Particular emphasis has been placed on the reconstruction of metabolic pathways present in this organism. For example, we provide evidence for (i) a complete pathway for lipid A biosynthesis; (ii) export of triose-phosphates from rhodoplasts; (iii) and absence of eukaryotic hexokinases.   abstract  = {Menschen wollen wissen, was wirklich ist. Kinder lassen sich gern eine Geschichte erz{\"a}hlen, aber sp{\"a}testens mit vier Jahren fragten meine, ob diese Geschichte so passiert sei oder nur erfunden. Das setzt sich fort: Auch unsere wissenschaftliche Neugier wird vom Interesse befeuert herauszufinden, was wirklich ist. Selbst dort, wo wir poetische Texte oder Tr{\"a}ume erforschen, tun wir es in der Absicht, die realen sprachlichen Strukturen bzw. die neurologischen Faktoren von bloß vermuteten zu unterscheiden. Im Idealfall k{\"o}nnen wir Ergebnisse pr{\"a}sentieren, die von anderen logisch nachvollzogen und empirisch wiederholbar sind. Meistens geht das aber nicht. Wir k{\"o}nnen nicht jedes Buch lesen und nicht in jedes Mikroskop schauen, nicht einmal innerhalb der eigenen Disziplin. Wie viel mehr sind wir in der Lebenswelt darauf angewiesen, den Ausf{\"u}hrungen anderer zu vertrauen, wenn wir wissen wollen, wo es zum Bahnhof geht oder ob es in Ulan Bator sch{\"o}n ist. Deshalb haben wir uns daran gew{\"o}hnt, anderen Glauben zu schenken, vom Freund bis zum Tagesschausprecher. Das ist kein kindliches Verhalten, sondern eine Notwendigkeit. Freilich ist das riskant, denn alle anderen k{\"o}nnten uns - wie in der „Truman- Show" - anl{\"u}gen. In der Wirklichkeit wissen wir uns erst dann, wenn wir unser Selbstbewusstsein verlassen und akzeptieren, dass wir erstens nicht nur Objekte, sondern Subjekte im Bewusstsein von anderen sind, und zweitens, dass alle unsere dialogischen Beziehungen noch einmal von einem Dritten betrachtet werden, der nicht Teil dieser Welt ist. F{\"u}r Religi{\"o}se ist das der Glaube. Glaube als Unterstellung, dass alle menschlichen Beziehungen erst dann wirklich, ernst und {\"u}ber Zweifel erhaben sind, wenn sie sich vor den Augen Gottes wissen. Erst vor ihm ist etwas als es selbst und nicht nur „f{\"u}r mich" oder „unter uns". Daher unterscheidet die biblische Sprache drei Formen des Glaubens: die Beziehung zur Ding-Welt („glauben, dass"), die Beziehung zur Subjekt-Welt („jemandem glauben") und die Annahme einer subjekthaften {\"u}berirdischen Wirklichkeit („glauben an"). Wissenschaftstheoretisch gesehen ist Glaube also eine Totalhypothese. Glaube ist nicht das Gegenteil von Wissen, sondern der Versuch, Wirklichkeit vor dem Zweifel zu retten, indem man die fragile empirische Welt als Ausdruck einer stabilen transzendenten Welt begreift. Oft wollen Studierende in Gespr{\"a}chen nicht nur wissen, was ich weiß, sondern, was ich glaube. Als Religionswissenschaftler und gleichzeitig gl{\"a}ubiger Katholik sitze ich zwischen den St{\"u}hlen: Einerseits ist es als Professor meine Aufgabe, alles zu bezweifeln, d.h. jeden religi{\"o}sen Text auf seine historischen Kontexte und soziologischen Funktionen zur{\"u}ckzuf{\"u}hren. Andererseits h{\"a}lt der Christ in mir bestimmte religi{\"o}se Dokumente - in meinem Fall die Bibel - zwar f{\"u}r einen interpretierbaren, aber doch irreversiblen, offenbarten Text, der vom Ursprung der Wirklichkeit handelt. Werktags ist das Neue Testament eine antike Schriftensammlung neben vielen anderen, am Sonntag ist es die Offenbarung. Beides kann klar unterschieden werden, aber es ist schwer zu entscheiden, ob das Zweifeln oder das Glauben wirklicher ist. Das vorliegende Heft geht diesem doppelten Verh{\"a}ltnis zum Glauben nach: Wie steht Wissenschaft zum Glauben - ob religi{\"o}s oder nicht? Wo bringt Wissenschaft Dinge ans Licht, die wir kaum glauben m{\"o}gen oder uns (wieder) glauben lassen? Was passiert, wenn Forschung irrige Annahmen oder Mythen aufkl{\"a}rt? Ist Wissenschaft in der Lage, Dingen auf den Grund zu gehen, die zwar {\"u}berzeugend, aber unerkl{\"a}rbar sind? Wie kann sie selbst glaubw{\"u}rdig bleiben und sich dennoch weiterentwickeln? In den Beitr{\"a}gen dieser „Portal Wissen" scheinen diese Fragen immer wieder auf. Sie bilden ein vielf{\"a}ltiges, spannendes und auch {\"u}berraschendes Bild der Forschungsprojekte und der Wissenschaftler an der Universit{\"a}t Potsdam.   abstract  = {Von einem „Kulturwandel" spricht die neue Bundesregierung und will f{\"u}r einen „echten Innovationsschub" Ausgr{\"u}ndungen aus Hochschulen vorantreiben. Eine Deutsche Agentur f{\"u}r Transfer und Innovation soll die anwendungsorientierte Forschung st{\"a}rken und mit der Wirtschaft zusammenbringen. Außerdem w{\"u}nscht sie sich mehr gesellschaftliche Perspektiven in der Wissenschaft durch b{\"u}rgerschaftliches Engagement. F{\"u}r die Universit{\"a}t Potsdam bedeutet all dies keinen Aufbruch ins Unbekannte. Im Gegenteil. Sie geh{\"o}rt zu den transferst{\"a}rksten Hochschulen Deutschlands und belegt im nationalen Gr{\"u}nderradar seit Jahren vordere Pl{\"a}tze. Die wendebedingten Strukturprobleme in Brandenburg vor Augen, hat sie bereits in den 1990er Jahren nach Wegen und Werkzeugen gesucht, um neues Wissen und technologische Entwicklungen aus der Forschung in die Praxis {\"u}berf{\"u}hren zu k{\"o}nnen. Eine Erfolgsgeschichte, die sich in vielen einzelnen Erfolgsgeschichten erz{\"a}hlen l{\"a}sst. Einige davon haben wir in diesem Magazin aufgeschrieben. So berichten wir von einem „universalen Probleml{\"o}ser", der als „KI made in Potsdam" seinen Siegeszug um die Welt antritt. Oder von dem sehr jungen Start-up Koppla, das mit digitalem Werkzeug den Bau revolutioniert und daf{\"u}r 2021 den Innovationspreis Berlin-Brandenburg erhielt. Dass nicht immer eine Firma gegr{\"u}ndet werden muss, um Erfindungen zu verwerten, zeigt das Beispiel eines in Potsdamer Labors entwickelten Tests zur Fr{\"u}herkennung von Darmkrebs, der patentiert und als Lizenz an ein Unternehmen vergeben wurde. Aber nicht nur der Transfer in die Wirtschaft z{\"a}hlt, sondern auch der des Wissens in die Gesellschaft. Der Gewinner des diesj{\"a}hrigen Better World Awards, Julian Risch, hat in seiner Doktorarbeit ein Tool entwickelt, mit dem sich Hasskommentare auf Online-Plattformen automatisch identifizieren lassen. Erste Redaktionen arbeiten damit. Wenn sich so der {\"o}ffentliche Diskurs im Internet sch{\"u}tzen und erhalten l{\"a}sst, hat der junge Forscher tats{\"a}chlich dazu beigetragen, die Welt ein wenig besser zu machen. So wie er sind viele, die in unserer zweiten Ausgabe der „Portal Transfer" zu Wort kommen, Absolventinnen und Absolventen unserer Alma Mater. Alumni-Referentin Juliane Seip will enger als bisher mit ihnen in Kontakt bleiben und ein belastbares Netzwerk kn{\"u}pfen, das gemeinsame F{\"o}rderprojekte tragen kann. Zu den Ehemaligen z{\"a}hlt sie {\"u}brigens auch diejenigen, die zeitweise oder viele Jahre an der Universit{\"a}t geforscht haben, so wie der Politologe Heinz Kleger, der sich f{\"u}r das Neue Potsdamer Toleranzedikt engagiert hat. Oder der Wirtschaftswissenschaftler Dieter Wagner, der die Transfereinrichtungen an der Hochschule mit aufgebaut hat und heute der Universit{\"a}tsgesellschaft Potsdam e.V. vorsteht. Die Vereinigung der Freunde und F{\"o}rderer ist in den vergangenen Jahren auf ein Maß angewachsen, dass sie mittlerweile einzelne Fachkapitel bilden kann. Das j{\"u}ngste widmet sich dem Sport, angef{\"u}hrt von der ehemaligen Chefin des Hochschulsports, Petra Bischoff-Krenzien. Die Entwicklung junger Athletinnen und Athleten im Studium liegt ihr besonders am Herzen. Es gibt bestimmt viele Gr{\"u}nde, sich f{\"u}r diese Universit{\"a}t stark zu machen. Der wichtigste ist jedoch, dass die Allgemeinheit dabei nur gewinnen kann: durch Erfindungen und Innovationen genauso wie mithilfe kluger Ideen und neuer Erkenntnisse.},

  abstract  = {Secretion in blowfly salivary glands is induced by the neurohormone serotonin and powered by a vacuolar-type H+- ATPase (V-ATPase) located in the apical membrane of the secretory cells. We have established a microfluorometric method for analysing pH changes at the luminal surface of the secretory epithelial cells by using the fluorescent dye 5-N- hexadecanoyl-aminofluorescein (HAF). After injection of HAF into the lumen of the tubular salivary gland, the fatty acyl chain of the dye molecule partitions into the outer leaflet of the plasma membrane and its pH-sensitive fluorescent moiety is exposed at the cell surface. Confocal imaging has confirmed that HAF distributes over the entire apical membrane of the secretory cells and remains restricted to this membrane domain. Ratiometric analysis of HAF fluorescence demonstrates that serotonin leads to a reversible dose-dependent acidification at the luminal surface. Inhibition by concanamycin A confirms that the serotonin-induced acidification at the luminal surface is due to H+ transport across the apical membrane via V-ATPase. Measurements with pH-sensitive microelectrodes corroborate a serotonin-induced luminal acidification and demonstrate that luminal pH decreases by about 0.4 pH units at saturating serotonin concentrations. We conclude that ratiometric measurements of HAF fluorescence provide an elegant method for monitoring V-ATPase-dependent H+ transport in the blowfly salivary gland in vivo and for analysing the spatiotemporal pattern of pH changes at the luminal surface},

  abstract  = {In this paper, we describe tools and resources for the study of African languages developed at the Collaborative Research Centre 632 "Information Structure". These include deeply annotated data collections of 25 sub-Saharan languages that are described together with their annotation scheme, as well as the corpus tool ANNIS, which provides unified access to a broad variety of annotations created with a range of different tools. With the application of ANNIS to several African data collections, we illustrate its suitability for the purpose of language documentation, distributed access, and the creation of data archives.},

  abstract  = {Recent research illuminated the links between Agency, Communion, trait emotional intelligence (TEL), and internalizing mental health difficulties (IMHDs). However, for a more complete picture, unmitigated Agency and Communion have also to be considered. Drawing on a sample of N = 405 female occupational therapists, the present study examined (a) the factorial validity of the German TEI Questionnaire Short Form, (b) the relations of TEL to Agency. Communion, and their unmitigated variants, and (c) a multiple predictor-TEI-IMHDs mediation model. The factor structure suggested by TEI theory fitted approximately to the data. Agency and Communion related positively and both unmitigated traits related negatively to TEL. Indirect effects via TEL on IMHDs emerged for all four traits. The results help to integrate TEI within the Agency-Communion framework and suggest that TEL is an important intervening variable that helps to clarify the links of agentic and communal traits to mental health. Objective To examine whether lockdown measures are associated with AC and consumption-related temporal and psychological within-person mechanisms. Design, Setting, and Participants This quantitative, intensive, longitudinal cohort study recruited 1743 participants from 3 sites from February 20, 2020, to February 28, 2021. Data were provided before and within the second lockdown of the COVID-19 pandemic in Germany: before lockdown (October 2 to November 1, 2020); light lockdown (November 2 to December 15, 2020); and hard lockdown (December 16, 2020, to February 28, 2021). Main Outcomes and Measures Daily ratings of AC (main outcome) captured during 3 lockdown phases (main variable) and temporal (weekends and holidays) and psychological (social isolation and drinking intention) correlates. Results Of the 1743 screened participants, 189 (119 [63.0\%] male; median [IQR] age, 37 [27.5-52.0] years) with at least 2 alcohol use disorder (AUD) criteria according to the Diagnostic and Statistical Manual of Mental Disorders (Fifth Edition) yet without the need for medically supervised alcohol withdrawal were included. These individuals provided 14 694 smartphone ratings from October 2020 through February 2021. Multilevel modeling revealed significantly higher AC (grams of alcohol per day) on weekend days vs weekdays (β = 11.39; 95\% CI, 10.00-12.77; P < .001). Alcohol consumption was above the overall average on Christmas (β = 26.82; 95\% CI, 21.87-31.77; P < .001) and New Year's Eve (β = 66.88; 95\% CI, 59.22-74.54; P < .001). During the hard lockdown, perceived social isolation was significantly higher (β = 0.12; 95\% CI, 0.06-0.15; P < .001), but AC was significantly lower (β = -5.45; 95\% CI, -8.00 to -2.90; P = .001). Independent of lockdown, intention to drink less alcohol was associated with lower AC (β = -11.10; 95\% CI, -13.63 to -8.58; P < .001). Notably, differences in AC between weekend and weekdays decreased both during the hard lockdown (β = -6.14; 95\% CI, -9.96 to -2.31; P = .002) and in participants with severe AUD (β = -6.26; 95\% CI, -10.18 to -2.34; P = .002). Conclusions and Relevance This 5-month cohort study found no immediate negative associations of lockdown measures with overall AC. Rather, weekend-weekday and holiday AC patterns exceeded lockdown effects. Differences in AC between weekend days and weekdays evinced that weekend drinking cycles decreased as a function of AUD severity and lockdown measures, indicating a potential mechanism of losing and regaining control. This finding suggests that temporal patterns and drinking intention constitute promising targets for prevention and intervention, even in high-risk individuals.},

  abstract  = {Oil palm (Elaeis guineensis Jacq.) is the most productive oil-producing crop per hectare of land. The oil that accumulates in the mesocarp tissue of the fruit is the highest observed among fruit-producing plants. A comparative analysis between high-, medium-, and low-yielding oil palms, particularly during fruit development, revealed unique characteristics. Metabolomics analysis was able to distinguish accumulation patterns defining of the various developmental stages and oil yield. Interestingly, high- and medium-yielding oil palms exhibited substantially increased sucrose levels compared to low-yielding palms. In addition, parameters such as starch granule morphology, granule size, total starch content, and starch chain length distribution (CLD) differed significantly among the oil yield categories with a clear correlation between oil yield and various starch parameters. These results provide new insights into carbohydrate and starch metabolism for biosynthesis of oil palm fruits, indicating that starch and sucrose can be used as novel, easy-to-analyze, and reliable biomarker for oil yield.},
