They were collected within the northeastern margin of the Lepontine dome and easterly adjacent areas of the Swiss Central Alps. Three-dimensional mapping of isotemperature contours in map and profile views shows that the isotemperature contours associated with the Miocene Barrow-type Lepontine metamorphic event cut across refolded nappe contacts, both along and across strike within the northeastern margin of the Lepontine dome and adjacent areas. Further to the northeast, the isotemperature contours reflect temperatures reached during the Late Eocene subduction-related blueschist-facies event and/or during subsequent near-isothermal decompression; these contours appear folded by younger, large-scale post-nappe-stacking folds. A substantial jump in the recorded maximum temperatures across the tectonic contact between the frontal Adula nappe complex and surrounding metasediments indicates that this contact accommodated differential tectonic movement of the Adula nappe with respect to the enveloping Bundnerschiefer after maximum temperatures were reached within the northern Adula nappe, i.e. after Late Eocene time. It is an integral part of the East African Rift System and has an average elevation of approximately 1,000 m. Its location coincides with a negative Bouguer gravity anomaly with a semi-circular shape, closely related to a mantle plume, which influences the Cenozoic crustal development since its impingement in Eocene-Oligocene time. The uplift of the East African Plateau, preceding volcanism, and rifting formed an important orographic barrier and tectonically controlled environment, which is profoundly influenced by climate driven processes. Its location within the equatorial realm supports recently proposed hypotheses, that topographic changes in this region must be considered as the dominant forcing factor influencing atmospheric circulation patterns and rainfall distribution. The uplift of this region has therefore often been associated with fundamental climatic and environmental changes in East Africa and adjacent regions. While the far-reaching influence of the plateau uplift is widely accepted, the timing and the magnitude of the uplift are ambiguous and are still subject to ongoing discussion. This dilemma stems from the lack of datable, geomorphically meaningful reference horizons that could record surface uplift. In order to quantify the amount of plateau uplift and to find evidence for the existence of significant relief along the East African Plateau prior to rifting, I analyzed and modeled one of the longest terrestrial lava flows; the 300-km-long Yatta phonolite flow in Kenya. This lava flow is 13.5 Ma old and originated in the region that now corresponds to the eastern rift shoulders. The phonolitic flow utilized an old riverbed that once drained the eastern flank of the plateau. Due to differential erosion this lava flow now forms a positive relief above the parallel-flowing Athi River, which is mimicking the course of the paleo-river. My approach is a lava-flow modeling, based on an improved composition and temperature dependent method to parameterize the flow of an arbitrary lava in a rectangular-shaped channel. The essential growth pattern is described by a one-dimensional model, in which Newtonian rheological flow advance is governed by the development of viscosity and/or velocity in the internal parts of the lava-flow front. Comparing assessments of different magma compositions reveal that length-dominated, channelized lava flows are characterized by high effusion rates, rapid emplacement under approximately isothermal conditions, and laminar flow. By integrating the Yatta lava flow dimensions and the covered paleo-topography (slope angle) into the model, I was able to determine the pre-rift topography of the East African Plateau. The modeling results yield a pre-rift slope of at least 0.2°, suggesting that the lava flow must have originated at a minimum elevation of 1,400 m. Hence, high topography in the region of the present-day Kenya Rift must have existed by at least 13.5 Ma. This inferred mid-Miocene uplift coincides with the two-step expansion of grasslands, as well as important radiation and speciation events in tropical Africa. Accordingly, the combination of my results regarding the Yatta lava flow emplacement history, its location, and its morphologic character, validates it as a suitable “paleo-tiltmeter” and has thus to be considered as an important topographic and volcanic feature for the topographic evolution in East Africa. N2 - Das Ostafrikanische Plateau ist ein eindrucksvolles Beispiel für aktive, kontinentale Grabenbildung, aber auch für geodynamische Hochebenenbildung mit weitreichendem klimatischen Einfluss auf die gesamte Region. Als integraler Bestandteil des Ostafrikanischen Grabensystems beläuft sich die mittlere Höhe des Plateaus auf durchschnittlich 1000 m ü.NN. Seine Lage korreliert gut mit der Präsenz einer halbkreisförmigen negativen Bouguer-Schwereanomalie, die an den Aufstieg eines Manteldiapirs im Untergrund gekoppelt ist. Dieser prägte die känozoische Krustenentwicklung seit seinem Aufstieg im Eozän-Oligozän. Die Hebungsgeschichte und topographische Entwicklung des Hochlandes steht seither in enger Beziehung zum einsetzenden Vulkanismus, der Bildung erster Abschiebungssysteme und führte schließlich zur Entwicklung des heutigen Vollgrabensystems. Neueste Hypothesen lassen den Schluss zu, dass topographische Veränderungen als dominierende Einflussgrößen atmosphärischer Zirkulationsmuster sowie der regionalen Niederschlagsverbreitung anzusehen sind. Zusätzlich werden diese Prozesse durch die äquatoriale Lage des Ostafrikanischen Plateaus verstärkt und die Hebung dieser Region oft mit wichtigen Klima- und Umweltveränderungen in Ostafrika und angrenzende Gebiete in Verbindung gebracht. Während der weitreichende klimatische Einfluss des Hochlandes größtenteils akzeptiert ist, sind Zeitpunkt und Ausmaß seiner Heraushebung nicht eindeutig bestimmt und daher noch immer Grundlage bestehender Diskussionen. Diese Zwangslage hat ihre Ursache im Fehlen aussagekräftiger und datierbarer Referenzhorizonte. Um den Hebungsbetrag zu quantifizieren und Beweise signifikanten Reliefs vor der Entwicklung des Grabensystems entlang des Ostafrikanischen Hochlandes zu erbringen, analysierte und modellierte ich einen der längsten terrestrischen Lavaströme. Dieser vor 13,5 Ma abgelagerte Yatta-Lavastrom hat mit 300 km Länge seinen Ursprung in der Region der heutigen östlichen Grabenschulter des zentralen Kenia-Rifts. Die phonolitische Lava ergoss sich entlang eines Flussbettes, das einst die östliche Flanke des Hochlandes entwässerte. Aufgrund unterschiedlicher Erosionspotentiale bildet der Lavastrom gegenwärtig ein positives Relief und befindet sich oberhalb des Athi Flusses, der parallel zum Paläofluß, und somit versetzt zu seinen früheren Verlauf, strömt. Mein Ansatz der Lavastrom-Modellierung basiert auf einer Methode, die das Fließverhalten einer beliebigen Lava in Abhängigkeit von Temperatur und Magmenzusammensetzung in einem rechtwinkligen Kanal berechnet. Die wesentlichen Wachstumsmuster des Lavastroms sind durch ein eindimensionales Modell beschrieben, wobei Newtonsches Fließverhalten im Innern hinter der Lavastromfront von der zeitlichen Entwicklung der Viskosität und/oder der Fließgeschwindigkeit bestimmt wird. Vergleiche meiner Resultate mit verschiedenen Magmenzusammensetzungen zeigen, dass sich lange, kanalisierte Lavaströme mit hohen Ergussraten und schneller Platznahme bilden können. Dies geschieht unter annähernd isothermalen Bedingungen und erfordert laminares Fließen. Die Integration der Yatta- Lavastrom-Dimensionen und der bedeckten Paläotopographie (Neigungswinkel) in das Modell, erlaubte es mir die Topographie des Ostafrikanischen Hochlandes vor der Grabenbildung zu modellieren. Das Ergebnis der Modellierung ergibt einen Neigungswinkel von mindestens 0,2° und impliziert, dass der Lavastrom seinen Ursprung in einer Höhe von mindestens 1400 m ü.NN gehabt haben muss. Somit existierte bereits vor 13,5 Ma hohe Topographie in der heutigen Region des zentralen Kenia-Rifts. Diese abgeleitete regionale Hebungsgeschichte im mittleren Miozän korreliert mit der zweistufigen Ausbreitung der Graslandschaften, sowie dem Aufkommen neuer Arten im tropischen Afrika. Die Kombination aus Fließverhalten, Entstehungsort und morphologischer Eigenschaften macht den Yatta-Lavastrom zu einem “Paläoneigungsmesser” und wichtigen vulkanischen Untersuchungsobjekt für die topographische Entwicklung in Ostafrika. The absolute configuration of the isolated enantiomers could be unambiguously determined by comparison of calculated and measured circular dichroism (CD) spectra. Furthermore, we analyzed ring strain phenomena of (1,5)naphthalenophanes 10. Depending on the length of the linker units, one can distinguish three classes of naphthalenophanes. Compounds 10a-c are highly strained (E-STR = 7-31 kcal/mol), and the strain is caused by small bond angles in the linker unit and deformation of the naphthalene moiety. Another type of strain is observed if the linker unit becomes relatively long (10g,h) originating from transannular interactions and is comparable with the well-known strain of medium sized rings. The naphthalenophanes 10d-f with a linker length of 10-14 atoms are only marginally strained. To clearly discriminate the different sources of strain, we defined two geometrical parameters (average central dihedral angle delta(C) and naphthalene thickness D-N) and demonstrated that they are well-suited to indicate naphthalene deformation of our naphthalenophanes 10 as well as of ten model naphthalenophanes (I-X) with different linker lengths and linking positions. In the ischemic brain, levels of endothelin-1, one of the most potent vasoconstrictors, are raised. Anti-inflammatory and neuroprotective effects of endothelin antagonists after stroke have been described in literature. Based on these findings, we investigated the protective effect of the endothelin converting enzyme/neutral endopeptidase blocker, SLV 338, in salt-loaded, stroke-prone, spontaneously hypertensive rats. Methods: Male, 8-week-old spontaneously hypertensive stroke-prone rats were put on a high salt diet and treated with either 30 mg/kg or 100 mg/kg SLV 338 or vehicle for 27 weeks. Blood pressure, neurological outcome, body weight, and mortality were investigated throughout treatment. In weeks 1 and 9, animals were housed in metabolic cages for collection of urinary and blood samples and assessment of salt water and food intake. In weeks 22 and 27, additional blood samples were taken. At the end of the study, all brains were analyzed using magnetic resonance imaging. Results: SLV 338 was well tolerated in all animals. Neurological outcome and infarct size were similar in all groups. Albuminuria was considerably delayed and the incidence of stroke significantly lowered in treated animals. In spontaneously hypertensive stroke-prone rats, treatment with SLV 338 significantly (P=0.01) improved survival in comparison to the vehicle treated group in a blood pressure-independent manner. Discussion: Our data in spontaneously hypertensive stroke-prone rats demonstrate that combined endothelin converting enzyme/neutral endopeptidase inhibition could offer a new therapeutic approach for primary stroke prevention and improvement of mortality. The mechanism seems to be blood pressure-independent. In this work, the enzyme structure in solution (native state) was studied by use of different scattering methods. The results are compared with those from hydrodynamic model calculations based on the DFPase crystal structure. Bicontinuous microemulsions made of sugar surfactants are discussed as host systems for the DFPase. The microemulsion remains stable in the presence of the enzyme, which is shown by means of scattering experiments. Moreover, activity assays reveal that the DFPase still has high activity in this complex reaction medium. To complement the scattering experiments cryo-SEM was also employed to study the microemulsion structure. The successive conversion of such block copolymers from a hydrophilic into a hydrophobic state includes intermediate amphiphilic states with a variable hydrophilic-to-lipophilic balance. As a result, the self-organization is not following an all-or-none principle but a multistep aggregation in dilute solution was observed. The synthesis of double thermoresponsive diblock copolymers as well as triple thermoresponsive triblock copolymers was realized using twofold-TMS labeled RAFT agents which provide direct information about the average molar mass as well as residual end group functionality from a routine proton NMR spectrum. First a set of double thermosensitive diblock copolymers poly(N-n-propylacrylamide)-b-poly(N-ethylacrylamide) was synthesized which differed only in the relative size of the two blocks. Depending on the relative block lengths, different aggregation pathways were found. Furthermore, the complementary TMS-labeled end groups served as NMR-probes for the self-assembly of these diblock copolymers in dilute solution. Reversible, temperature sensitive peak splitting of the TMS-signals in NMR spectroscopy was indicative for the formation of mixed star-/flower-like micelles in some cases. Moreover, triple thermoresponsive triblock copolymers from poly(N-n-propylacrylamide) (A), poly(methoxydiethylene glycol acrylate) (B) and poly(N-ethylacrylamide) (C) were obtained from sequential RAFT polymerization in all possible block sequences (ABC, BAC, ACB). Their self-organization behavior in dilute aqueous solution was found to be rather complex and dependent on the positioning of the different blocks within the terpolymers. Especially the localization of the low-LCST block (A) had a large influence on the aggregation behavior. Above the first cloud point, aggregates were only observed when the A block was located at one terminus. Once placed in the middle, unimolecular micelles were observed which showed aggregation only above the second phase transition temperature of the B block. Carrier abilities of such triple thermosensitive triblock copolymers tested in fluorescence spectroscopy, using the solvatochromic dye Nile Red, suggested that the hydrophobic probe is less efficiently incorporated by the polymer with the BAC sequence as compared to ABC or ACB polymers above the first phase transition temperature. In addition, due to the problem of increasing loss of end group functionality during the subsequent polymerization steps, a novel concept for the one-step synthesis of multi thermoresponsive block copolymers was developed. This allowed to synthesize double thermoresponsive di- and triblock copolymers in a single polymerization step. The copolymerization of different N-substituted maleimides with a thermosensitive styrene derivative (4-vinylbenzyl methoxytetrakis(oxyethylene) ether) led to alternating copolymers with variable LCST. Consequently, an excess of this styrene-based monomer allowed the synthesis of double thermoresponsive tapered block copolymers in a single polymerization step. N2 - Die Selbstorganisation von mehrfach thermisch schaltbaren Blockcopolymeren in verdünnter wässriger Lösung wurde mittels Trübungsphotometer, dynamischer Lichtstreuung, TEM Messungen, NMR sowie Fluoreszenzspektroskopie untersucht. Die schrittweise Überführung eines hydrophilen in ein hydrophobes Blockcopolymer beinhaltet ein oder mehr amphiphile Zwischenstufen mit einstellbarem hydrophilen zu lipophilen Anteil (HLB). Dies führt dazu, dass die Selbstorganisation solcher Polymere in Lösung nicht nur einem Alles-oder-nichts-Prinzip folgt sondern ein mehrstufiges Aggregationsverhalten beobachtet wird. Die Synthese von doppelt thermisch schaltbaren Diblockcopolymeren und dreifach thermisch schaltbaren Triblockcopolymeren wurde durch sequenzielle RAFT Polymerisation realisiert. Dazu wurden zweifach TMS-markierte RAFT Agentien verwendet, welche die Bestimmung der molaren Masse sowie der verbliebenen Endgruppenfunktionalität direkt aus einem Protonen NMR Spektrum erlauben. Mit diesen RAFT Agentien wurde zunächst eine Serie von doppelt thermisch schaltbaren Diblockcopolymeren aus Poly(N-n-propylacrylamid)-b-Poly(N-ethylacrylamid), welche sich lediglich durch die relativen Blocklängen unterscheiden, hergestellt. In Abhängigkeit von der relativen Blocklänge wurde ein unterschiedliches Aggregationsverhalten der Diblockcopolymere in verdünnter wässriger Lösung beobachtet. Des Weiteren wirken die komplementär TMS-markierten Endgruppen als NMR-Sonden während der schrittweisen Aggregation dieser Polymere. Reversible, temperaturabhängige Peakaufspaltung der TMS-Signale in der NMR Spektroskopie spricht für eine Aggregation in gemischte stern-/blumenartige Mizellen, in denen ein Teil der hydrophoben Endgruppen in den hyrophoben Kern zurückfaltet. Obendrein wurden dreifach thermisch schaltbare Triblockcopolymere aus Poly(N-n-propylacrylamid) (A), Poly(methoxydiethylen glycol acrylat) (B) und Poly(N-ethylacrylamid) (C) in allen möglichen Blocksequenzen (ABC, BAC, ACB) durch schrittweisen Aufbau mittels RAFT Polymerisation erhalten. Das Aggregationsverhalten dieser Polymere in verdünnter wässriger Lösung war relativ komplex und hing stark von der Position der einzelnen Blöcke in den Triblockcopolymeren ab. Besonders die Position des Blocks mit der niedrigsten LCST (A) war ausschlaggebend für die resultierenden Aggregate. So wurde oberhalb der ersten Phasenübergangstemperatur nur Aggregation der Triblockcopolymere beobachtet, wenn der A Block an einem der beiden Enden der Polymere lokalisiert war. Wurde der A Block hingegen in der Mitte der Polymere positioniert, entstanden unimere Mizellen zwischen der ersten und zweiten Phasenübergangstemperatur, welche erst aggregierten, nachdem der zweite Block (B) seinen Phasenübergang durchlief. Die Transportereigenschaften dieser Triblockcopolymere wurden mittels Fluoreszenzspektroskopie getestet. Dazu wurde die Einlagerung eines hydrophoben, solvatochromen Fluoreszenzfarbstoffes, Nilrot, in Abhängigkeit der Temperatur untersucht. Im Gegensatz zu den Polymeren mit der Blocksequenz ABC oder ACB, zeigten die Polymere mit der Sequenz BAC eine verminderte Aufnahmefähigkeit des hydrophoben Farbstoffes oberhalb des ersten Phasenübergangs, was auf die fehlende Aggregation und die damit verbundenen relativ kleinen hydrophoben Domänen der unimolekularen Mizellen zwischen der ersten und zweiten Phasenübergangstemperatur zurückzuführen ist. Aufgrund des zunehmenden Verlustes von funktionellen Endgruppen während der RAFT Synthese von Triblockcopolymeren wurde ein neuartiges Konzept zur Einschrittsynthese von mehrfach schaltbaren Blockcopolymeren entwickelt. Dieses erlaubt die Synthese von mehrfach schaltbaren Diblock- und Triblockcopoylmeren in einem einzelnen Reaktionsschritt. Die Copolymeriation von verschiedenen N-substituierten Maleimiden mit einem thermisch schaltbaren Styrolderivat (4-Vinylbenzylmethoxytetrakis(oxyethylene) ether) ergab alternierende Copolymere mit variabler LCST. Bioinformatic analysis of the microviridin biosynthesis gene cluster suggests a novel type of processing machinery, which could rationalize the challenging in vivo/in vitro reconstitution of the pathway. In this work, we report the establishment of a minimal expression system for microviridins. Through bioinformatics and mutational analysis of the MdnA leader peptide we identified and characterized a strictly conserved binding motif that is specific for microviridin ligases. Furthermore, we showed that the ABC transporter MdnE is crucial for cyclization and processing of microviridins and demonstrated that MdnE is essential for stability of the microviridin biosynthesis complex. We performed laboratory experiments with flagellates and ciliates from three acidic mining lakes (AML, pH similar to 2.7) to investigate if similar habitats may affect similar organisms differently. Such man-made lakes provide suitable ecosystem models to test for the significance of strong habitat selection. To this end, we analyzed the growth response of three protist taxa (three strains of the phytoflagellate Chlamydomonas acidophila, two isolates of the phytoflagellate Ochromonas and two species of the ciliate genus Oxytricha) by exposing them to lake water of their origin and from the two other AML in a cross-factorial design. Population growth rates were measured as a proxy for their fitness. Results revealed significant effects of strain, lake (= habitat), and strain X habitat interaction. In the environmentally most adverse AML, all three protist taxa were locally adapted. In conclusion, our study demonstrates that (1) the same habitat may affect strains of the same species differently and that (2) similar habitats may harbor ecophysiologically different strains or species. Polymers of differing block sequences, ABC, BAC, and ACB, with increasing phase transition temperatures in the order A < B < C were prepared. Their aggregation behavior in dilute aqueous solution was investigated using dynamic light scattering, turbidimetry, and NMR spectroscopy. The self-organization of such polymers was found to dependent strongly on the block sequence. While polymers with a terminal low-LCST (lower critical solution temperature) block undergo aggregation above the first phase transition temperature at 20-25 degrees C, triblock copolymers with the low-LCST block in the middle show aggregation only above the second phase transition. The collapse of the middle block is not sufficient to induce aggregation but produces instead stable, unimolecular micelles with a collapsed middle block, as supported by NMR and fluorescence probe data. Continued heating of all copolymers led to two additional thermal transitions at 40-55 and 70-80 degrees C, which could be correlated to the phase transitions of the B and C blocks, respectively. All polymers show a high tendency for cluster formation, once aggregation is induced. The carrier abilities of the triple responsive triblock copolymers for hydrophobic agents were probed with the solvatochromic fluorescence dye Nile Red. With passing through the first thermal transition, the block copolymers are capable of solubilizing Nile Red. In the case of block copolymers with sequences ABC or ACB, which bear the low-LCST block at one terminus, notable amounts of dye are solubilized already at this stage. In contrast, the hydrophobic probe is much less efficiently incorporated by the BAC triblock copolymer, which forms unimolecular micelles. Only after the collapse of the B block, when reaching the second phase transition at about 45 degrees C, does aggregation occur and solubilization becomes efficient. Their temperature-dependent self-organisation in dilute aqueous solution was studied by turbidimetry, dynamic light scattering, transmission electron microscopy, and (1)H NMR spectroscopy. The copolymers show distinct, two-step self-organisation behaviour with respect to transition temperatures, aggregate type and size, which can be correlated to the relative lengths of the low and high LCST blocks. For polymers having short blocks with low LCST, the first thermal transition induces the formation of individual micelles. Further heating above the second thermal transition results reversibly either in a shrink of the micelle size or in aggregation of the micelles, with hydrodynamic diameters below 250 nm. In contrast in the case of polymers having a long block with low LCST, the first thermal transition already leads to clusters of micelles, while the second thermal transition makes the clusters shrink. Noteworthy, the twofold TMS-labeled end groups report not only on the molar masses of the polymers, but can simultaneously serve as NMR-probes for the self-assembly process. It is based on an error-controlled solution of the partial differential equation (PDE) that describes the evolution of the probability density function associated with the input uncertainty/variability. The density yields a more accurate estimate of the output uncertainty/variability, where not only some observables (such as mean and variance) but also structural properties (e.g., skewness, heavy tails, bi-modality) can be resolved up to a selected accuracy. For the adaptive solution of the PDE Cauchy problem we use the Rothe method with multiplicative error correction, which was originally developed for the solution of parabolic PDEs. We show that, unlike in parabolic problems, conservation properties necessitate a coupling of temporal and spatial accuracy to avoid accumulation of spatial approximation errors over time. We provide convergence conditions for the numerical scheme and suggest an implementation using approximate approximations for spatial discretization to efficiently resolve the coupling of temporal and spatial accuracy. The performance of the method is studied by means of low-dimensional case studies. The favorable properties of the spatial discretization technique suggest that this may be the starting point for an error-controlled sensitivity analysis in higher dimensions. The accompanying structural evolution can, in principle, be monitored by femtosecond stimulated Raman spectroscopy (FSRS). The method is used here to characterize the excited-state properties of riboflavin and flavin adenine dinucleotide in polar solvents. Raman modes are observed in the range 90-1800 cm(-1) for the electronic ground state S-0 and upon excitation to the S-1 state, and modes >1000 cm(-1) of both states are assigned with the help of quantum-chemical calculations. Line shapes are shown to depend sensitively on resonance conditions. They are affected by wavepacket motion in any of the participating electronic states, resulting in complex amplitude modulation of the stimulated Raman spectra. Wavepackets in S-1 can be marked, and thus isolated, by stimulated-emission pumping with the picosecond Raman pulses. Excited-state absorption spectra are obtained from a quantitative comparison of broadband transient fluorescence and absorption. In this way, the resonance conditions for FSRS are determined. Early differences of the emission spectrum depend on excess vibrational energy, and solvation is seen as dynamic Stokes shift of the emission band. In process and service management, consistency verification of behavioural models has manifold applications. For instance, a business process model used as system specification and a corresponding workflow model used as implementation have to be consistent. Another example would be the analysis to what degree a process log of executed business operations is consistent with the corresponding normative process model. Typically, existing notions of behaviour equivalence, such as bisimulation and trace equivalence, are applied as consistency notions. Still, these notions are exponential in computation and yield a Boolean result. In many cases, however, a quantification of behavioural deviation is needed along with concepts to isolate the source of deviation. In this article, we propose causal behavioural profiles as the basis for a consistency notion. These profiles capture essential behavioural information, such as order, exclusiveness, and causality between pairs of activities of a process model. Consistency based on these profiles is weaker than trace equivalence, but can be computed efficiently for a broad class of models. In this article, we introduce techniques for the computation of causal behavioural profiles using structural decomposition techniques for sound free-choice workflow systems if unstructured net fragments are acyclic or can be traced back to S-or T-nets. In order to judge on compliance of the business processing, the degree of behavioural deviation of a case, i.e., an observed execution sequence, is quantified with respect to a process model (referred to as fitness, or recall). Recently, different compliance measures have been proposed. Still, nearly all of them are grounded on state-based techniques and the trace equivalence criterion, in particular. As a consequence, these approaches have to deal with the state explosion problem. In this paper, we argue that a behavioural abstraction may be leveraged to measure the compliance of a process log - a collection of cases. To this end, we utilise causal behavioural profiles that capture the behavioural characteristics of process models and cases, and can be computed efficiently. We propose different compliance measures based on these profiles, discuss the impact of noise in process logs on our measures, and show how diagnostic information on non-compliance is derived. As a validation, we report on findings of applying our approach in a case study with an international service provider. Checking the consistency of such corresponding models is a major challenge for process modeling theory and practice. In this paper, we take the inappropriateness of existing strict notions of behavioral equivalence as a starting point. Our contribution is a concept called behavioral profile that captures the essential behavioral constraints of a process model. We show that these profiles can be computed efficiently, i.e., in cubic time for sound free-choice Petri nets w.r.t. their number of places and transitions. We use behavioral profiles for the definition of a formal notion of consistency which is less sensitive to model projections than common criteria of behavioral equivalence and allows for quantifying deviation in a metric way. The derivation of behavioral profiles and the calculation of a degree of consistency have been implemented to demonstrate the applicability of our approach. Conceptual models are of central importance for BPM. Most prominently, process models define the behaviour that is performed to achieve a business value. In essence, a process model is a mapping of properties of the original business process to the model, created for a purpose. Different modelling purposes, therefore, result in different models of a business process. Against this background, the misalignment of process models often observed in the field of BPM is no surprise. Even if the same business scenario is considered, models created for strategic decision making differ in content significantly from models created for process automation. Despite their differences, process models that refer to the same business process should be consistent, i.e., free of contradictions. Apparently, there is a trade-off between strictness of a notion of consistency and appropriateness of process models serving different purposes. Existing work on consistency analysis builds upon behaviour equivalences and hierarchical refinements between process models. Hence, these approaches are computationally hard and do not offer the flexibility to gradually relax consistency requirements towards a certain setting. This thesis presents a framework for the analysis of behaviour consistency that takes a fundamentally different approach. As a first step, an alignment between corresponding elements of related process models is constructed. Then, this thesis conducts behavioural analysis grounded on a relational abstraction of the behaviour of a process model, its behavioural profile. Different variants of these profiles are proposed, along with efficient computation techniques for a broad class of process models. Using behavioural profiles, consistency of an alignment between process models is judged by different notions and measures. The consistency measures are also adjusted to assess conformance of process logs that capture the observed execution of a process. Further, this thesis proposes various complementary techniques to support consistency management. It elaborates on how to implement consistent change propagation between process models, addresses the exploration of behavioural commonalities and differences, and proposes a model synthesis for behavioural profiles. Die Striktheit des Konsistenzbegriffs steht hierbei in Konflikt mit der Eignung der Prozessmodelle für einen bestimmten Zweck. Existierende Ansätze zur Analyse von Verhaltenskonsistenz basieren auf Verhaltensäquivalenzen und nehmen an, dass Prozessmodelle in einer hierarchischen Verfeinerungsrelation stehen. Folglich weisen sie eine hohe Berechnungskomplexität auf und erlauben es nicht, den Konsistenzbegriff graduell für einen bestimmten Anwendungsfalls anzupassen. Die vorliegende Arbeit stellt einen Ansatz für die Analyse von Verhaltenskonsistenz vor, welcher sich fundamental von existierenden Arbeiten unterscheidet. Zunächst werden korrespondierende Elemente von Prozessmodellen, welche den gleichen Geschäftsprozess darstellen, identifiziert. Auf Basis dieser Korrespondenzen wird ein Ansatz zur Konsistenzanalyse vorgestellt. Jener basiert auf einer relationalen Verhaltensabstraktion, dem Verhaltensprofil eines Prozessmodells. Die Arbeit führt verschiedene Varianten dieses Profils ein und zeigt wie sie für bestimmte Modellklassen effizient berechnet werden. Mithilfe von Verhaltensprofilen werden Konsistenzbegriffe und Konsistenzmaße für die Beurteilung von Korrespondenzen zwischen Prozessmodellen definiert. Weiterhin werden die Konsistenzmaße auch für den Anwendungsfall der Konformität angepasst, welcher sich auf beobachtete Abläufe in Form von Ausführungsdaten bezieht. Darüber hinaus stellt die Arbeit eine Reihe von Methoden vor, welche die Analyse von Verhaltenskonsistenz ergänzen. They are depleted in LILE elements compared to MORB, have a negative Nb-anomaly and flat to enriched REE patterns, suggesting that their protoliths evolved in a subduction related tectonic setting. They show island-arc affinities and are similar to primitive island-arc rocks described in the Caribbean. The geochemical characteristics are comparable to low-grade greenschists from the nearby Etpana Terrane, which are interpreted as part of a Cretaceous intra-oceanic arc. These data support evidence that the eclogites and the Etpana terrane rocks formed from the same volcano-sedimentary sequence. Part of this sequence was accreted onto the margin and another was incorporated into the subduction channel and metamorphosed at eclogite facies conditions. Ar-40-Ar-39 ages of 79.2 +/- 1.1Ma and 82.2 +/- 2.5Ma determined on white micas, separated from two eclogite samples, are interpreted to be related to the cooling of the main metamorphic event. The formation of a common volcano-sedimentary protolith and subsequent metamorphism of these units record the ongoing Late Cretaceous continental subduction of the South American margin within the Caribbean intra-oceanic arc subduction zone. This gave way to an arc-continent collision between the Caribbean and the South American plates, where this sequence was exhumed after the Campanian. Anecdotal observations suggest that they disperse also by rhizome fragments. For testing their resprouting ability, rhizome fragments of different sizes from both species were buried at four different soil depths (0, 5, 10 and 20 cm, respectively) in a common garden. Rhizome fragments of S. canadensis ranged 3-6 cm in length, those of S. gigantea 5-20 cm in length. Resprouting plants were harvested after 3 months and growth related traits measured to assess their vitality. Resprouting rate in S. gigantea was far higher than in S. canadensis (85 and 19%, respectively). In S. gigantea, fragments of all sizes resprouted from all soil depths whereas none rhizome of S. canadensis emerged from 20 cm burial depth. In S. gigantea, size related traits showed significant interactions between fragment size and burial depth, but not relative shoot growth rate. At all burial depths, vitality of plants emerging from small rhizomes was lower than plants emerging from large rhizomes. Effects of rhizome size became stronger with increasing burial depth. The results show that both species are able to resprout from buried rhizome fragments, and that succesful regeneration is more likely to occur in S. gigantea. Managing these species should avoid any activities promoting rhizome fragmentation and dispersal of fragments. Our results show that both plant and reptile species richness as well as plant cover and reptile abundance was significantly higher in the protected KTP than in the non-protected farmland. The higher proportion of shrub but lower proportions of perennial grass cover, herb cover, and herb species richness in the farmland can be explained by higher stocking rates and the differences in feeding behaviour between native wild ungulates (e.g. Antidorcas marsupialis, Oryx gazella) and livestock (mainly sheep). The reptile's prey availability and microhabitats (perennial grass tussocks and rodent burrows) for thermoregulation and protection against predators were significantly lower in the farmland. To conclude, our results clearly show that long term effects of different land use options (wildlife in protected KTP versus extensive livestock production in the non-protected farmland) even within the same habitat type have led to significant changes in vegetation composition, availability of microhabitat structures and in the reptile community. However, since the relative proportions of fossil pollen taxa are largely influenced by individual pollen productivities and the dispersal characteristics, such inferences on vegetation have the potential to be considerably biased. We therefore examined the modern pollen-vegetation relationships for four common pollen species on the Tibetan Plateau, using Extended R-value (ERV) models. Assuming an average radius of 100 m for the sampled lakes, we estimated the relevant source area of pollen (RSAP) to be 2200 m (which represents the distance from the lake). Using Poaceae as the reference taxa (Pollen Productivity Estimate, PPE = 1), ERV Submodel 2 derived relative high PPEs for the steppe and desert taxa: 2.079 +/- 0.432 for Artemisia and 5.379 +/- 1.077 for Chenopodiaceae. Low PPEs were estimated for the Cyperaceae (1.036 +/- 0.012). whose plants are characteristic of the alpine Kobresia meadows. Applying these PPEs to four fossil pollen sequences since the Late Glacial, the plant abundances on the central and north-eastern Tibetan Plateau were quantified using the "Regional Estimates of Vegetation Abundance from Large Sites" (REVEALS) model. The proportions of Artemisia and Chenopodiaceae were greatly reduced compared to their original pollen percentages in the reconstructed vegetation, owing to their high productivities and their dispersal characteristics, while Cyperaceae showed a relative increase in the vegetation reconstruction. The reconstructed vegetation assemblages of the four pollen sequence sites always yielded smaller compositional species turnovers than suggested by the pollen spectra, as revealed by Detrended Canonical Correspondence Analyses (DCCA) of the Holocene sections. The strength of the previously reported vegetation changes may therefore have been overestimated, which indicates the importance of taking into account pollen-vegetation relationships when discussing the potential drivers (such as climate, land use, atmospheric CO(2) concentrations) and implications (such as for land surface-climate feedbacks, carbon storage, and biodiversity) of vegetation change. As a major influence on the climate in this region, the Asian Summer Monsoon (ASM) and its evolutionary history are of vital importance for accurate predictions. However, neither the evolutionary pattern of the summer monsoon nor the driving mechanisms behind it are yet clearly understood. For this research, I first synthesized previously published Late Glacial to Holocene climatic records from monsoonal central Asia in order to extract the general climate signals and the associated summer monsoon intensities. New climate and vegetation sequences were then established using improved quantitative methods, focusing on fossil pollen records recovered from Tibetan lakes and also incorporating new modern datasets. The pollen-vegetation and vegetation-climate relationships on the TP were also evaluated in order to achieve a better understanding of fossil pollen records. The synthesis of previously published moisture-related palaeoclimate records in monsoonal central Asia revealed generally different temporal patterns for the two monsoonal subsystems, i.e. the Indian Summer Monsoon (ISM) and East Asian Summer Monsoon (EASM). The ISM appears to have experienced maximum wet conditions during the early Holocene, while many records from the area affected by the EASM indicate relatively dry conditions at that time, particularly in north-central China where the maximum moisture levels occurred during the middle Holocene. A detailed consideration of possible driving factors affecting the summer monsoon, including summer solar insolation and sea surface temperatures, revealed that the ISM was primarily driven by variations in northern hemisphere solar insolation, and that the EASM may have been constrained by the ISM resulting in asynchronous patterns of evolution for these two subsystems. This hypothesis is further supported by modern monsoon indices estimated using the NCEP/NCAR Reanalysis data from the last 50 years, which indicate a significant negative correlation between the two summer monsoon subsystems. By analogy with the early Holocene, intensification of the ISM during coming decades could lead to increased aridification elsewhere as a result of the asynchronous nature of the monsoon subsystems, as can already be observed in the meteorological data from the last 15 years. A quantitative climate reconstruction using fossil pollen records was achieved through analysis of sediment core recovered from Lake Donggi Cona (in the north-eastern part of the TP) which has been dated back to the Last Glacial Maximum (LGM). A new data-set of modern pollen collected from large lakes in arid to semi-arid regions of central Asia is also presented herein. The concept of "pollen source area" was introduced to modern climate calibration based on pollen from large lakes, and was applied to the fossil pollen sequence from Lake Donggi Cona. Extremely dry conditions were found to have dominated the LGM, and a subsequent gradually increasing trend in moisture during the Late Glacial period was terminated by an abrupt reversion to a dry phase that lasted for about 1000 years and coincided with the first Heinrich Event of the northern Atlantic region. Subsequent periods corresponding to the warm Bølling-Allerød period and the Younger Dryas cold event were followed by moist conditions during the early Holocene, with annual precipitation of up to about 400 mm. A slightly drier trend after 9 cal ka BP was then followed by a second wet phase during the middle Holocene that lasted until 4.5 cal ka BP. Relatively steady conditions with only slight fluctuations then dominated the late Holocene, resulting in the present climatic conditions. In order to investigate the relationship between vegetation and climate, temporal variations in the possible driving factors for vegetation change on the northern TP were examined using a high resolution late Holocene pollen record from Lake Kusai. Moving-window Redundancy Analyses (RDAs) were used to evaluate the correlations between pollen assemblages and individual sedimentary proxies. These analyses have revealed frequent fluctuations in the relative abundances of alpine steppe and alpine desert components, and in particular a decrease in the total vegetation cover at around 1500 cal a BP. The climate was found to have had an important influence on vegetation changes when conditions were relatively wet and stable. However, after the 1500 cal a BP threshold in vegetation cover was crossed the vegetation appears to have been affected more by extreme events such as dust storms or fluvial erosion than by the general climatic trends. In addition, pollen spectra over the last 600 years have been revealed by Procrustes analysis to be significantly different from those recovered from older samples, which is attributed to an increased human impact that resulted in unprecedented changes to the composition of the vegetation. Theoretical models that have been developed and widely applied to the European area (i.e. the Extended R-Value (ERV) model and the Regional Estimates of Vegetation Abundance from Large Sites (REVEALS) model) have been applied to the high alpine TP ecosystems in order to investigate the pollen-vegetation relationships, as well as for quantitative reconstructions of vegetation abundance. The modern pollen–vegetation relationships for four common pollen species on the TP have been investigated using Poaceae as the reference taxa. The ERV Submodel 2 yielded relatively high PPEs for the steppe and desert taxa (Artemisia Chenopodiaceae), and low PPEs for the Cyperaceae that are characteristic of the alpine Kobresia meadows. The plant abundances on the central and north-eastern TP were quantified by applying these PPEs to four post-Late Glacial fossil pollen sequences. The reconstructed vegetation assemblages for the four pollen sequences always yielded smaller compositional species turnovers than suggested by the pollen spectra, indicating that the strength of the previously-reported vegetation changes may therefore have been overestimated. In summary, the key findings of this thesis are that (a) the two ASM subsystems show asynchronous patterns during both the Holocene and modern time periods, (b) fossil pollen records from large lakes reflect regional signals for which the pollen source areas need to be taken into account, (c) climate is not always the main driver for vegetation change, and (d) previously reported vegetation changes on the TP may have been overestimated because they ignored inter-species variations in pollen productivity. Dies gilt insbesondere für das Holozän, für welches große Kimaschwankungen und regionale Diskrepanzen weithin belegt sind. Deshalb habe ich zuerst holozäne Klimadaten zusammengefasst. Bereits veröffentlichte Publikationen aus den Monsungebieten Zentralasiens dienten als Grundlage, um die wichtigsten Klimasignale und die zugehörigen Intensitäten des Sommermonsuns heraus zu arbeiten. Anhand von Pollensequenzen aus tibetischen Seen erzeugte ich neue Klima- und Vegetationssequenzen, welche auf verbesserten quantitativen Methoden und rezenten Datensätzen beruhen. Außerdem wurden die Verhältnisse Pollen-Vegetation und Vegetation-Klima bewertet, um Schlussfolgerungen fossiler Pollensequenzen zu verbessern. Die Zusammenfassung der zuvor veröffentlichten, niederschlagsbezogenen Paläoklimadaten im Monsungebiet Zentralasiens ergab generell unterschiedliche Muster für die zwei Teilsysteme des ASMs, den Indischen Sommermonsun (ISM) und den Ostasiatischen Sommermonsun (OASM). Der ISM weist maximale feuchte Bedingungen während des frühen Holozöns auf, während viele Datensätze aus dem Gebiet des OASMs einen relativ trockenen Zustand anzeigen, besonders im nördlichen Zentralchina, wo maximale Niederschläge während des mittleren Holozäns registriert wurden. Genaue Betrachtungen der Antriebsfaktoren des Sommermonsuns ergaben, dass der ISM hauptsächlich durch Veränderungen der Sonneneinstrahlung auf der Nordhemisphäre angetrieben wird, während der OASM potentiell durch den ISM beherrscht wird - dies führt zu asynchronen Entwicklungen. Diese Hypothese wird durch rezente Monsunindizes gestützt. Sie weisen eine signifikant negative Korrelation zwischen den beiden Sommermonsun-Teilsystemen auf. Für die quantitative Klimarekonstruktion von Pollensequenzen wurde ein Sedimentkern aus dem See Donggi Cona im Nordosten der HT analysiert, der bis zum letzten glazialen Maximum (LGM) zurückdatiert wurde. Aufgrund der Tatsache, dass Donggi Cona ein relativ großer See ist, wird hiermit ein neuer Pollen-Klima-Kalibrierungsdatensatz auf Grundlage großer Seen in ariden und semiariden Regionen Zentralasiens vorgelegt. Das Konzept des Pollenherkunftsgebietes wurde in diese rezente, pollenbasierte Klimakalibrierung eingebracht und auf die Pollensequenz von Donggi Cona angewendet. Die Auswertung ergab, dass extrem trockene Bedingungen während des LGM (ca. 100 mm/yr) vorherrschten. Ein ansteigender Trend von Niederschlägen während des späten Glazials wurde durch einen abrupten Rückgang zu einer etwa 1000-jährigen Trockenphase beendet, welche mit Heinrich-Ereignis 1 in der Nordatlantik-Region übereinstimmt. Danach entsprechen die Klimaperioden dem warmen Bølling/Allerød und dem Kälteereignis der Jüngeren Dryas. Anschließend herrschten feuchte Bedingungen im frühen Holozän (bis zu 400 mm/yr). Ein etwas trockenerer Trend nach dem Holozänen Klimaoptimum wurde dann von einer zweiten Feuchtphase abgelöst, welche bis 4,5 cal. ka vor heute andauerte. Relativ gleichmäßige Bedingungen dominierten das späte Holozän bis heute. Die Klimadynamik seit dem LGM wurde vor allem durch Entgletscherung und Intensitätsschwankungen des ASM bestimmt. Bei der Betrachtung des Vegetation-Klima-Verhältnisses habe ich die zeitlichen Variationen der bestimmenden Faktoren hinsichtlich der Vegetationsdynamik auf der nördlichen HT untersucht. Dabei wurden hochauflösende holozäne Pollendaten des Kusai-Sees verwendet. Eine Redundanzanalyse (RDA) wurde angewendet um die Korrelation zwischen Pollenvergesellschaftungen und individuellen sedimentären Klimaanzeigern als auch die damit verbundene Signifikanz zu bewerten. Es stellte sich heraus, dass das Klima einen wichtigen Einfluss auf den Veränderungen in der Vegetation besaß, wenn die Bedingungen relativ warm und feucht waren. Trotzdem scheint es, dass, dass die Vegetation bei zu geringer Bedeckung stärker durch Extremereignisse wie Staubstürme oder fluviale Erosion beeinflusst wurde. Pollenspektren der vergangen 600 Jahre erwiesen sich als signifikant unterschiedlich verglichen mit den älterer Proben, was auf verstärkten anthropogenen Einfluss hindeutet. Dieser resultierte in einem beispiellosen Wandel in der Zusammensetzung der Vegetation. In Hinsicht auf das Pollen-Vegetation-Verhältnis und der quantitativen Rekonstruktion der Vegetationshäufigkeit habe ich theoretische Modelle, welche für europäische Regionen entwickelt und weithin angewendet wurden, respektive die Modelle "Extended R-Value" (ERV) sowie "Regional Estimates of Vegetation Abundance from Large Sites" (REVEALS), auf die hochalpinen Ökosysteme der HT überführt. Dafür wurden rezente Pollen-Vegetations-Verhältnisse von vier weit verbreiteten Pollen-Arten der HT überprüft. Poaceae wurden als Referenztaxa verwendet. Bei der Anwendung dieser Verhältnisse auf vier Pollensequenzen, welche die Paläoumweltbedingungen seit dem letzten Glazial widerspiegeln, wurden die Häufigkeiten von Pflanzen auf der zentralen und nordöstlichen HT quantifiziert. Anteile von Artimisia und Chenopodiaceae waren dabei im Vergleich zu ihren ursprünglichen Pollenprozenten deutlich verringert. Cyperaceae hingegen wies eine relative Zunahme in dieser Vegetationsrekonstruktion auf. Die rekonstruierten Vegetationsvergesellschaftungen an den Standorten der vier Pollensequenzen ergaben stets geringere Umwälzungen in der Artenzusammensetzung, als durch die Pollenspektren zu vermuten gewesen wäre. Dies kann ein Hinweis darauf sein, dass die Intensität der bislang angenommenen Vegetationsveränderungen überschätzt worden ist. Zusammengefasst sind die Hauptresultate dieser Dissertation, dass (a) die zwei ASM Teilsysteme asynchrone Muster während des Holozäns und heute aufweisen, dass (b) fossile Pollensequenzen großer Seen regionale Klimasignale widerspiegeln sofern die Herkunftsgebiete der Pollen berücksichtigt werden, dass (c) Klima nicht immer der Haupteinflussfaktor für Vegetationswandel ist und dass (d) das Ausmaß von Vegetationsveränderungen in zuvor veröffentlichten Studien auf der Hochebene von Tibet überschätzt worden sein kann, weil Diskrepanzen der Pollenproduktivität zwischen den Arten nicht einbezogen wurden. The basic units for distributing these services are the web sites scattered throughout the world. However, the extreme fragility of web services and content, the high competence between similar services supplied by different sites, and the wide geographic distributions of the web users drive the urgent requirement from the web managers to track and understand the usage interest of their web customers. This thesis, "X-tracking the Usage Interest on Web Sites", aims to fulfill this requirement. "X" stands two meanings: one is that the usage interest differs from various web sites, and the other is that usage interest is depicted from multi aspects: internal and external, structural and conceptual, objective and subjective. "Tracking" shows that our concentration is on locating and measuring the differences and changes among usage patterns. This thesis presents the methodologies on discovering usage interest on three kinds of web sites: the public information portal site, e-learning site that provides kinds of streaming lectures and social site that supplies the public discussions on IT issues. On different sites, we concentrate on different issues related with mining usage interest. The educational information portal sites were the first implementation scenarios on discovering usage patterns and optimizing the organization of web services. In such cases, the usage patterns are modeled as frequent page sets, navigation paths, navigation structures or graphs. However, a necessary requirement is to rebuild the individual behaviors from usage history. We give a systematic study on how to rebuild individual behaviors. Besides, this thesis shows a new strategy on building content clusters based on pair browsing retrieved from usage logs. The difference between such clusters and the original web structure displays the distance between the destinations from usage side and the expectations from design side. Moreover, we study the problem on tracking the changes of usage patterns in their life cycles. The changes are described from internal side integrating conceptual and structure features, and from external side for the physical features; and described from local side measuring the difference between two time spans, and global side showing the change tendency along the life cycle. A platform, Web-Cares, is developed to discover the usage interest, to measure the difference between usage interest and site expectation and to track the changes of usage patterns. E-learning site provides the teaching materials such as slides, recorded lecture videos and exercise sheets. We focus on discovering the learning interest on streaming lectures, such as real medias, mp4 and flash clips. Compared to the information portal site, the usage on streaming lectures encapsulates the variables such as viewing time and actions during learning processes. The learning interest is discovered in the form of answering 6 questions, which covers finding the relations between pieces of lectures and the preference among different forms of lectures. We prefer on detecting the changes of learning interest on the same course from different semesters. The differences on the content and structure between two courses leverage the changes on the learning interest. We give an algorithm on measuring the difference on learning interest integrated with similarity comparison between courses. A search engine, TASK-Moniminer, is created to help the teacher query the learning interest on their streaming lectures on tele-TASK site. Social site acts as an online community attracting web users to discuss the common topics and share their interesting information. Compared to the public information portal site and e-learning web site, the rich interactions among users and web content bring the wider range of content quality, on the other hand, provide more possibilities to express and model usage interest. We propose a framework on finding and recommending high reputation articles in a social site. We observed that the reputation is classified into global and local categories; the quality of the articles having high reputation is related with the content features. Based on these observations, our framework is implemented firstly by finding the articles having global or local reputation, and secondly clustering articles based on their content relations, and then the articles are selected and recommended from each cluster based on their reputation ranks. Das Web bietet verschiedene Dienste (z. B. Um solche Dienste anzubieten, werden weltweite, vereinzelte Websites als Basiseinheiten definiert. Aber die extreme Fragilität der Web-Services und -inhalte, die hohe Kompetenz zwischen ähnlichen Diensten für verschiedene Sites bzw. die breite geographische Verteilung der Web-Nutzer treiben einen dringenden Bedarf für Web-Manager und das Verfolgen und Verstehen der Nutzungsinteresse ihrer Web-Kunden. Die Arbeit zielt darauf ab, dass die Anforderung "X-tracking the Usage Interest on Web Sites" erfüllt wird. "X" hat zwei Bedeutungen. Die erste Bedeutung ist, dass das Nutzungsinteresse von verschiedenen Websites sich unterscheidet. Durch das Scenario können Nutzungsmuster gefunden und die Organisation von Web-Services optimiert werden. In solchen Fällen wird das Nutzungsmuster als häufige Pagemenge, Navigation-Wege, -Strukturen oder -Graphen modelliert. Eine notwendige Voraussetzung ist jedoch, dass man individuelle Verhaltensmuster aus dem Verlauf der Nutzung (Usage History) wieder aufbauen muss. Deshalb geben wir in dieser Arbeit eine systematische Studie zum Nachempfinden der individuellen Verhaltensweisen. Außerdem zeigt die Arbeit eine neue Strategie, dass auf Page-Paaren basierten Content-Clustering aus Nutzungssite aufgebaut werden. Das Lerninteresse wird erfasst, wenn wir Antworten zu sechs Fragen gehandelt haben. Diese Fragen umfassen verschiedene Themen, wie Erforschung der Relation zwischen Teilen von Lehrveranstaltungen oder die Präferenz zwischen den verschiedenen Formen der Lehrveranstaltungen. Wir bevorzugen die Aufdeckung der Veränderungen des Lerninteresses anhand der gleichen Kurse aus verschiedenen Semestern. Der Differenz auf den Inhalt und die Struktur zwischen zwei Kurse beeinflusst die Änderungen auf das Lerninteresse. Ein Algorithmus misst die Differenz des Lerninteresses im Bezug auf einen Ähnlichkeitsvergleich zwischen den Kursen. Die Suchmaschine „Task-Moniminer“ wird entwickelt, dass die Lehrkräfte das Lerninteresse für ihre Streaming-Vorlesungen über das Videoportal tele-TASK abrufen können. Social Websites dienen als eine Online-Community, in den teilnehmenden Web-Benutzern die gemeinsamen Themen diskutieren und ihre interessanten Informationen miteinander teilen. Im Vergleich zur Public Informationsportal-Website und E-Learning Website bietet diese Art von Website reichhaltige Interaktionen zwischen Benutzern und Inhalten an, die die breitere Auswahl der inhaltlichen Qualität bringen. Allerdings bietet eine Social-Website mehr Möglichkeiten zur Modellierung des Nutzungsinteresses an. Wir schlagen ein Rahmensystem vor, die hohe Reputation für Artikel in eine Social-Website empfiehlt. Unsere Beobachtungen sind, dass die Reputation in globalen und lokalen Kategorien klassifiziert wird. Außerdem wird die Qualität von Artikeln mit hoher Reputation mit den Content-Funktionen in Zusammenhang stehen. Durch die folgenden Schritte wird das Rahmensystem im Bezug auf die Überwachungen implementiert. Der erste Schritt ist, dass man die Artikel mit globalen oder lokalen Reputation findet. This work provides new possibilities for the specific activation of immune cells in order to use them for the generation of antibodies which are not of murine origin. Bone marrow-derived dendritic cells were loaded with antigen and co-cultured with naive T and B lymphocytes of non-immunized mice. The interaction and activation of the different cell types were investigated by measuring the expression of specific cell surface markers, the release of activation-dependent interleukins and the secretion of antigen-specific antibodies. We could demonstrate that dendritic cells process and present antigen fragments and activate T cells, that T cells proliferate and release activation-induced interleukins, and that B cells maturate under the influence of activated T cells and secrete antigen-specific antibodies. While biogeographical studies on alien invasive plant species have indicated coevolution with soil biota in their native distribution range, it is unknown whether adaptation to soil biota varies among populations within the native distribution range. The question of local adaptation between plants and their soil biota has important implications for conservation of biodiversity and may justify the use of seed material from local provenances in restoration campaigns. Methodology/Principal Findings: We studied soil biota effects in ten populations of the steppe grass Stipa capillata from two distinct regions, Europe and Asia. We tested for local adaptation at two different scales, both within (ca. 10-80 km) and between (ca. 3300 km) regions, using a reciprocal inoculation experiment in the greenhouse for nine months. Generally, negative soil biota effects were consistent. However, we did not find evidence for local adaptation: both within and between regions, growth of plants in their 'home soil' was not significantly larger relative to that in soil from other, more distant, populations. Conclusions/Significance: Our study suggests that negative soil biota effects can prevail in different parts of a plant species' range. Absence of local adaptation points to the possibility of similar rhizosphere biota composition across populations and regions, sufficient gene flow to prevent coevolution, selection in favor of plasticity, or functional redundancy among different soil biota. From the point of view of plant - soil biota interactions, our findings indicate that the current practice of using seeds exclusively from local provenances in ecosystem restoration campaigns may not be justified. With great regularity, particular species establish only poorly. More reliable establishment could improve outcome of restoration projects and increase cost-effectiveness. We investigated the abiotic germination niche of ten poorly establishing calcareous grassland species by simultaneously exploring the effects of moisture and light availability and temperature fluctuation on percentage germina- tion and speed of germination. We also investigated the effects of three different pre-treatments used to enhance seed germination – cold-stratification, osmo- tic priming and priming in combination with gibberellic acid (GA 3 ) – and how these affected abiotic germination niches. Species varied markedly in width of abiotic germination niche, ranging from Carex flacca with very strict abiotic requirements, to several species reliably germinating across the whole range of abiotic conditions. Our results suggest pronounced differ- ences between species in gap requirements for establishment. Germination was improved in most species by at least one pre-treatment. Evidence for positive effects of adding GA 3 to seed priming solutions was limited. In several species, pre-treated seeds germinated under a wider range of abiotic conditions than untreated seeds. Improved knowledge of species-specific germination niches and the effects of seed pre-treatments may help to improve species establishment by sowing, and to identify species for which sowing at a later stage of restoration or introduction as small plants may represent a more viable strategy. There are far more works with a focus on Jewish life and agency, and on the social fabric’s structure, than were to be expected after reading the secondary literature on that period. An academic approach that takes the Jews as acting subjects and does not present them as a nameless mass of victims has thus been present quite early. Also, there were quite differentiated statements on the Jewish leadership’s policy. Die Messung der kognitiven Aspekte des geschlechtlichen Selbstkonzepts (Maskulinität und Femininität) erfolgte mittels der GERO-Skala von Brengelmann und Hendrich (1990). Zur Erfassung der emotionalen Aspekte und Identifikationsmuster der Entwicklung des geschlechtlichen Selbstkonzepts wurden die Werte für die Variablen Maskulinität und Femininität zuerst mittels der computergesteuerten Methodik IDEXMONO und IDEXIDIO, die auf der Identitätsstrukturanalyse (Identity Structure Analysis) von Weinreich (2003) basiert, aufgearbeitet und weiter interferenzstatistisch ausgewertet. Weiterhin wurden der Fragebogen zur Messung normativer Geschlechtsrollenorientierung (NGRO) von Athenstaedt (2000) sowie ein ad hoc entworfener demographischer Fragebogen eingesetzt. Die Ergebnisse zeigen, dass der Verlauf der psychosexuellen Entwicklung einen starken Einfluss auf die Integration der Geschlechterstereotypen in die geschlechtliche Selbst- und Fremdwahrnehmung hat. Im kognitiven Bereich, bezogen auf die persönliche Identität (Grad der Selbstzuschreibung männlicher und weiblicher Merkmale), stellt die Maskulinität eine stabile und erstrebenswerte Variable zur Herausbildung des geschlechtlichen Selbstkonzepts bei allen Gruppen dar. Die Femininität trägt am meisten zur Differenzierung zwischen den Hetero-, Homo- und Transsexuellen bei. Sie wird, je nach der Entwicklungsphase, unterschiedlich in das geschlechtliche Selbstkonzept integriert. Hinsichtlich der sozialen Identität (Zugehörigkeitsgefühl) lassen sich die Gruppen bezüglich der wahrgenommenen Ähnlichkeiten sowohl mit männlichen als auch weiblichen Personen, je nach der Entwicklungsphase, unterscheiden. Die soziale Wahrnehmung von Männern und Frauen (Fremdwahrnehmung), ist bei Transsexuellen traditioneller als die der Hetero- und Homosexuellen. Bei der Selbst- und Fremdbewertung ergaben sich keine signifikanten Unterschiede. Bei der Internalisierung der sozialen Normen des geschlechtsspezifischen Verhaltens zeigt sich, dass Heterosexuelle der Ausübung der Geschlechterrollen gegenüber egalitärer eingestellt sind als Trans- und Homosexuelle. Bei den Sozialisationsfaktoren ist hervorzuheben, dass generell weibliche Identifikationspersonen einen stärkeren Einfluss auf die Herausbildung des geschlechtlichen Selbstkonzeptes hatten als männliche Identifikationspersonen. Es scheint jedoch, dass Homosexuelle bei der Entwicklung ihres geschlechtlichen Selbstkonzepts stärker unter dem Einfluss der Frauen stehen als die anderen zwei erforschten Gruppen. N2 - The aim of the present explorative-comparative study was to find out to which extent the biologically determined differences as well as socialization conditions in the psychosexual development of hetero-, homo- and transsexual men (N = 191) affect the integration of the gender stereotypes in cognitive (self-perception und perception of the others), emotional (appraisal of self and the others), and behavioral aspects (norms of gender behavior) of gender identity, as well as to deduce identification patterns in the development of the gender self-concept. The GERO-scale by Brengelmann und Hendrich (1990) was used to measure the cognitive aspects of the self-concept of gender (masculinity and femininity). The collected data of masculinity and femininity were further processed using the computer-operated Identity Structure Analysis based method IDEXMONO and IDEXIDIO (Weinreich, 2003) to detect the emotional aspects and identification patterns of the development of the self-concept of gender. Furthermore, the questionnaire of normative gender role orientation by Athenstaedt (2000) and an ad hoc designed demographic questionnaire were applied. The results show that the course of the psychosexual development has a strong impact upon the integration of the gender stereotypes in gender related self-perception and perception of others. As for cognitive aspects related to personal identity (grade of self-attribution of masculine and feminine traits), the masculinity represents a stable and desirable variable in the construction of the self-concept of gender in all samples. The femininity contributes the most to the differentiation between hetero-, homo- and trans-sexuals. Femininity has been integrated in the gender self-concept to a different extent, depending on the phase of the psychosexual development. As for social identity (feeling of social affiliation), there are differences among the samples in self-perceived similarities to both masculine and feminine persons, depending on the phase of the psychosexual development. The social perception of men and women (perception of the others) is more traditionally oriented in transsexuals than in hetero- and homosexuals. There are no significant differences in the appraisal of self and the others. As for the internalization of the social norms of gender specific behavior, heterosexuals are more egalitarian in their attitude towards the practice of gender roles than homo- and transsexuals. The examination of the socialization conditions shows that in general, feminine identification persons have a stronger impact upon the construction of self-concept of gender than masculine ones. However, it seems as though women have a stronger impact upon the construction of self-concept of gender in homosexuals than in the other groups. A discriminant analysis was computed to answer the question which self-related variables and developmental factors have the strongest statistical importance for the division and prediction of the samples. The variables “stereotypic perception of masculine persons” and “ego-involvement with feminine persons” have the strongest discriminant importance for the discriminant function 1 (division of transsexuals from hetero- and homosexuals) and the variables “empathic identification with masculine persons in the past” and “growth of empathic identification with feminine persons” for the discriminant function 2 (division of heterosexuals from homosexuals). FT-IR spectra and DSC measurements indicate that the polymer end groups strongly affect the crystallinity of the material. The role of MoeBR and MoeZR was studied in detail for the interaction with the two MoaD-homologs involved in Moco biosynthesis, MoaD1 and MoaD2, in addition to the CysO protein involved in cysteine biosynthesis. We show that both proteins have a role in Moco biosynthesis, while only MoeZR, but not MoeBR, has an additional role in cysteine biosynthesis. MoeZR and MoeBR were able to complement an E. coli moeB mutant strain, but only in conjunction with the Mycobacterial MoaD1 or MoaD2 proteins. Both proteins were able to sulfurate MoaD1 and MoaD2 in vivo, while only MoeZR additionally transferred the sulfur to CysO. Our in vivo studies show that Mycobacteria have acquired several homologs to maintain Moco biosynthesis. Viele Stoffwechselwege haben Reaktionen in mehr als einem Kompartiment. Sowohl die Saccharose-phosphat-synthase, also auch die Saccharose-phosphatase sind in vivo weit von einem Gleichgewichtszustand entfernt. In Wildtyp Arabidopsis thaliana Columbia-0 Blättern wurde der gesamte Pool von ADPGlc im Chloroplasten gefunden. Das Enzyme ADPGlc pyrophosphorylase ist im Chloroplasten lokalisiert und synthetisiert ADPGlc aus ATP und Glc1P. Dieses Verteilungsmuster spricht eindeutig gegen die Hypothese von Pozueta-Romero und Kollegen, dass ADPGlc im Zytosol durch ADP vermittelte Spaltung von Saccharose durch die Saccharose Synthase erzeugt wird. N2 - Metabolism in plant cells is highly compartmented, with many pathways involving reactions in more than one compartment. For example, during photosynthesis in leaf mesophyll cells, primary carbon fixation and starch synthesis take place in the chloroplast, whereas sucrose is synthesized in the cytosol and stored in the vacuole. These reactions are tightly regulated to keep a fine balance between the carbon pools of the different compartments and to fulfil the energy needs of the organelles. I applied a technique which fractionates the cells under non-aqueous conditions, whereby the metabolic state is frozen at the time of harvest and held in stasis throughout the fractionation procedure. With the combination of non-aqueous fractionation and mass spectrometry based metabolite measurements (LC-MS/MS, GC-MS) it was possible to investigate the intracellular distributions of the intermediates of photosynthetic carbon metabolism and its products in subsequent metabolic reactions. With the knowledge about the in vivo concentrations of these metabolites under steady state photosynthesis conditions it was possible to calculate the mass action ratio and change in Gibbs free energy in vivo for each reaction in the pathway, to determine which reactions are near equilibrium and which are far removed from equilibrium. The Km value and concentration of each enzyme were compared with the concentrations of its substrates in vivo to assess which reactions are substrate limited and so sensitive to changes in substrate concentration. Several intermediates of the Calvin-Benson cycle are substrates for other pathways, including dihydroxyacetone-phosphate (DHAP,sucrose synthesis), fructose 6-phosphate (Fru6P, starch synthesis), erythrose 4-phosphate (E4P,shikimate pathway) and ribose 5-phosphate (R5P, nucleotide synthesis). Several of the enzymes that metabolise these intermediates, and so lie at branch points in the pathway, are triose-phosphate isomerase (DHAP), transketolase (E4P, Fru6P), sedoheptulose-1,7-bisphosphate aldolase (E4P) and ribose-5-phosphate isomerase (R5P) are not saturated with their respective substrate as the metabolite concentration is lower than the respective Km value. In terms of metabolic control these are the steps that are most sensitive to changes in substrate availability, while the regulated irreversible reactions of fructose-1,6-bisphosphatase and sedoheptulose-1,7-bisphosphatase are relatively insensitive to changes in the concentrations of their substrates. In the pathway of sucrose synthesis it was shown that the concentration of the catalytic binding site of the cytosolic aldolase is lower than the substrate concentration of DHAP, and that the concentration of Suc6P is lower than the Km of sucrose-phosphatase for this substrate. Both the sucrose-phosphate synthase and sucrose-phosphatase reactions are far removed from equilibrium in vivo. In wild type A. thaliana Columbia-0 leaves, all of the ADPGlc was found to be localised in the chloroplasts. ADPglucose pyrophosphorylase is localised to the chloroplast and synthesises ADPGlc from ATP and Glc1P. This distribution argues strongly against the hypothesis proposed by Pozueta-Romero and colleagues that ADPGlc for starch synthesis is produced in the cytosol via ADP-mediated cleavage of sucrose by sucrose synthase. Based on this observation and other published data it was concluded that the generally accepted pathway of starch synthesis from ADPGlc produced by ADPglucose pyrophosphorylase in the chloroplasts is correct, and that the alternative pathway is untenable. Within the pathway of starch synthesis the concentration of ADPGlc was found to be well below the Km value of starch synthase for ADPGlc, indicating that the enzyme is substrate limited. A general finding in the comparison of the Calvin-Benson cycle with the synthesis pathways of sucrose and starch is that many enzymes in the Calvin Benson cycle have active binding site concentrations that are close to the metabolite concentrations, while for nearly all enzymes in the synthesis pathways the active binding site concentrations are much lower than the metabolite concentrations. Nevertheless, treatment interruptions inevitably result in viral rebounds from persistent, latently infected cells, necessitating lifelong treatment. Virological failure due to resistance development is a frequent event and the major threat to treatment success. Currently, it is recommended to change treatment after the confirmation of virological failure. However, at the moment virological failure is detected, drug resistant mutants already replicate in great numbers. They infect numerous cells, many of which will turn into latently infected cells. This pool of cells represents an archive of resistance, which has the potential of limiting future treatment options. The objective of this study was to design a treatment strategy for treatment-naive patients that decreases the likelihood of early treatment failure and preserves future treatment options. We propose to apply a single, pro-active treatment switch, following a period of treatment with an induction regimen. The main goal of the induction regimen is to decrease the abundance of randomly generated mutants that confer resistance to the maintenance regimen, thereby increasing subsequent treatment success. Treatment is switched before the overgrowth and archiving of mutant strains that carry resistance against the induction regimen and would limit its future re-use. In silico modelling shows that an optimal trade-off is achieved by switching treatment at & 80 days after the initiation of antiviral therapy. Evaluation of the proposed treatment strategy demonstrated significant improvements in terms of resistance archiving and virological response, as compared to conventional HAART. While continuous pro-active treatment alternation improved the clinical outcome in a randomized trial, our results indicate that a similar improvement might also be reached after a single pro-active treatment switch. The clinical validity of this finding, however, remains to be shown by a corresponding trial. Several influential eye-tracking studies on syntactic reanalysis (Frazier & Rayner, 1982; Meseguer, Carreiras, & Clifton, 2002; Mitchell, Shen, Green, & Hodgson, 2008) have addressed this question by examining scanpaths, i.e., sequential patterns of eye fixations. However, in the absence of a suitable method for analyzing scanpaths, these studies relied on simplified dependent measures that are arguably ambiguous and hard to interpret. We address the theoretical question of repair strategy by developing a new method that quantifies scanpath similarity. Our method reveals several distinct fixation strategies associated with reanalysis that went undetected in a previously published data set (Meseguer et al., 2002). One prevalent pattern suggests re-parsing of the sentence, a strategy that has been discussed in the literature (Frazier & Rayner, 1982); however, readers differed tremendously in how they orchestrated the various fixation strategies. Our results suggest that the human parsing system non-deterministically adopts different strategies when confronted with the disambiguating material in garden-path sentences. Generally speaking, 'poetic justice' succeeds in establishing coherence and propel the narrative forward. The constituents of societal norms and of laws are nevertheless relentlessly questioned - to such an extent that the narrative inquiry occasionally departs from the common understanding of justice. With its focus on morality, especially the presence or absence of faith, Loher und Muller is primarily concerned with the potential for conflict inherent in medieval constructions of legality and justice. In doing so, the epic opens up a narrative playground unencumbered by legal constraints as - after all - literature need not comply with medieval jurisdiction and its claims to the validity and scope of its writings. It is literature's privilege to facilitate unfamiliar ways of looking. The playful - but by no means inconsequential - casuistry played out in Loher und Maller gives rise to a 'probable' world tangential to historical reality and its understanding of justice and the law. Methods Entrepreneurs (n = 632), teachers (n = 5,196), and physicians (n = 549) were surveyed in a cross-sectional design. The questionnaire Work-related Behavior and Experience Patterns (AVEM) was used for all professions and, in addition, two scales (health prevention and self-confidence) from the Checklist for Entrepreneurs in the sample of entrepreneurs. Results The largest proportion of the entrepreneurs (45%) presented with a healthy pattern (compared with 18.4% teachers and 18.3% physicians). Thirty-eight percent of entrepreneurs showed a risk pattern of overexertion and stress, followed by teachers (28.9%) and physicians (20.6%). Unambitious or burnout patterns were seen in only 9.3/8.2% of entrepreneurs, respectively, and 25.3/27.3% of teachers, and 39.6/21.5% of physicians. While the distribution of patterns in teachers and physicians differed significantly between genders, a gender difference was not found among entrepreneurs. Entrepreneurs with the risk pattern of overexertion scored significantly (P < 0.01) lower in self-confidence and health care than those with the healthy pattern. Conclusions The development of a successful enterprise depends, in part, on the health of the entrepreneur. The large proportion of entrepreneurs with the healthy pattern irrespective of gender may support the notion that self-selection effects of healthy individuals in this special career might be important. At the same time, a large proportion was at risk for overexertion and might benefit from measures to cope with professional demands and stress and promote a healthy behavior pattern. The results show larger male strength than female strength. The concept of Public Service Motivation states that employees working in the public sector are primarily motivated by serving the common good. Based on an empirical study this construct was applied to public employees in Germany. Moreover the thesis collects evidence for the relationship between Public Service Motivation and process theories of motivation. Based on information processing approaches to intelligence, we tested a mediation hypothesis, which states that the complex cognitive abilities of reasoning and divergent thinking mediate the influence of the basic cognitive abilities of mental speed and short-term memory on achievement. We administered a comprehensive test battery and analyzed the data through structural equation modeling while controlling for the cluster structure of the data. Our findings support the notion that mental speed and short-term memory, as ability factors reflecting basic cognitive processes, exert an indirect influence on academic achievement by affecting reasoning and divergent thinking (total indirect effects: beta=.22 and .24. respectively). Short-term memory also directly affects achievement (beta=.22).
