Parental plants which are two different inbred (pure) lines that have desired traits are crossed to obtain hybrids. Maximum heterosis is observed in the first generation (F1) of crosses. Heterosis has been utilised in plant and animal breeding programs for at least 90 years: by the end of the 21st century, 65% of worldwide maize production was hybrid-based. Generally, it is believed that an understanding of the molecular basis of heterosis will allow the creation of new superior genotypes which could either be used directly as F1 hybrids or form the basis for the future breeding selection programmes. Two selected accessions of a research model plant Arabidopsis thaliana (thale cress) were crossed to obtain hybrids. These typically exhibited a 60-80% increase of biomass when compared to the average weight of both parents. This PhD project focused on investigating the role of selected regulatory genes given their potentially key involvement in heterosis. In the first part of the project, the most appropriate developmental stage for this heterosis study was determined by metabolite level measurements and growth observations in parents and hybrids. At the selected stage, around 60 candidate regulatory genes (i.e. differentially expressed in hybrids when compared to parents) were identified. Of these, the majority were transcription factors, genes that coordinate the expression of other genes. Subsequent expression analyses of the candidate genes in biomass-heterotic hybrids of other Arabidopsis accessions revealed a differential expression in a gene subset, highlighting their relevance for heterosis. Moreover, a fraction of the candidate regulatory genes were found within DNA regions closely linked to the genes that underlie the biomass or growth heterosis. Additional analyses to validate the role of selected candidate regulatory genes in heterosis appeared insufficient to establish their role in heterosis. This uncovered a need for using novel approaches as discussed in the thesis. Taken together, the work provided an insight into studies on the molecular mechanisms underlying heterosis. Although studies on heterosis date back to more than one hundred years, this project as many others revealed that more investigations will be needed to uncover this phenomenon. N2 - „Heterosis“ ist ein in der Genetik und der Züchtung verwendeter Begriff, der die Hybridwüchsigkeit oder die Überlegenheit der Hybriden über ihre Eltern in Bezug auf Eigenschaften wie Größe, Wachstumsrate, Biomasse, Fruchtbarkeit, Ertrag, Nährstoffgehalt, Widerstand gegen Krankheiten oder Toleranz in Bezug auf biotischen oder abiotischen Stress bezeichnet. Um Hybriden zu erzeugen, werden aus zwei verschiedenen Inzuchtlinien (reine Linien) bestehende Elternpflanzen, welche die gewünschten Eigenschaften besitzen, miteinander gekreuzt. Der stärkste Heterosiseffekt wird in der ersten Kreuzungsgeneration (F1) beobachtet. Heterosis wird in Pflanzen- und Tierzuchtprogrammen schon seit mindestens 90 Jahren genutzt. So beruhte zum Ende des 20. Jahrhunderts 65% der weltweiten Maisproduktion auf Hybridzüchtung. Es wird angenommen, dass ein Verständnis der molekularen Grundlagen der Heterosis die Schaffung neuer, überlegener Genotypen erlaubt, die dann direkt als F1-Hybriden verwendet, oder als Grundlage für zukünftige Zucht- und Selektionsprogramme dienen können. Zwei ausgewählte Akzessionen der Modellpflanze Arabidopsis thaliana (Ackerschmalwand) wurden miteinander gekreuzt, um Hybriden zu erzeugen. Verglichen mit dem durchschnittlichen Gewicht ihren beiden Elternlinien zeigten diese eine 60-80%ige Zunahme an Biomasse. Diese Doktorarbeit befasst sich damit, die Rolle ausgewählter, regulatorischer Gene und ihre mögliche Schlüsselrolle bei der Heterosis zu untersuchen. Im ersten Teil der Arbeit wurde anhand der Gehaltsbestimmung ausgewählter Stoffwechselprodukte und Wachstumsbeobachtungen bei den Eltern und Hybriden das günstigste Entwicklungsstadium für diese Heterosisstudie bestimmt. In diesem Entwicklungsstadium wurden ungefähr 60 regulatorische Gene (d.h. Expressionsunterschiede zwischen Hybriden und Elternlinien) als Kandidaten identifiziert. Ein Großteil dieser Kandidaten waren Transkriptionsfaktoren, also Gene, die die Expression anderer Gene regulieren. Die nachfolgende Expressionsanalyse dieser Kandidatengene in Biomasse-Heterosis Hybriden anderer Arabidopsis Akzessionen zeigte bei einem Teil dieser Gene Expressionsunterschiede, die ihre Bedeutung bei der Heterosis betonen. Darüber hinaus wurde ein Teil dieser regulatorischen Kandidatengene innerhalb von DNS-Regionen gefunden, die eng mit Biomasse- oder Wachstumsheterosis in Verbindung stehen, und somit ihre Wichtigkeit in Bezug auf Heterosis unterstreichen. Weitergehende Analysen um die Rolle dieser ausgewählten regulatorischen Kandidatengene bei der Heterosis aufzuklären, waren nicht aussagekräftig genug, um ihre Rolle bei der Heterosis zu bestätigen. In der Doktorarbeit wird die Notwendigkeit neue Wege zur Aufklärung der Heterosis zu finden, diskutiert. Zusammenfassend gibt diese Doktorarbeit einen Einblick über Studien der molekularen Mechanismen, die der Heterosis zugrunde liegen. Diese Arbeit zeigt, dass obwohl Heterosis bereits seit mehr als hundert Jahren studiert wird, weitere Untersuchungen zur Aufklärung dieses Phänomens notwendig sind. Lignin is known as the most abundant aromatic polymer in nature and is isolated from the lignocellulosic component of plants by different possible extraction treatments. Both the biomass source and the extraction method affect the structure of the isolated lignin, therefore influencing its further application. Lignin was extracted from beech wood by two different hydrothermal alkaline treatments, which use NaOH and Ba(OH)2 as base and by an acid-catalyzed organosolv process. Moreover, lignin was isolated from bamboo, beech wood and coconut by soda treatment of the biomasses. A comparison of the structural features of such isolated lignins was performed through the use of a wide range of analytical methods. Alkaline lignins resulted in a better candidate as carbon precursor and macromonomers for the synthesis of polymer than organosolv lignin. In fact, alkaline lignins showed higher residual mass after carbonization and higher content of the reactive hydroxy functionalities. In contrast, the lignin source turned out to slightly affect the lignin hydroxyl content. One of the most common lignin modifications is its deconstruction to obtain aromatic molecules, which can be used as starting materials for the synthesis of fine chemicals. Lignin deconstruction leads to a complex mixture of aromatic molecules. A gas chromatographic analytical method was developed to characterize the mixture of products obtained by lignin deconstruction via heterogeneous catalytic hydrogenolysis. The analytical protocol allowed the quantification of three main groups of molecules by means of calibration curves, internal standard and a preliminary silylation step of the sample. The analytical method was used to study the influence of the hydrogenolysis catalyst, temperature and system (flow and batch reactor) on the yield and selectivity of the aromatic compounds. Lignin extracted from beech wood by a hydrothermal process using Ba(OH)2 as base, was functionalized by aromatic nitration in order to add nitrogen functionalities. The final goal was the synthesis of a nitrogen doped carbon. Nitrated lignin was reduced to the amino form in order to compare the influence of different nitrogen functionalities on the porosity of the final carbon. The carbons were obtained by ionothermal treatment of the precursors in the presence of the eutectic salt mixture KCl/ZnCl2 Such synthesized carbons showed micro-, macro- and mesoporosity and were tested for their electrocatalytic activity towards the oxygen reduction reaction. Mesoporous carbon derived from nitro lignin displayed the highest electrocatalytic activity. Lignins isolated from coconut, beech wood and bamboo were used as macromonomers for the synthesis of biobased polyesters. A condensation reaction was performed between lignin and a hyper branched poly(ester-amine), previously obtained by condensation of triethanolamine and adipic acid. The influence of the lignin source and content on the thermochemical and mechanical properties of the final material was investigated. The prepolymer showed adhesive properties towards aluminum and its shear strength was therefore measured. The gluing properties of such synthesized glues turned out to be independent from the lignin source but affected by the amount of lignin in the final material. This work shows that, although still at a laboratory scale, the valorization of lignin can overcome the critical issues of lignin´s structure variability and complexity. N2 - Das Thema dieser Arbeit ist Lignin als alternative Quelle für aromatische Moleküle einzusetzen, welche sonst aus fossilen Rohstoffen hergestellt werden. Lignin ist das am häufigsten vorkommende aromatische Polymer in der Natur. Es wird durch verschiedene Extraktionsmethoden aus pflanzlicher Lignocellulose isoliert. Sowohl der Rohstoff als auch das Extraktionsverfahren beeinflussen die Struktur des isolierten Lignins und bestimmen dadurch die weiteren Anwendungsmöglichkeiten. In der vorliegenden Arbeit die wurde Lignin durch zwei alkalisch hydrothermale Extraktionsmethoden aus Buchenholz extrahiert, wobei entweder NaOH oder Ba(OH)2 verwendet wurde. Eine dritte Methode bedient sich der sogenannten säuerekatalysierten Organosolv-Extraktion. Das Weiteren wurden drei Lignine durch eine Soda Extraktionsmethode aus Bambus, Buchenholz und Kokosnussschalen isoliert. Die Strukturen dieser wurden mit Hilfe verschiedener analytischer Techniken analysiert und verglichen. Während alkalische Lignine sich besser als Ausgangsstoff für poröse Kohlenstoffmaterialien und Makromonomere für die Synthese von Polymeren eignen, zeigen Lignine aus dem Organosolv Prozess weniger gute Eigenschaften hinsichtlich dieser Applikationen. Die bis heute häufigste Modifikation von Lignin beschäftigt sich mit der Aufschließung zu aromatischen Molekülen, welche die Grundlage für die weitere Herstellung von Chemikalien darstellen. Um die Aufschlussprodukte der Hydrogenolyse aus Lignin vergleichen zu können, wurde während dieser Arbeit wurde eine Gaschromatographiemethode entwickelt. In einem weiteren Schritt wurde isoliertes Lignin aus Buchenholzspänen durch Nitrierung mit zusätzlichem Stickstoff funktionalisiert. Das modifizierte Lignin konnte direkt al Präkursor für stickstoffdotierte kohlen dienen, welche mittels ionothermaler Synthese mit Hilfe von KCl/ZnCl2 als eutektischem Salz hergestellt wurden. Diese Materialien wurden erfolgreich in der Sauerstoffreduktion als Katalysator zum Einsatz gebracht. Die gewonnen Lignine aus Kokosnussschalen, Bambus und Buchenholz wurden als Makromonomere für die Herstellung von biobasiertem Polyester verwendet. Sowohl die thermochemischen als auch die mechanischen Eigenschaften wurden durch die unterschiedlichen Lignine der verschiedenen Pflanzen beeinflusst. Das hergestellte Polymer zeigte gute Eigenschaften als Biomass-basierter Kleber. Mit dieser Arbeit konnte gezeigt werden, dass Lignin, obwohl es eine sehr variable und komplexe Struktur aufweis, in verschiedenen Anwendungsbereichen zum Einsatz kommen kann, auch wenn diese bis heute nur im Labormaßstab verwirklicht wurden. Whether common mechanisms underpin the size structure of unicellular and multicellular autotrophs is, however, unknown. Using a global data compilation, we show that individual body masses in tree and phytoplankton communities follow power-law distributions and that the average exponents of these individual size distributions (ISD) differ. Phytoplankton communities are characterized by an average ISD exponent consistent with three-quarter-power scaling of metabolism with body mass and equivalence in energy use among mass classes. Tree communities deviate from this pattern in a manner consistent with equivalence in energy use among diameter size classes. Our findings suggest that whilst universal metabolic constraints ultimately underlie the emergent size structure of autotroph communities, divergent aspects of body size (volumetric vs. linear dimensions) shape the ecological outcome of metabolic scaling in forest vs. pelagic ecosystems. However, sustainable sources of energy, such as wind-, solar- and hydro-energy, produce primarily electrical energy and can not just be poured in canister like many fosil fuels, creating necessity for rechragable batteries. However, modern Li-ion batteries are made from toxic heavy metals and sustainable alternatives are needed. Here we show that naturally abundant catecholic and guaiacyl groups can be utilised to replace heavy metals in Li-ion batteries. Foremost vanillin, a naturally occurring food additive that can be sustainably synthesised from industrial biowaste, lignin, was utilised to synthesise materials that showed extraordinary performance as cathodes in Li-ion batteries. Furthermore, behaviour of catecholic and guiacyl groups in Li-ion system was compared, confirming usability of guiacayl containing biopolymers as cathodes in Li-ion batteries. Lastly, naturally occurring polyphenol, tannic acid, was incorporated in fully bioderived hybrid material that shows performance comparable to commercial Li-ion batteries and good stability. This thesis presents an important advancement in understanding of biowaste derived cathode materials for Li-ion batteries. Further research should be conducted to better understand behaviour of guaiacyl groups during Li-ion battery cycling. Lastly, challenges of incorporation of lignin, an industrial biowaste, have to be addressed and lignin should be incorporated as a cathode material in Li-ion batteries. N2 - Diese Dissertation untersucht, wie nachhaltige Kathoden (Kathodenmaterialien) für Lithium-Ionen-Batterien aus Holzabfällen hergestellt werden können. In den letzten Jahren hat die Menschheit erkannt, wie wenig nachhaltig unsere moderne Gesellschaft ist und große Mengen an Energie verbraucht, welche zum größten Teil aus fossilen Brennstoffen gewonnen werden. Daher versucht man jetzt die Energie aus hauptsächlich erneuerbaren Quellen wie Sonne und Wind zu gewinnen. Allerdings kann elektrische Energie nicht einfach wie Öl in einen Kanister gegossen werden, sondern muss in wieder aufladbaren Batterien gespeichert werden. In den letzten Jahren wurden Lithium-Ionen-Batterien entwickelt, die leistungsstark und allgegenwärtig sind, da sie zum Beispiel in Handys und sogar Autos Verwendung finden. Lithium-Ionen-Batterien verwenden jedoch Trägermaterialien aus giftigen Schwermetallen, die abgebaut werden müssen, was sich negativ auf die Umwelt auswirkt. In diesem Zusammenhang ist insbesondere das Schwermetall Kobalt zu erwähnen, welches in den meisten modernen Kathoden verwendet wird. Nach dem Bekanntwerden von Sklaverei und Kinderarbeit beim Kobaltabbau im Kongo, folgten große Kontroversen, da Kobalt praktisch in jedem Gerät führender Unternehmen wie zum Beispiel Apple und Microsoft zu finden ist. Idealerweise müssen wir von nicht erneuerbaren Schwermetallen zu erneuerbaren organischen Molekülen wechseln. Daher verwende ich in meiner Forschung Vanillin, ein Molekül, das hinsichtlich der Elektronenspeicherung ähnliche Eigenschaften wie Schwermetalle aufweist, jedoch viele Vorteile bietet. Erstens erkennt man Vanillin am spezifischen Geruch, da es einer der Hauptbestandteile von Vanille und daher ein natürlich vorkommendes Molekül ist. Zweitens kann es aus Holzabfällen oder aus Abfällen vieler Industrien hergestellt werden, die Holz als Rohstoff verwenden, wie beispielsweise der Papierindustrie. Durch milde chemische Reaktionen in Lösemitteln wie Wasser, Essig und Alkohol haben wir Vanillin zu einem Material modifiziert, welches hervorragende Eigenschaften zur Verwendung in Lithium-Ionen-Batterien hat und die bisher verwendeten Schwermetelle ersetzen kann. Diese Batterien wären somit erneuerbar und können uns der nachhaltigen Welt einen Schritt näher bringen. Darüber hinaus wurde Tanninsäure, ein natürlich vorkommendes Polymer in Holzrinde, verwendet, um vollständig aus Bioabfällen bestehende Batterien herzustellen. Yet, most candidates are not truly sustainable, i.e., not derived from renewable feedstock or made in benign reactions. Here an attempt is reported to resolve this issue by synthesizing an organic cathode material from tannic acid and microporous carbon derived from biomass. All constituents, including the redox-active material and conductive carbon additive, are made from renewable resources. Using a simple, sustainable fabrication method, a hybrid material is formed. The low cost and ecofriendly material shows outstanding performance with a capacity of 108 mAh g(-1) at 0.1 A g(-1) and low capacity fading, retaining approximately 80% of the maximum capacity after 90 cycles. With approximately 3.4 V versus Li+/Li, the cells also feature one of the highest reversible redox potentials reported for biomolecular cathodes. Finally, the quinone-catecholate redox mechanism responsible for the high capacity of tannic acid is confirmed by electrochemical characterization of a model compound similar to tannic acid but without catecholic groups. In the boreal zone, fires are an important factor in climate-vegetation interactions and biogeochemical cycles. Fire regimes are characterized by small, frequent, low-intensity fires within summergreen boreal forests dominated by larch, whereas evergreen boreal forests dominated by spruce and pine burn large areas less frequently but at higher intensities. Here, we explore the potential of the monosaccharide anhydrides (MA) levoglucosan, mannosan and galactosan to serve as proxies of low-intensity biomass burning in glacial-to-interglacial lake sediments from the high northern latitudes. We use sediments from Lake El'gygytgyn (cores PG 1351 and ICDP 5011-1), located in the far north-east of Russia, and study glacial and interglacial samples of the last 430 kyr (marine isotope stages 5e, 6, 7e, 8, 11c and 12) that had different climate and biome configurations. Combined with pollen and non-pollen palynomorph records from the same samples, we assess how far the modern relationships between fire, climate and vegetation persisted during the past, on orbital to centennial timescales. We find that MAs attached to particulates were well-preserved in up to 430 kyr old sediments with higher influxes from low-intensity biomass burning in interglacials compared to glacials. MA influxes significantly increase when summergreen boreal forest spreads closer to the lake, whereas they decrease when tundra-steppe environments and, especially, Sphagnum peatlands spread. This suggests that low-temperature fires are a typical characteristic of Siberian larch forests also on long timescales. The results also suggest that low-intensity fires would be reduced by vegetation shifts towards very dry environments due to reduced biomass availability, as well as by shifts towards peatlands, which limits fuel dryness. In addition, we observed very low MA ratios, which we interpret as high contributions of galactosan and mannosan from biomass sources other than those currently monitored, such as the moss-lichen mats in the understorey of the summergreen boreal forest. Overall, sedimentary MAs can provide a powerful proxy for fire regime reconstructions and extend our knowledge of long-term natural fire-climate-vegetation feedbacks in the high northern latitudes.
