T1 - Avoiding irreducible CSC conflicts by internal communication
N2 - Resynthesis of handshake specifications obtained e. g. from BALSA or TANGRAM with speed-independent logic synthesis from STGs is a promising approach. To deal with state-space explosion, we suggested STG decomposition; a problem is that decomposition can lead to irreducible CSC conflicts. Here, we present a new approach to solve such conflicts by introducing internal communication between the components. We give some first, very encouraging results for very large STGs concerning synthesis time and circuit area.

T1 - Tuning the thickness of polymer brushes grafted from nonlinearly growing multilayer assemblies
N2 - A new versatile method for tuning the thickness of surface-tethered polymer brushes is introduced. It is based on the combination of polyelectrolyte multilayer deposition and surface-initiated atom transfer radical polymerization. To control the thickness of the brushes, the nonlinear growth of certain polyelectrolyte multilayer systems is exploited. The method is demonstrated to work with different polyelectrolytes and different monomers. The relevance for applications is demonstrated by cell adhesion experiments oil grafted thermoresponsive polymer layers with varying thickness.

T1 - Conditions and timing of pumpellyite-actinolite-facies metamorphism in the early Mesozoic frontal accretionary prism of the Madre de Dios Archipelago (latitude 50 degrees 20'S; Southern Chile)
N2 - The Madre de Dios Metamorphic Complex (MDMC) in southern Chile is a fossil frontal accretionary prism, which is mainly composed of metapsammopelitic rocks, intercalations of oceanic rocks (greenstone and metachert) and platform carbonate. We concentrated on the metabasite to decipher the metamorphic evolution. This rock type contains assemblages of the pumpellyite-actinolite facies: pumpellyite +/- actinolite-chlorite +/- grandite +/- phengite +/- epidote-albite- quartz-titanite +/- K-feldspar +/- calcite. The metamorphic phases mainly grew by prograde hydration reactions during various episodes of restricted fluid influx. Fundamental phase relations of the pumpellyite-actinolite facies and adjacent facies were reproduced by pseudosections calculated for the system K2O-Na2O-CaO-FeO-O-2-MgO-Al2O3-TiO2-SiO2-H2O- CO2 at 200-400 degrees C and 1-9 kbar. The calculated stability fields of the metamorphic assemblages as realized in the MDMC metabasite indicate highest metamorphic conditions restricted to 290-310 degrees C, 4-6 kbar for the MDMC, presumably as a result of the main fluid influx at these conditions. Nevertheless, earlier local equilibria are still preserved as a result of strongly kinetically controlled mineral reactions and a lack of recrystallization and compositional homogenization at thin-section scale. Hence, thermodynamic calculations of local multivariant mineral equilibria using the entire compositional variation of minerals in the MDMC show that the prograde PT path evolved from 4 +/- 1 kbar, 200-220 degrees C to 5 +/- 1 kbar, 290-330 degrees C. The prograde PT path reflects nearly horizontal particle paths after reaching the maximum depth typical for frontal accretionary prisms. Long residence at maximum depth resulted in thermal re-equilibration. Ar-40/Ar-39 spot ages were measured by in situ UV laser ablation of local phengite concentrations in a deformed metapelite at 233 center dot 2 +/- 1 center dot 8 Ma and in an undeformed metabasite at 200 center dot 8 +/- 2 center dot 4 Ma. Whereas the first age represents an age of accretion, the latter age can be attributed to mineral growth either during a younger stage of accretion or during a retrograde stage. Ar-40/Ar-39 isotopic analyses of two further metabasite samples reflect a prominent resetting of ages at 152 center dot 0 +/- 2 center dot 2 Ma and white mica growth during external fluid access triggered by either a local intrusion or a late Jurassic extensional episode.

T1 - Specific interactions between four Molybdenum-binding proteins contribute to Mo-dependent gene regulation in Rhodobacter capsulatus
N2 - The phototrophic purple bacterium Rhodobacter capsulatus encodes two transcriptional regulators, MopA and MopB, with partially overlapping and specific functions in molybdate-dependent gene regulation. Both MopA and MopB consist of an N-terminal DNA-binding helix-turn-helix domain and a C-terminal molybdate-binding di-MOP domain. They formed homodimers as apo-proteins and in the molybdate-bound state as shown by yeast two-hybrid (Y2H) studies, glutaraldehyde cross-linking, gel filtration chromatography, and copurification experiments. Y2H studies suggested that both the DNA- binding and the molybdate-binding domains contribute to dimer formation. Analysis of molybdate binding to MopA and MopB revealed a binding stoichiometry of four molybdate oxyanions per homodimer. Specific interaction partners of MopA and MopB were the molybdate transporter ATPase ModC and the molbindin-like Mop protein, respectively. Like other molbindins, the R. capsulatus Mop protein formed hexamers, which were stabilized by binding of six molybdate oxyanions per hexamer. Heteromer formation of MopA and MopB was shown by Y2H studies and copurification experiments. Reporter gene activity of a strictly MopA-dependent mop-lacZ fusion in mutant strains defective for either mopA, mopB, or both suggested that MopB negatively modulates expression of the mop promoter. We propose that depletion of the active MopA homodimer pool by formation of MopA-MopB heteromers might represent a fine-tuning mechanism controlling mop gene expression.

T1 - Protein interactions with cyanidin-3-glucoside and its influence on alpha-amylase activity
N2 - BACKGROUND: Recent studies indicate that the bioavailability of anthocyanins is extremely low. One of the possible reasons could be their binding to proteins. Therefore, the binding affinity of cyanidin-3-glucoside (Cy3glc) to HSA and alpha-amylase was investigated by the quenching of protein tryptophan fluorescence. From data obtained, the binding constants and the free Gibbs energy were calculated. The changes in conformation of the proteins tested were studied with circular dichroism and the influence of binding on alpha-amylase activity determined. RESULTS: Cy3glc quenched the tryptophan fluorescence and upon ligand binding a change in protein structure was observed related to the corresponding decrease in the et-amylase activity. The association constants of 25 to 77 x 10(3) L mol(-1) were calculated for different proteins, indicating weak interactions of non-covalent nature. Competitive binding with HSA in the presence of 8-anilino-1-naphthalene sulfonic acid suggest involvement of hydrophobic interactions, in the case of HSA the possible site being subdomain IIA. CONCLUSION: The strongest affinity of Cy3glc for HSA being at pH 7 underlines its potential in transport and distribution of the phenolic compounds in organisms. An influence on salivary amylase activity is possible when drinking berry juices with high anthocyanins content.

T1 - Kiezdeutsch as a test case for the interaction between grammar and information structure
N2 - This paper deals with Kiezdeutsch, a way of speaking that emerged among adolescents in multiethnic urban neighbourhoods of Germany. We show that, in Kiezdeutsch, we find evidence for both grammatical reduction and new developments in the domain of information structure, and hypothesise that this points to a systematic interaction between grammar and information structure, between weakened grammatical constraints and a more liberal realisation of information-structural preferences. We show that Kiezdeutsch can serve as an interesting test case for such an interaction, that this youth language is a multiethnolect, that is, a new variety that is spoken by speakers from a multitude of ethnic backgrounds, including German, and forms a dynamic linguistic system of its own, thus allowing for systematic developments on grammatical levels and their interfaces with extragrammatical domains.

T1 - Grammatical innovation in multiethnic urban Europe : new linguistic practices among adolescents
N2 - This paper discusses a phenomenon that has recently been observed in areas with a large migrant population in European cities: the rise of new linguistic practices among adolescents in multiethnic contexts. The main grammatical characteristics that have been described for them are (1) phonological/phonctic and lexical influences from migrant languages and (2) morpho-syntactic reductions and simplifications. In this paper, I show that from a grammatical point of view, morpho-syntactic reductions are only part of the story. Using 'Kiezdeutsch' as an example. the German instance of such a youth language (which may be the one with most speakers), I discuss several phenomena that provide evidence for linguistic productivity and show that they evolve from a specific interplay of grammatical and pragmatic features that is typical for contact languages: grammatical reductions go hand-in-hand with productive elaborations that display a systematicity that can lead to the emergence of new constructions, indicating the innovative grammatical power of these muitiethnolects.

T1 - Applied tracers for the observation of subsurface stormflow at the hillslope scale
N2 - Rain fall-runoff response in temperate humid headwater catchments is mainly controlled by hydrolo gical processes at the hillslope scale. Applied tracer experiments with fluore scent dye and salt tracers are well known tools in groundwater studies at the large scale and vadose zone studies at the plot scale, where they provide a means to characterise subsurface flow. We extend this approach to the hillslope scale to investigate saturated and unsaturated flow path s concertedly at a forested hill slope in the Austrian Alps. Dye staining experiments at the plot scale revealed that crack s and soil pipe s function as preferential flow path s in the fine-textured soils of the study area, and these preferenti al flow structures were active in fast subsurface transport of tracers at the hillslope scale. Breakthrough curves obtained under steady flow conditions could be fitted well to a one-dimensional convection-dispersion model. Under natural rain fall a positive correlation of tracer concentrations to the transient flows was observed. The results of this study demon strate qualitative and quantitative effects of preferential flow feature s on subsurface stormflow in a temperate humid headwater catchment. It turn s out that , at the hill slope scale, the interaction s of structures and processes are intrinsically complex, which implies that attempts to model such a hillslope satisfactorily require detailed investigation s of effective structures and parameters at the scale of interest.

T1 - Synergistic acceleration of thyroid hormone degradation by phenobarbital and the PPAR alpha agonist WY14643 in rat hepatocytes
N2 - Energy balance is maintained by controlling both energy intake and energy expenditure. Thyroid hormones play a crucial role in regulating energy expenditure. Their levels are adjusted by a tight feed back-control led regulation of thyroid hormone production/incretion and by their hepatic metabolism. Thyroid hormone degradation has previously been shown to be enhanced by treatment with phenobarbital or other antiepileptic drugs due to a CAR-dependent induction of phase 11 enzymes of xenobiotic metabolism. We have recently shown, that PPAR alpha agonists synergize with phenobarbital to induce another prototypical CAR target gene, CYP2B1. Therefore, it was tested whether a PPAR alpha agonist could enhance the phenobarbital-dependent acceleration of thyroid hormone elimination. In primary cultures of rat hepatocytes the apparent half-life of T3 was reduced after induction with a combination of phenobarbital and the PPARa agonist WY14643 to a larger extent than after induction with either Compound alone. The synergistic reduction of the half-life could be attributed to a synergistic induction of CAR and the CAR target genes that code for enzymes and transporters involved in the hepatic elimination of T3, such as OATP1A1, OATP1A3, UGT1A3 and UCT1A10. The PPAR alpha-dependent CAR induction and the subsequent induction of T3-eliminating enzymes might be of physiological significance for the fasting- incluced reduction in energy expenditure by fatty acids as natural PPARa ligands. The synergism of the PPAR alpha agonist WY14643 and phenobarbital in inducing thyroid hormone breakdown might serve as a paradigm for the synergistic disruption of endocrine control by other combinations of xenobiotics.

T1 - Alpine orogenic evolution from subduction to collisional thermal overprint : the Ar-40/Ar-39 age constraints from the Valaisan Ocean, central Alps
N2 - The investigated HP/LT metasedimentary units of the Valaisan and adjacent European domains occupy a key position in the Alpine belt for understanding the transition from early subduction-related HP/LT metamorphism to collision-related Barrovian overprint and the evolution of mountain belts in general. The timing of high-pressure metamorphism, subsequent retrogression and following Barrow-type overprint was studied by Ar-40/Ar-39 dating of biotite and several white mica generations that are well characterized in terms of mineral chemistry, texture and associated mineral assemblages. Four distinct age populations of white mica record peak pressure conditions (42-40 Ma) and several stages of subsequent retrograde metamorphic evolution (36-25 Ma). Biotite isotopic analyses yield consistent apparent ages that cluster around 18-16 Ma for the Barrow-type thermal overprint. The recorded isotopic data reveal a significant time gap in the order of some 20 Ma between subduction-related HP/LT metamorphism and collision-related Barrovian overprint, supporting the notion of a polymetamorphic evolution associated with a bimodal P-T path.

T1 - Is the normal heart rate "chaotic" due to respiration?
N2 - The incidence of cardiovascular diseases increases with the growth of the human population and an aging society, leading to very high expenses in the public health system. Therefore, it is challenging to develop sophisticated methods in order to improve medical diagnostics. The question whether the normal heart rate is chaotic or not is an attempt to elucidate the underlying mechanisms of cardiovascular dynamics and therefore a highly controversial topical challenge. In this contribution we demonstrate that linear and nonlinear parameters allow us to separate completely the data sets of the three groups provided for this controversial topic in nonlinear dynamics. The question whether these time series are chaotic or not cannot be answered satisfactorily without investigating the underlying mechanisms leading to them. We give an example of the dominant influence of respiration on heart beat dynamics, which shows that observed fluctuations can be mostly explained by respiratory modulations of heart rate and blood pressure (coefficient of determination: 96%). Therefore, we recommend reformulating the following initial question: "Is the normal heart rate chaotic?" We rather ask the following: " Is the normal heart rate 'chaotic' due to respiration?"

N2 - While it is well known that Charles Darwin and Alexander von Humboldt appreciated each other´s work, Humboldt’s influence on Darwin has not yet been thoroughly examined. The following paper shows to what extent Darwin was inspired by Humboldt’s publications while writing his Origin of Species. Darwin paid special attention to Humboldt’s observations on plant migration and throughout his whole scientific career he used Humboldt’s books as a major source of information. In fact, Darwin’s last annotations in his copy of the Personal Narrative of travels to the Equinoctial Regions of the New continent during the Years 1799-1804 date back to the last weeks of his life. On the other hand, Darwin rejected many of Humboldt’s scientific conclusions. The two authors differed in their assessment of Heinrich Bronn’s biological research. Finally, Darwin was dissatisfied with Humboldt’s last work Cosmos because it did not contain the information he expected. Darwin’s collaboration with Humboldt’s travel companion Christian Gottfried Ehrenberg proved to be similarly disappointing. Darwin sent 183 samples of infusoria to Berlin, but the results of Ehrenberg’s analysis did not support Darwin’s theory on evolution.

T1 - Testgütekriterien des Fragebogens VKS zur Erfassung volitionaler Komponenten im Sport
N2 - In this paper the psychometric quality of the sport-specific Volitional Components Questionnaire (VKS) is examined. After an item analysis, a factor analysis was undertaken resulted in the four factors self optimization, lack of activation, loss of focus, and self impediment. The questionnaire exhibits satisfactory internal consistency (Cronbach's alpha) and retest reliability. In line with theory (Kuhl, 2001), correlations with the general questionnaire VCQ II (Kuhl & Fuhrmann, 1998) and sport-specific action orientation as measured with the HOSP questionnaire (Beckmann, 2003) indcate construct validity. Validity can also be shown by correlations with the external criterion of expert rating by coaches. In addition, the VKS is able to differentiate between athletes who perform well and those who perforin poorly.

T1 - Construction and characterisation of transgenic mice for human sulfotransferases with polymorphic SULT1A genes
N2 - In humans, SULT1A1 and its polymorphic variants play an important role in xenobiotic metabolism and display a broad tissue distribution and high expression level. This enzyme is expressed in almost every human organ whereas in mice SULT1A1 can only be detected in liver, lung and colon. The most common polymorphism of this gene leads to an amino acid substitution from arginine to histidine at the position 213. In platelets, the allele encoding histidine (also designated as SULT1A1*2) is associated with both low activity and low thermal stability of the SULT protein. However, so far only little is known about the significance of these allelic variants in the other tissues with hSULT1A1 expression. Previous epidemiological studies have made attempts to correlate SULT1A1 allelic variants and cancer development, their data, however, have been contradictory for an appropriate cancer risk assessment. In this thesis, we addressed the effect of the hSULT1A1 genetic variability on the susceptibility to nutritional and environmental carcinogens using transgenic (tg) mouse models. We generated tg mice carrying the most common allelic variants of the human SULT1A1 gene. The coding region and large flanking human sequences upstream and downstream of the hSULT1A1 gene were integrated randomly into the mouse genome by microinjection. Several tg mouse lines were generated. Two of them, line (li) 31 with the SULT1A1*1 allele and li 28 with the SULT1A1*2 allele, were analysed in detail. At first, an identical transgene copy number was detected in both lines. Furthermore, biochemical characterization of li 28 showed that the tissue distribution, the cellular and subcellular localisation of the protein were very similar to those in humans. In contrast, li 31 exhibited differences in tissue distribution and cellular localisation of the human protein compared to li 28. The protein expression level in the tg line with SULT1A1*2 (li 28) was generally higher than in SULT1A1*1 (li 31) mice. These results were surprising since the SULT1A1*1 allele in human platelets usually leads to a higher amount of SULT1A1 protein compared to the SULT1A1*2 allele. To investigate these differences, we sequenced the cDNA and 5´-flanking region of the SULT1A1 gene. In both tg mouse lines, the cDNA sequence was identical to the reference sequence from the gene databank (Pubmed). We subsequently analysed the common polymorphisms of the 5´-flanking region, and determined different haplotypes at position -624 and -396 in the tg mouse lines. According to the literature, the haplotype associated with a higher SULT1A1 enzyme activity, we detected in li 31. We analyzed the possible correlation between gene transcription and protein expression by measuring RNA expression levels of the coding and the non-coding region (with alternative exons 1B and 1A). We detected a higher RNA expression level of the coding region and exon 1B in li 28 compared to li 31, whereas RNA for exon 1A was only found in li 28 in all investigated tissues, but only in lung in li 31. Furthermore we detected with exon 1A-primers larger RNA in both lines. These differences in exon 1A expression accompanied by potential splicing variants could be responsible for the different expression and activity of the human SULT1A1 protein in both tg mouse lines. In order to validate our generated and characterized tg mouse models as toxicological in vivo models, we used them for the evaluation of the heterocyclic aromatic amine 2-amino-1-methyl-6-phenylimidazo-[4,5-b]pyridine (PhIP). PhIP is typically generated during heating and roasting of meat and fish and is suggested to be associated with an increased colon cancer incidence in the western world. We measured the impact of the additionally expressed human SULT proteins on the PhIP-DNA adduct level by 32P-postlabelling. We detected significantly higher DNA adduct levels in tg compared to wildtype mice, which correlated positively with the expression pattern of the human SULT1A1 protein in the tg mice. In conclusion, in this thesis, we have successfully generated and validated the transgenic mouse lines carrying the most common allelic variants of the human SULT1A1 gene. Interestingly, these lines exhibited differences in both the SULT1A1 RNA and protein levels. Using these transgenic mouse models as in vivo toxicological tools we have shown that the expression of human SULT1A1 in mice has a decisive impact on the strength and the target tissue of DNA adducts.

T1 - Activated carbon may have undesired side effects for testing allelopathy in invasive plants
N2 - Activated carbon has become a widely used tool to investigate root-mediated allelopathy of plants, especially in plant invasion biology, because it adsorbs and thereby neutralizes root exudates. Allelopathy has been a controversially debated phenomenon for years, which revived in plant invasion biology as one possible reason for the success of invasive plants. Noxious plant exudates may harm other plants and provide an advantage to the allelopathic plant. However, root exudates are not always toxic, but may stimulate the microbial community and change nutrient availability in the rhizosphere. In a greenhouse experiment, we investigated the interacting effects of activated carbon, arbuscular mycorrhiza and plant competition between the invasive Senecio inaequidens and the native Artemisia vulgaris. Furthermore, we tested whether activated carbon showed any undesired effects by directly affecting mycorrhiza or soil chemistry. Contrary to the expectation, S. inaequidens was a weak competitor and we could not support the idea that allelopathy was involved in the competition. Activated carbon led to a considerable increase in the aboveground biomass production and reduced the infection with arbuscular mycorrhiza of both plant species. We expected that arbuscular mycorrhiza promotes plant growth by increasing nutrient availability, but we found the contrary when activated carbon was added. Chemical analyses of the substrate showed, that adding activated carbon resulted in a strong increase in plant available phosphate and in a decrease of the C-organic/N-total ration both of which suggest stimulated microbial activity. Thus, activated carbon not only reduced potential allelopathic effects, but substantially changed the chemistry of the substrate. These results show that activated carbon should be handled with great care in ecological experiments on allelopathy because of possible confounding effects on the soil community.

N2 - “Mare Liberum” was published by Hugo Grotius in March 1609. 400 years later, a panel discussion in Potsdam focused on the classical text as such and then dealt with the reception of “Mare Liberum” both in Grotius’ time and today. Furthermore, it was discussed how Grotius himself was influenced by Francisco de Vitoria. These three papers are included in the present publication; a fourth paper on the current problems of fighting piracy by the European Union was presented but is not included here because we wanted to preserve the historical approach of the publication.
