Mapping of surface fault ruptures and high-resolution geodetic observations are increasingly revealing complex fault geometries near the surface and accurate locations of aftershocks often indicate geometrical complexities at depth. With better geodetic data and observations of fault ruptures, more details of complex fault geometries can be estimated resulting in more realistic fault models of large earthquakes. To address this topic, we here parametrize non-planar fault geometries with a set of polynomial parameters that allow for both along-strike and down-dip variations in the fault geometry. Our methodology uses Bayesian inference to estimate the non-planar fault parameters from geodetic data, yielding an ensemble of plausible models that characterize the uncertainties of the non-planar fault geometry and the fault slip. The method is demonstrated using synthetic tests considering slip spatially distributed on a single continuous finite non-planar fault surface with varying dip and strike angles both in the down-dip and along-strike directions. The results show that fault-slip estimations can be biased when a simple planar fault geometry is assumed in presence of significant non-planar geometrical variations. The overarching goal was to elucidate the mechanisms leading to sedimentation and deposition of microplastics in such environments. This is of importance, as large amounts of initially buoyant microplastics are found in reservoir sediments worldwide. However, the transport processes which lead to microplastics accumulation in sediments, were up to now understudied. The impact of biofilm formation on the density and subsequent sedimentation of microplastics was investigated in the eutrophic Bautzen reservoirs (Chapter 2). Biofilms are complex microbial communities fixed to submerged surfaces through a slimy organic film. The mineral calcite was detected in the biofilms, which led to the sinking of the overgrown microplastic particles. The calcite was of biogenic origin, most likely precipitated by sessile cyanobacteria within the biofilms. Biofilm formation was also studied in the mesotrophic Malter reservoir. Unlike in Bautzen reservoir, biofilm formation did not govern the sedimentation of different microplastics in Malter reservoir (Chapter 3). Instead autumnal lake mixing led to the formation of sinking aggregates of microplastics and iron colloids. Such colloids form when anoxic, iron-rich water from the hypolimnion mixes with the oxygenated epilimnetic waters. The colloids bind organic material from the lake water, which leads to the formation of large and sinking iron-organo flocs. Hence, iron-organo floc formation and their influence on the buoyancy or burial of microplastics into sediments of Bautzen reservoir was studied in laboratory experiments (Chapter 4). Microplastics of different shapes (fiber, fragment, sphere) and sizes were readily incorporated into sinking iron-organo flocs. By this initially buoyant polyethylene microplastics were transported on top of sediments from Bautzen reservoir. Shortly after deposition, the microplastic bearing flocs started to subside and transported the pollutants into deeper sediment layers. The microplastics were not released from the sediments within two months of laboratory incubation. The stability of floc microplastic deposition was further investigated employing experiments with the iron reducing model organism Shewanella oneidensis (Chapter 5). It was shown, that reduction or re-mineralization of the iron minerals did not affect the integrity of the iron-organo flocs. The organic matrix was stable under iron reducing conditions. Hence, no incorporated microplastics were released from the flocs. As similar processes are likely to take place in natural sediments, this might explain the previous described low microplastic release from the sediments. This thesis introduced different mechanisms leading to the sedimentation of initially buoyant microplastics and to their subsequent deposition in freshwater reservoirs. Novel processes such as the aggregation with iron-organo flocs were identified and the understudied issue of biofilm densification through biogenic mineral formation was further investigated. The findings might have implications for the fate of microplastics within the river-reservoir system and outline the role of freshwater reservoirs as important accumulation zone for microplastics. Microplastics deposited in the sediments of reservoirs might not be transported further by through flowing river. They can serve as a real threat for fish or mammals and can restrict the use of lakes or rivers for recreational purposes or as a source of drinking water, when they form blooms. One of the most abundant bloom-forming cyanobacteria is Microcystis aeruginosa. In the first part of the study, the role and possible dynamics of RubisCO in M. aeruginosa during high-light irradiation were examined. Its response was analyzed on the protein and peptide level via immunoblotting, immunofluorescence microscopy and with high performance liquid chromatography (HPLC). It was revealed that large amounts of RubisCO were located outside of carboxysomes under the applied high light stress. RubisCO aggregated mainly underneath the cytoplasmic membrane. There it forms a putative Calvin-Benson-Bassham (CBB) super complex together with other enzymes of photosynthesis. This complex could be part of an alternative carbon-concentrating mechanism (CCM) in M. aeruginosa, which enables a faster, and energy saving adaptation to high light stress of the whole bloom. Furthermore, the re-localization of RubisCO was delayed in the microcystin-deficient mutant ΔmcyB and RubisCO was more evenly distributed over the cell in comparison to the wild type. Since ΔmcyB is not harmed in its growth, possibly other produced cyanopeptides as aeruginosin or cyanopeptolin also play a role in the stabilization of RubisCO and the putative CBB complex, especially in the microcystin-free mutant. In the second part of this work, the possible role of microcystin as an extracellular signaling peptide during the diurnal cycle was studied. HPLC analysis showed a strong increase of extracellular microcystin in the wild type when the population entered nighttime and it resumed into the next day as well. Together with the increase of extracellular microcystin, a strong decrease of protein-bound intracellular microcystin was observed via immunoblot analysis. Interestingly, the signal of the large subunit of RubisCO (RbcL) also diminished when high amounts of microcystin were present in the surrounding medium. Microcystin addition experiments to M. aeruginosa WT and ΔmcyB cultures support this observation, since the immunoblot signal of both subunits of RubisCO and CcmK, a shell protein of carboxysomes, diminished after the addition of microcystin. In this study, we investigated the potential of fungi to degrade kelp detritus, which is a major source of cellulose in marine systems. Given that kelp detritus can be transported considerable distances in the marine environment, we were specifically interested in the capability of endophytic fungi, which are transported with detritus, to ultimately contribute to kelp detritus degradation. We isolated 10 species and two strains of endophytic fungi from the kelp Ecklonia radiata. We then used a dye decolorization assay to assess their ability to degrade organic polymers (lignin, cellulose, and hemicellulose) under both oxic and anoxic conditions and compared their degradation ability with common terrestrial fungi. Under oxic conditions, there was evidence that Ascomycota isolates produced cellulose-degrading extracellular enzymes (associated with manganese peroxidase and sulfur-containing lignin peroxidase), while Mucoromycota isolates appeared to produce both lignin and cellulose-degrading extracellular enzymes, and all Basidiomycota isolates produced lignin-degrading enzymes (associated with laccase and lignin peroxidase). Under anoxic conditions, only three kelp endophytes degraded cellulose. We concluded that kelp fungal endophytes can contribute to cellulose degradation in both oxic and anoxic environments. Here, we review the use of spatially explicit models for decision support and to identify key gaps in current modelling in conservation and restoration. Of 650 reviewed publications, 217 publications had a clear management application and were included in our quantitative analyses. Overall, modelling studies were biased towards static models (79\%), towards the species and population level (80\%) and towards conservation (rather than restoration) applications (71\%). Correlative niche models were the most widely used model type. Dynamic models as well as the gene-to-individual level and the community-to-ecosystem level were underrepresented, and explicit cost optimisation approaches were only used in 10\% of the studies. We present a new model typology for selecting models for animal conservation and restoration, characterising model types according to organisational levels, biological processes of interest and desired management applications. This typology will help to more closely link models to management goals. Additionally, future efforts need to overcome important challenges related to data integration, model integration and decision-making. We conclude with five key recommendations, suggesting that wider usage of spatially explicit models for decision support can be achieved by 1) developing a toolbox with multiple, easier-to-use methods, 2) improving calibration and validation of dynamic modelling approaches and 3) developing best-practise guidelines for applying these models. Further, more robust decision-making can be achieved by 4) combining multiple modelling approaches to assess uncertainty, and 5) placing models at the core of adaptive management. Ergebnisse: Empirische Befunde zeigen, dass SP psychische Storungen authentisch darstellen konnen. Voraussetzung dafur sind beispielsweise die Auswahl geeigneter SP, detaillierte Rollenanleitungen, spezifisches Training, Feedback und Nachschulungen. Auch wenn einige Forschungsfragen, wie zur vergleichenden Wirksamkeit des Einsatzes von SP, noch unbeantwortet sind, lassen sich praktische Implikationen fur SP-Programme in Lehre, Prufung und Forschung ableiten, die in einem Ablaufschema dargestellt werden. Schlussfolgerungen: Der Einsatz von SP bietet gro ss es Potenzial fur die klinisch-psychologische Lehre und Ausbildungsforschung. Dyer and Greenhill proved a dichotomy stating that the tractability of non-modular counting graph homomorphisms depends on the structure of the target graph. Many intractable cases in non-modular counting become tractable in modular counting due to the common phenomenon of cancellation. In subsequent studies on counting modulo 2, however, the influence of the structure of H on the tractability was shown to persist, which yields similar dichotomies.
Our main result states that for every tree H and every prime p the problem \#pHOMSTOH is either polynomial time computable or \#P-p-complete. This relates to the conjecture of Faben and Jerrum stating that this dichotomy holds for every graph H when counting modulo 2. In contrast to previous results on modular counting, the tractable cases of \#pHOMSTOH are essentially the same for all values of the modulo when H is a tree. To prove this result, we study the structural properties of a homomorphism. As an important interim result, our study yields a dichotomy for the problem of counting weighted independent sets in a bipartite graph modulo some prime p. This thesis experimentally addresses specific quasi-particle interactions, namely interactions be- tween electrons and between electrons and phonons. It describes the consequential effects of those processes on the electronic structure and the core-hole relaxation pathways in 3d metals. Despite the great amount of experimental and theoretical studies of these interactions and their impact on the behavior of solid-state matter, there are still open questions concerning the cor- responding physical, chemical and mechanical properties of solid-state matter. Especially, the study of 3d metals and their compounds is a great experimental challenge, since those exhibit a variety of spectral features originating from many-body effects such as multiplet splitting, shake up/off satellites, vibrationally excited states or more complex effects like superconductivity and ultrafast demagnetization. In X-ray spectroscopy, these effects often produce overlapping fea- tures, complicating the analysis and limiting the understanding. In this thesis, to overcome the limitations set by conventional X-ray spectroscopy, two different experimental approaches were successfully refined, namely Auger electron photoelectron coincidence spectroscopy (APECS) and temperature-dependent X-ray emission spectroscopy (tXES), which enabled the separation of different core-hole relaxation pathways and the isolation of the impact of specific many-body interactions in the experimental spectra. APECS was utilized at the new Coincidence electron spectroscopy for chemical analysis (Co- ESCA) station at BESSY II to study the core-hole decay and electron-correlation effects in single- crystal Ni, Cu and Co. The observation of photoelectrons in coincidence with Auger electrons allows for the separation of the initial and final state effects in the Auger electron spectra. The results show that a Cu LV V Auger spectrum can be represented by broadened atomic multiplets confirming the localized nature of the intermediate core-hole states. In contrast, the Co LV V Auger spectrum is band-like and can be represented by the self-convolution of the valence band. Ni behaves mixed, localized and itinerant. Thus, the Ni Auger spectrum can only be represented by a mixture of atomic multiplet peaks and the self-convoluted valence band. In the case of Ni, the LV V Auger electrons in coincidence with the 6 eV satellite photoelectrons were also stud- ied. Utilizing the core-hole clock method, the lifetime of the localized double-hole intermediate 2 p53d9 states of 1.8 fs could be determined. However, a fraction of these states delocalizes before the Auger decay contributing to the main peak. A similar delocalization was observed for the double-hole states produced by the L2L3M4,5 Coster-Kronig process. Additionally, the influence of surface oxidation on the Ni(111) 3p levels was studied with APECS. The Ni 3p PES spectrum is broad and featureless, due to overlapping many-body effects and gives little chance for exact analysis using conventional photoelectron spectroscopy. Utilizing APECS or precisely the final state selectivity of the method, the spectral width of the 3p levels could be narrowed and their positions and the spin-orbit splitting were determined. Moreover, due to the surface sensitivity of the method, the chemically shifted 3p photoelectron peaks originating from the oxidized surface and the bulk Ni were disentangled. For the study of the atomic electron-phonon spin-flip scattering in 3d metals as a spin-relaxation channel, the tXES method at the SolidFlexRIXS station was developed. The atomic spin-flip scat- tering was studied in single-crystal Ni, Cu, Co and in FeNi alloys, which show considerable dif- ferences in their behavior. The scattering rate in Ni increases with temperature, whereas the rate in Cu and Co remains constant within the measured temperature range up to 1000 K. In FeNi alloys, our results reveal that the spin-flip scattering is restricted by sublattice exchange energies J. The electron-phonon scattering driven spin-flips only appear in the case where the thermal energy ex- ceeds the exchange energy kT > J. This thresholding is an important microscopic process for the description of the sublattice dynamics in alloys, but as shown also relevant for elemental magnetic systems. Overall, the results strongly indicate that the spin-flip probability is correlated with the exchange energy, which might become an important parameter in the ultrafast demagnetization debate. Taken together, the applied experimental approaches allowed to study complex many-body effects in 3d metals. The results show that utilizing APECS enabled the distinction and clear assignment of otherwise overlapping features in AES or PES spectra of Ni, Cu, Co and NiO. This is of fundamental importance for the basic understanding of photoionization and core-hole decay processes but also for the chemical analysis in applied science. The measurement of the atomic electron-phonon spin-flip scattering rate utilizing tXES shows that the electron-phonon spin-flip scattering is a relevant atomic process for the macroscopic demagnetization process. We perform two stick-slip experiments on fractured (rough) and cut (smooth) Westerly granite samples to explore fault roughness effects on acoustic emission (AE) source parameters. We separate large stick-slip events that generally saturate the seismic recording system from populations of smaller AE events which are sensitive to fault stresses prior to slip. AE event populations show many similarities to natural seismicity and may be interpreted as laboratory equivalent of natural microseismic events. We then compare the temporal evolution of mechanical data such as measured stress release during slip to temporal changes in stress drops derived from Alis using the spectral ratio technique. We report on two primary observations: (1) In contrast to most case studies for natural earthquakes, we observe a strong increase in seismic stress drop with AE size. (2) The scaling of stress drop with magnitude is governed by fault roughness, whereby the rough fault shows a more rapid increase of the stress drop magnitude relation with progressing large stick-slip events than the smooth fault. The overall range of AE sizes on the rough surface is influenced by both the average grain size and the width of the fault core. The magnitudes of the smallest AE events on smooth faults may also be governed by grain size. However, AEs significantly grow beyond peak roughness and the width of the fault core. Unlike previous interpretations, we ascribe spatial electron-localization to the laser creation of a rovibronic wavepacket that involves field-free molecular eigenstates with opposite space-inversion symmetry i.e., even and odd parity. Space-inversion symmetry breaking would then lead to an asymmetric distribution of the (space-fixed) electronic density over the forward and backward hemisphere. However, owing to the simultaneous coexistence of two indistinguishable molecular orientational isomers, our analytical and computational results show that the post-dissociation electronic density along a specified space-fixed axis is equally shared between the two identical nuclei-a result that is in perfect accordance with the principle of the indistinguishability of identical particles. In each instance, these measurements helped in unravelling the geodynamics of the region and seismic hazard assessment, e.g., the discovery of a plate boundary fault in the Indo-Burmese wedge, rate and mode of strain accumulation and its spatial variation in the Garhwal-Kumaun and Kashmir region of the Himalayan arc, the influence of non-tectonic deformation on tectonic deformation in the Himalayan arc, nature of crustal deformation through earthquake cycle in the Andaman Sumatra subduction zone, and localised deformation in the intraplate region and across the paleo rift regions. Besides these, GNSS measurements initiated in the Antarctica region have helped in understanding the plate motion and influence of seasonal variations on deformation. Another important by-product of the GNSS observations is the capabilities of these observations in understanding the ionospheric variations due to earthquake processes and also due to solar eclipse. We isolated 10 species and two strains of endophytic fungi from the kelp Ecklonia radiata. We then used a dye decolorization assay to assess their ability to degrade organic polymers (lignin, cellulose, and hemicellulose) under both oxic and anoxic conditions and compared their degradation ability with common terrestrial fungi. Under oxic conditions, there was evidence that Ascomycota isolates produced cellulose-degrading extracellular enzymes (associated with manganese peroxidase and sulfur-containing lignin peroxidase), while Mucoromycota isolates appeared to produce both lignin and cellulose-degrading extracellular enzymes, and all Basidiomycota isolates produced lignin-degrading enzymes (associated with laccase and lignin peroxidase). Under anoxic conditions, only three kelp endophytes degraded cellulose. We concluded that kelp fungal endophytes can contribute to cellulose degradation in both oxic and anoxic environments. It has been shown previously that HSP60 is downregulated in brains of type 2 diabetic (T2D) mice and patients, causing mitochondrial dysfunction and insulin resistance. As HSP60 is also decreased in peripheral tissues in T2D animals, this thesis investigated the effect of overall reduced HSP60 in the development of obesity and associated co-morbidities. To this end, both female and male C57Bl/6N control (i.e. without further alterations in their genome, Ctrl) and heterozygous whole-body Hsp60 knock-out (Hsp60+/-) mice, which exhibit a 50 \% reduction of HSP60 in all tissues, were fed a normal chow diet (NCD) or a highfat diet (HFD, 60 \% calories from fat) for 16 weeks and were subjected to extensive metabolic phenotyping including indirect calorimetry, NMR spectroscopy, insulin, glucose and pyruvate tolerance tests, vena cava insulin injections, as well as histological and molecular analysis. Interestingly, NCD feeding did not result in any striking phenotype, only a mild increase in energy expenditure in Hsp60+/- mice. Exposing mice to a HFD however revealed an increased body weight due to higher muscle mass in female Hsp60+/- mice, with a simultaneous decrease in energy expenditure. Additionally, these mice displayed decreased fasting glycemia. Opposingly, male Hsp60+/- compared to control mice showed lower body weight gain due to decreased fat mass and an increased energy expenditure, strikingly independent of lean mass. Further, only male Hsp60+/- mice display improved HOMA-IR and Matsuda insulin sensitivity indices. Despite the opposite phenotype in regards to body weight development, Hsp60+/- mice of both sexes show a significantly higher cell number, as well as a reduction in adipocyte size in the subcutaneous and gonadal white adipose tissue (sc/gWAT). Curiously, this adipocyte hyperplasia - usually associated with positive aspects of WAT function - is disconnected from metabolic improvements, as the gWAT of male Hsp60+/- mice shows mitochondrial dysfunction, oxidative stress, and insulin resistance. Transcriptomic analysis of gWAT shows an up regulation of genes involved in macroautophagy. Confirmatory, expression of microtubuleassociated protein 1A/1B light chain 3B (LC3), as a protein marker of autophagy, and direct measurement of lysosomal activity is increased in the gWAT of male Hsp60+/- mice. In summary, this thesis revealed a novel gene-nutrient interaction. After presenting our field site and details regarding our data acquisition and processing, we calculate and filter 3D texture attributes to generate a database comprising the dominant texture features of our GPR data. Then, we perform a dimensionality reduction of this database to obtain meta texture attributes, which we analyze and integrate using composite imaging and (also considering additional geometric information) fuzzy c-means cluster analysis resulting in a classified GPR facies model. Considering our facies model and a corresponding GPR facies chart, we interpret our GPR data set in terms of near-surface sedimentary units, the corresponding depositional environments, and the recent formation history at our field site. Thus, we hypothesized that a reduction of Hsp10 in hypothalamic neurons will impair mitochondrial function and impact brain insulin action. Therefore, we investigated the role of chaperone Hsp10 by introducing a lentiviral-mediated Hsp10 knockdown (KD) in the hypothalamic cell line CLU-183 and in the arcuate nucleus (ARC) of C57BL/6N male mice. Temperature was a more important determinant of ring growth at the drier site (Kuzikus), while rainfall was more important at the wetter site (Waterberg). Growth ring responsiveness to rainfall was not immediate but showed a rather lagged pattern. We conclude that D. cinerea differentially responds to variations in rainfall and temperature across short climatic gradients. Previous studies investigating how functional diversity influences the response of food webs to disturbances have mainly considered systems with at most two functionally diverse trophic levels. We investigated the effects of functional diversity on the robustness, that is, resistance, resilience, and elasticity, using a tritrophic—and thus more realistic—plankton food web model. We compared a non-adaptive food chain with no diversity within the individual trophic levels to a more diverse food web with three adaptive trophic levels. The species fitness differences were balanced through trade-offs between defense/growth rate for prey and selectivity/half-saturation constant for predators. We showed that the resistance, resilience, and elasticity of tritrophic food webs decreased with larger perturbation sizes and depended on the state of the system when the perturbation occurred. Importantly, we found that a more diverse food web was generally more resistant and resilient but its elasticity was context-dependent. Particularly, functional diversity reduced the probability of a regime shift toward a non-desirable alternative state. The basal-intermediate interaction consistently determined the robustness against a nutrient pulse despite the complex influence of the shape and type of the dynamical attractors. This relationship was strongly influenced by the diversity present and the third trophic level. Currently, medical and pharmaceutical work is a complicated process of performing professional functions connected with the observance of established standards and requirements of its organization. Most medical tests and manipulations of prevention, research, diagnostic, treatment or rehabilitation character are regulated by formal protocols which could, in some cases, prevent a qualified doctor from saving a patient's life and in others - inflict forced harm on their health. Both of these situations require a legal assessment of the actions, the mechanism of which has not yet been fully determined. This circumstance could lead to a criminal prosecution of a medical professional whose fault is absent (or nonobvious). On the other hand, a structurally complex professional activity, for which there are no recognized methods of legal assessment, creates preconditions for various violations and abuses on the part of medical professionals. Changing relationships between a doctor and a patient, as well as the commercialization of modern medical practices have made the healthcare system one of the most delicto- and even criminally-oriented.
These factors act as causes for the growing complexity of legislation (in the wide sense of the word) on criminal liability of medical professionals and the controversial law enforcement practice which, in its turn, leads to the interest of researchers in these problems. The results of such research often remain unconnected with other achievements of the criminal law science; due to this, it is necessary to study the development of law, including the practice of law enforcement and the doctrine, on criminal liability of medical professionals - medical criminal law. We focused on the expression levels of chromatin modifiers and SNPs in regulatory sequences. We identified alternative splicing events in islets of diabetes-susceptible mice amongst others in genes linked to insulin secretion, endocytosis or ubiquitin-mediated proteolysis pathways. The expression pattern of 54 histones and chromatin modifiers, which may modulate splicing, were markedly downregulated in islets of diabetic animals. Furthermore, diabetes-susceptible mice carry SNPs in RNA-binding protein motifs and in splice sites potentially responsible for alternative splicing events. They also exhibit a larger exon skipping rate, e.g., in the diabetes gene Abcc8, which might affect protein function. Expression of the neuronal splicing factor Srrm4 which mediates inclusion of microexons in mRNA transcripts was markedly lower in islets of diabetes-prone compared to diabetes-resistant mice, correlating with a preferential skipping of SRRM4 target exons. The repression of Srrm4 expression is presumably mediated via a higher expression of miR-326-3p and miR-3547-3p in islets of diabetic mice. The level of HSP21 is negatively controlled by plastid-localized metalloprotease FtsH6 during heat stress recovery. Here, we demonstrate that autophagy, a cellular recycling mechanism, exerts additional control over HSP21 degradation. Genetic and chemical disruption of both metalloprotease activity and autophagy trigger superior HSP21 accumulation, thereby improving memory. Furthermore, we provide evidence that autophagy cargo receptor ATG8-INTERACTING PROTEIN1 (ATI1) is associated with heat stress memory. ATI1 bodies co-localize with both autophagosomes and HSP21, and their abundance and transport to the vacuole increase during heat stress recovery. Latent Transition Analyses showed that motivational beliefs became more hierarchical over time. Gendered changes in profile membership occurred, with boys experiencing a process of specialization into mathematics domains. One proxy (crown defoliation) solely captured Seca-dependent effects, one proxy solely captured Seca-independent effects (tree dimensions such as diameter and height), while the third proxy (tree vigor) captured overall canopy effects. We then used the best-performing proxies to assess canopy effects on key ecosystem services (ESs) such as aboveground net primary production (ANPP), grass and legume biomass, species diversity, litter decomposition rates, and a combined index of ecosystem multifunctionality.
We found that both types of canopy effects (i.e. Seca-dependent and Seca-independent effects) were related, indicating that ANPP was disproportionally more affected by Seca when defoliated trees were large. Responses of other ESs were mostly not significant, although lower species diversity was found under trees with intermediate vigor. Our results underline that a Seca-related decline in canopy density triggered a homogenization of ecosystem service delivery on the ecosystem scale. The ecosystem functions (EFs) under trees of low vigor are similar to that in adjacent open microsites indicating that the presence of vigorous (i.e. old and vital) trees is critical for maintaining EFs at a landscape level. Overall, weight gains ranged from 21\% to 42\% after five days of feeding. The larval death rate significantly increased in all groups after 10 days of feeding (p < 0.05), whereas the pupation rate was about 25\% among larvae fed with rice (Oryza sativa L.) and Siyazan/Esperya wheat meals, and only 8\% and 14\% among those fed with Damougari and S35 sorghum meals. As determined using the Lowry method, the protein contents of the sodium phosphate extracts ranged from 7.80 ± 0.09 to 9.42 ± 0.19 mg/mL and those of the ammonium bicarbonate/urea reached 19.78 ± 0.16 to 37.47 ± 1.38 mg/mL. The total protein contents of the larvae according to the Kjeldahl method ranged from 44.0 and 49.9 g/100 g. The relative abundance of α-amylase, CLIP domain-containing serine protease, modular serine protease zymogen and C1 family cathepsin significantly decreased in the larvae, whereas dipeptidylpeptidase I and chymotrypsin increased within the first hours after feeding (p < 0.05). Trypsin content was found to be constant independently of time or feed material. All three groups performed worse in the non-food condition only for number of errors and raw score. Conclusions and implications: Planning abilities were impaired in PWS adults. Results also showed that intellectual level plays a role in participants' performance. The waveform data of a subset of local earthquakes with magnitudes in the range of 1-4.2 M-L were recorded by the dense, temporary SWATH-D network and selected stations of the AlpArray network between September 2017 and the end of 2018. The first arrival times of P and S waves of earthquakes are determined by a semi-automatic procedure. We applied a Markov chain Monte Carlo inversion method to simultaneously calculate robust hypocenters, a 1-D velocity model, and station corrections without prior assumptions, such as initial velocity models or earthquake locations. A further advantage of this method is the derivation of the model parameter uncertainties and noise levels of the data. The precision estimates of the localization procedure is checked by inverting a synthetic travel time dataset from a complex 3-D velocity model and by using the real stations and earthquakes geometry. The location accuracy is further investigated by a quarry blast test. The average uncertainties of the locations of the earthquakes are below 500m in their epicenter and similar to 1.7 km in depth. The earthquake distribution reveals seismicity in the upper crust (0-20 km), which is characterized by pronounced clusters along the Alpine frontal thrust, e.g., the Friuli-Venetia (FV) region, the Giudicarie-Lessini (GL) and Schio-Vicenza domains, the Austroalpine nappes, and the Inntal area. Some seismicity also occurs along the Periadriatic Fault. The general pattern of seismicity reflects head-on convergence of the Adriatic indenter with the Alpine orogenic crust. A prominent approach in linguistic theory argues that these types of exposure effects (satiation effects) show direct evidence of a generative approach to linguistic knowledge: only some sentences improve under repeated exposure, and which sentences do improve can be predicted by a model of linguistic competence that yields natural syntactic classes. However, replications of the original findings have been inconsistent, and it remains unclear whether satiation effects can be reliably induced in an experimental setting at all. Here we report four findings regarding satiation effects in wh-questions across German and English. First, the effects pertain to zone of well-formedness rather than syntactic class: all intermediate ratings, including calibrated fillers, increase at the beginning of the experimental session regardless of syntactic construction. Second, though there is satiation, ratings asymptote below maximum acceptability. Third, these effects are consistent across judgments of superiority effects in English and German. Fourth, wh-questions appear to show similar profiles in English and German, despite these languages being traditionally considered to differ strongly in whether they show effects on movement: violations of the superiority condition can be modulated to a similar degree in both languages by manipulating subject-object initiality and animacy congruency of the wh-phrase. We improve on classic satiation methods by distinguishing between two crucial tests, namely whether exposure selectively targets certain grammatical constructions or whether there is a general repeated exposure effect. We conclude that exposure effects can be reliably induced in rating experiments but exposure does not appear to selectively target certain grammatical constructions. As such, the open-circuit voltage of the PM6:Y6 blend is determined by the energetics and kinetics of the charge-transfer (CT) state. Moreover, we find that no information on the energetics of the CT state manifold can be gained from the low-energy tail of the photovoltaic external quantum efficiency spectrum, which is dominated by the excitation spectrum of the Y6 exciton. Results: To address this issue, we formalize protein complexes as biclique spanned subgraphs, which include both sparse and dense subgraphs. We then cast the problem of protein complex prediction as a network partitioning into biclique spanned subgraphs with removal of minimum number of edges, called coherent partition. Since finding a coherent partition is a computationally intractable problem, we devise a parameter-free greedy approximation algorithm, termed Protein Complexes from Coherent Partition (PC2P), based on key properties of biclique spanned subgraphs. Through comparison with nine contenders, we demonstrate that PC2P: (i) successfully identifies modular structure in networks, as a prerequisite for protein complex prediction, (ii) outperforms the existing solutions with respect to a composite score of five performance measures on 75\% and 100\% of the analyzed PPI networks and gold standards in yeast and human, respectively, and (iii,iv) does not compromise GO semantic similarity and enrichment score of the predicted protein complexes. Oxygen uptake in absolute (VO2) and relative to body weight (rel. VO2), lactate, and the rate of perceived exertion (RPE) were measured before and after the test. WB-EMS intensity was adjusted individually according to the feedback of the participant. The descriptive statistics were given in mean ± SD. For the statistical analyses, one-factorial ANOVA for repeated measures and two-factorial ANOVA [factors include EMS, W/NW, and factor combination (EMS*W/NW)] were performed (α = 0.05). Significant effects were found for EMS and W/NW factors for the outcome variables VO2 (EMS: p = 0.006, r = 0.736; W/NW: p < 0.001, r = 0.870), relative VO2 (EMS: p < 0.001, r = 0.850; W/NW: p < 0.001, r = 0.937), and lactate (EMS: p = 0.003, r = 0.771; w/NW: p = 0.003, r = 0.764) and both the factors produced higher results. However, the difference in VO2 and relative VO2 is within the range of biological variability of ± 12\%. The factor combination EMS*W/NW is statistically non-significant for all three variables. WB-EMS resulted in the higher RPE values (p = 0.035, r = 0.613), RPE differences for W/NW and EMS*W/NW were not significant. The as-fabricated CuNPs-incorporated PCL membrane [PCL-(MI-hPG)-CuNPs] was confirmed with effective antibacterial performance via in vitro antibacterial tests against Staphylococcus aureus (S. aureus), Escherichia coli (E. coli), and multi-resistant E. coli. In addition, the in vitro results demonstrated that osteogenic property of PCL-(MI-hPG)-CuNPs was realized by upregulating the osteoblast-related gene expressions and protein activity. Using data from the modern period (1947-2012), we show that the established understanding (i.e. deep-water formation in the North Atlantic cools the deep ocean and warms the surface) is correct, but our analysis is not based on this fact. Secondly, C\&T claim that our results are based on a statistical analysis of only one cycle of data which was furthermore incorrectly detrended. This, too, is not true. Our conclusion that a weaker AMOC delays the current surface warming rather than enhances it, is based on several independent lines of evidence. The data we show to support this covers more than one cycle and the detrending (which was performed to avoid spurious correlations due to a common trend) does not affect our conclusion: the correlation between AMOC strength and GMST is positive. In 2015, we examined the vegetation change in 140 semi-permanent plots in managed forests in the Elbe valley in the NE German lowlands (Saxony-Anhalt, Brandenburg). The first survey took place from 1956 to 1963. The releves cover an almost uniquely broad spectrum of different site conditions, ranging from wet forests (alluvial, swamp and bog forests of Alnion incanae, Alnion glutinosae and Betulion pubescentis) to acidic mixed oak forests (Quercion roboris) up to acidic, mostly dry pine forests with different nutrient status (Dicrano-Pinion). We analyzed the changes in the vegetation with the help of forest stand data, winner and loser species, alpha- and beta-diversity as well as the Ellenberg indicator values for nitrogen, reaction, moisture and light. In contrast to previous resurvey studies, areas were also taken into account on which a complete change of forest stand had taken place before the second survey. Particularly in the wet forests and acidic forests with a moderately good nutrient supply, changes in the main tree species have been recorded, and many pine stands have been newly established in the meantime. The species richness has decreased overall and in almost all forest types, but the beta-diversity has remained unchanged or has increased. The Ellenberg values indicate a decrease in soil moisture in the wet forests, while the acidic pine forests in particular have become darker, richer in nutrients and more humid. The number of loser species is more than twice as high as that of the winner species, but with different developments in the individual forest types. In particular, the wet forests, the acidic mixed oak forests and the lichen-pine forests have lost most of their characteristic species. The resurvey after more than 50 years shows a different development of the individual forest types. Vegetation changes in the wet forests are mainly due to local groundwater level drawdown and the resulting increased availability of nutrients. The alluvial forests were also strongly influenced by forest interventions. The reasons for the trend towards more humid and more nutrient-rich conditions in formerly dry acidic pine and oak forests are nitrogen depositions and a succession after the abandonment of historical forms of forest use (litter raking, forest pasture). Although the individual forest types have developed differently, eutrophication, falling groundwater levels and silviculture are the most important causes for the changes in vegetation. We address this question by exploring the sensitivity of a multivariate logistic regression-one of the most widely used susceptibility models-to data sampled from different portions of landslides in two independent inventories (i.e. a historic and a multi-temporal) covering parts of the eastern rim of the Fergana Basin, Kyrgyzstan. We find that considering only areas on lower parts of landslides, and hence most likely their deposits, can improve the model performance by >10\% over the reference case that uses the entire landslide areas, especially for landslides of intermediate size. Hence, using landslide toe areas may suffice for this particular model and come in useful where landslide scars are vague or hidden in this part of Central Asia. The model performance marginally varied after progressively updating and adding more landslides data through time. We conclude that landslide susceptibility estimates for the study area remain largely insensitive to changes in data over about a decade. Spatial or temporal stratified sampling contributes only minor variations to model performance. The recent discovery of a UHE white dwarf that is both spectroscopically and photometrically variable led to the speculation that the UHE lines might be created in a shock-heated circumstellar magnetosphere. Aims. We aim to gain a better understanding of these mysterious objects by studying the photometric variability of the whole population of UHE white dwarfs, and white dwarfs showing only the He II line problem, as both phenomena are believed to be connected. Methods. We investigate (multi-band) light curves from several ground- and space-based surveys of all 16 currently known UHE white dwarfs (including one newly discovered) and eight white dwarfs that show only the He II line problem. Results. We find that 75(-13)(+8) \% of the UHE white dwarfs, and 75(-19)(+9)\% of the He II line problem white dwarfs are significantly photometrically variable, with periods ranging from 0.22 d to 2.93 d and amplitudes from a few tenths to a few hundredths of a magnitude. The high variability rate is in stark contrast to the variability rate amongst normal hot white dwarfs (we find 9(2)(+4)\%), marking UHE and He II line problem white dwarfs as a new class of variable stars. The period distribution of our sample agrees with both the orbital period distribution of post-common-envelope binaries and the rotational period distribution of magnetic white dwarfs if we assume that the objects in our sample will spin-up as a consequence of further contraction. Conclusions. We find further evidence that UHE and He II line problem white dwarfs are indeed related, as concluded from their overlap in the Gaia HRD, similar photometric variability rates, light-curve shapes and amplitudes, and period distributions. The lack of increasing photometric amplitudes towards longer wavelengths, as well as the nondetection of optical emission lines arising from the highly irradiated face of a hypothetical secondary in the optical spectra of our stars, makes it seem unlikely that an irradiated late-type companion is the origin of the photometric variability. In recent years, the single degenerate helium (He) donor channel, where a white dwarf star accretes He-rich matter from a hydrogen-depleted companion, has emerged as a promising candidate progenitor scenario for these events. An unresolved question in this scenario is the fate of the companion star, which would be evident as a runaway hot subdwarf O/B stars (He sdO/B) in the aftermath of the SN event. Aims Previous studies have shown that the kinematic properties of an ejected companion provide an opportunity to closer examine the properties of an SN progenitor system. However, with the number of observed objects not matching predictions by theory, the viability of this mechanism is called into question. In this study, we first synthesize a population of companion stars ejected by the aforementioned mechanism, taking into account predicted ejection velocities, the inferred population density in the Galactic mass distribution, and subsequent kinematics in the Galactic potential. We then discuss the astrometric properties of this population. Methods We present 10(6) individual ejection trajectories, which were numerically computed with a newly developed, lightweight simulation framework. Initial conditions were randomly generated, but weighted according to the Galactic mass density and ejection velocity data. We then discuss the bulk properties (Galactic distribution and observational parameters) of our sample. Results Our synthetic population reflects the Galactic mass distribution. A peak in the density distribution for close objects is expected in the direction of the Galactic centre. Higher mass runaways should outnumber lower mass ones. If the entire considered mass range is realised, the radial velocity distribution should show a peak at 500 km s(-1). If only close US 708 analogues are considered, there should be a peak at (similar to 750-850) km s(-1). In either case, US 708 should be a member of the high-velocity tail of the distribution. Cyanobacterial composition, species richness, and evenness was not significantly different among the pelagic depths, sediment traps and surface sediments (p > 0.05), indicating that the cyanobacteria in the sediments reflected the cyanobacterial assemblage in the water column. However, total cyanobacterial abundances (qPCR) decreased from the metalimnion down the water column. The aggregate-forming (Aphanizomenon) and colony-forming taxa (Snowella) showed pronounced sedimentation. In contrast, Planktothrix was only very poorly represented in sediment traps (meta- and hypolimnion) and surface sediments, despite its highest relative abundance at the thermocline (10 m water depth) during periods of lake stratification (May-October). We observed significant seasonal variation in the cyanobacterial community composition (p < 0.05) in the epi- and metalimnion layers, but not in the hypolimnion. In winter-when the water column is mixed-picocyanobacteria (Synechococcus and Cyanobium) were dominant. With the onset of stratification in late spring, we observed potential niche specialization and coexistence among the cyanobacteria taxa driven mainly by light and nutrient dynamics. Specifically, ASVs assigned to picocyanobacteria and the genus Planktothrix were the main contributors to the formation of deep chlorophyll maxima along a light gradient. While Synechococcus and different Cyanobium ASVs were abundant in the epilimnion up to the base of the euphotic zone from spring to fall, Planktothrix mainly occurred in the metalimnetic layer below the euphotic zone where also overall cyanobacteria abundance was highest in summer. Our data revealed two potentially psychrotolerant (cold-adapted) Cyanobium species that appear to cope well under conditions of lower hypolimnetic water temperature and light as well as increasing sediment-released phosphate in the deeper waters in summer. The potential cold-adapted Cyanobium species were also dominant throughout the water column in fall and winter. Furthermore, Snowella and Microcystis-related ASVs were abundant in the water column during the onset of fall turnover. The ability of an atmospheric model to realistically simulate the stratospheric dynamics is essential in order to move forward in the field of seasonal predictions in midlatitudes. Earlier studies with the ICOsahedral Nonhydrostatic atmospheric model (ICON) point out that stratospheric westerlies in ICON are underestimated. This is the first extensive study on the evaluation of Northern Hemisphere stratospheric winter circulation with ICON in numerical weather prediction (NWP) mode. Seasonal experiments with the default setup are able to reproduce the basic climatology of the stratospheric polar vortex. However, westerlies are too weak and major stratospheric warmings too frequent in ICON. Both a reduction of the nonorographic, and a reduction of the orographic gravity wave and wake drag lead to a strengthening of the stratospheric vortex and a bias reduction, in particular in January. However, the effect of the nonorographic gravity wave drag scheme on the stratosphere is stronger. Stratosphere-troposphere coupling is intensified and more realistic due to a reduced gravity wave drag. Furthermore, an adjustment of the subgrid-scale orographic drag parameterization leads to a significant error reduction in the mean sea level pressure. As a result of these findings, we present our current suggested improved setup for seasonal experiments with ICON-NWP.
Plain Language Summary Although seasonal forecasts for midlatitudes have the potential to be highly beneficial to the public sector, they are still characterized by a large amount of uncertainty. Exact simulations of the circulation in the stratosphere can help to improve tropospheric predictability on seasonal time scales. For this reason, we investigate how well the new German atmospheric model is able to simulate the stratospheric circulation. The model reproduces the basic behavior of the Northern Hemisphere stratospheric polar vortex, but the westerly circulation in winter is underestimated. The stratospheric circulation is influenced by gravity waves that exert drag on the flow. These processes are only partly physically represented in the model, but are very important and are hence parameterized. By adjusting the parameterizations for the gravity wave drag, the stratospheric polar vortex is strengthened, thereby yielding a more realistic stratospheric circulation. In addition, the altered parameterizations improve the simulated surface pressure pattern. Blue horizontal-branch stars evolve from low-mass stars that have completed their main-sequence lifetimes and undergone a helium flash at the end of their red-giant phase. As such, blue horizontal-branch stars are very old objects that can be used as markers in studies of the Galactic structure and formation history. To create a clean sky catalogue of blue horizontal-branch stars, we cross-matched the Gaia data release 2 (DR2) dataset with existing reference catalogues to define selection criteria based on Gaia DR2 parameters. Following the publication of Gaia early data release 3 (EDR3), these methods were verified and subsequently applied to this latest release. Aims. Previous catalogues of blue horizontal-branch stars were developed using spectral analyses or were restricted to individual globular clusters. The purpose of this catalogue is to identify a set of blue horizontal-branch star candidates that have been selected using photometric and astrometric observations and exhibits a low contamination rate. This has been deemed important as the success of the Gaia mission has changed the way that targets are selected for large-scale spectroscopic surveys, meaning that far fewer spectra will be acquired for blue horizontal-branch stars in the future unless they are specifically targeted.
Methods. We cross-matched reference blue horizontal-branch datasets with the Gaia DR2 database and defined two sets of selection criteria. Firstly, in Gaia DR2 - colour and absolute G magnitude space, and secondly, in Gaia DR2 - colour and reduced proper motion space. The main-sequence contamination in both subsets of the catalogue was reduced, at the expense of completeness, by concentrating on the Milky Way's Galactic halo, where relatively young main-sequence stars were not expected. The entire catalogue is limited to those stars with no apparent neighbours within 5 arcsec. These methods were verified and subsequently applied to the Gaia EDR3. Results. We present a catalogue, based on Gaia EDR3, of 57 377 blue horizontal-branch stars. In a greenhouse experiment, we exposed the offspring of these populations to a climate change scenario for central Europe and revealed the population structure through whole-genome sequencing. Plants were grown under two temperatures (18°C and 21°C) and three precipitation regimes (65, 75, and 90 mm) to measure their response in biomass and fecundity-related traits. To reveal the population genetic structure, ddRAD sequencing was employed for a whole-genome approach. We found three major genetic clusters in S. vulgaris from Europe: one cluster comprising Southern European populations, one cluster of Western European populations, and another cluster containing central European populations. Population genetic diversity decreased with increasing latitude, and a Mantel test revealed significant correlations between FST and geographic distances as well as between genetic and environmental distances. Our trait analysis showed that the genetic clusters significantly differed in biomass-related traits and in the days to flowering. However, half of the traits showed parallel response patterns to the experimental climate change scenario. In image classification, this limitation has been mitigated by powerful oversampling techniques such as data augmentation. Using this technique, this work evaluates to what extent transforming inertial sensor data into movement trajectories and into 2D heatmap images can be advantageous for HAR when data are scarce. A convolutional long short-term memory (ConvLSTM) network that incorporates spatiotemporal correlations was used to classify the heatmap images. Evaluation was carried out on Deep Inertial Poser (DIP), a known dataset composed of inertial sensor data. Prior to the investigation of the effect of various load situation, the reproducibility of scapular kinematics and scapular muscle activity were evaluated under maximum effort arm movements. Healthy asymptomatic adults performed unloaded and maximal loaded concentric and eccentric isokinetic shoulder flexion and extension movements in the scapular plane while scapular kinematics and scapular muscle activity were simultaneously assessed. A 3D motion capture system (infra-red cameras \& reflective markers) was utilized to track scapular and humerus motion in relation to the thorax. 3D scapular position angles were given for arm raising and lowering between humerus positions of 20° and 120° flexion. To further characterize the scapular pattern, the scapular motion extent and scapulohumeral rhythm (ratio of scapular and humerus motion extent) were determined. Muscle activity of the upper and lower trapezius and the serratus anterior were assessed with surface electromyography. Amplitudes were calculated for the whole ROM and four equidistant movement phases. Reliability was characterized by overall moderate to good reproducibility across the load conditions. Irrespective of applied load, scapular kinematics followed a motion pattern of continuous upward rotation, posterior tilt and external rotation during arm elevation and a continuous downward rotation, anterior tilt and internal rotation during arm lowering. However, kinematics were altered between maximal loaded and unloaded conditions showing increased upward rotation, reduced posterior tilt and external rotation. Further, the scapulohumeral rhythm was decreased and scapular motion extent increased under maximal loaded movements. Muscle activity during maximum effort were of greater magnitude and differed in their pattern in comparison to the continuous increase and decrease of activity during unloaded shoulder flexion and extension. Relationships between scapular kinematics and their underlying scapular muscle activity could only be identified for a few isolated combinations, whereas the majority showed no associations. Scapular kinematics and scapular muscle activity pattern alter according to the applied load. Alterations between the load conditions comply in magnitude and partially in direction with differences seen between symptomatic and asymptomatic individuals. Even though long-term effects of identified adaptations in response to maximum load are so far unclear, deviations from typical scapular motion or muscle activation should not per se be seen as indicators of shoulder impairment. When combined with treadmill walking, a large number of steps can be collected in a controlled environment without the need of a dedicated gait analysis laboratory. In order to evaluate existing and novel IMU-based gait analysis algorithms for treadmill walking, a reference dataset that includes IMU data as well as reliable ground truth measurements for multiple participants and walking speeds is needed. This article provides a reference dataset consisting of 15 healthy young adults who walked on a treadmill at three different speeds. Data were acquired using seven IMUs placed on the lower body, two different reference systems (Zebris FDMT-HQ and OptoGait), and two RGB cameras. Additionally, in order to validate an existing IMU-based gait analysis algorithm using the dataset, an adaptable modular data analysis pipeline was built. Our results show agreement between the pressure-sensitive Zebris and the photoelectric OptoGait system (r = 0.99), demonstrating the quality of our reference data. As a use case, the performance of an algorithm originally designed for overground walking was tested on treadmill data using the data pipeline. The accuracy of stride length and stride time estimations was comparable to that reported in other studies with overground data, indicating that the algorithm is equally applicable to treadmill data. The Python source code of the data pipeline is publicly available, and the dataset will be provided by the authors upon request, enabling future evaluations of IMU gait analysis algorithms without the need of recording new data.}, language = {en} }
