Based on new empirical evidence from five Gur and Kwa languages, we claim that these focus expressions have to be analysed as biclausal constructions even though they do not represent clefts containing restrictive relative clauses. First, we relativize the partly overgeneralized assumptions about structural correspondences between the out-of-focus part and relative clauses, and second, we show that our data do in fact support the hypothesis of a clause coordinating pattern as present in clause sequences in narration. It is argued that we deal with a non-accidental, systematic feature and that grammaticalization may conceal such basic narrative structures. Complementary to these structures, Foodo knows an elliptic form consisting of the focused constituent and a predication marker gɛ́. KSM-P15 ist mit 197 Aminosäuren eines der kleinsten, bekannten β-3-Solenoidproteine. Sie spaltet Polygalakturonsäurederivate in einem Ca2+-abhängigen β-Eliminierungsprozess. Wie bei allen Proteinen dieser Enzymfamilie ist auch die Polypeptidkette von Pel-15 zu einer einsträngigen, rechtsgängigen, parallelen β-Helix aufgewunden. In diesem Strukturmotiv enthält jede Windung drei β-Stränge, die jeweils durch flexible Schleifenbereiche miteinander verbunden sind. Insgesamt acht Windungen stapeln sich in Pel-15 übereinander und bilden entlang der Helixachse flächige, parallele β-Faltblätter aus. Im Bereich dieser β-Faltblätter existiert ein ausgedehntes Netzwerk von Wasserstoffbrückenbindungen, durch das der hydrophobe Kern, der sich im Inneren der β-Helix befindet, vom umgebenden Lösungsmittel abgeschirmt wird. Besondere Abschlussstrukturen an beiden Enden der β-Helix, wie sie typischerweise bei anderen Ver-tretern dieser Strukturklasse ausgeprägt werden, sind in Pel-15 nicht zu beobachten. Stattdessen sind die terminalen Bereiche der β-Helix über Salzbrücken und hydrophobe Seitenkettenkontakte stabilisiert. In der vorliegenden Dissertation wurde die Pektatlyase Pel-15 hinsichtlich ihres Faltungsgleichgewichtes, ihrer enzymatischen Aktivität und der Kinetik ihrer Strukturbildung charakterisiert. In eine evolutionär konservierte Helixwindung wurden destabilisierende Mutationen eingeführt, und deren Auswirkungen mittels spektroskopischer Methoden analysiert. Die Ergebnisse zeigen, dass Pel-15 in Gegenwart des Denaturierungsmittels Guanidiniumhydrochlorid einen hyperfluoreszenten Gleichgewichtsustand (HF) populiert, der nach Messungen von Faltungs- und Entfaltungskinetiken ein konformationelles Ensemble aus den Zuständen HFslow und HFfast darstellt. Diese HF-Zustände sind durch eine hohe Aktivierungsbarriere voneinander getrennt. In Rückfaltungsexperimenten populieren nur etwa 80 % der faltenden Moleküle den Zwischenzustand HFslow, der mit einer Zeitkonstante von ca. 100 s zu HFfast weiterreagiert. Die Denaturierungsmittelabhängigkeit dieser Reaktion ist sehr gering, was eine trans-/cis-Prolylisomerisierung als geschwindigkeitslimitierenden Schritt nahelegt. Die Existenz eines cis-Peptides in der nativen Struktur macht es erforderlich, den denaturierten Zustand als ein Ensemble kinetisch separierter Konformationen, kurz: DSE, zu betrachten, das durch die Spezies Ufast und Uslow populiert wird. Nach dem in dieser Arbeit aufgestellten „Minimalmodell der Pel-15 Faltung“ stehen die HF-Spezies (HFslow, HFfast) mit den Konformationen des DSE in einem thermodynamischen Kreisprozess. Das Modell positioniert HFfast und die native Konformation N auf die „native Seite“ der Aktivierungsbarriere und trägt damit der Tatsache Rechnung, dass die Gleichgewichtseinstellung zwischen diesen Spezies zu schnell ist, um mit manuellen Techniken erfasst zu werden. Die hochaffine Bindung von Ca2+ (Kd = 10 μM) verschiebt sich das Faltungsgleichgewicht bereits in Gegenwart von 1 mM CaCl2 soweit auf die Seite des nativen Zustandes, das HFfast nicht länger nachweisbar ist. Entgegen anfänglicher Vermutungen kommt einer lokalen, evolutionär konservierten Disulfidbrücke im Zentrum der β-Helix eine wichtige Stabilisierungsfunktion zu. Die Disulfidbrücke befindet sich in einem kurzen Schleifenbereich der β-Helix nahe dem aktiven Zentrum. Obwohl ihr Austausch gegen die Reste Val und Ala die freie Stabilisierungsenthalpie des Proteins um ca. 10 kJ/mol reduziert, lässt die Struktur im Bereich der Mutationsstelle keine gravierende Veränderung erkennen. Auch die katalytisch relevante Ca2+-Bindungsaffinität bleibt unbeeinflusst; dennoch zeigen Enzymaktivitätstests für VA-Mutanten eine Reduktion der enzymatischen Aktivität um fast 50 % an. Die evolutionär konservierte Helixwindung im Allgemeinen und die in ihr enthaltene Disulfidbrücke im Besonderen müssen nach den vorliegenden Ergebnissen also eine zentrale Funktion sowohl für die Struktur des katalytischen Zentrums als auch für die Strukturbildung der β-Helix während der Faltungsreaktion besitzen. Die Ergebnisse dieser Arbeit finden in mehreren Punkten Anklang an Faltungseigenschaften, die für andere β -Helixproteine beschrieben wurden. Vor allem aber prädestinieren sie Pel-15 als ein neues, β-helikales Modellprotein. Aufgrund seiner einfachen Topologie, seiner niedrigen Windungszahl und seiner hohen thermodynamischen Stabilität ist Pel-15 sehr gut geeignet, die Determinanten von Stabilität und Strukturbildung des parallelen β-Helix-Motivs in einer Auflösung zu studieren, die aufgrund der Komplexität bestehender β-helikaler Modellsysteme bislang nicht zur Verfügung stand. N2 - Pectate lyase Pel-15 was isolated from alcaliphlic Bacillus spec. strain KSM-P15. Like all pectate lyases Pel-15 binds and subsequently cleaves polygalacturonic acid, the main pectic compound in plant cell walls and middle lamellae, in a Ca2+ dependent beta-elimination reaction. With 197 amino acids and a molecular mass of only 21 kDa the protein is one of the smallest right-handed parallel beta-helical proteins known today. Polypeptide chains that are classified into this structural family adopt super-helical folds in which each “solenoid stack” consists of three beta-structured regions that are connected by flexible turn segments. Along its longitudinal axis the right-handed parallel beta-helix thus comprises three elongated parallel beta-sheets that are stabilized by an extensive network of hydrogen bonds wrapping around the densely packed hydrophobic core. Together with the shield-like arrangement of hydrogen bonds this hydrophobic core is considered as the main contributor to an exceptionally high stability that is a common feature of all beta-helical proteins. In contrast to most right-handed parallel beta-helices, Pel-15 is devoid of any terminal capping domains and laterally associated secondary structure. Therefore, this protein is considered to be a promising model protein of a pure beta-helix which will help to understand the determinants of both parallel beta-sheet formation and stability. In the dissertation at hand optical spectroscopic methods were used to assess the enzymatic activity, the folding/unfolding equilibrium and the kinetic mechanism of structure formation in neutral buffered solutions. Results indicate that Pel-15 populates a hyper-fluorescent equilibrium intermediate (HF) that is effectively populated in presence of the denaturing agent guanidinium hydrochloride (GdmCl). According to kinetic folding and unfolding experiments HF is not only an essential on-pathway intermediate but has to be considered as a conformational ensemble in which several hyperfluorescent states are in thermodynamic equilibrium with each other. According to their existence in kinetic folding trajectories these different HF-species were termed HFslow and HFfast. The activation energy between both states is remarkably high leading to a time constant of about 100 seconds for the reaction HFslow ⇆ HFfast. Since native Pel-15 contains an energetically disfavoured cis-prolyl peptide between A59 and P60 it is proposed that HFslow and HFfast differ in their prolyl peptide conformations. Two main results emerge from this dissertation. First, an extensive study of the Pel-15 folding- and unfolding behaviour facilitated the proposal of a “minimal folding model”. According to this model the HF-states and the according denatured species Uslow and Ufast are aligned into a thermodynamic circle. This implies that unfolded polypeptide chains reach the HF-ensemble via parallel folding trajectories. Since the native conformation N together with HFfast are on the same side of the activation barrier, it is the reaction HFslow ⇆ HFfast that is the rate limiting step in the folding reaction of Pel-15. Second, the importance of an evolutionarily conserved disulfide bond in the central region of Pel-15 was tested by site directed mutagenesis and subsequent spectroscopic characterization. The exchange of the disulfide against a hydrophobic pair of alanine and valine decreases the folding free energy by about 10 kJ/mol. Although this value is unexpectedly high, structural perturbations around both mutational positions are small as was deduced from X-Ray crystallography. Interestingly, the stability decrease is accompanied by a major loss of enzymatic activity while the Ca2+ binding affinity is not significantly affected. It is therefore concluded that the allosterically relevant disulfide bond stabilizes long-range interactions that stabilize several adjacent solenoid turns near the N-terminus of the protein. Indeed, planar stacking interactions are perturbed and flexibility of N-terminal loops is increased once the disulfide bond is removed. This dissertation establishes Pel-15 as a novel beta-helical model protein and – even more important – smoothes the way for a generally accepted perspective on the formation and stability of parallel beta-sheet proteins. In Arabidopsis (Arabidopsis thaliana), TREHALOSE-6-PHOSPHATE SYNTHASE1 (TPS1) catalyzes the synthesis of the sucrose-signaling metabolite trehalose 6-phosphate (Tre6P) and is essential for embryogenesis and normal postembryonic growth and development. To understand its molecular functions, we transformed the embryo-lethal tps1-1 null mutant with various forms of TPS1 and with a heterologous TPS (OtsA) from Escherichia coli, under the control of the TPS1 promoter, and tested for complementation. TPS1 protein localized predominantly in the phloem-loading zone and guard cells in leaves, root vasculature, and shoot apical meristem, implicating it in both local and systemic signaling of Suc status. The protein is targeted mainly to the nucleus. Restoring Tre6P synthesis was both necessary and sufficient to rescue the tps1-1 mutant through embryogenesis. However, postembryonic growth and the sucrose-Tre6P relationship were disrupted in some complementation lines. A point mutation (A119W) in the catalytic domain or truncating the C-terminal domain of TPS1 severely compromised growth. Despite having high Tre6P levels, these plants never flowered, possibly because Tre6P signaling was disrupted by two unidentified disaccharide-monophosphates that appeared in these plants. The noncatalytic domains of TPS1 ensure its targeting to the correct subcellular compartment and its catalytic fidelity and are required for appropriate signaling of Suc status by Tre6P. We expressed tagged forms of TREHALOSE-6-PHOSPHATE SYNTHASE1 (TPS1) to determine where Tre6P is synthesized in arabidopsis (Arabidopsis thaliana), and investigated the impact of localized changes in Tre6P levels, in axillary buds or vascular tissues, on shoot branching in wild-type and branching mutant backgrounds. TPS1 is expressed in axillary buds and the subtending vasculature, as well as in the leaf and stem vasculature. Expression of a heterologous Tre6P phosphatase (TPP) to lower Tre6P in axillary buds strongly delayed bud outgrowth in long days and inhibited branching in short days. TPP expression in the vasculature also delayed lateral bud outgrowth and decreased branching. Increased Tre6P in the vasculature enhanced branching and was accompanied by higher expression of FLOWERING LOCUS T (FT) and upregulation of sucrose transporters. Increased vascular Tre6P levels enhanced branching in branched1 but not in ft mutant backgrounds. These results provide direct genetic evidence of a local role for Tre6P in regulation of axillary bud outgrowth within the buds themselves, and also connect Tre6P with systemic regulation of shoot branching via FT. Identifying classes of programs for which the consistency problem is in NP is of interest from the theoretical standpoint and can potentially lead to improvements in the design of answer set programming solvers. One of such classes consists of dual-normal programs, where the number of positive body atoms in proper rules is at most one. Unlike other classes of programs, dual-normal programs have received little attention so far. In this paper we study this class. We relate dual-normal programs to propositional theories and to normal programs by presenting several inter-translations. With the translation from dual-normal to normal programs at hand, we introduce the novel class of body-cycle free programs, which are in many respects dual to head-cycle free programs. We establish the expressive power of dual-normal programs in terms of SE- and UE-models, and compare them to normal programs. We also discuss the complexity of deciding whether dual-normal programs are strongly and uniformly equivalent. The chosen model taxon, the genus Campylomormyrus, exhibits a wide diversity of electric organ discharge (EOD) waveform types. Their EOD is age, sex, and species specific and is an important character for discriminating among species that are otherwise cryptic. After having established a complementary set of molecular markers, I examined the radiation of Campylomormyrus by a combined approach of molecular data (sequence data from the mitochondrial cytochrome b and the nuclear S7 ribosomal protein gene, as well as 18 microsatellite loci, especially developed for the genus Campylomormyrus), observation of ontogeny and diversification of EOD waveform, and morphometric analysis of relevant morphological traits. I built up the first convincing phylogenetic hypothesis for the genus Campylomormyrus. Taking advantage of microsatellite data, the identified phylogenetic clades proved to be reproductively isolated biological species. This way I detected at least six species occurring in sympatry near Brazzaville/Kinshasa (Congo Basin). By combining molecular data and EOD analyses, I could show that there are three cryptic species, characterised by their own adult EOD types, hidden under a common juvenile EOD form. In addition, I confirmed that adult male EOD is species-specific and is more different among closely related species than among more distantly related ones. This result and the observation that the EOD changes with maturity suggest its function as a reproductive isolation mechanism. As a result of my morphometric shape analysis, I could assign species types to the identified reproductively isolated groups to produce a sound taxonomy of the group. Besides this, I could also identify morphological traits relevant for the divergences between the identified species. Among them, the variations I found in the shape of the trunk-like snout, suggest the presence of different trophic specializations; therefore, this trait might have been involved in the ecological radiation of the group. In conclusion, I provided a convincing scenario envisioning an adaptive radiation of weakly electric fish triggered by sexual selection via assortative mating due to differences in EOD characteristics, but caused by a divergent selection of morphological traits correlated with the feeding ecology. N2 - Das übergreifende Ziel dieser Arbeit ist das bessere Verständnis der Bedeutung der schwachen Elektrizität für die adaptive Radiation der Mormyriden Afrikas. Das gewählte Modell-Taxon, die Mormyriden-Gattung Campylomormyrus, zeigt eine große Vielfalt an elektrischen Entladungsformen. Diese Entladungsformen sind alters-, geschlechts-, sowie artspezifisch und ein wichtiges Unterscheidungskriterium von ansonsten kryptischen Arten. Ich untersuchte die Radiation der Gattung Campylomormyrus anhand eines kombinierten Ansatzes aus molekularen Daten (Sequenzdaten des mitochondrialen Cytochrom b Gens und des nukleären S7 ribosomalen Protein-Gens, sowie 18 Mikrosatelliten, speziell von mir entwickelt für die Gattung Campylomormyrus), Beobachtungen der Ontogenie und der Diversifikation der Entladungsform, sowie morphometrische Auswertungen der Gestalt relevanter morphologischer Merkmale. Ich erstellte eine erste phylogenetische Hypothese für die Gattung Campylomormyrus. Durch meine Mikrosatellitendaten, die als unabhängiger Beweis dienten, konnte ich zeigen, dass die identifizierten phylogenetischen Gruppen reproduktiv isolierte biologische Arten sind. Auf diese Weise konnte ich mindesten sechs Arten nachweisen, die in Sympatrie nahe Brazzaville/Kinshasa (Kongo-Becken) vorkommen. Durch die Übereinstimmung von molekularen Daten und Entladungsformen konnte ich drei kryptische Arten unterscheiden, die sich hinter einheitlichen juvenilen Entladungsformen verbergen, sich aber zu verschiedenen adulten Formen entwickelten. Des Weiteren konnte ich zeigen, dass die adulten männlichen Entladungsformen artspezifisch sind und, dass der Unterschied in der Entladungsform zwischen nah verwandten Arten deutlicher ausgeprägt ist als zwischen entfernter verwandten Arten. Dieses Ergebnis und die Beobachtung, dass sich die Entladungsform bei der Geschlechtsreife ändert, weisen darauf hin, dass die Entladungsform als reproduktiver Isolationsmechanismus dient. In einer morphometrischen Gestalt-Analyse verglich ich das Typen-Material der beschriebenen Arten mit den zuvor ermittelten reproduktiv isolierten Gruppen, um auf diese Weise deren Art zu bestimmen. Überdies konnte ich maßgebliche morphologische Unterscheidungsmerkmale identifizieren. Diese äußern sich hauptsächlich in der Gestalt der rüsselartigen Schnauze, könnten daher mit einer trophischen Spezialisierung einhergehen und eine ökologische Artbildung ermöglichen. Zusammenfassend entwickelte ich, in Übereinstimmung mit anderen Untersuchungen und theoretischen Überlegungen, eine plausible Hypothese einer adaptiven Radiation der schwach-elektrischen Fische Afrikas, ausgelöst durch sexuelle Selektion. Diese wirkt durch assortative Verpaarung, basierend auf Charakteristika der elektrischen Entladungsform. Verursacht wird der Prozess der adaptiven Radiation jedoch durch divergierende Selektion morphologischer Merkmale, die in Bezug zur Nahrungsökologie stehen. Fourier reprsentations of velocity, pressure and magnetic field have been used to transform the original partial differential equations into systems of ordinary differential equations (ODE), to which then special numerical methods for the qualitative analysis of systems of ODE have been applied, supplemented by the simulative calculation of solutions for selected initial conditions. In a part of the calculations, in order to reduce the number of modes to be retained, the concept of approximate inertial manifolds has been applied. For varying (incereasing from zero) strength of the imposed forcing, or varying Reynolds number, respectively, time-asymptotic states, notably stable stationary solutions, have been traced. A primary non-magnetic steady state loses, in a Hopf bifurcation, stability to a periodic state with a non-vanishing magnetic field, showing the appearance of a generic dynamo effect. From now on the magnetic field is present for all values of the forcing. The Hopf bifurcation is followed by furhter, symmetry-breaking, bifurcations, leading finally to chaos. We pay particular attention to kinetic and magnetic helicities. The dynamo effect is observed only if the forcing is chosen such that a mean kinetic helicity is generated; otherwise the magnetic field diffuses away, and the time-asymptotic states are non-magnetic, in accordance with traditional kinematic dynamo theory. Bifurcation-analysis techniques have been applied to explore the qualitative behavior of solution branches. Due to the symmetry of the forcing, the equations are equivariant with respect to a group of transformations isomorphic to the octahedral group, and we have paid special attention to symmetry-breaking effects. As the Reynolds number is increased, the primary nonmagnetic steady state, the ABC flow, loses its stability to a periodic magnetic state, showing the appearance of a generic dynamo effect; the critical value of the Reynolds number for the instability of the ABC flow is decreased compared to the purely hydrodynamic case. The bifurcating magnetic branch in turn is subject to secondary, symmetry-breaking bifurcations. We have traced periodic and quasi- periodic branches until they end up in chaotic states. In particular detail we have analyzed the subgroup symmetries of the bifurcating periodic branches, which are closely related to the spatial structure of the magnetic field. Fourier representations of velocity and pressure have been used to approximate the original partial differential equations by a finite-dimensional system of ordinary differential equations, which then has been studied by means of bifurcation-analysis techniques. A special route into chaos observed for increasing Reynolds number or strength of the imposed forcing is described. It includes several steady states, traveling waves, modulated traveling waves, periodic and torus solutions, as well as a period-doubling cascade for a torus solution. Lyapunov exponents and Kaplan-Yorke dimensions have been calculated to characterize the chaotic branch. While studying the dynamics of the system in Fourier space, we also have transformed solutions to real space and examined the relation between the different bifurcations in Fourier space and toplogical changes of the streamline portrait. N2 - The thesis deals with the synthesis of carbon nitride based materials in eutectic mixtures of bivalent metal chlorides. The focus is on ZnCl2, SnCl2 and CoCl2 containing eutectic mixtures in which the condensation reaction of classical organic precursors is carried out. In this context, it is investigated how to control molecular structure and other materials properties, like morphology, crystallinity and specific surface areas by using salt melts of different Lewis acidity. Moreover optical properties of the materials are discussed and in this connection their suitability as photocatalysts for dye degradation and photocatalytic water reduction and oxidation is investigated. The only between-group difference was CLI: one group consistently produced significantly more errors of CLI on both tasks than the other (thereby replicating our findings from a bilingual picture naming task). This striking group difference in language control ability can only be explained by differences in cognitive control, not in language proficiency or language mode. Dazu gehören u. a. Prüfungen zur Vorhersage des embryotoxischen Potentials, die am lebenden Organismus durchgeführt werden. Mit dem Ziel die Anzahl der Tierversuche zu verringern, die notwendig sind um das toxikologische Profil einer Prüfsubstanz zu bestimmen, wurde der Embryonale Stammzelltest (EST) entwickelt. Als Grundlage des EST dienen embryonale Stammzellen (ES-Zellen) einer Zelllinie. ES-Zellen sind Zellen, die sich in der frühen embryonalen Entwicklung in die Zellen der Keimblätter entwickeln können. Daraus wiederum differenzieren die vielen verschiedenen, unterschiedlich spezialisierten Zelltypen des komplexen Organismus. Im EST wird die Konzentration einer Prüfsubstanz bestimmt, bei der die Differenzierung von ES-Zellen zu Herzmuskelzellen zu 50 % inhibiert wird. Zusätzlich wird die Konzentration der Prüfsubstanz bestimm°t, bei der 50 % der ES-Zellen (IC50D3) bzw. Fibroblastenzellen (IC503T3) absterben. Die allgemeine Toxizität ist damit von der spezifischen Toxizität der Prüfsubstanz auf die ES-Zellen und deren Differenzierung unterscheidbar. Die Parameter fliessen in ein biostatistisches Modell zur Prädiktion des embryotoxischen Potentials der Prüfsubstanzen ein. Es wurde ein Versuchsprotokoll entwickelt, wonach die ES-Zellen sich verstärkt zu Endothelzellen differenzieren. Die Endothelzellen, die im lebenden Organismus die Wand der späteren Blutgefässe, wie Venen und Arterien bilden, wurden mittels molekularbiologischer Methoden auf der RNA- und der Protein-Ebene nachgewiesen und quantifiziert. Verschiedene Zellkulturmethoden, Wachstumsfaktoren, als auch Wachstumsfaktorkonzentrationen wurden auf deren Vermögen die Differenzierung der ES-Zellen zu Endothelzellen zu induzieren, untersucht. Nach der Etablierung des Differenzierungsprotokolls wurden sieben Substanzen auf deren Vermögen geprüft, die Differenzierung von ES-Zellen zu Endothelzellen zu inhibieren. Die Endothelzellen wurden dabei über die Expression der RNA von zwei endothelzellspezifischen Genen quantifiziert. Im Vergleich dazu wurden die IC50D3 und die IC503T3 der Prüfsubstanz bestimmt, um eine Abschätzung des embryotoxischen Potentials der Prüfsubstanz zu ermöglichen. Die Ergebnisse zeigten, dass eine Abschätzung des embryotoxischen Potentials der sieben Prüfsubstanzen in nicht-, schwach- oder stark embryotoxisch vorgenommen werden konnte. Es ist zu schlussfolgern, dass der weiterentwickelte in vitro Embryotoxizitätsassay sensitiv und reproduzierbar ist. Mit der Verwendung von verschiedenen Differenzierungsendpunkten kann die Prädiktionskraft des Assays deutlich verbessert, und die Anzahl von Tierversuchen verringert werden. Durch die Verwendung von molekularbiologischen Markern kann der Assay einem Hochdurchsatzscreening zugängig gemacht werden und damit die Anzahl von Prüfsubstanzen deutlich erhöht werden. N2 - Compounds of the pharmaceutical and chemical industry need to be tested for their toxicological potential with regard to humans and environment following international guidelines. Tests for the prediction of the embryotoxic potential executed on living organisms are examples of these guidelines. In order to reduce the number of animal experiments necessary for the assessment of the toxicological profile of compounds the embryonic stem cell test (EST) was developed. Embryonic stem cells (ES-cells) of a cell line are used as the basis of the EST. ES-cells are cells which develop at the early embryonic development into cells of the germ layers. Out of these the many different specialized cell types of the complex organism can differentiate. With the EST this concentration of a test compound will be determined where a 50 % inhibition of the differentiation of ES-cells into cardiomyocytes can be detected. Additionally, the concentration of a test compound which is cytotoxic to 50 % of the ES-cells (IC50D3) and to 50 % of fibroblasts (IC503T3) will be determined. Therefore, general toxicity caused by the test compound on ES-cells and its differentiation can be distinguished from specific toxicity of the test compound. Determined parameters will be included into a biostatistical model for the subsequent predicition of the embryotoxic potential of test compounds. A protocol was developed whereby the differentiation of ES-cells into endothelial cells is induced. Endothelial cells which make up the walls of blood vessels, such as arteries and veins, were detected and quantified at the RNA and the protein level applying molecular biological methods. Different cell culture methods, growth factors and growth factor concentrations were studied for their ability to induce the differentiation of ES-cells into endothelial cells. Applying the developed differentiation protocol seven compounds were tested for their potential to inhibit the differentiation of ES-cells into endothelial cells. Endothelial cells were quantified by the RNA-expression of two endothelial-specific genes. In comparison to the expression levels the IC50D3 and the IC503T3 were determined in order to assess the embryotoxic potential of the test compound. The results showed that an assessment of the embryotoxic potential of the seven test compounds into non-, weakly- and strongly embroytoxic was possible. It can be concluded that the improved in vitro embryotoxicity assay is sensitive and reproducible. With the use of different differentiation endpoints the power of predicitivity of this assay can be significantly increased and the number of animal experiments can be reduced. With the application of molecular biological markers this assay can be applied as a high througput screening and therefore the number of test compounds can be strongly increased. This study aims (a) to develop a comprehensive multi-objective calibration framework using remote sensing vegetation data and hydrological signatures (flow duration curve - FDC, and baseflow index) in addition to discharge, and (b) to apply this framework for calibration of the Soil and Water Assessment Tool (SWAT) in a typical Andean catchment. Overall, our calibration approach outperformed traditional discharge-based and FDC signature-based calibration strategies in terms of vegetation, streamflow, and flow partitioning simulation. New hydrological insights for the region are the following: baseflow is the main component of the streamflow sustaining the long dry-season flow, and pasture areas offer higher water yield and baseflow than other land-cover types. Methods One-hundred and nineteen young male handball players (under (U)-13 (U13) [n = 85]) and U15 [n = 34]) volunteered to participate in this study. The participating athletes had a mean background of sytematic handball training of 5.5 ± 2.8 years and they exercised on average 540 ± 10.1 min per week including sport-specific team handball training and strength and conditioning programs. Players were tested for passive shoulder range-of-motion (ROM) for both internal (IR) and external rotation (ER) and isometric strength (i.e., IR and ER) of the dominant/non-dominant shoulders, overhead medicine ball throw (OMB), hip isometric abductor (ABD) and adductor (ADD) strength, hip ROM, jumps (countermovement jump [CMJ] and triple leg-hop [3H] for distance), linear sprint test, modified 505 change-of-direction (COD) test and handball throwing speed (7 m [HT7] and 9 m [HT9]). Results U15 players outperformed U13 in upper (i.e., HT7 and HT9 speed, OMB, absolute IR and ER strength of the dominant and non-dominant sides; Cohen’s d: 0.76–2.13) and lower body (i.e., CMJ, 3H, 20-m sprint and COD, hip ABD and ADD; d: 0.70–2.33) performance measures. Regarding shoulder ROM outcomes, a lower IR ROM was found of the dominant side in the U15 group compared to the U13 and a higher ER ROM on both sides in U15 (d: 0.76–1.04). It seems that primarily anthropometric characteristics (i.e., body height, body mass) and upper body strength/power (OMB distance) are the most important factors that explain the throw speed variance in male handball players, particularly in U13. Conclusions Findings from this study imply that regular performance monitoring is important for performance development and for minimizing injury risk of the shoulder in both age categories of young male handball players. Besides measures of physical fitness, anthropometric data should be recorded because handball throwing performance is related to these measures. Their size distribution is thought to be a result of competing accretion and fragmentation processes. While growth is naturally limited in tidal environments, frequent collisions among these objects may contribute to both accretion and fragmentation. As ring particles are primarily made of water ice attractive surface forces like adhesion could significantly influence these processes, finally determining the resulting size distribution. Here, we derive analytic expressions for the specific self-energy Q and related specific break-up energy Q⋆ of aggregates. These expressions can be used for any aggregate type composed of monomeric constituents. We compare these expressions to numerical experiments where we create aggregates of various types including: regular packings like the face-centered cubic (fcc), Ballistic Particle Cluster Aggregates (BPCA), and modified BPCAs including e.g. different constituent size distributions. We show that accounting for attractive surface forces such as adhesion a simple approach is able to: a) generally account for the size dependence of the specific break-up energy for fragmentation to occur reported in the literature, namely the division into “strength” and “gravity” regimes, and b) estimate the maximum aggregate size in a collisional ensemble to be on the order of a few meters, consistent with the maximum aggregate size observed in Saturn’s rings of about 10m. N2 - Die Ringe des Saturns bestehen aus Myriaden von Teilchen, deren Größe von Mikrometern (Staub) bis hin zu Hunderten von Metern reicht. Die Ringteilchen bestehen hauptsächlich aus Eis, wobei attraktive Oberflächenkräfte wie Adhäsion und Gravitation zur Bildung von Aggregaten führen kann. Das Wachstum der Aggregate wird durch die Wirkung der Gezeitenkräfte und auch durch Kollisionen der Ringteilchen untereinander auf natürliche Weise begrenzt. Die Kollisionen der Ringteilchen führen zu Akkretion und Fragmentation, welche die resultierende Größenverteilung der Agglomerate schließlich bestimmen. Diese Ausdrücke können für alle aus monomeren Teilchen bestehenden Agglomerate verwendet werden. Die analytisch gewonnenen Ergebnisse wurden mit numerischen Experimenten verglichen. In den numerischen Experimenten wurden verschiedene Agglomerattypen erzeugt: (i) Agglomerate mit kubischem Kristallsystem, (ii) ballistische Teilchenaggregate und (iii) modifiziert ballistische Teilchenaggregate. Für die ballistischen Teilchenaggregate wurden verschiedene Größenverteilungen der Konstituenten verwendet. Als Ergebnis lassen sich die erzeugten Aggregate gemäß ihrer Größe in zwei Gruppen einteilen. Während die kleinen Aggregate hauptsächlich durch die Kontaktkräfte (Adhäsion) zusammengehalten werden, dominiert bei großen Aggregaten (größer als einige Meter) die Gravitationskraft. D.h. wächst aus kleinen Teilchen ein Aggregat, so wird dieses zunächst durch die haftenden Kontakte zwischen den Teilchen zusammengehalten. Wächst das Agglomerat über eine bestimmte Größe, so ist es die Eigengravitation, die den Körper zusammenhält. Damit kann die maximale Gesamtgröße der Aggregate im Kollisionsensemble abgeschätzt werden. The enzyme catalyzed the penultimate step of juvenile hormone biosynthesis in the insects. Culturing corpora allata in the presence of sinefungin greatly suppressed juvenile hormone production. The following in vivo effects were visible after injection of the inhibitor: increase in mortality and reduction of total haemolymph protein liter and ovary fresh weight, as well as length of terminal oocytes. Attempts to reverse these effects by topical application of the juvenile hormone analog ZR-515 (methoprene) were only partly successful. Therefore, the in vivo effects may be due to a general inhibition of methyltransferase enzymes in the insect. In this study, we identify and quantify the ENSO contribution to the eastern African rainfall variability to forecast future eastern African vegetation response to rainfall variability related to a predicted intensified ENSO. To differentiate the vegetation variability due to ENSO, we removed the ENSO signal from the climate data using empirical orthogonal teleconnection (EOT) analysis. Then, we simulated the ecosystem carbon and water fluxes under the historical climate without components related to ENSO teleconnections. We found ENSO-driven patterns in vegetation response and confirmed that EOT analysis can successfully produce coupled tropical Pacific sea surface temperature-eastern African rainfall teleconnection from observed datasets. We further simulated eastern African vegetation response under future climate change as it is projected by climate models and under future climate change combined with a predicted increased ENSO intensity. Our EOT analysis highlights that climate simulations are still not good at capturing rainfall variability due to ENSO, and as we show here the future vegetation would be different from what is simulated under these climate model outputs lacking accurate ENSO contribution. We simulated considerable differences in eastern African vegetation growth under the influence of an intensified ENSO regime which will bring further environmental stress to a region with a reduced capacity to adapt effects of global climate change and food security. Ein Studium seiner Gegenwart und seiner Zukunft wiederum hilft uns dabei, die global bedeutende Artenvielfalt und die ökosystemaren Dienstleistungen Ostafrikas zu schützen. Eine zentrale Rolle spielt dabei spielt die ostafrikanische Vegetation, deren Dynamiken in dieser Dissertation durch Computersimulationen quantifiziert werden sollen. Über Computersimulationen lassen sich frühere Rahmenbedingungen reproduzieren, Voraussagen treffen oder Simulationsexperimente durchführen, die durch Feldforschung nicht möglich wären. Zuallererst muss jedoch ihre Leistungsfähigkeit überprüft werden. Die von dem Modell anhand der heutigen Inputs gelieferten Ergebnisse stimmten weitgehend mit heutigen Beobachtungen ostafrikanischer Vegetation überein. Als nächstes wurde die frühere Vegetation simuliert, für die fossile Pollen-Daten zum Abgleich vorliegen. Über Computermodelle lassen sich Wissenslücken zwischen Standorten überbrücken, bei denen wir über fossile Pollen-Daten verfügen, sodass ein vollständigeres Bild der Vergangenheit entsteht. Zusätzlich validiert wurde die Leistungsfähigkeit des Modells durch die hohe Übereinstimmung zwischen Modell und Pollen-Daten, wo sie im Raum überlappen. Nachdem das Modell getestet und für die Region validiert war, konnte eine der seit langem offenen Fragen über die ostafrikanische Vegetation angegangen werden, nämlich wie Ostafrika seines Tropenwaldes verlustig gehen konnte. In den Tropen wird die heutige Vegetation weltweit hauptsächlich von Wäldern dominiert, mit Ausnahme der Tropengebiete Ostafrikas, wo Wälder nur noch stellenweise an der Küste und im Hochland vorkommen. Durch eine Reihe von Simulationsexperimenten konnte aufgezeigt werden, unter welchen Bedingungen jene Waldgebiete früher zusammenhingen und schließlich fragmentiert wurden. Die Studie hat erwiesen, wie empfindlich die ostafrikanische Vegetation für die Klimaschwankungen ist, die durch den künftigen Klimawandel zu erwarten sind. Weitere Auswirkungen auf das ostafrikanische Klima ergeben sich aus dem El Niño/Southern Oscillation-Phänomen (ENSO), das aus Temperaturfluktuationen zwischen dem Ozean und der Atmosphäre herrührt und künftig an Intensität zunehmen dürfte. Die derzeitigen Klimamodelle sind allerdings noch nicht gut genug beim Erfassen solcher Ereignismuster. In einer Studie wurde der Einfluss des ENSO-Phänomens auf die ostafrikanische Vegetation quantifiziert und dabei aufgezeigt, wie sehr sich die künftige Vegetation von den heute simulierten Ergebnissen unterscheiden könnte, bei denen der genaue ENSO-Beitrag nicht berücksichtigt werden kann. Bei der Berechnung der künftigen weltweiten CO2-Bilanz und den zu treffenden Entscheidungen stellt dies einen zusätzlichen Unsicherheitsfaktor dar. Computer models help us recreate past settings, forecast into the future or conduct simulation experiments that we cannot otherwise perform in the field. But before all that, one needs to test their performance. The outputs that the model produced using the present day-inputs, agreed well with present-day observations of East African vegetation. Next, I simulated past vegetation for which we have fossil pollen data to compare. With computer models, we can fill the gaps of knowledge between sites where we have fossil pollen data from, and create a more complete picture of the past. Good level of agreement between model and pollen data where they overlapped in space further validated our model performance. Once the model was tested and validated for the region, it became possible to probe one of the long standing questions regarding East African vegetation: How did East Africa lose its tropical forests? The present-day vegetation in the tropics is mainly characterized by continuous forests worldwide except in tropical East Africa, where forests only occur as patches. In a series of simulation experiments, I was able to show under which conditions these forest patches could have been connected and fragmented in the past. This study showed the sensitivity of East African vegetation to climate change and variability such as those expected under future climate change. El Niño Southern Oscillation (ENSO) events that result from the fluctuations in temperature between the ocean and atmosphere, bring further variability to East African climate and are predicted to increase in intensity in the future. But climate models are still not good at capturing the pattens of these events. In a study where I quantified the influence of ENSO events on East African vegetation, I showed how different the future vegetation could be from what we currently predict with these climate models that lack accurate ENSO contribution. Consideration of these discrepancies is important for our future global carbon budget calculations and management decisions. Although the conjoint occurrence between brain-structural and brain-functional changes, the decrease of plasticity, and changes in cognitive abilities suggests a certain causality between these processes, concrete evidence for the relation between brain development, language processing, and language performance is rare. Therefore, the current dissertation investigates the tripartite relationship between behavior (in the form of language performance and cognitive maturation as prerequisite for language processing), brain structure (in the form of gray matter maturation), and brain function (in the form of brain activation evoked by complex sentence processing). Previous developmental studies indicate a missing increase of activation in accordance to sentence complexity (functional selectivity) in language-relevant brain areas in children. To determine the factors contributing to the functional development of language-relevant brain areas, different methodologies and data acquisition techniques were used to investigate the processing of center-embedded sentences in 5- and 6-year-old children, 7- and 8-year-old children, and adults. Behavioral results indicate that children between 5 and 8 years show difficulties in processing double embedded sentences and that their performance for these type of sentences is positively correlated with digit span. In 7- and 8-year-old children, it was found that especially the processing of long-distance relations between the initial phrase and its corresponding verb appears to be associated with the subject’s verbal working memory capacity. In contrast, children’s performance for double embedded sentences in the younger age group positively correlated with their performance in a standardized sentence comprehension test. This finding supports the hypothesis that processing difficulties in this age group may be mainly attributed to difficulties in processing case marking information. These findings are discussed with respect to current accounts of language and working memory development. A second study aimed at investigating the structural maturation of brain areas involved in sentence comprehension. To do this, whole-brain magnetic resonance images from 59 children between 5 and 8 years were collected and children’s gray matter was analyzed by using voxel-based morphometry. Children’s grammatical proficiency was assessed by a standardized sentence comprehension test. A confirmatory factory analysis corroborated a grammar-relevant and a verbal working memory-relevant factor underlying the measured performance. While children’s ability to assign thematic roles is positively correlated with gray matter probability (GMP) in the left inferior temporal gyrus and the left inferior frontal gyrus, verbal working memory-related performance is positively correlated with GMP in the left parietal operculum extending into the posterior superior temporal gyrus. These areas have been previously shown to be differentially engaged in adults’ complex sentence processing. Thus, the findings of the second study suggest a specific correspondence between children’s GMP in language-relevant brain regions and differential cognitive abilities which underlie complex sentence comprehension. In a third study, functional brain activity during the processing of center-embedded sentences was investigated in three different age groups (5–6 years, 7–8 years, and adults). Although all age groups engage a qualitatively comparable network of the left pars opercularis (PO), the left inferior parietal lobe extending into the posterior superior temporal gyrus (IPL/pSTG), the supplementary motor area (SMA) and the cerebellum, functional selectivity of these regions was only observable in adults. However, functional activation of the language-related regions (PO and IPL/pSTG) predicted sentence comprehension performance for all age groups. To solve the question of the complex interplay between different maturational factors, a fourth study analyzed the predictive power of gray matter probability, verbal working memory capacity, and behavioral differences in performance for simple and complex sentence for the functional selectivity of each activated region. These analyses revealed that the establishment of the adult-like functional selectivity for complex sentences is predicted by a reduction of the left PO’s gray matter probability across age groups while that of the IPL/pSTG is additionally predicted by verbal working memory capacity. Taken all findings together, the current thesis provides evidence that both structural brain maturation and verbal working memory expansion provide the basis for the emergence of functional selectivity in language-related brain regions leading to more efficient sentence processing during development. Sie werden als allgemeines Phänomen bei unterschiedlichsten astronomischen Quellen gefunden - bei Objekten die sich sowohl in der Grössenskala als auch im Energieumsatz um viele Grössenordnungen unterscheiden. Jets werden beobachtet bei jungen stellaren Objekten (etwa TTauri-Sternen oder eingebettete IR-Quellen), bei sogenannten Mikroquasaren und bei aktiven galaktischen Kernen (etwa Radiogalaxien oder Quasare). So unterschiedlich die Jetquellen von ihrer Erscheinung sein mögen, zwei Tatsachen scheinen sie zu vereinen: Alle Jetquellen zeigen ebenfalls Hinweise auf die Existenz einer Akkretionsscheibe und von Magnetfeldern. Damit sind die wichtigsten Punkte einer Theorie der Jetentstehung schon umrissen. Sie muss sowohl die komplexe Struktur der Jetquelle berücksichtigen - ein System bestehend aus einem Zentralobjekt, der es umgebenden Scheibe, und dem Jet - als auch die magnetohydrodynamische Wechselwirkung zwischen diesen Komponenten. Die magnetohydrodynamischen Gleichungen für solch ein Problem sind derart kompliziert, dass sie meist nur numerisch, also nur mit dem Computer zu lösen sind. Zusätzlich sind viele vereinfachende Annahmen notwendig, da sonst auch der Computer überfordert waere. Im allgemeinen sind folgende Fragestellungen zu lösen: - Die Frage, wie ein Scheibenwind langsamer Geschwindigkeit beschleunigt wird und in einen Jet kollimiert wird ("jet formation"). - Die Frage, wie ein Ausfluss aus der Akkretionsscheibe überhaupt entsteht, d.h. die Frage wie die akkretierende Materie der Scheibe in den Scheibenwind umgelenkt wird ("jet launching"). - Die Frage, wie und wo das Magnetfeld, das zur Jetentstehung notwendig scheint, erzeugt wird. - Die Frage der Stabilität des asymptotischen Jets über weite Laengenskalen und die Rolle der Strahlungsprozesse. Die vorliegende Arbeit konzentriert sich auf die erste Frage. Der Versuch ihrer Beantwortung wird auf verschiedene Weise angestrebt. Zum einen durch Lösung der zeitunabhängigen Gleichungen, mit deren Hilfe das gesamte Jetentstehungsgebiet numerisch erfasst werden kann, zum anderen durch zeitabhängige Simulationen, die zwar nur einen Ausschnitt auflösen, dafür aber die zeitliche Entwicklung des Jets liefern koennen. Es werden relativistische und nicht-relativistische Lösungen diskutiert, Jets, die einen magnetisierten Stern im Ursprung haben und solche, wo dort ein schwarzes Loch existiert. Insgeamt sind grundlegenden Resultate aber allgemein gültig. Sie bestätigen die Vorstellung der magnetohydrodynamischen Entstehung astrophysikalischer Jets aus Akkretionsscheiben. N2 - Highly collimated, high velocity streams of hot plasma – the jets – are observed as a general phenomenon being found in a variety of astrophysical objects regarding their size and energy output. Known as jet sources are protostellar objects (T Tauri stars, embedded IR sources), galactic high energy sources ("microquasars"), and active galactic nuclei (extragalactic radio sources and quasars). Within the last two decades our knowledge regarding the processes involved in astro-physical jet formation has condensed in a kind of standard model. This is the scenario of a magnetohydrodynamically accelerated and collimated jet stream launched from the innermost part of an accretion disk close to the central object. Traditionally, the problem of jet formation is divided in two categories. One is the question how to collimate and accelerate an uncollimated low velocity disk wind into a jet. The second is the question how to initiate that outflow from a disk, i.e. how to turn accretion of matter into an ejection as a disk wind. My own work is mainly related to the first question, the collimation and acceleration process. Due to the complexity of both, the physical processes believed to be responsible for the jet launching and also the spatial configuration of the physical components of the jet source, the enigma of jet formation is not yet completely understood. On the theoretical side, there has been a substantial advancement during the last decade from purely station-ary models to time-dependent simulations lead by the vast increase of computer power. Observers, on the other hand, do not yet have the instruments at hand in order to spatially resolve observe the very jet origin. It can be expected that also the next years will yield a substantial improvement on both tracks of astrophysical research. Three-dimensional magnetohydrodynamic simu-lations will improve our understanding regarding the jet-disk interrelation and the time-dependent character of jet formation, the generation of the magnetic field in the jet source, and the interaction of the jet with the ambient medium. Another step will be the combina-tion of radiation transfer computations and magnetohydrodynamic simulations providing a direct link to the observations. At the same time, a new generation of telescopes (VLT, NGST) in combination with new instrumental techniques (IR-interferometry) will lead to a "quantum leap" in jet observation, as the resolution will then be sufficient in order to zoom into the innermost region of jet formation. In this paper we investigate how syntactic constraints and gender congruence interact to guide memory retrieval during the resolution of subject pronouns. Subject pronouns are always technically ambiguous, and the application of syntactic constraints on their interpretation depends on properties of the antecedent that is to be retrieved. While pronouns can freely corefer with non-quantified referential antecedents, linking a pronoun to a quantified antecedent is only possible in certain syntactic configurations via variable binding. We report the results from a judgment task and three online reading comprehension experiments investigating pronoun resolution with quantified and non-quantified antecedents. Results from both the judgment task and participants' eye movements during reading indicate that comprehenders freely allow pronouns to corefer with non-quantified antecedents, but that retrieval of quantified antecedents is restricted to specific syntactic environments. We interpret our findings as indicating that syntactic constraints constitute highly weighted cues to memory retrieval during anaphora resolution. The results show that both L1 and L2 speakers of English are sensitive to extraction islands during processing, suggesting that memory storage limitations affect L1 and L2 comprehenders in essentially the same way. Furthermore, these results show that the timing of island effects in L1 compared to L2 sentence comprehension is affected differently by the type of cue (semantic fit versus filled gaps) signaling whether dependency formation is possible at a potential gap site. Even though L1 English speakers showed immediate sensitivity to filled gaps but not to lack of semantic fit, proficient German-speaking learners of English as a L2 showed the opposite sensitivity pattern. This indicates that initial wh-dependency formation in L2 processing is based on semantic feature matching rather than being structurally mediated as in L1 comprehension.
