T1 - Products of in vitro oxidation of N-acetyldopamine as possible components in the sclerotization of insect cuticle N2 - [1-14C]-N-Acetyldopamine (NADA) was oxidized in the presence of methyl [3-3H]-β-alanate with mushroom tyrosinase. The complex mixture of reaction products was partly resolved by chromatographic procedures and analyzed by spectroscopic methods. Methyl-β-alanate is incorporated to only a small extent into oxidation products of NADA which inter alia are presumed to be oligomeric hydroxyquinones. After oxidation of [1-14C, 2-3H]-NADA with preparations from tanning Manduca sexta pupal cuticle, N-acetylnoradrenalin was identified as one of the products. Binding of radioactivity to melanin-like material was also observed. These results suggest that oxidation products different from those formulated usually for the crosslinkages between protein amino groups and N-acetyldopaquinone are deposited in darkly brown coloured insect cuticles during sclerotization. The enzyme catalyzed the penultimate step of juvenile hormone biosynthesis in the insects. Culturing corpora allata in the presence of sinefungin greatly suppressed juvenile hormone production. The following in vivo effects were visible after injection of the inhibitor: increase in mortality and reduction of total haemolymph protein liter and ovary fresh weight, as well as length of terminal oocytes. Attempts to reverse these effects by topical application of the juvenile hormone analog ZR-515 (methoprene) were only partly successful. Therefore, the in vivo effects may be due to a general inhibition of methyltransferase enzymes in the insect. Sinefungin appeared to be of potential interest as the first representative of a new class of insect growth regulators. The Kd value of the protein for racemic JH-III is 1.3 ± 0.2 × 10−7 M. It has a lower affinity for racemic JH-I and it does not bind JH-III-diol or JH-III-acid. The binding protein does discriminate between the enantiomers of synthetic, racemic JH-III as was determined by stereochemical anaysis of the bound and the free JH-III. Incubation of racemic JH-III with crude haemolymph results in preferential formation of (10S)-JH-III-acid, the unnatural configuration. The JH-esterase present in L. decemlineata haemolymph is not enantioselective. It is concluded that the most important function of the binding protein is that of a specific carrier, protecting the natural hormone against degradation by esterases. The carrier does not protect JH-I as efficiently as the lower homologue. When the spacer concept was used, efficient decoupling of the motions of the polymeric chain and the amphiphilic side groups is achieved. Thus, the often found loss of the important fluid phases by polymerization is avoided. Polymeric monolayers of the spacer lipid, prepared either by polymerization in the monolayer or by spreading of prepolymerized lipid, exhibit nearly identical surface pressure-area diagrams. Most distinctly, the successful decoupling of the motions of the polymer main chain and the membrane forming amphiphilic side groups is demonstrated by the self-organization of bulk polymerized spacer lipids to polymeric liposomes. In addition, spacer lipids are able to build polymeric Langmuir-Blodgett multilayers. The decoupling of the polymer main chain and the membrane-forming amphiphilic side groups enables the deposition of already polymeric monolayers onto supports to form defined multilayers. If, alternatively, monomeric monolayers are deposited and polymerized on the support, defects in the layers due to structural changes during the polymerization are avoided by the flexible spacer group. These lipid-like compounds with photoreactive units undergo self-organization to form monolayers at the gas-water interface and bilayer structures (vesicles) in aqueous solutions. The photoreaction of the cinnamic acid moiety induced by 254 nm UV light was investigated in the crystalline state, in monolayers, in vesicles and in solution in organic solvents. The single-chain amphiphiles undergo dimerization to yield photoproducts with twice the molecular weight of the corresponding monomers in organized systems. The photoreaction of amphiphiles containing two cinnamic acid groups occurs via two mechanisms: The intramolecular dimerization produces bicycles, with retention of the molecular weight of the corresponding monomer. The intermolecular reaction leads to oligomeric and polymeric photoproducts. The introduction of hydrophilic spacer groups into the polymeric amphiphiles allowed the formation of highly ordered systems (monolayers, liposomes, multilayers) from prepolymerized amphiphiles: due to the partial decoupling of the different mobilities and orientation tendencies of the polymer chain and the amphiphilic side groups, these polymers are able to self-organize. In monolayer experiments the high order of these membranes could be demonstrated by their surface pressure area-diagrams. In addition the combination of order and mobility of these spacer groups containing polymeric amphiphiles allowed the formation of Langmuir-Blodgett-multilyers with a high layer correlation. All amphiphiles investigated form monolayers. However, only acid and alcohol derivatives were able to build up multilayers. Those multilayers are rapidly photopolymerized in the layers via a two-step process: Irradiation with long-wavelength UV light yields soluble polymers, whereas additional irradiation with sfiort-wavelength UV light produces insoluble and presumably cross-linked polymers. The reaction meclianism is discussed according to the polymer characterization by UV spectroscopy, small-angle X-ray scattering, NMR spectroscopy, and gel permeation chromatography. All multilayers undergo structural changes during the polymerization; substantial changes result in defects in the polymerized layers as observed by scanning electron microscopy. In contrast to the acids and alcohols, the deposition of monolayers of the aldehyde derivatives did not yield well-ordered multilayers, but rather amorphous films. In this different film structure, the photopolymerization process differs from the one observed in multilayers. Als sekundäre Naturstoffe wirken sie häufig antibiotisch, cytotoxisch, aber auch pathogen, und eine Reihe von Pflanzen und Tieren benutzt chinoide Substanzen als Abwehrstoffe, oft mit spektakulärem Erfolg. Auf makromolekularer Ebene spielen Chinonmethide im Pflanzenreich eine Schlüsselrolle bei der Biosynthese von Lignin, während die Bildung von Melanoproteinen ein Beispiel für Reaktionen von o-Chinonen im Tierreich ist. Bei den Insekten dienen Chinone und Chinonmethide zur Bildung des lebensnotwendigen Exoskeletts. Die Reaktivität von Chinonen in biologischen Systemen hat auch für den Menschen unmittelbare Bedeutung in pharmazeutischer, toxikologischer und technologischer Hinsicht. Den Beispielen in diesem Aufsatz liegt ein gemeinsames Prinzip zugrunde, nämlich die chemische Modifikation von Biopolymeren durch Chinone und Chinonmethide. Wie sich besonders bei einer detaillierteren Betrachtung der Reaktionen zeigt, die zur Sklerotisierung der Insektencuticula führen, sind in den letzten Jahren wichtige neue Erkenntnisse hinzugekommen, die vor allem durch die modernen Methoden der Stofftrennung und der Festkörper-NMR-Spektroskopie ermöglicht worden sind. Coupling of (Ac4Gal)3-TRIS-Gly hydrogen succinate (3f) with Gly-O-Chol (5d) by means of EEDQ yields 6d. Monomers and polymers were investigated with respect to their aggregation behavior in aqueous systems and compared to analogous monomers and polymers lacking spacer groups. In the case of the monomeric surfactants, the spacer groups depress both the Kraffttemperature and the critical micelle concentration. the area occupied per molecule at the air-water interface is substantially enlarged by the spacers, whereas the depression of surface tension is nearly constant. Although the monomers with and without spacers are true surfactants, all the polymers are water-insoluble, but form monomolecular layers at the air-water interface. In analogy to the monomer behavior, the incorporation of the spacer groups increases the area occupied per repeat unit at the air-water interface substantially, but hardly affects the surface activity. Two such adducts, NAH-NADA-I and NAH NADA-II, have been isolated and their structures determined. In one of the adducts the link connecting the two residues occurs between the I-position (ß-position) in the NADA side chain and the 1-N atom (τ-N) in the imidazole ring of histidine. Diphenoloxidase activity alone is not sufficient for formation of this adduct, whereas extracts containing both diphenoloxidase and o-quinone-p-quinone methide isomerase activities catalyze the coupling reaction. The adduct consists of a mixture of two diastereomers and they are presumably formed by spontaneous reaction between enzymatically produced NADA-p-quinone methide and N-acetylhistidine. The other adduct has been identified as a ring addition product of N-acetylhistidine and NADA. In contrast to the former adduct it can be formed by incubation of the two substrates with mushroom tyrosinase alone. An adduct between N-acetylhistidine and the benzodioxan-type NADA-dimer is produced in vitro, when the N-acetylhistidine-NADA adduct is incubated with NADA and locust cuticle containing a 1,2-dehydro-NADA generating enzyme system. Trimeric NADA-polymerization products of the substituted benzodioxan-type have been obtained from in vivo sclerotized locust cuticle, confirming the ability of cuticle to produce NADA-oligomers. The results indicate that some insect cuticles contain enzymes promoting linkage of oxidized NADA to histidine residues. It is suggested that histidine residues in the cuticular proteins can serve as acceptors for oxidized NADA and that further addition of NADA-residues to the phenolic groups of bound NADA can occur, resulting in formation of protein-linked NADA-oligomers. The coupling reactions identified may be an important step in natural cuticular sclerotization. These polymer systems were complemented by alternating copolymers of appropriate zwitterionic vinyl compounds. Thus, polymers with reduced (as compared with simple vinylic homopolymers, or statistical copolymers) and well defined density of surfactant side groups are obtained. The solubilities found for these polymers are dominated by polymer geometry rather than by the balance of hydrophilic and hydrophobic fragments, thus corroborating a main-chain spacer model proposed recently. All water-soluble polymers exhibit characteristic features of classical polysoaps, as shown by surface tension measurements and by solubilization of hydrophobic dyes. In contrast, the water-insoluble copolymers are capable to form stable monolayers at the air-water interface. A1 - Kremer, Friedrich A1 - Köberle, Peter A1 - Laschewsky, André T1 - Relaxation and charge transport in mixtures of zwitterionic polymers and inorganic salts N2 - Dielectric spectroscopy is employed to analyze the molecular dynamics and the charge transport in mixtures of zwitterionic polymers of the type poly{3 [N(-methacryloyloxyalkyl)] N, [N-dimethylammonio propanesulfonate] with sodium iodide in the frequency range of 10²Hz-10(up)7 Hz and in the temperature range of 110 K-400 K. The amount of inorganic salt added varies from 0-200 mol-% relative to the number of zwitterionic groups present in the polymer, contributing strongly to the conductivity. One relaxation process is observed whose relaxation rate depends strongly on the length of the aliphatic spacer between the polymethacrylate main chain and the zwitterionic group. Exhibiting an Arrhenius-like temperature depence with activation energy EA = 47 KJ/mol, this relaxation process is assigned to fluctuation of the quaternary ammonium groups in the side chains. At higher temperatures, the dielectric properties and the conductivity are primarily dominated by the mobile inorganic ions: conductivity strongly depends on the salt concentration, showing a pronounced electrode polarization effect. The frequency and salt concentration, dependences of the conductivity can be quantitatively described as hopping of charge carriers being subject to spatially randomly varying energy barriers. For the low-frequency regime and for the critical frequency marking the onset of the conductivity's dispersion, the Barton-Nakajima-Namikawa (BNN) relationship is fulfilled. Es konnte gezeigt werden, daß auf diese Weise detaillierte Aussagen zu den Bindungsverhältnissen der Metallchelate getroffen werden können. Die Berechnung der Molekülstrukturen gelingt mit exzellenter Übereinstimmung mit den Kristallstrukturen der Komplexe. Die molekülmechanischen Berechnungen erfolgen auf der Grundlage des Extensible Systematic Force Field ESFF und des Consistent Force Field 91 (CFF91). Dabei konnte die hohe Flexibilität und Zuverlässigkeit des regelbasierten ESFF für eine Vielzahl verschiedenster Metallchelate nachgewiesen werden. Aufgrund der mangelhaften Ergebnisse für trigonal-prismatische Komplexgeometrien mit dem ESFF wurden eine Anpassung des CFF91 für derartige Vanadiumkomplexe vorgenommen. Auf Grundlage von theoretischen Ergebnissen wurden die alternativen Strukturen von isoelektronischen Vanadiumkomplexen berechnet und in Übereinstimmung mit experimentellen Daten, theoretischen Modellen der Komplexchemie und empirischen Fakten eine Hypothese für die Ursache der strukturellen Differenzen erarbeitet. Der hier vorgestellte, kombinierte Algorithmus aus kraftfeldbasierter Geometrieoptimierung und single-point-Rechnung an diesen Strukturen ist ein zuverlässiger und relativ schneller Weg Molekülgeometrien von Metallkomplexen zu berechnen. Er kann somit zur Voraussagen von Komplexstrukturen und zur gezielten Modellierung definierter Koordinationsgeometrien verwendet werden. N2 - In this work the molecular structures and the electronic properties of Vanadium(IV)- and Oxo-Rhenium(V)-chelates have been investigated to quantify steric and electronic effects of complexation. It has been shown, that in this way detailed insight can be gained into the bonding conditions of that metal complexes. Molecular mechanic calculations based on the Extensible Systematic Force Field (ESFF) and the Consistent Force Field 91 (CFF91) have been carried out. High flexibility and reliability of the rule based ESFF has been proven for a large variety of different metal chelates. Due to the poor ESFF-results for trigonal-prismatic complex geometries, a fit of the CFF91 for that species was done. Based on the theoretical results the alternative structure of isoelectronical vanadium(IV)- complexes have been calculated and a hypothesis on the reason for the structural differnces have been stated in accordance with experimental results, theoretical models of complex chemistry, and empirical facts. This combined approach of force field based geometry optimization and single point calculation at these structures has been proven to be a reliable and fast way to get molecular structures of metal complexes. Diese Parameter beeinflussten jedoch nicht den Grad der Deacetylierung (DD). Wichtig ist jedoch die Quelle des Ausgangsmaterials: Chitin aus Pandalus borealis ist ein guter Rohstoff für die Herstellung von Chitosan mit niedrigem DD und gleichzeitig hoher mittlerer Mv, während Krill-Chitin (Euphausia superba) ein gutes Ausgangsmaterial zur Herstellung von Chitosan mit hohem DD und niedrigem Mv ist. Chitosan, das aus Insekten (Calliphora erythrocephala), unter milden Bedingungen (Temperatur: 100°C, NaOH-Konzentration: 40 %, Zeit: 1-2h ) hergestellt wurde, hatte die gleichen Eigenschaften hinsichtlich DD und Mv wie das aus Krill hergestellte Chitosan. Der Bedarf an Zeit, Energie und NaOH ist für die Herstellung von Insekten-Chitosan geringer als für crabshell-Chitosan vergleichbare Resultaten für DD und Mv. 2. Chitosan wurde durch den Schimmelpilz Aspergillus fumigatus zu Chitooligomeren fermentiert. Die Ausbeute beträgt 25%. Die Chitooligomere wurden mit Hilfe von HPLC und MALDI-TOF-Massenspektrmetrie identifiziert. Die Fermentationsmischung fördert die Immunität von Pflanzen gegen Bakterien und Virusinfektion. Die Zunahme der Immunität schwankt jedoch je nach System Pflanze-Pathogen. Die Fermentation von Chitosan durch Aspergillus fumigatus könnte eine schnelle und billige Methode zur Herstellung von Chitooligomeren mit guter Reinheit und Ausbeute sein. Eine partiell aufgereinigte Fermentationsmischung dieser Art könnte in der Landwirtschaft als Pathogeninhibitor genutzt werden. Durch kontrollierte Fermentation, die Chitooligomere in definierter Zusammensetzung (d.h. definierter Verteilung des Depolymerisationsgrades) liefert, könnte man zu Mischungen kommen, die für die jeweilige Anwendung eine optimale Bioaktivität besitzen. 3. Die aus Chitosan-Dispersionen hergestellten MCChB-Filme weisen bessere mechanische Eigenschaften (Bruchfestigkeit, Dehnung) und eine höhere Wasseraufnahmefähigkeit auf als Filme, die nach herkömmlichen Methoden aus sauerer Lösung hergestellt werden. Die Einführung von Proteinen ändert die mechanischen Eigenschaften der MCChB-Filme abhängig von der Art, der Proteine sowie des DD und der Mv des eingesetzte Chitosan. Die Zugabe von Protein beschleunigt den biologischen Abbau der MCChB-Filme. Aus den untersuchten MCChB-Filmen mit Proteinzusatz können leichte, reißfeste und dennoch elastische Materialen hergestellt werden. 4. Mit Hilfe von MCChB-Dispersion kann Papier modifiziert werden. Dadurch werden die mechanischen Eigenschaften verbessert und die Wasseraufnahme wird verringert. Die Zugabe von Proteinen verringert das Wasseraufnahmevermögen noch weiter. Ein geringes Wasseraufnahmevermögen ist der bedeutendste Faktor bei der Papierherstellung. Auch Papier, das mit einem MCChB-Protein-Komplexe modifiziert wurde, zeigt gute mechanische Eigenschaften. 5. Wird Chitosan durch unmittelbare Einführung von MCChB auf Cellulose-Fasern aufgebracht, so erhält man eine netzartige Struktur, während durch Ausfällung aufgebrachtes Chitosan eine dünne Schicht auf den Cellulose-Fasern bildet. Die netzartige Struktur erleichtert die Bioabbaubarkeit, während die Schichtstruktur diese erschwert. 6. Die guten mechanischen Eigenschaften, die geringe Wasseraufnahmefähigkeit und die mit Cellulose vergleichbare Bioabbaubarkeit von Papier, das mit MCChB modifiziert wurde, lassen MCChB für die Veredlung von Papier nützlich erscheinen. N2 - 1. Deacetylation of the crustacean chitosan causes drastically decrease in the Mv with increasing reaction temperature and time as well as the concentration of sodium hydroxide. However, the DD are relatively less affected. Pandalus borealis is a good source for production of chitosan having high Mv and low DD, whereas chitosan of medium to low Mv can ideally be prepared using krill chitin. Insect chitosan is prepared under milder condition as compared with the crustacean chitosan, showed similar Mv and DD. Moreover, the consumption of time, energy and sodium hydroxide is much lower than for crustacean chitosan used. The properties of chitin (type of source, crystallinity, DD, Mv, swelling properties, particle size) affect the deacetylated polymer parameters. 2. Fermentation of chitosan using fungus Aspergillus fumigatus resulted in a composition of oligosaccharides with controlled molecular weight and yield at least 25 wt%. The product of fermentation effectively inhibited the viral and/or bacterial infection of the plant. This method can be an excellent, inexpensive system for preparation of bioactive agent. The preliminary purified fermentation mixture due to its antiviral and antibacterial behaviour is capable to be used as a natural, plant protection agent. The controlled degradation of chitosan connected with the production of various oligosaccharides having specified molecular weight allows obtaining the product with optimum bioactivity for suitable applications. 3. The films formed form microcrystalline chitosan (MCChB) gel-like dispersion demonstrate the better mechanical properties and higher swelling behaviour than typical films prepared using acidic solution of chitosan. The introduction of proteins significantly changes the mechanical strength and swelling behaviour. Addition of proteins causes the increase in their biodecomposition. The blended films containing proteins could be the base for formation of the resistant materials showed excellent elongation at break. 4. The application of MCChB in a paper formation as a modificator of the fibre-water interactions allows producing the paper sheets indicating the high increase in the mechanical properties and significant decrease in swelling properties. The introduction of MCChB with proteins causes a slight decrease in paper mechanical strength, if determined at low relative humidity. However, the mechanical strength measured at high relative humidity differ less than for paper sheet containing only MCChB. 5. Direct introduction of MCChB to a paper pulp forms the "web-like" structure of cellulose fibres and MCChB. The "web-like" structure of MCChB enables the faster biodecomposition of formed paper sheets. The precipitation of MCChB as wells as introduction of MCChB with proteins causes the "coat-like" structure. MCChB creates a thin layers coated the cellulose fibres lowering a biodecomposition rate. 6. The properties of paper sheets modified by MCChB such as: similar to cellulose biodegradation, excellent mechanical properties at rel. high humidity and the decrease in swelling properties as well as various possibilities to introduce MCChB allow to apple microcrystalline chitosan with or without proteins as the modificator of the fibre-water interactions in paper. Aufgrund der Anwendbarkeit zahlreicher analytischer Methoden sind schwimmende unlösliche Monoschichten als Modellsysteme sehr geeignet, um Ordnung und Strukturbildung sowie den Transport von Materie in zwei Dimensionen oder die Wechselwirkung von Molekülen an der Grenzfläche mit Teilchen in Lösung (Stichwort “molekulare Erkennung”) zu studieren. Aus dem Verhalten von Monoschichten lassen sich z. B. Rückschlüsse ziehen auf die Eigenschaften von Lipidschichten auf festen Substraten oder in biologischen Membranen. Diese Arbeit befasst sich mit spezifischen und fundamentalen Wechselwirkungen in Monoschichten sowohl auf molekularer als auch auf mikroskopischer Ebene und deren Beziehung zu Gitterstruktur, Aussehen und thermodynamischem Verhalten von Monoschichten an der Wasser/Luft Grenzfläche. Als Modellsystem werden hauptsächlich Monoschichten langkettiger Fettsäuren verwendet, da in ihnen die molekularen Wechselwirkungen durch Änderung des Subphasen-pH-Werts über den Dissoziationsgrad gezielt und schrittweise verändert werden können. Ausser über die Subphasenzusammensetzung werden die molekularen Wechselwirkungen auch über die Temperatur und die Monoschichtzusammensetzung systematisch variiert. Mit Hilfe von Isothermen- und Oberflächenpotentialmessungen, Brewsterwinkel-Mikroskopie, Röntgenbeugung unter streifendem Einfall und polarisationsmodulierter Infrarot-Reflexions-Absorptions-Spektroskopie wird die Änderung der Monoschichteigenschaften als Funktion eines äusseren Parametern analysiert. Dabei werden aus den Röntgenbeugungsdaten quantitative Masse für die molekularen Wechselwirkungen und für die Kettenkonformationsordnung in Monoschichten abgeleitet. Zu den interessantesten Ergebnissen dieser Arbeit zählen die Aufklärung des Ursprungs von regelmässigen polygonalen und dendritischen Domänenformen, die vielfältige Wirkung von Cholesterin auf die Molekülpackung und Gitterordnung langkettiger Amphiphile, sowie die Entdeckung einer abrupten Änderung in den Kopfgruppenbindungswechselwirkungen, der Kettenkonformationsordnung und des Phasenübergangsdrucks zwischen geneigten Monoschichtphasen in Fettsäuremonoschichten nahe pH 9. N2 - Research on monolayers of amphiphilic lipids on aqueous solution is of basic importance in surface science. Due to the applicability of a variety of surface sensitive techniques, floating insoluble monolayers are very suitable model systems for the study of order, structure formation and material transport in two dimensions or the interactions of molecules at the interface with ions or molecules in the bulk (headword 'molecular recognition'). From the behavior of monolayers conclusions can be drawn on the properties of lipid layers on solid substrates or in biological membranes. This work deals with specific and fundamental interactions in monolayers both on the molecular and on the microscopic scale and with their relation to the lattice structure, morphology and thermodynamic behavior of monolayers at the air-water interface. As model system especially monolayers of long chain fatty acids are used, since there the molecular interactions can be gradually adjusted by varying the degree of dissociation by means of the suphase pH value. For manipulating the molecular interactions besides the subphase composition also temperature and monolayer composition are systematically varied. The change in the monolayer properties as a function of an external parameter is analyzed by means of isotherm and surface potential measurements, Brewster-angle microscopy, X-ray diffraction at grazing incidence and polarization modulated infrared reflection absorption spectroscopy. For this a quantitative measure for the molecular interactions and for the chain conformational order is derived from the X-ray data. The most interesting results of this work are the elucidation of the origin of regular polygonal and dendritic domain shapes, the various effects of cholesterol on molecular packing and lattice order of long chain amphiphiles, as well as the detection of an abrupt change in the head group bonding interactions, the chain conformational order and the phase transition pressure between tilted phases in fatty acid monolayers near pH 9. For the interpretation of the latter point a model of the head group bonding structure in fatty acid monolayers as a function of the pH value is developed. N2 - This work describes the synthesis and characterization of linear and branched polypeptide block copolymers having amphiphilic character. The studies focused on the impact of the block copolymers' conformation and architecture on the supramolecular and colloidal properties. The polypeptide block copolymers were prepared in a multi-step process involving the anionic synthesis of (poly)amino-functional polymers (polystyrene and polybutadiene) which where used as macroinitiators for the ring-opening polymerization of N-carboxyanhydrides (NCA) of protected a-aminoacids. Supramolecular structures of the block copolymers in the solid state as well as in diluted aqueous solution were investigated using scattering (SAXS, WAXS, DLS) and microscopic (TEM) methods. Both linear and branched polypeptide block copolymers were used as polymeric stabilizers in the emulsion polymerization of styrene to yield polypeptide-decorated latexes. Biocompatible block copolymers were used as drug-delivery systems in lymphatic cancer therapy. Darüber hinaus wurden einige dieser Polymere mit einer geeigneten 'Lochtransport'-Funktionalität (Triphenylamin-Derivate) ausgestattet, um die Ladungstransporteigenschaften und das Molekulargewicht dieser Substanzen zu kontrollieren. Die thermische Orientierung dieser neuen Polymere auf geriebenen Polyimid-Schichten führte zu hoch anisotropen Filmen mit großen dichroischen Verhältnissen (Absorption parallel und senkrecht zur Reibungsrichtung gemessen). Ferner wurde eine Gruppe chiraler Polyfluorene synthetisiert und hinsichtlich ihrer chiroptischen Eigenschaften untersucht. Der mit diesen chiralen, konjugierten Polymeren erreichte Grad der Anisotropie in Absorption (CD), circular polarisierter Photolumineszenz (CPPL) und Elektrolumineszenz (CPEL) überstieg die bis dahin bekannten Werte um einen Faktor von 200. N2 - A series of 9,9-dialkyl-poly(fluorene-2,7-diyl)s containing linear and branched alkyl substituents with a molecular weight of up to 200000 g/mol has been synthesized and characterized. Moreover, some of the polymers were end capped with a suitable hole transport functionality, such as a triphenylamine derivative, to improve their charge transport properties and to control the molecular weight. The thermal alignment of these novel polymers on a rubbed polyimide layer led to highly anisotropic film formation with large dichroic ratios (absorption parallel and perpendicular to the rubbing direction). Additionally a set of chiral polyfluorenes has been synthesized and investigated with respect to their chiroptical properties. Die Darstellung der 1-Alkylcyclohexa-2,5-dien-1-carbonsäuren erfolgte mittels Birch-Reduktion in flüssigem Ammoniak und anschließender Umsetzung der intermediär entstehenden Dianionen mit Alkylhalogeniden. So konnte ausgehend von verschiedenen Benzoesäurederivaten eine Reihe interessanter Cyclohexadiene in sehr guten Ausbeuten synthetisiert werden. Erstmals gelangen säurekatalysierte Decarbonylierungen von Cyclohexadiencarbonsäuren, was die einfache Synthese substituierter Aromaten in ausgezeichneten Ausbeuten ermöglichte. In dieser Arbeit wird der Reaktionsmechanismus vorgestellt, welcher durch den Nachweis von Kohlenmonoxid in der Gasphase der Reaktionslösung durch IR-Spektroskopie untermauert wird. Bei der säurekatalysierten Umsetzung von 3-alkylsubstituierten Cyclohexadien-carbonsäuren entstanden neben den erwarteten Aromaten Lactone in ca. 50% Ausbeute. Schließlich zeigen die untersuchten Singulettsauerstoff-En-Reaktionen, der im ersten Teil dargestellten Cyclohexadiene und Lactone, durchweg hohe Regioselektivitäten und lieferten durch elektrostatische Wechselwirkungen und konformative Effekte zum Teil sehr gute Diastereoselektivitäten. Die auxiliarkontrollierte Photooxygenierungen von Cyclohexenon welches mit verschiedenen Weinsäureestern ketalisiert wurde, zeigten jedoch keine bevorzugte p-Facialität des Singulettsauerstoffs. N2 - In this dissertation thesis it was possible to extend the synthetical potential of cyclohexadienes and cyclohexenes with partially newly developed and very efficient synthetic pathways. The alkylcyclohexadiene carboxylic acids were synthesized in good yields via well known Birch-reduction procedure in liquid ammonia from a broad variety of substituted benzoic acid derivatives. A wide range of alkylhalides were subsequently added to the resulting dianion. We were able to decarbonylize the cyclohexadienes through acid catalyses, which leads to ipso-substituted aromatics in excellent yields. The study presents the postulated mechanism, which is supported by the proof of carbon monoxide in the gas phase of the reactionmixture with IR-spectroscopy. When 3-alkylsubstituted cyclohexadiene carboxylic acids were subjected to the acid catalyses they gave lactones in up to 50% yield as side products of the aromatization product.The cyclohexadienes and lactones were used as substrates in singlet oxygen-en-reactions with photochemically induced singulet oxygen. The products showed high regioselectivitiy and partially high diastereoselectivity which is caused by electrostatic interaction and conformativ effects of the reaction intermediates. Experiments to induce auxiliary controlled photooxygenations of cyclohexenones, which were ketalysed with different chiral tartaric acid esters leaded to poor diastereoselectivity and a diverse mixture of isomers. Using standard 2D emission and total luminescence spectra, fluorescence maxima, the width of the fluorescence band and a relative fluorescence quantum efficiency were determined. Different trends for fulvic acids and humic acids were observed indicating differences in the heterogeneity of the sample fractions. The complexity of the fluorescence decay of humic substances is discussed and compared to simple model compounds. The effect of oxidation of humic substances on their fluorescence properties is discussed as well.
