Data on the flora, climate and soil characteristics of these slopes near Ornate at 1600-2600 m a.s.l. are provided. Fifty-nine angiosperm species established themselves on the pumice slopes in the past ca. 400 years, with the bulk of the small and herbaceous species and several species new records for Peru. Three Ornate sites were sampled in both a dry and a wet year and species numbers differed widely (14 versus 45 spp.). Among areas compared floristic composition is most similar to the Lomas de Tacna, and has less in common with geographically closer Lomas or Sierra formations. Nine species represent highly disjunct populations (200->700 km) from their nearest known living populations in central Peru, Chile, or Argentina/Bolivia and appear to have reached the area via long-distance dispersal. Abiotic conditions may have played an important role in limiting the establishment of species from the neighboring vegetation. The concentration change in the chamber is monitored over time, and the flux is usually calculated by the slope of a linear regression function. Theoretically, the gas exchange cannot be constant over time but has to decrease, when the concentration gradient between chamber headspace and soil air decreases. In this study, we test whether we can detect this non- linearity in the concentration change during the chamber closure with six air samples. We expect generally a low concentration gradient on dry sites (hummocks) and thus the occurrence of exponential concentration changes in the chamber due to a quick equilibrium of gas concentrations between peat and chamber headspace. On wet (flarks) and sedge- covered sites (lawns), we expect a high gradient and near-linear concentration changes in the chamber. To evaluate these model assumptions, we calculate both linear and exponential regressions for a test data set (n = 597) from a Finnish mire. We use the Akaike Information Criterion with small sample second order bias correction to select the best-fitted model. 13.6%, 19.2% and 9.8% of measurements on hummocks, lawns and flarks, respectively, were best fitted with an exponential regression model. A flux estimation derived from the slope of the exponential function at the beginning of the chamber closure can be significantly higher than using the slope of the linear regression function. Non-linear concentration-overtime curves occurred mostly during periods of changing water table. This could be due to either natural processes or chamber artefacts, e.g. initial pressure fluctuations during chamber deployment. To be able to exclude either natural processes or artefacts as cause of non-linearity, further information, e.g. CH4 concentration profile measurements in the peat, would be needed. If this is not available, the range of uncertainty can be substantial. We suggest to use the range between the slopes of the exponential regression at the beginning and at the end of the closure time as an estimate of the overall uncertainty. The Arabidopsis thaliana NAC transcription factor ANAC092 (also called AtNAC2 and ORE1) has recently been shown to control age-dependent senescence, but its mode of action has not been analysed yet. To explore the regulatory network administered by ANAC092 we performed microarray-based expression profiling using estradiol-inducible ANAC092 overexpression lines. Approximately 46% of the 170 genes up-regulated upon ANAC092 induction are known senescence-associated genes, suggesting that the NAC factor exerts its role in senescence through a regulatory network that includes many of the genes previously reported to be senescence regulated. We selected 39 candidate genes and confirmed their time-dependent response to enhanced ANAC092 expression by quantitative RT-PCR. We also found that the majority of them (24 genes) are up-regulated by salt stress, a major promoter of plant senescence, in a manner similar to that of ANAC092, which itself is salt responsive. Furthermore, 24 genes like ANAC092 turned out to be stage-dependently expressed during seed growth with low expression at early and elevated expression at late stages of seed development. Disruption of ANAC092 increased the rate of seed germination under saline conditions, whereas the opposite occurred in respective overexpression plants. We also detected a delay of salinity-induced chlorophyll loss in detached anac092-1 mutant leaves. Promoter-reporter (GUS) studies revealed transcriptional control of ANAC092 expression during leaf and flower ageing and in response to salt stress. Our novel assay technology uses the newly developed monoclonal antibody (G24-BA9) to quench the luminescence of europium trisbipyridine (EuTBP). We performed a competitive biotin immunoassay including an EuTBP-biotin conjugate, the anti-EuTBP antibody G24-BA9 and streptavidin as assay components. Steric hindrance allows only the binding of either G24-BA9 (to the EuTBP moiety) or streptavidin (to the biotin moiety) to the EuTBP-biotin conjugate. Addition of the analyte biotin resulted in the binding of streptavidin to biotin and a concomitant preferred binding of G24-BA9 to EuTBP-biotin. Since G24-BA9 quenches the luminescence of EuTBP within the conjugate, the luminescence signal could be used to indicate and quantify the presence of free biotin in the system. These inaccuracies can lead, among other things, to spurious long-range correlations, which can be eliminated by Schur-product-based localization techniques. In this article, we propose a new technique for implementing such localization techniques within the class of ensemble transform/square-root Kalman filters. Our approach relies on a continuous embedding of the Kalman filter update for the ensemble members, i.e. we state an ordinary differential equation (ODE) with solutions that, over a unit time interval, are equivalent to the Kalman filter update. The ODE formulation forms a gradient system with the observations as a cost functional. Besides localization, the new ODE ensemble formulation should also find useful application in the context of nonlinear observation operators and observations that arrive continuously in time. A multitude of empirical studies, enhancing our knowledge of the antecedents and consequences of the different performance aspects, have recently been summarised through various meta-analyses. We use this as all occasion for taking stock in order to identify new areas of theorising and empirical research. Focusing in particular oil proactive performance aspects, the present paper identifies three themes that could inspire new research and model development. Such overyielding occurs when a community performs better than its component monocultures due to the complementarity or dominance effect and is mostly detected in substrate-bound plant communities (terrestrial plants or submerged macrophytes) where resource use complementarity can be enhanced due to differences in rooting architecture and depth. Here, we investigated whether these findings arc generalizeable for free-floating phytoplankton with little potential for spatial differences in resource use. We performed aquatic microcosm experiments with eight phytoplankton species belonging to four functional groups to determine the manner in which species and community biovolume varies in relation to the number of functional groups and hypothesized that an increasing number of functional groups within a community promotes overyielding. Unexpectedly, we did not detect overyielding in any algal community. Instead. total community biovolume tended to decrease with all increasing, number of functional groups. This underyielding was mainly caused by the negative dominance effect that originated from a trade-off between growth rate and filial biovolume. In monoculture, slow-groing species built up higher biovolumes that fast-growing ones, whereas in mixture a fast-growing but low-productive species monopolized most of the nutrients and prevented competing species from developing high biovolumes expected from monocultures. Our results indicated that the Magnitude of the community biovolume was largely determined by the identify of one species. Functional diversity and resource use complementarity were of minor Importance among free-floating phytoplankton, possibly reflecting the lack of spatially heterogeneous resource distribution. We performed a meta-analysis of 117 field or experimental-garden studies that measured pair-wise trait differences of a total of 125 invasive and 196 non-invasive plant species in the invasive range of the invasive species. We tested whether invasiveness is associated with performance-related traits (physiology, leaf-area allocation, shoot allocation, growth rate, size and fitness), and whether such associations depend on type of study and on biogeographical or biological factors. Overall, invasive species had significantly higher values than non-invasive species for all six trait categories. More trait differences were significant for invasive vs. native comparisons than for invasive vs. non-invasive alien comparisons. Moreover, for comparisons between invasive species and native species that themselves are invasive elsewhere, no trait differences were significant. Differences in physiology and growth rate were larger in tropical regions than in temperate regions. Trait differences did not depend on whether the invasive alien species originates from Europe, nor did they depend on the test environment. We conclude that invasive alien species had higher values for those traits related to performance than non-invasive species. This suggests that it might become possible to predict future plant invasions from species traits. T1 - A minimal cysteine motif required to activate the SKOR K+ channel of Arabidopsis by the reactive oxygen species H2O2 N2 - Reactive oxygen species (ROS) are essential for development and stress signaling in plants. They contribute to plant defense against pathogens, regulate stomatal transpiration, and influence nutrient uptake and partitioning. Although both Ca2+ and K+ channels of plants are known to be affected, virtually nothing is known of the targets for ROS at a molecular level. Here we report that a single cysteine (Cys) residue within the Kv-like SKOR K+ channel of Arabidopsis thaliana is essential for channel sensitivity to the ROS H2O2. We show that H2O2 rapidly enhanced current amplitude and activation kinetics of heterologously expressed SKOR, and the effects were reversed by the reducing agent dithiothreitol (DTT). Both H2O2 and DTT were active at the outer face of the membrane and current enhancement was strongly dependent on membrane depolarization, consistent with a H2O2-sensitive site on the SKOR protein that is exposed to the outside when the channel is in the open conformation. Cys substitutions identified a single residue, Cys(168) located within the S3 alpha-helix of the voltage sensor complex, to be essential for sensitivity to H2O2. The same Cys residue was a primary determinant for current block by covalent Cys S-methioylation with aqueous methanethiosulfonates. These, and additional data identify Cys168 as a critical target for H2O2, and implicate ROS-mediated control of the K+ channel in regulating mineral nutrient partitioning within the plant. A1 - Mischke, Steffen A1 - Zhang, Chengjun A1 - Böhner, Jürgen T1 - A modern pollen-climate calibration set based on lake sediments from the Tibetan Plateau and its application to a Late Quaternary pollen record from the Qilian Mountains N2 - Aim: Fossil pollen spectra from lake sediments on the Tibetan Plateau have been used for qualitative climate reconstruction, but no modern pollen-climate calibration set based on lake sediments is available to infer past climate quantitatively. This study aims to develop such a dataset and apply it to fossil data. Location: The Tibetan Plateau, between 30 and 40 degrees N and 87 and 103 degrees E. Methods: We collected surface sediments from 112 lakes and analysed them palynologically. The lakes span a wide range of mean annual precipitation (P-ann; 31-1022 mm), mean annual temperature (T-ann; -6.5 to 1 degrees C), and mean July temperature (T-July; 2.6-19.7 degrees C). Redundancy analysis showed that the modern pollen spectra are characteristic of their respective vegetation types and local climate. Transfer functions for P-ann, T-ann and T-July were developed with weighted averaging partial least squares. Model performance was assessed by leave-one-out cross-validation. Results: The root mean square errors of prediction (RMSEP) were 104 mm (P-ann), 1.18 degrees C (T-ann) and 1.17 degrees C (T-July). The RMSEPs, when expressed as percentages of the gradient sampled, were 10.6% (P-ann), 15.7% (T-ann) and 11.9% (T-July). These low values indicate the good performance of our models. An application of the models to fossil pollen spectra covering the last c. 50 kyr yielded realistic results for Luanhaizi Lake in the Qilian Mountains on the north-eastern Tibetan Plateau (modern P-ann 480 mm; T-ann-1 degrees C). T-ann and P-ann values similar to present ones were reconstructed for late Marine Isotope Stage 3, with minimum values for the Last Glacial Maximum (c. 300 mm and 2 degrees C below present), and maximum values for the early Holocene (c. 70 mm and 0.5 degrees C greater than present). O-acetylserine (thiol) lyase (OAS-TL) catalyses the final step of cysteine biosynthesis. OAS-TL enzyme isoforms are localised in the cytoplasm, the plastids and mitochondria but the contribution of individual OAS-TL isoforms to plant sulphur metabolism has not yet been fully clarified. Results: The seedling lethal phenotype of the Arabidopsis onset of leaf death3-1 (old3-1) mutant is due to a point mutation in the OAS-A1 gene, encoding the cytosolic OAS-TL. The mutation causes a single amino acid substitution from Gly(162) to Glu(162), abolishing old3-1 OAS-TL activity in vitro. The old3-1 mutation segregates as a monogenic semidominant trait when backcrossed to its wild type accession Landsberg erecta (Ler-0) and the Di-2 accession. Consistent with its semi- dominant behaviour, wild type Ler-0 plants transformed with the mutated old3-1 gene, displayed the early leaf death phenotype. However, the old3-1 mutation segregates in an 11: 4: 1 (wild type: semi-dominant: mutant) ratio when backcrossed to the Colombia-0 and Wassilewskija accessions. Thus, the early leaf death phenotype depends on two semi- dominant loci. The second locus that determines the old3-1 early leaf death phenotype is referred to as odd-ler (for old3 determinant in the Ler accession) and is located on chromosome 3. The early leaf death phenotype is temperature dependent and is associated with increased expression of defence-response and oxidative-stress marker genes. Independent of the presence of the odd-ler gene, OAS-A1 is involved in maintaining sulphur and thiol levels and is required for resistance against cadmium stress. Kilimanjaro were evaluated describing the ecological niche of the new species. This interdisciplinary approach allows not only a profound characterisation of the ecological demands of the new genus but also predictions of the potential distribution area, which is tested for the first time for an African bush cricket species. Lunidia viridis n. gen. n. sp. occurs within humid and perhumid forests and Chagga home gardens, avoiding subhumid conditions on the mountain. It is found from 1,330 m upwards on the southern slopes, whereas the same ecological conditions are expressed from 1,930 m upwards on the drier northern slopes. Lunidia viridis has an unusually complex and variable song, which is described from field and laboratory recordings. The FISH technique for characterizing chromosomes is applied for the first time for an African species; L. viridis exhibits a karyotype typical for most Tettigoniidae. The members of this subtribe are small conocephaline bush crickets, confined to Africa. Melanoscirtes is erected on Phlesirtes kibonotensis, a species restricted to forest clearings and forest edge in the submontane and montane zones of Mt. Kilimanjaro. A subspecies, M. kibonotensis uguenoensis, is described from the North Pare mountains, a mountain range of the Eastern Arc adjacent to Mt. Kilimanjaro. Further species of Melanoscirtes occur on other mountain ranges of the northern branch of the Eastern Arc mountains of northern Tanzania and southern Kenya. The South Pare mountains harbour M. shengenae; the West Usambaras, M. usambarensis, and the Taita Hills, M. taitensis. All species and subspecies of Melanoscirtes exhibit a similar morphology and occupy analogous habitats on the respective mountains. The song patterns for all species found within this genus are very similar, and this, together with evidence from molecular data, suggests that allopatric speciation is the reason for the biogeographic pattern found in this genus. Our spectroscopic follow-up of the central point source associated with the nebula showed that it is a WN7h star (we named it WR121b). We analysed the spectrum of WR 121b by using the Potsdam Wolf-Rayet model atmospheres, obtaining a stellar temperature of similar or equal to 50 kK. The stellar wind composition is dominated by helium with similar to 20 per cent of hydrogen. The stellar spectrum is highly reddened [E(B - V) = 2.85 mag]. Adopting an absolute magnitude of M-v = 5.7, the star has a luminosity of log L/L-circle dot = 5.75 and a mass-loss rate of 10(-4.7)M(circle dot)yr(-1), and resides at a distance of 6.3 kpc. We searched for a possible parent cluster of WR 121b and found that this star is located at similar or equal to 1 degrees from the young star cluster embedded in the giant HII region W43 (containing a WN7+a/OB? star - WR121a). We also discovered a bow shock around the O9.5III star ALS 9956, located at similar or equal to 0 degrees.5 from the cluster. We discuss the possibility that WR121b and ALS 9956 are runaway stars ejected from the cluster in W43. The insertion sites of six independent isolates mapped within the same gene predicted to code for a permease of unknown function located in the cytoplasmic membrane. During growth under Mo-replete conditions, the wild-type strain accumulated considerably more Mo than the permease mutant. For mutants defective for the permease, the high-affinity molybdate importer ModABC, or both transporters, in vivo Mo-dependent nitrogenase (Mo-nitrogenase) activities at different Mo concentrations suggested that ModABC and the permease import molybdate in nanomolar and micromolar ranges, respectively. Like the permease mutants, a mutant defective for ATP sulfurylase tolerated high Mo concentrations, suggesting that ATP sulfurylase is the main target of Mo inhibition in R. capsulatus. Sulfate-dependent growth of a double mutant defective for the permease and the high-affinity sulfate importer CysTWA was reduced compared to those of the single mutants, implying that the permease plays an important role in sulfate uptake. After presenting the approach, some case studies of the display and management of affectivity in storytelling in telephone and face-to-face conversations are presented. The analysis reconstructs the display and handling of affectivity by both storyteller and story recipient. In particular, I describe the following kinds of resources: the verbal and segmental display: Rhetorical, lexico-semantic, syntactic, phonetic-phonological resources; the prosodic and suprasegmentalvocal display: Resources from the realms of prosody and voice quality; visual or "multimodal" resources from the realms of body posture and its changes, head movements, gaze, and hand movements and gestures. It is shown that the display of affectivity is organized in orderly ways in sequences of storytelling in conversation. I reconstruct (a) how verbal, vocal and visual cues are deployed in co-occurrence in order to make affectivity in general and specific affects in particular interpretable for the recipient and (b) how in turn the recipient responds and takes up the displayed affect. As a result, affectivity is shown to be managed by teller and recipient in storytelling sequences in conversation, involving both the reporting of affects from the story world as well as the negotiation of in-situ affects in the here-and-now of the storytelling situation. O. T1 - All reversible dynamics in maximally nonlocal theories are trivial N2 - A remarkable feature of quantum theory is nonlocality ( Bell inequality violations). However, quantum correlations are not maximally nonlocal, and it is natural to ask whether there are compelling reasons for rejecting theories in which stronger violations are possible. To shed light on this question, we consider post-quantum theories in which maximally nonlocal states ( nonlocal boxes) occur. We show that reversible transformations in such theories are trivial: they consist solely of local operations and permutations of systems. In particular, no correlations can be created; nonlocal boxes cannot be prepared from product states and classical computers can efficiently simulate all such processes.
