This new trajectory is characterized by rising risks of triggering irreversible and unmanageable shifts in Earth system functioning. We urgently need a new global approach to safeguard critical Earth system regulating functions more effectively and comprehensively. The global commons framework is the closest example of an existing approach with the aim of governing biophysical systems on Earth upon which the world collectively depends. Derived during stable Holocene conditions, the global commons framework must now evolve in the light of new Anthropocene dynamics. This requires a fundamental shift from a focus only on governing shared resources beyond national jurisdiction, to one that secures critical functions of the Earth system irrespective of national boundaries. We propose a new framework—the planetary commons—which differs from the global commons frame­work by including not only globally shared geographic regions but also critical biophysical systems that regulate the resilience and state, and therefore livability, on Earth. The new planetary commons should articulate and create comprehensive stewardship obligations through Earth system governance aimed at restoring and strengthening planetary resilience and justice. Among other things, it is related to followers' perceived meaningfulness of their work. However, little is known about whether uncertainty in the workplace affects visionary leadership's effects. Given that uncertainty is rising in many, if not most, workplaces, it is vital to understand whether this development influences the extent to which visionary leadership is associated with followers' perceived meaningfulness. In a two-source, lagged design field study of 258 leader-follower dyads from different settings, we show that uncertainty moderates the relation between visionary leadership and followers' perceived meaningfulness such that this relation is more strongly positive when uncertainty is high, rather than low. Moreover, we show that with increasing uncertainty, visionary leadership is more negatively related to followers' turnover intentions via perceived meaningfulness. This research broadens our understanding of how visionary leadership may be a particularly potent tool in times of increasing uncertainty. It is often claimed that men negotiate better than women, yet it is simultaneously emphasized that results strongly depend on context. Through the use of qualitative methods such as content analysis and critical mixed-methods review, the research question: "Are women truly inferior negotiators compared to men?" is addressed. The study comprises a descriptive and an interpretive part. The descriptive section illuminates various interpretations of gender-specific negotiation theory among citing authors, with 67% arguing for a general superiority of men. However, given the high variance in gender-specific differences, the focus should instead be on the context-dependency of negotiation performance. Generalized statements can be made within contexts, but not across them. In the interpretive section, several factors contributing to this misinterpretation are highlighted, including discrepancies in the definition of negotiation performance and distortions in research communication.. From a scientific perspective, this study underscores the need for a nuanced sociological analysis and warns against the one-sided acceptance of inaccurate scientific interpretations. From a practical standpoint, it amplifies the voices of women affected by biased research paradigms. Overall, the dissertation clarifies the theory of gender-specific negotiation performance and advocates for the elimination of biases in scientific discourse. However, while the impact of emotions and psychological needs on students' achievements has been a focus of research, the impact of their physiological needs has been under studied. In this explorative study, I examine what impact the physiological and psychological needs of student teachers have on their feelings, motivation, and interest in different learning settings. Approach The research method used was the daily reconstruction method and included the Felix-App, a new digital research and feedback tool that allows the measurement of feelings, needs, motivation, and interest in real time. Findings The results suggest the importance of physiological needs for perceived emotions, motivation, and interest in the learning subject. The psychological needs, on the other hand, are of less importance. Originality The Felix-App is an innovative tool to learn more about learners' emotions and needs in real learning settings. The importance of physiological needs has been known since Maslow, but should be considered much more in the context of teaching and learning research in the future. In contrast, many rabbis and other Jewish officials still concentrate on legal aspects alone. The need for resolving important issues on the verge of wedlock is too often left to secular experts in law, psychology, or counseling. However, in recent years, this lack of formal training for marriage preparation has also been acknowledged by the Jewish clergy in order to incorporate it in the preparatory period before the bond is tied. This case study focuses on Jewish and Roman Catholic conceptions of marriage, past and present. We intend to do a comparative analysis of the prerequisites of religious marriage based on the assumption that both Judaism and the Roman Catholic Church have a distinct legal framework to assess marriage preparation. To fulfil the Paris Agreement, the European Union (EU) needs a rapid transition to a fully decarbonised power production system. Presumably, such a system will be largely based on renewables. So far, many EU countries have supported a shift towards renewables such as solar and wind power using support schemes, but the economic and political context is changing. Renewables are now cheaper than ever before and have become cost-competitive with conventional technologies. Therefore, European policymakers are striving to better integrate renewables into a competitive market and to increase the cost-effectiveness of the expansion of renewables. The first step was to replace previous fixed-price schemes with competitive auctions. In a second step, these auctions have become more technology-open. Finally, some governments may phase out any support for renewables and fully expose them to the competitive power market. However, such policy changes may be at odds with the need to rapidly expand renewables and meet national targets due to market characteristics and investors’ risk perception. Without support, price risks are higher, and it may be difficult to meet an investor’s income expectations. Furthermore, policy changes across different countries could have unexpected effects if power markets are interconnected and investors able to shift their investments. Finally, in multi-technology auctions, technologies may dominate, which can be a risk for long-term power system reliability. Therefore, in my thesis, I explore the effects of phasing out support policies for renewables, of coordinating these phase-outs across countries, and of using multi-technology designs. I expand the public policy literature about investment behaviour and policy design as well as policy change and coordination, and I further develop an agent-based model. The main questions of my thesis are what the cost and deployment effects of gradually exposing renewables to market forces would be and how coordination between countries affects investors’ decisions and market prices.. In my three contributions to the academic literature, I use different methods and come to the following results. In the first contribution, I use a conjoint analysis and market simulation to evaluate the effects of phasing out support or reintroducing feed-in tariffs from the perspective of investors. I find that a phase-out leads to investment shifts, either to other still-supported technologies or to other countries that continue to offer support. I conclude that the coordination of policy changes avoids such shifts.. In the second contribution, I integrate the empirically-derived preferences from the first contribution in to an agent-based power system model of two countries to simulate the effects of ending auctions for renewables. I find that this slows the energy transition, and that cross-border effects are relevant. Consequently, continued support is necessary to meet the national renewables targets. In the third contribution, I analyse the outcome of past multi-technology auctions using descriptive statistics, regression analysis as well as case study comparisons. I find that the outcomes are skewed towards single technologies. This cannot be explained by individual design elements of the auctions, but rather results from context-specific and country-specific characteristics. Based on this, I discuss potential implications for long-term power system reliability. The main conclusions of my thesis are that a complete phase-out of renewables support would slow down the energy transition and thus jeopardize climate targets, and that multi-technology auctions may pose a risk for some countries, especially those that cannot regulate an unbalanced power plant portfolio in the long term. If policymakers decide to continue supporting renewables, they may consider adopting technology-specific auctions to better steer their portfolio. In contrast, if policymakers still want to phase out support, they should coordinate these policy changes with other countries. Otherwise, overall transition costs can be higher, because investment decisions shift to still-supported but more expensive technologies. N2 - Die Stromproduktion trägt zu einem bedeutenden Anteil der Treibhausgasemissionen in Europa bei und ist damit ein wichtiger Treiber des Klimawandels. Um das Pariser Abkommen zu erfüllen, muss die Europäische Union (EU) eine Wende hin zu einem vollständig treibhausgasneutralen Stromerzeugungssystem durchlaufen. Vermutlich wird ein solches System zu einem Grossteil auf erneuerbaren Energien beruhen. Obwohl viele EU-Länder die Umstellung auf erneuerbare Energien wie Solar- und Windenergie bisher durch Förderprogramme unterstützt haben, ändert sich der wirtschaftliche und politische Kontext. Erneuerbare Energien sind heute billiger als je zuvor und konkurrenzfähig gegenüber konventionellen Technologien. Daher sind Entscheidungsträger der europäischen Regierungen bestrebt, die erneuerbaren Energien besser in einen wettbewerbsfähigen Markt zu integrieren und die Kosteneffizienz des Ausbaus der erneuerbaren Energien zu erhöhen. In einem ersten Schritt wurden Festpreisvergütungen durch wettbewerbsorientierte Auktionen ersetzt. In einem zweiten Schritt wurden diese Auktionen technologieoffener gestaltet. Schließlich könnten einige Regierungen die Unterstützung für erneuerbare Energien beenden und diese vollständig dem wettbewerbsorientierten Strommarkt aussetzen. Solche Reformen könnten jedoch aufgrund von Marktmerkmalen und der Risikowahrnehmung von Investoren im Widerspruch zu der Notwendigkeit stehen, die erneuerbaren Energien rasch auszubauen, um die nationalen Ziele zu erreichen. Ohne Förderinstrumente steigen Preisrisiken und es könnte schwierig werden, die Einkommenserwartungen der Investoren zu erfüllen. Darüber hinaus könnten Reformen in verschiedenen Ländern unerwartete Auswirkungen haben, insbesondere wenn deren Strommärkte gekoppelt sind und die Investoren ihre Anlagen in andere Länder verlagern. Schließlich könnten technologieoffene Auktionen zu einer Dominanz einzelner Technologien führen, was ein Risiko für die langfristige Zuverlässigkeit des Stromsystems darstellen würde. In meiner Dissertation untersuche ich daher die Auswirkungen des Ausstiegs aus der Förderung erneuerbarer Energien und deren länderübergreifenden Koordinierung. Außerdem beleuchte ich die Resultate von Multitechnologie-Auktionen. Mit dieser Dissertation erweitere ich die wissenschaftliche Literatur über Investitionsverhalten und Policydesign sowie über Politikänderungen und -koordinierung und entwickle ein agentenbasiertes Modell weiter. Die Hauptforschungsfragen meiner Arbeit sind, wie sich die schrittweise Annäherung der erneuerbaren Energien hin zu Marktkräften auf die Kosten und deren Ausbau auswirken würde und wie die Koordinierung zwischen den Ländern die Entscheidungen der Investoren und die Marktpreise beeinflusst. In meinen drei wissenschaftlichen Artikeln verwende ich unterschiedliche Methoden und gelange zu den folgenden Ergebnissen. Im ersten Artikel wende ich eine Conjoint-Analyse an, um die Auswirkungen einer Beendigung der Förderung oder der Wiedereinführung von Einspeisevergütungen aus der Sicht der Investoren zu bewerten. Ich komme zu dem Ergebnis, dass ein Ende der Förderung zu Investitionsverlagerungen führt, entweder hin zu anderen, weiterhin geförderten Technologien oder in andere Länder, die weiterhin eine Förderung anbieten. Die Koordinierung dieser Politikänderungen kann eine solche Verlagerung jedoch verhindern. Im zweiten Artikel integriere ich die empirisch-erhobenen Präferenzen aus dem ersten Artikel in ein agentenbasiertes Energiesystemmodell für zwei Länder, um die Auswirkungen einer Abschaffung von Auktionen für erneuerbare Energien zu simulieren. Ich stelle fest, dass dies die Energiewende insgesamt verlangsamt und dass grenzübergreifende Effekte von Bedeutung sind. Es wäre eine kontinuierliche Förderung notwendig, um die nationalen Ziele zum Ausbau der Erneuerbaren zu erreichen. Im dritten Artikel analysiere ich die Ergebnisse vergangener Multi-Technologie-Auktionen mittels deskriptiver Statistik, Regressionsanalysen sowie Fallstudienvergleichen. Ich stelle fest, dass die Resultate zugunsten einzelner Technologien ausfallen. Dies lässt sich nicht durch einzelne Designmerkmale der Auktionen erklären, sondern ist vielmehr auf kontext- und länderspezifische Merkmale zurückzuführen. Darauf aufbauend, diskutiere ich mögliche Auswirkungen auf die langfristige Zuverlässigkeit des Stromsystems. Die Hauptaussagen meiner Arbeit sind, dass ein vollständiges Auslaufen der Förderung der Erneuerbaren Energien die Energiewende verlangsamen und damit die Klimaziele gefährden würde, sowie, dass Multi-Technologie-Auktionen ein Risiko für einige Länder darstellen können, insbesondere für solche, die ein unausgewogenes Kraftwerksportfolio nicht regeln können. We first derive hypotheses regarding patterns of knowledge-sharing relationships by which workers pass on and exchange tacit and codified knowledge within and across organizational hierarchies to address the challenges that underpin contemporary knowledge work. We use survey data and apply exponential random graph models to test our hypotheses. We then extend the quantitative network analysis with insights from qualitative interviews and demonstrate that the identified knowledge-sharing patterns are the micro-foundational traces of collective coordination resulting from two underlying coordination mechanisms which we label ‘invisible iterations’ and ‘bringing in the big guns’. These mechanisms and, by extension, the associated knowledge-sharing patterns enable knowledge workers to perform in a setting that is characterized by complexity, uncertainty and ambiguity. Our research contributes to theory on the interplay between formal and informal organization for coordination by showing how self-directed, informal action is supported by the formal organizational hierarchy. In doing so, it also extends understanding of the role that hierarchy plays for knowledge-intensive work. We refer to this phenomenon as self-referential information (SRI) consumption and examine its consequences and the mechanism behind them. Building on self-awareness research and the strength model of self-control, we argue that SRI consumption heightens the state of self-awareness and thereby depletes participants’ mental resources, eventually undermining virtual meeting (VM) outcomes. Our findings from a European employee sample revealed contrary effects of SRI consumption across speaker vs listener roles. Engagement with self-view is positively associated with self-awareness, which, in turn, is negatively related to satisfaction with VM process, perceived productivity, and enjoyment. Looking at the self while listening to others exhibits adverse direct and indirect (via self-awareness) effects on VM outcomes. However, looking at the self when speaking exhibits positive direct effects on satisfaction with VM process and enjoyment. When these curricula are designed by education specialists, entrepreneurs are usually not consulted. To explore practitioners’ curricular recommendations, we conducted 73 semi-structured interviews with entrepreneurs with at least five years of professional experience. We collected 49 items for teaching and learning objectives, 37 for contents, 28 for teaching methods, and 17 for assessment methods. The respondents are convinced that students should acquire solid knowledge in business and management, legal issues, and entrepreneurship. For the latter, only some core aspects are provided. The entrepreneurs put greater emphasis on entrepreneurial skills and attitudes and consider experiential learning designs as most suitable, both in the secure setting of the classroom and in real life. The findings can help reflect on current entrepreneurship curriculum designs. Drawing on the case of sexual violence against refugees in European refugee camps, this paper argues that there is a form of testimonial injustice—willful testimonial injustice—that is deliberate. To do so, the paper argues (a) that the hearer intentionally utilizes negative identity prejudices for a particular purpose and (b) that the hearer is aware of the fact that the intentionally used prejudices are in fact prejudices. Furthermore, the paper shows how testimonial injustice relates to recognition failures both in terms of a causal as well as a constitutive claim. In fact, introducing willful testimonial injustice can support the constitutive claim of such a relation that has so far received little attention. Besides arguing for a novel form of testimonial injustice and contributing to the recent debate on the relation between epistemic injustice and recognition failures, this paper is also motivated by the attempt to draw attention to the inhumane conditions for refugees at the border of Europe as well as elsewhere. Digital tools can constrain discretionary power, like the curtailment thesis proposed, or serve as action resources, like the enablement thesis suggested. This article assesses empirical evidence of the impact of digital tools on street-level work and decision-making in service-oriented and regulation-oriented organisations based on a systematic literature review and thematic qualitative content analysis of 36 empirical studies published until 2021. The findings demonstrate different effects with regard to the role of digital tools and the core tasks of the public administration, depending on political and managerial goals and consequent system design. Leading or decisive digital tools mostly curtail discretion, especially in service-oriented organisations. In contrast, an enhanced information base or recommendations for actions enable decision-making, in particular in regulation-oriented organisations. By showing how street-level bureaucrats actively try to resist the curtailing effects caused by rigid design to address individual circumstances, for instance by establishing ways of coping like rule bending or rule breaking, using personal resources or prioritising among clients, this study demonstrates the importance of the continuation thesis and the persistently crucial role of human judgement in policy implementation. This coincides with the enhanced formation of bone marrow adipose tissue (BMAT), suggesting a negative effect of bone marrow adipocytes on skeletal health. Increased BMAT formation is also observed in pathologies such as obesity, type 2 diabetes and osteoporosis. However, a subset of bone marrow adipocytes forming the constitutive BMAT (cBMAT), arise early in life in the distal skeleton, contain high levels of unsaturated fatty acids and are thought to provide a physiological function. Regulated BMAT (rBMAT) forms during aging and obesity in proximal regions of the bone and contain a large proportion of saturated fatty acids. Paradoxically, BMAT accumulation is also enhanced during caloric restriction (CR), a life-span extending dietary intervention. This indicates, that different types of BMAT can form in response to opposing nutritional stimuli with potentially different functions. To this end, two types of nutritional interventions, CR and high fat diet (HFD), that are both described to induce BMAT accumulation were carried out. CR markedly increased BMAT formation in the proximal tibia and led to a higher proportion of unsaturated fatty acids, making it similar to the physiological cBMAT. Additionally, proximal and diaphyseal tibia regions displayed higher adiponectin expression. In aged mice, CR was associated with an improved trabecular bone structure. Taken together, these findings demonstrate, that the type of BMAT that forms during CR might provide beneficial effects for local bone stem/progenitor cells and metabolic health. The HFD intervention performed in this thesis showed no effect on BMAT accumulation and bone microstructure. RNA Seq analysis revealed alterations in the composition of the collagen-containing extracellular matrix (ECM). In order to investigate the effects of glucose homeostasis on osteogenesis, differentiation capacity of immortalized multipotent mesenchymal stromal cells (MSCs) and osteochondrogenic progenitor cells (OPCs) was analyzed. Insulin improved differentiation in both cell types, however, combination of with a high glucose concentration led to an impaired mineralization of the ECM. In the MSCs, this was accompanied by the formation of adipocytes, indicating negative effects of the adipocytes formed during hyperglycemic conditions on mineralization processes. However, the altered mineralization pattern and structure of the ECM was also observed in OPCs, which did not form any adipocytes, suggesting further negative effects of a hyperglycemic environment on osteogenic differentiation. In summary, the work provided in this thesis demonstrated that differentiation commitment of bone-resident stem cells can be altered through nutrient availability, specifically glucose. Surprisingly, both high nutrient supply, e.g. the hyperglycemic cell culture conditions, and low nutrient supply, e.g. CR, can induce adipogenic differentiation. However, while CR-induced adipocyte formation was associated with improved trabecular bone structure, adipocyte formation in a hyperglycemic cell-culture environment hampered mineralization. BT - observational and experimental evidence from Germany N2 - Does working in a gender-atypical occupation reduce individuals’ likelihood of finding a different-sex romantic partner, and do such occupational partnership penalties contribute to occupational gender segregation? To answer this question, we theorized partnership penalties for working in gender-atypical occupations by drawing on insights from evolutionary psychology, social constructivism, and rational choice theory and exploited the stability of occupational pathways in Germany. In Study 1, we analyzed observational data from a national probability sample (N= 1,634,944) to assess whether individuals in gender-atypical occupations were less likely to be partnered than individuals who worked in gender typical occupations. To assess whether the observed partnership gaps found in Study 1 were causally related to the gender typicality of men’s and women’s occupations, we conducted a field experiment on a dating app (N = 6,778). Because the findings from Study 2 suggested that young women and men indeed experienced penalties for working in a gender-atypical occupation (at least when they were not highly attractive), we employed a choice-experimental design in Study 3 (N = 1,250) to assess whether women and men were aware of occupational partnership penalties and showed that anticipating occupational partnership penalties may keep young and highly educated women from working in gender-atypical occupations. Our main conclusion therefore is that that observed penalties and their anticipation seem to be driven by unconscious rather than conscious processes. The ability to cope with rapidly changing customer requirements and unexpected internal and external events guarantees robust and efficient production processes, requiring a dedicated control concept at the shop floor level. Yet in today's practice, conventional control approaches remain in use, which may not keep up with the dynamic behaviour due to their scenario-specific and rigid properties. To address this challenge, deep learning methods were increasingly deployed due to their optimization and scalability properties. However, these approaches were often tested in specific operational applications and focused on technical performance indicators such as order tardiness or total throughput. In this paper, we propose a deep reinforcement learning based production control to optimize combined techno-financial performance measures. Based on pre-defined manufacturing modules that are supplied and operated by multiple agents, positive effects were observed in terms of increased revenue and reduced penalties due to lower throughput times and fewer delayed products. The combined modular and multi-staged approach as well as the distributed decision-making further leverage scalability and transferability to other scenarios. Both require information and involve search efforts, such as localizing prey or finding potential mates. In non-migrating bat species, playback studies showed that listening to vocalizations of other bats, both con-and heterospecifics, may help a recipient bat to find foraging patches and mating sites. However, we are still unaware of the degree to which migrating bats depend on con-or heterospecific vocalizations for identifying potential feeding or mating opportunities during nightly transit flights. Here, we investigated the vocal responses of Nathusius’ pipistrelle bats, Pipistrellus nathusii, to simulated feeding and courtship aggregations at a coastal migration corridor. We presented migrating bats either feeding buzzes or courtship calls of their own or a heterospecific migratory species, the common noctule, Nyctalus noctula. We expected that during migratory transit flights, simulated feeding opportunities would be particularly attractive to bats, as well as simulated mating opportunities which may indicate suitable roosts for a stopover. However, we found that when compared to the natural silence of both pre-and post-playback phases, bats called indifferently during the playback of conspecific feeding sounds, whereas P. nathusii echolocation call activity increased during simulated feeding of N. noctula. In contrast, the call activity of P. nathusii decreased during the playback of conspecific courtship calls, while no response could be detected when heterospecific call types were broadcasted. Our results suggest that while on migratory transits, P. nathusii circumnavigate conspecific mating aggregations, possibly to save time or to reduce the risks associated with social interactions where aggression due to territoriality might be expected. This avoidance behavior could be a result of optimization strategies by P. nathusii when performing long-distance migratory flights, and it could also explain the lack of a response to simulated conspecific feeding. However, the observed increase of activity in response to simulated feeding of N. noctula, suggests that P. nathusii individuals may be eavesdropping on other aerial hawking insectivorous species during migration, especially if these occupy a slightly different foraging niche. Different approaches are used to analyze the data. First, univariate regression (UVR) is used. However, due to the strong influence of matrix effects, this is not suitable for the quantitative analysis of copper grades. Second, the multivariate method of partial least squares regression (PLSR) is used, which is more suitable for quantification. They are considered sympatric species, because their distribution area overlaps. These species generate species-specific electric organ discharges (EODs) varying in waveform characteristics, including duration, polarity, and phase number. They exhibit also pronounced divergence in their snout, i.e. the length, thickness, and curvature. The diversifications in these two phenotypical traits (EOD and snout) have been proposed as key factors promoting adaptive radiation in Campylomormyrus. The role of EODs as a pre-zygotic isolation mechanism driving sympatric speciation by promoting assortative mating has been examined using behavioral, genetical, and histological approaches. However, the evolutionary effects of the snout morphology and its link to species divergence have not been closely examined. Hence, the main objective of this study is to investigate the effect of snout morphology diversification and its correlated EOD to better understand their sympatric speciation and evolutionary drivers. Moreover, I aim to utilize the intragenus and intergenus hybrids of Campylomormyrus to better understand trait divergence as well as underlying molecular/genetic mechanisms involved in the radiation scenario. To this end, I utilized three different approaches: feeding behavior analysis, diet assessment, and geometric morphometrics analysis. I performed feeding behavior experiments to evaluate the concept of the phenotype-environment correlation by testing whether Campylomormyrus species show substrate preferences. The behavioral experiments showed that the short snout species exhibits preference to sandy substrate, the long snout species prefers a stone substrate, and the species with intermediate snout size does not exhibit any substrate preference. The experiments suggest that the diverse feeding apparatus in the genus Campylomormyrus may have evolved in adaptation to their microhabitats. I also performed diet assessments of sympatric Campylomormyrus species and a sister genus species (Gnathonemus petersii) with markedly different snout morphologies and EOD using NGS-based DNA metabarcoding of their stomach contents. The diet of each species was documented showing that aquatic insects such as dipterans, coleopterans and trichopterans represent the major diet component. The results showed also that all species are able to exploit diverse food niches in their habitats. However, comparing the diet overlap indices showed that different snout morphologies and the associated divergence in the EOD translated into different prey spectra. These results further support the idea that the EOD could be a ‘magic trait’ triggering both adaptation and reproductive isolation. Geometric morphometrics method was also used to compare the phenotypical shape traits of the F1 intragenus (Campylomormyrus) and intergenus (Campylomormyrus species and Gnathonemus petersii) hybrids relative to their parents. The hybrids of these species were well separated based on the morphological traits, however the hybrid phenotypic traits were closer to the short-snouted species. In addition, the likelihood that the short snout expressed in the hybrids increases with increasing the genetic distance of the parental species. The results confirmed that additive effects produce intermediate phenotypes in F1-hybrids. It seems, therefore, that morphological shape traits in hybrids, unlike the physiological traits, were not expressed straightforward. To this area of inquiry, we introduce networking speed—the time that it takes for individuals to form a network tie—as an important but so far largely overlooked aspect. We develop a theory of networking speed that explains how different catalysts enable professionals to introduce variation into the speed with which they form interpersonal network ties. We discuss how such variation in the speed with which ties have been formed influences relational outcomes and the network returns that these ties generate. This discussion illustrates that high networking speed can entail advantages as well as pitfalls. We also explore temporal implications of networking speed—for instance, the persistence of the effects of speed over time. Overall, we conceptualize networking speed as a constitutive element of how interpersonal networks function in professional settings, and we propose a future research program for the integration of this novel concept into organizational network research. The comprehensive analysis of the literature and the current state of research provide the basis of this paper. Theoretical models, such as the Technology-Acceptance-Model (TAM) and the Unified Theory of Acceptance and Use of Technology 2 (UTAUT 2) enable the evaluation of expectations and acceptance, as well as the assessment of possible inhibitory factors for the subsequent descriptive and statistical examination of the acceptance of digital technologies in fashion stores. The research on this subject was examined in a quantitative way. The key findings show that customers do accept digital technologies in fashion stores. The final part of this contribution describes the innovative Digitalization 4 Sustainability Framework which shows that digital technologies at the point of sale (PoS) in fashion stores could have a positive impact on sustainability. Overall, this paper shows that it is particularly important for fashion stores to concentrate on their individual strengths and customer needs as well as to indicate a more sustainable way by using digital technologies, in order to achieve added value for the customers and to set themselves apart from the competition while designing a more sustainable future. Clinical risk factors are numerous, but it has been suggested that altered biomechanics might be associated with AT. No systematic review has been conducted investigating these biomechanical alterations in specifically athletic populations. Therefore, the aim of this systematic review was to compare the lower-limb biomechanics of athletes with AT to athletically matched asymptomatic controls. Databases were searched for relevant studies investigating biomechanics during gait activities and other motor tasks such as hopping, isolated strength tasks, and reflex responses. Inclusion criteria for studies were an AT diagnosis in at least one group, cross-sectional or prospective data, at least one outcome comparing biomechanical data between an AT and healthy group, and athletic populations. Studies were excluded if patients had Achilles tendon rupture/surgery, participants reported injuries other than AT, and when only within-subject data was available.. Effect sizes (Cohen's d) with 95% confidence intervals were calculated for relevant outcomes. The initial search yielded 4,442 studies. After screening, twenty studies (775 total participants) were synthesised, reporting on a wide range of biomechanical outcomes. Females were under-represented and patients in the AT group were three years older on average. Biomechanical alterations were identified in some studies during running, hopping, jumping, strength tasks and reflex activity. Equally, several biomechanical variables studied were not associated with AT in included studies, indicating a conflicting picture. Kinematics in AT patients appeared to be altered in the lower limb, potentially indicating a pattern of “medial collapse”. Muscular activity of the calf and hips was different between groups, whereby AT patients exhibited greater calf electromyographic amplitudes despite lower plantar flexor strength. Overall, dynamic maximal strength of the plantar flexors, and isometric strength of the hips might be reduced in the AT group. This systematic review reports on several biomechanical alterations in athletes with AT. With further research, these factors could potentially form treatment targets for clinicians, although clinical approaches should take other contributing health factors into account. During evolution, many genes are lost from the organellar genomes and get integrated in the nuclear genome, in what is known as intracellular/endosymbiotic gene transfer (IGT/EGT). IGT has been reproduced experimentally in Nicotiana tabacum at a gene transfer rate (GTR) of 1 event in 5 million cells, but, despite its centrality to eukaryotic evolution, there are no genetic factors known to influence the frequency of IGT in higher eukaryotes. The focus of this work was to determine the role of different DNA repair pathways of double strand break repair (DSBR) in the integration step of organellar DNA in the nuclear genome during IGT. Here, a CRISPR/Cas9 mutagenesis strategy was implemented in N. tabacum, with the aim of generating mutants in nuclear genes without expected visible phenotypes. This strategy led to the generation of a collection of independent mutants in the LIG4 (necessary for non-homologous end joining, NHEJ) and POLQ genes (necessary for microhomology mediated end joining, MMEJ). Targeting of other DSBR genes (KU70, KU80, RPA1C) generated mutants with unexpectedly strong developmental phenotypes.. These factors have telomeric roles, hinting towards a possible relationship between telomere length, and strength of developmental disruption upon loss of telomere structure in plants. The mutants were made in a genetic background encoding a plastid-encoded IGT reporter, that confers kanamycin resistance upon transfer to the nucleus. Through large scale independent experiments, increased IGT from the chloroplast to the nucleus was observed in lig4 mutants, as well as lines encoding a POLQ gene with a defective polymerase domain (polqΔPol). This shows that NHEJ or MMEJ have a double-sided relationship with IGT: while transferred genes may integrate using either pathway, the presence of both pathways suppresses IGT in wild-type somatic cells, thus demonstrating for the first time the extent on which nuclear genes control IGT frequency in plants. The IGT frequency increases in the mutants are likely mediated by increased availability of double strand breaks for integration. Additionally, kinetic analysis reveals that gene transfer (GT) events accumulate linearly as a function of time spent under antibiotic selection in the experiment, demonstrating that, contrary to what was previously thought, there is no such thing as a single GTR in somatic IGT experiments. Furthermore, IGT in tissue culture experiments appears to be the result of a "race against the clock" for integration in the nuclear genome, that starts when the organellar DNA arrives to the nucleus granting transient antibiotic resistance. GT events and escapes of kanamycin selection may be two possible outcomes from this race: those instances where the organellar DNA gets to integrate are recovered as GT events, and in those cases where timely integration fails, antibiotic resistance cannot be sustained, and end up considered as escapes. In the mutants, increased opportunities for integration in the nuclear genome change the overall ratio between IGT and escape events. The resources generated here are promising starting points for future research: (1) the mutant collection, for the further study of processes that depend on DNA repair in plants (2) the collection of GT lines obtained from these experiments, for the study of the effect of DSBR pathways over integration patterns and stability of transferred genes and (3) the developed CRISPR/Cas9 workflow for mutant generation, to make N. tabacum meet its potential as an attractive model for answering complex biological questions. Dazu wurde in N. tabacum eine CRISPR/Cas9-basierte Mutagenesestrategie angewandt, mit dem Ziel, Mutanten in Kerngenen zu erzeugen, für die keine sichtbaren Phänotypen zu erwarten sind. Diese Strategie führte zur Erzeugung einer Reihe unabhängiger Mutanten im LIG4-Gen (notwendig für „non-homologous end joining“, die nicht-homologe Verbindung von Enden, NHEJ) und POLQ (notwendig für „microhomology mediated end joining“, die Mikrohomologie-vermittelte Verbindung von Enden, MMEJ). Die gezielte Beeinflussung anderer DSBR-Gene (KU70, KU80, RPA1C) führte zu Mutanten mit unerwartet starken Entwicklungsphänotypen. Diese Gene spielen eine Rolle beim Erhalt der Telomere, was auf einen möglichen Zusammenhang zwischen der Telomerlänge und der Stärke der Entwicklungsstörung bei Verlust der Telomerstruktur in Pflanzen hindeutet. Die Mutanten wurden in einem genetischen Hintergrund erzeugt, der über einen in den Plastiden lokalisierten IGT-Reporter verfügt, der nach Übertragung in den Zellkern Kanamycin-Resistenz vermittelt. In groß angelegten unabhängigen Experimenten wurde in lig4-Mutanten sowie in Linien, die für ein POLQ-Gen mit einer defekten Polymerase-Domäne (polqΔPol) kodieren, eine erhöhte GTR vom Chloroplasten zum Zellkern beobachtet. Dies zeigt, dass NHEJ oder MMEJ eine zweischneidige Beziehung zum IGT haben: Während übertragene Gene folglich über jeden der beiden Mechanismen integriert werden können, unterdrückt das gleichzeitige Vorhandensein beider Wege IGT in somatischen Wildtyp-Zellen, wodurch zum ersten Mal gezeigt wird, in welchem Ausmaß Kerngene die IGT-Häufigkeit in Pflanzen kontrollieren. Die erhöhte Verfügbarkeit von Doppelstrangbrüchen für die Integration könnte für die erhöhte IGT-Häufigkeit in den Mutanten verantwortlich sein. Darüber hinaus zeigt die Analyse des Zeitverlaufs, dass Gentransferereignisse (GT) in Abhängigkeit von der Zeit, die im Experiment unter Antibiotikaselektion verbracht wurde, linear akkumulieren, was beweist, dass es, anders als bisher angenommen, in somatischen IGT-Experimenten keine statische GTR gibt. Darüber hinaus scheint IGT in Gewebekulturexperimenten das Ergebnis eines Wettlaufs mit der Zeit um die Integration in das Kerngenom zu sein, der beginnt, wenn die Organellen-DNA in den Zellkern gelangt und eine vorübergehende Antibiotikaresistenz gewährt. Echte GT-Ereignisse und „Escapes“ (scheinbare Resistenz, ein vorläufiges Ausweichen vor der Kanamycin-Selektion) können zwei mögliche Ergebnisse dieses Wettlaufs sein: Die Fälle, in denen die Organellen-DNA integriert wird, werden als GT-Ereignisse gewertet, und in den Fällen, in denen die rechtzeitige Integration scheitert, kann die Antibiotikaresistenz nicht aufrechterhalten werden und sie werden als „Escape“ betrachtet. In den Mutanten verändern sich die Möglichkeiten zur Integration in das Kerngenom, wodurch sich das Gesamtverhältnis zwischen IGT- und „Escape“-Ereignissen ändert. Die hier erzeugten Ressourcen sind vielversprechende Ausgangspunkte für künftige Forschungen: (1) die Mutantensammlung, für die Untersuchung von weiteren Prozessen, die von der DNA-Reparatur in Pflanzen abhängen, (2) die Sammlung von GT-Linien, die aus den hier beschriebenen Experimenten gewonnen wurden, für die Untersuchung der Auswirkungen von DSBR-Mechanismen auf Integrationsmuster und Stabilität übertragener Gene und (3) der hier entwickelte Arbeitsablauf für die Mutantenerzeugung mittels CRISPR/Cas9, damit N. tabacum sein Potenzial als attraktives Modell für die Beantwortung komplexer biologischer Fragestellungen erfüllen kann. Given the ubiquitous expression of GPER1, it is likely that the receptor plays a role in a variety of malignancies, not only in the classic hormonally regulated tissues (e.g., breast, ovary, and prostate), but also in the colon. As colorectal cancer (CRC) is the third most common cancer in both men and women worldwide and environmental factors and dietary habits are important risk factors, it is increasingly recognized that natural and synthetic hormones and their associated receptors might play a role in CRC. Through oral consumption, environmental contaminants with endocrine activity are in contact with the gastrointestinal mucosa, where they might exert their toxic effects. Although GPER1 has been shown to be engaged in physiological and pathophysiological processes, its role in CRC remains poorly understood. Thus, pro- as well as anti-tumorigenic effects are described in the literature. This thesis has uncovered novel roles of GPER1 in mediating major CRC-associated phenotypes in transformed and non-transformed colon cell lines. Exposure to the estrogens 17β-estradiol (E2), bisphenol-A (BPA) and diethylstilbestrol (DES) but also the androgen dihydrotestosterone (DHT) resulted in GPER1-dependent induction of supernumerary centrosomes, whole chromosomal instability (w-CIN) and aneuploidy. Indeed, both knockdown and inhibition of GPER1 attenuated the generation of (xeno)hormone-driven supernumerary centrosomes and karyotype instability. Mechanistically, (xeno)hormone-induced centrosome amplification was associated with transient multipolar mitosis and the generation of so called anaphase “lagging” chromosomes. The results of this thesis propose a GPER1/PKA/AKAP9-pathway in regulating centrosome numbers in colorectal cancer cells and the involvement of the centriolar protein centrin. Remarkably, exposure to (xeno)hormones resulted in atypical enlargement and unexpected phosphorylation of the centriole marker centrin in interphase. These findings provide a novel role for GPER1 in key CRC-prone lesions and shed light on underlying mechanisms that involve GPER1 function in the colon. Elucidating to what extent centrosomal proteins are involved in the GPER1-mediated aneugenic effect will be an important task for future studies. The present study was intended to lay a first foundation to understand the molecular basis and potential risk factors of CRC which might help to reduce the use of laboratory animals. Since numerous animal experiments are conducted in biomedical research, the development of alternative methods is indispensable. Obwohl gezeigt wurde, dass GPER1 an physiologischen und pathophysiologischen Prozessen beteiligt ist, ist seine Rolle im Darmkrebs nach wie vor unzureichend geklärt. In nature, many factors influence light availability for photosynthesis on different time scales, from shading by leaves within seconds up to seasonal changes over months. Variability of light energy supply for photosynthesis can limit a plant´s biomass accumulation. Plants have evolved multiple strategies to cope with strongly fluctuation light (FL). These range from long-term optimization of leaf morphology and physiology and levels of pigments and proteins in a process called light acclimation, to rapid changes in protein activity within seconds. Therefore, uncovering how plants deal with FL on different time scales may provide key ideas for improving crop yield. Photosynthesis is not an isolated process but tightly integrates with metabolism through mutual regulatory interactions. We thus require mechanistic understanding of how long-term light acclimation shapes both, dynamic photosynthesis and its interactions with downstream metabolism. To approach this, we analyzed the influence of growth light on i) the function of known rapid photosynthesis regulators KEA3 and VCCN1 in dynamic photosynthesis (Chapter 2-3) and ii) the interconnection of photosynthesis with photorespiration (PR; Chapter 4). We approached topic (i) by quantifying the effect of different growth light regimes on photosynthesis and photoprotection by using kea3 and vccn1 mutants. Firstly, we found that, besides photosynthetic capacity, the activities of VCCN1 and KEA3 during a sudden high light phase also correlated with growth light intensity. This finding suggests regulation of both proteins by the capacity of downstream metabolism. Secondly, we showed that KEA3 accelerated photoprotective non-photochemical quenching (NPQ) kinetics in two ways: Directly via downregulating the lumen proton concentration and thereby de-activating pH-dependent NPQ, and indirectly via suppressing accumulation of the photoprotective pigment zeaxanthin. For topic (ii), we analyzed the role of PR, a process which recycles a toxic byproduct of the carbon fixation reactions, in metabolic flexibility in a dynamically changing light environment. For this we employed the mutants hpr1 and ggt1 with a partial block in PR. We characterized the function of PR during light acclimation by tracking molecular and physiological changes of the two mutants. Our data, in contrast to previous reports, disprove a generally stronger physiological relevance of PR under dynamic light conditions. Additionally, the two different mutants showed pronounced and distinct metabolic changes during acclimation to a condition inducing higher photosynthetic activity. This underlines that PR cannot be regarded purely as a cyclic detoxification pathway for 2PG. Instead, PR is highly interconnected with plant metabolism, with GGT1 and HPR1 representing distinct metabolic modulators. In summary, the presented work provides further insight into how energetic and metabolic flexibility is ensured by short-term regulators and PR during long-term light acclimation. Fluktuationen in der Lichtenergieverfügbarkeit in der Natur kann die Biomasseakkumulation der Pflanzen limitieren. Pflanzen haben verschiedene Strategien entwickelt, um stark fluktuierendes Licht nutzen zu können. Diese reichen von der langfristigen Optimierung der Blattmorphologie und Physiologie und des Gehalts an Pigmenten und Proteinen in dem Prozess der Lichtakklimatisierung bis hin zu schnellen Veränderungen der Proteinaktivität innerhalb von Sekunden. Im ersten Themenkomplex (i) wurden die Auswirkungen verschiedener Wachstumslicht-bedingungen auf Photosynthese und Photoprotektion anhand von kea3- und vccn1-Mutanten quantifiziert. Zum einen konnten wir zeigen, dass neben der photosynthetischen Kapazität auch die Aktivitäten von VCCN1 und KEA3 während eines Hochlichtpulses mit der Wachstumslichtintensität korrelierten. Dies deutet auf eine Regulierung beider Proteine durch die Kapazität des Downstream-Metabolismus hin. Zum anderen beschleunigte KEA3 die Kinetik des photoprotektiven nicht-photochemischen Quenchings (NPQ) auf zweifache Weise: Direkt über die Herabregulierung der lumenalen Protonenkonzentration, was den pH-abhängigen NPQ deaktivierte, und indirekt über die Unterdrückung der Akkumulation des photoprotektiven Pigments Zeaxanthin. Für das zweite Thema (ii) untersuchten wir die Rolle des photorespiratorischen Metabolismus (PR), welcher ein toxisches Nebenprodukt der Kohlenstofffixierungsreaktionen recycelt, in der metabolischen Flexibilität in einer sich dynamisch verändernden Lichtumgebung. Dazu verwendeten wir die Mutanten hpr1 und ggt1 mit teilweise blockiertem PR Flux. Unsere Daten widerlegen, im Gegensatz zu früheren Berichten, eine allgemein größere physiologische Bedeutung von PR unter dynamischen Lichtbedingungen. Die beiden Mutanten zeigten ausgeprägte und distinkte metabolische Veränderungen während der Akklimatisierung an eine Bedingung mit höherer photosynthetischer Aktivität. Dies zeigt, dass PR nicht ausschließlich als zyklischer Entgiftungsweg für 2PG angesehen werden kann. Vielmehr ist PR tief in den pflanzlichen Stoffwechsel eingebettet, wobei GGT1 und HPR1 als distinkte Stellschrauben des Downstream-Metabolismus agieren. Among them, photosystem I (PSI), acting as plastocyanin-ferderoxin oxidoreductase, catalyzes the final reaction. Together with light-harvesting antenna I, PSI forms a high-molecular-weight supercomplex of ~600 kDa, consisting of eighteen subunits and nearly two hundred co-factors. Assembly of the various components into a functional thylakoid membrane complex requires precise coordination, which is provided by the assembly machinery. Although this includes a small number of proteins (PSI assembly factors) that have been shown to play a role in the formation of PSI, the process as a whole, as well as the intricacy of its members, remains largely unexplored. In the present work, two approaches were used to find candidate PSI assembly factors. First, EnsembleNet was used to select proteins thought to be functionally related to known PSI assembly factors in Arabidopsis thaliana (approach I), and second, co-immunoprecipitation (Co-IP) of tagged PSI assembly factors in Nicotiana tabacum was performed (approach II). Here, the novel PSI assembly factors designated CO-EXPRESSED WITH PSI ASSEMBLY 1 (CEPA1) and Ycf4-INTERACTING PROTEIN 1 (Y4IP1) were identified. A. thaliana null mutants for CEPA1 and Y4IP1 showed a growth phenotype and pale leaves compared with the wild type. Biophysical experiments using pulse amplitude modulation (PAM) revealed insufficient electron transport on the PSII acceptor side. Biochemical analyses revealed that both CEPA1 and Y4IP1 are specifically involved in PSI accumulation in A. thaliana at the post-translational level but are not essential. Consistent with their roles as factors in the assembly of a thylakoid membrane protein complex, the two proteins localize to thylakoid membranes. Remarkably, cepa1 y4ip1 double mutants exhibited lethal phenotypes in early developmental stages under photoautotrophic growth. Finally, co-IP and native gel experiments supported a possible role for CEPA1 and Y4IP1 in mediating PSI assembly in conjunction with other PSI assembly factors (e.g., PPD1- and PSA3-CEPA1 and Ycf4-Y4IP1). The fact that CEPA1 and Y4IP1 are found exclusively in green algae and higher plants suggests eukaryote-specific functions. Although the specific mechanisms need further investigation, CEPA1 and Y4IP1 are two novel assembly factors that contribute to PSI formation. Erstens wurde EnsembleNet verwendet, um Proteine auszuwählen, von denen angenommen wird, dass sie funktionell mit bekannten PSI-Assemblierungsfaktoren in Arabidopsis thaliana verwandt sind (Ansatz I), und zweitens wurde eine Co-Immunopräzipitation (Co-IP) von markierten PSI-Assemblierungsfaktoren in Nicotiana tabacum durchgeführt (Ansatz II). Dabei wurden die neuartigen PSI-Assemblierungsfaktoren mit der Bezeichnung CO-EXPRESSED WITH PSI ASSEMBLY 1 (CEPA1) und Ycf4-INTERACTING PROTEIN 1 (Y4IP1) identifiziert. A. thaliana Nullmutanten für CEPA1 und Y4IP1 zeigten einen Wachstumsphänotyp und blasse Blätter im Vergleich zum Wildtyp. Biophysikalische Experimente unter Verwendung der Pulsamplitudenmodulation (PAM) zeigten einen unzureichenden Elektronentransport auf der PSII-Akzeptorseite. Biochemische Analysen ergaben, dass sowohl CEPA1 als auch Y4IP1 spezifisch an der PSI-Akkumulation in A. thaliana auf posttranslationaler Ebene beteiligt, jedoch nicht essentiell sind. Entsprechend ihrer Rolle als Faktoren für den Aufbau eines Thylakoidmembran-Proteinkomplexes sind die beiden Proteine an Thylakoidmembranen lokalisiert. Bemerkenswerterweise wiesen cepa1 y4ip1-Doppelmutanten in frühen Entwicklungsstadien unter photoautotrophem Wachstum tödliche Phänotypen auf. Schließlich untermauerten Co-IP- und native Gelexperimente eine mögliche Rolle von CEPA1 und Y4IP1 bei der Vermittlung des PSI-Aufbaus in Verbindung mit anderen PSI-Aufbaufaktoren (z. B. While data inter-calibration techniques often yield continuous and reliable data for trend analysis, less attention is generally paid to maintaining higher-order statistics such as variance and autocorrelation. A growing body of work uses these metrics to quantify the stability or resilience of a system under study and potentially to anticipate an approaching critical transition in the system. Exploring the degree to which changes in resilience indicators such as the variance or autocorrelation can be attributed to non-stationary characteristics of the measurement process – rather than actual changes in the dynamical properties of the system – is important in this context. In this work we use both synthetic and empirical data to explore how changes in the noise structure of a data set are propagated into the commonly used resilience metrics lag-one autocorrelation and variance. We focus on examples from remotely sensed vegetation indicators such as vegetation optical depth and the normalized difference vegetation index from different satellite sources. We find that time series resulting from mixing signals from sensors with varied uncertainties and covering overlapping time spans can lead to biases in inferred resilience changes. These biases are typically more pronounced when resilience metrics are aggregated (for example, by land-cover type or region), whereas estimates for individual time series remain reliable at reasonable sensor signal-to-noise ratios. In addition to the ternary systems An(X)/GLU/C-S-H, also binary systems An(X)/C-S-H, GLU/C-S-H, and An(X)/GLU were studied. Complementary analytical techniques were applied to address the different specific aspects of the binary and ternary systems. Time-resolved laser-induced luminescence spectroscopy (TRLFS) was applied in combination with parallel factor analysis (PARAFAC) to identify retained species and to monitor species-selective sorption kinetics. ¹³C and ²⁹Si magic-angle-spinning (MAS) nuclear magnetic resonance (NMR) spectroscopy and X-ray photoelectron spectroscopy (XPS) were applied to determine the bulk structure and the composition of the C-S-H surface, respectively, in the absence and presence of GLU. The interaction of Th(IV) with GLU in different electrolytes was studied by capillary electrophoresis-inductively coupled plasma mass spectrometry (CE-ICP-MS). The influence of GLU on An(X) retention was investigated for a large concentration range up to 10⁻² M. The results showed that GLU had little to no effect on the overall An(X) retention by C-S-H with C/S of 0.8, regardless of the oxidation state of the actinides. For Eu(III), the TRLFS investigations additionally implied the formation of a Eu(III)-bearing precipitate with dissolved constituents of the C-S-H phase, which becomes structurally altered by the presence of GLU. For U(VI) sorption on the C-S-H phase, only a small influence of GLU could be established in the luminescence spectroscopic investigations, and no precipitation of U(VI)-containing secondary phases could be identified. From a scientific point of view, there is no definite consensus on what the Metaverse will be like. This paper collects, analyzes, and synthesizes scientific definitions and the accompanying major characteristics of the Metaverse using the methodology of a Systematic Literature Review (SLR). Two revised definitions for the Metaverse are presented, both condensing the key attributes, where the first one is rather simplistic holistic describing “a three-dimensional online environment in which users represented by avatars interact with each other in virtual spaces decoupled from the real physical world”. In contrast, the second definition is specified in a more detailed manner in the paper and further discussed. These comprehensive definitions offer specialized and general scholars an application within and beyond the scientific context of the system science, information system science, computer science, and business informatics, by also introducing open research challenges. Mesenchymal stromal cells (MSCs) play an important role in musculoskeletal homeostasis due to their broad differentiation potentials and their ability to support osteogenic and myogenic tissue maintenance and regeneration. In the bone, MSCs differentiate either into osteochondrogenic progenitors to form osteocytes and chondrocytes, or increasingly with age into adipogenic progenitors which give rise to bone-resident adipocytes. In skeletal muscle, during healthy regeneration MSCs provide regulatory signals that activate local, tissue-specific stem cells, known as satellite cells, which regenerate contractile myofibres. This process involves a significant cross-talk to immune cells stemming from both lymphoid and myeloid lineages. During ageing, muscle-resident MSCs undergo increased adipogenic lineage commitment, causing niche changes that contribute to fatty infiltration in muscles. These shifts in cell populations in bone lead to the loss of osteogenic cells and subsequently osteoporosis, or in muscle to impaired regeneration and to the development of sarcopenia. However, the signals that drive transition of MSCs into their respective cellular fates remain elusive. This thesis aims to elucidate the transcriptional shifts modulating cell states and cell types in musculoskeletal MSC fate determination. Single-cell RNA-sequencing (scRNA-seq) was used to characterise cell type-specific transcript regulation. State-of-the-art bioinformatics tools were combined with different analytical platforms that include both droplet-based scRNA-seq for large heterogeneous populations, and microfluidics-based scRNA-seq to assess small, rare subpopulations. For each platform, distinct computational pipelines were established including filtering steps to exclude low-quality cells, and data visualisation was performed by dimensionality reduction. Downstream analysis included clustering, cell type annotation, and differential gene expression to investigate transcriptional states in defined cell types during ageing and injury in the muscle and bone. Finally, a novel tool to assess publication activities in defined areas of research for the identified marker genes was developed. The results in the bone indicate that ageing MSCs increasingly commit towards an adipogenic fate at the expense of osteogenic specialisation. The data also suggests that significant cell population shifts of MSC-type fibro-adipogenic progenitors during muscle ageing underlie the pathologies observed in homeostatic and post-injury regenerative conditions. High-throughput visualisation of publication activity for candidate genes enabled more effective biological evaluation of scRNA-seq data. These results expose critical age-related changes in the stem cell niches of skeletal muscle and bone, highlight their respective sensitivity to nutrition and pathology, and elucidate novel factors that modulate stem cell-based regeneration. Targeting these processes might improve musculoskeletal health in the context of ageing and prevent the negative effects of pathological lineage determination. Mesenchymale Stromazellen (MSCs) spielen aufgrund ihres breiten Differenzierungspotenzials und ihrer Fähigkeit, myogene bzw. osteogene Regenerationsprozesse zu unterstützen, eine wichtige Rolle in der muskuloskelettalen Homöostase. Im Knochen differenzieren MSCs entweder zu osteochondrogenen Vorläufern, um Knochen- bzw. Knorpelzellen zu bilden. Oder mit zunehmendem Alter werden vermehrt adipogene Vorläufer gebildet, aus denen Knochen-Fettzellen entstehen. Im Skelettmuskel sezernieren MSCs während der Muskelregeneration beispielsweise regulatorische Signale, die lokale, gewebespezifische Stammzellen, sogenannte Satellitenzellen, aktivieren, und diese daraufhin die kontraktilen Muskelfasern regenerieren. Dieser Prozess umfasst bedeutsame Wechselwirkung von Stammzellen mit Immunzellen sowohl der lymphoiden als auch aus myeloischen Abstammungslinien. Während des Alterns erhalten muskelresidente MSCs jedoch ein erhöhtes adipogenenes Potential, welches Nischenveränderung verursacht und damit zu einer Fettinfiltration in den Muskeln beitragen kann. Die Verschiebungen der Zellpopulationen verursachen einerseits den Verlust von osteogenen Vorläufern und fördern degenerative Prozesse im Knochengewebe, die Osteoporose zur Folge haben, oder beeinträchtigen die Regeneration im Muskel sowie dessen Funktionalität, und können damit zur altersbedingten Sarkopenie beitragen. MSCs durchlaufen einen Entscheidungsprozess um final zu differenzieren, der jedoch bislang nur unzureichend charakterisiert ist. Um diesen Aspekt zu beleuchten, untersucht diese Dissertation die diesem Prozess zugrundeliegende Veränderung der Transkriptionsprofile, welche die Zellzustände und Zelltypen bei der Differenzierung von muskuloskelettalen MSCs steuern. Einzelzell-RNA-Sequenzierung (scRNA-Seq) wurde verwendet, um die zelltyp-spezifische Transkriptionsregulation zu charakterisieren. Moderne bioinformatische Analyse-Tools und -Plattformen wurden kombiniert, die sowohl droplet-basierte (für große heterogene Populationen) als auch mikrofluidik-basierte scRNA-seq (für kleine, seltene Subpopulationen), umfassten. Recent molecular research has focused on numerous approaches to increase recombinant protein yield in P. pastoris. For example, the design of expression vectors and synthetic genetic elements, gene copy number optimization, or co-expression of helper proteins (transcription factors, chaperones, etc.). However, high clonal variability of transformants and low screening throughput have hampered significant success. To enhance screening capacities, display-based methodologies inherit the potential for efficient isolation of producer clones via fluorescence-activated cell sorting (FACS). Therefore, this study focused on developing a novel clone selection method that is based on the non-covalent attachment of Fab fragments on the P. pastoris cell surface to be applicable for FACS. Initially, a P. pastoris display system was developed, which is a prerequisite for the surface capture of secreted Fabs. A Design of Experiments approach was applied to analyze the influence of various genetic elements on antibody fragment display. The combined P. pastoris formaldehyde dehydrogenase promoter (PFLD1), Saccharomyces cerevisiae invertase 2 signal peptide (ScSUC2), - agglutinin (ScSAG1) anchor protein, and the ARS of Kluyveromyces lactis (panARS) conferred highest display levels. Subsequently, eight single-chain variable fragments (scFv) specific for the constant part of the Fab heavy or light chain were individually displayed in P. pastoris. Among the tested scFvs, the anti-human CH1 IgG domain scFv allowed the most efficient Fab capture detected by flow cytometry. Irrespective of the Fab sequence, exogenously added as well as simultaneously secreted Fabs were successfully captured on the cell surface. Furthermore, Fab secretion capacities were shown to correlate to the level of surface-bound Fabs as demonstrated for characterized producer clones. Flow-sorted clones presenting high amounts of Fabs showed an increase in median Fab titers (factor of 21 to 49) compared to unsorted clones when screened in deep-well plates. For selected candidates, improved functional Fab yields of sorted cells vs. unsorted cells were confirmed in an upscaled shake flask production. Since the scFv capture matrix was encoded on an episomal plasmid with inherently unstable autonomously replicating sequences (ARS), efficient plasmid curing was observed after removing the selective pressure. Hence, sorted clones could be immediately used for production without the need to modify the expression host or vector. The resulting switchable display/secretion system provides a streamlined approach for the isolation of Fab producers and subsequent Fab production. Genetic diversities were similar among areas and a haplotype network did not show any geographic structure, while an analysis of molecular variance found evidence of genetic structure in this species. Pairwise FST and G'ST estimates and heterogeneity tests attributed this structure to weak but significant differentiation between areas 1W/1E and 2. A Mantel test and a high-resolution analysis of genetic diversity statistics showed a weak spatial cline of genetic differentiation. These findings could be reconciled by two possible stock structure scenarios: (1) a single population with kin-association affecting feeding ground preference and (2) two populations with feeding ground preference for either area 1W or area 2. An estimated dispersal rate between areas 1W and 2 indicates that both scenarios should be considered as a precautionary principle in stock assessments. Remarkably, these two developmental processes are controlled by similar molecular regulators and mechanisms. Given that FLC and SVP genes act as key flowering regulators in the model species Arabidopsis and in various other crop species, this study aimed at identifying FLC and SVP homologs in potato and investigating their roles in the regulation of plant development, with a particular focus on flowering and tuberization. Our analysis demonstrated that there are five FLC-like and three SVP like proteins encoded in the potato genome. The expression profiles of StFLCs and StSVPs throughout potato development and the detected interactions between their proteins indicate tissue specificity of the individual genes and distinct roles of a variety of putative protein complexes. In particular, we discovered that StFLC-D, as well as StFLC-B, StSVP-A, and StSVP-B play a complex role in the regulation of flowering time, as not only increased but also decreased levels of their transcripts promote earlier flowering. Most importantly, StFLC-D has a marked impact on tuberization under non-inductive conditions and susceptibility to temperature-induced tuber malformation, also known as second growth. Plants with decreased levels of StFLC-D demonstrated a strong ability to produce tubers under long days and appeared to be insensitive to temperature-induced second growth. Lastly, our data also suggests that StFLCs and StSVPs may be involved in the nitrogen-dependent regulation of potato development. Based on content analysis of 103 studies, we propose “relational harmony,” “task complementarity,” and “relational scope” as three categories to integrate the mechanisms and arguments used in the literature to theorize about the implications of multiplexity. We then survey the literature in light of this typology to show how it is also useful in revealing patterns of theorizing; for example, with respect to the types of relationships that are studied in relation to multiplexity. We conclude with suggestions for future research directions, focusing on how these can be pursued based on our integrative typology. While the question of ensuring individual negotiation success has been the subject and topic of multidisciplinary research for a long time, the question of how organizations can implement and ensure continuous negotiation success remains largely unexplored. This dissertation therefore aims to investigate how companies enable their employees to consistently achieve better negotiation outcomes. It is significant that, in the corporate context, negotiators do not act as individuals but as embedded representatives of an organization, and that negotiations are not one-time events but recurring necessities for the existence of the organization instead. In organizations, those recurring processes with a similar fundamental structure are handled by routines. A planned improvement of routines is often forced by new artifacts. In this context, artifacts refer to human-created technologies with which humans interact within routines and therefore artifacts have a central influence on executing the routine. If negotiation activities in companies are represented by organizational routines, one central issue for improving companies’ negotiation performance is the artifacts’ incorporation into organizational negotiation routines that facilitate the efficient application of the insights from negotiation research. The dissertation consists of three studies that were written as research papers to examine artifacts in the organizational negotiation context. The first study focuses on the pre-negotiation stage and presents four tools to assist negotiation practitioners in efficiently preparing for negotiation. The study examines the negotiation preparation’s effectiveness and efficiency and the negotiation outcome in a case-based experiment. The second study is devoted to a closer examination of the barriers that inhibit the adoption of negotiation support systems (NSSs) as one kind of organizational negotiation artifact. The investigation is conducted using a structural equation model based on information from participating practitioners. The third study is concerned with the future of negotiation support system research. An exploratory study based on qualitative in-depth interviews with proven and published experts in the field aims to evaluate the current state of research. Currently, various hypotheses are circulating to explain the exceptionally favourable transport properties of LHPs. Seeking an explanation for the low non-radiative recombination rates and long carrier lifetimes is particularly interesting to the halide perovskites research community. The first part of this work investigates the two main hypotheses that are believed to play a significant role: the existence of a giant Rashba effect and large polarons. The experimental method of ARPES is mainly applied to verify their credibility. The first hypothesis presumes that a giant Rashba effect restricts the recombination losses of the charge carriers by making the band gap slightly indirect. The Rashba effect is based on a strong SOC that could appear in LHPs thanks to incorporating the heavy element Pb in their structure. Earlier experimental work had pointed out this effect at the VBM of a hybrid LHP as a viable explanation for the long lifetimes of the charge carriers. My systematic ARPES studies on hybrid MAPbBr3 and spin-resolved ARPES studies on the inorganic CsPbBr3 disprove the presence of any Rashba effect in the VBM of the reported order of magnitude. Therefore, neither the spin texture nor an indirect band gap character at the VBM in the bulk or at the surface can explain the high efficiency of LHP. In case of existence, this effect is in terms of the Rashba parameter at least a factor of a hundred smaller than previously assumed. The second hypothesis proposes large polaron formation in the electronic structure of LHPs and attributes it to their high defect tolerance and low non-radiative recombination rate. Because the perovskite structure consists of negative and positive ions, polarons of this kind can be expected due to the Coulomb interaction between carriers and the polar lattice at intermediate electron-phonon coupling strength. Their existence is proposed to screen the carriers and defects to avoid recombination and trapping, thus leading to long carrier lifetimes. ARPES results by one group supported this assumption, reporting a 50% effective mass enhancement over the theoretical effective mass for CsPbBr3 in the orthorhombic structure. The current thesis examines this hypothesis experimentally by photon-energy-dependent ARPES spectra and theoretically by GW band calculations of CsPbBr3 perovskites. The investigation is based on the fact that a polaron contribution in charge transport can become evident by an increase of the effective mass as measured by ARPES over the calculated one without polaron effects. However, my experiments on crystalline CsPbBr3 did not imply a larger effective mass for which one could postulate large polarons. In fact, the effective masses determined from ARPES agree with that of theoretical predictions. The second part of my thesis thoroughly investigates the possibility of spontaneously magnetizing LHPs by introducing Mn2+ ions. Mn doping was reported to cause ferromagnetism in one of the most common LHPs, MAPbI3, mediated by super-exchange. The current work investigates the magnetic properties of a wide concentration range of Mn-doped MAPbI3 and triple-cation films by XAS, XMCD, and SQUID measurements. Based on the XAS line shape and a sum-rule analysis of the XMCD spectra, a pure Mn2+ configuration has been confirmed. Negative Curie temperatures are extracted from fitting the magnetization with a Curie-Weiss law. However, a remanent magnetization, which would be an indication of the absence of ferromagnetism down to 2K. As far as the double exchange is concerned, the element-specific XAS excludes a sufficient amount of Mn3+ as a prerequisite for this mechanism. All the findings show no evidence of significant double exchange or ferromagnetism in Mn-doped LHPs. The magnetic behavior is paramagnetic rather than ferromagnetic. In the dissertation's last chapter, orthorhombic features of CsPbBr3 are revealed by ARPES, including an extra VBM at the Γ-point. The VBM of CsPbBr3 shows a temperature-dependent splitting, which decreases by 190 meV between 38K and 300K and tracks a shift of a saddle point at the cubic M-point. It is possible to reproduce the energy shift using an atomic model with a larger unit cell for room temperature, allowing local inversion symmetry breaking. This indicates the importance of electric dipoles for the inorganic LHPs, which may contribute to their high efficiency by breaking inversion symmetry and a Berry-phase effect. Derzeit kursieren zahlreiche Hypothesen, um die außergewöhnlich guten Transporteigenschaften von LHPs zu erklären. Die Suche nach einer Erklärung für die niedrigen strahlungslosen Rekombinationsraten und die langen Ladungsträgerlebensdauern ist von besonderem Interesse für die Forschercommunity der Halogenidperowskite. Der erste Teil dieser Arbeit untersucht die beiden Haupthypothesen, von denen angenommen wird, dass sie eine wichtige Rolle spielen: die Existenz eines riesigen Rashba-Effekts und großer Polaronen. Hier wird hauptsächlich die experimentelle Methode der ARPES angewandt, um ihre Glaubwürdigkeit zu überprüfen. Die erste Hypothese geht davon aus, dass ein riesiger Rashba-Effekt die Rekombinationsverluste der Ladungsträger einschränkt, indem er die Bandlücke leicht indirekt macht. Der Rashba-Effekt basiert auf einer starken Spin-Bahn-Wechselwirkung, die in LHPs, dank der Präsenz des schweren Elements Pb in ihrer Struktur, potenziell auftreten könnte. Eine frühere experimentelle Arbeit hatte diesen Effekt am VBM eines hybriden LHP als mögliche Erklärung für die langen Lebensdauern der Ladungsträger vorgeschlagen. Meine systematischen ARPES-Studien am hybriden MAPbBr3 sowie die winkel- und spinaufgelösten ARPES-Studien am anorganischen CsPbBr3 widerlegen das Vorhandensein eines riesigen Rashba-Effekts im VBM in der angegebenen Größenordnung. Daher können weder die Spin-Textur noch der indirekte Bandlückencharakter am VBM im Volumen oder an der Oberfläche die hohe Effizienz von LHP erklären. Dieser Effekt ist, falls er existiert, mindestens um einen Faktor hundert kleiner als bisher angenommen. Die zweite Hypothese geht von der Bildung großer Polaronen in der elektronischen Struktur von LHPs aus, welche zu ihrer hohen Defekttoleranz und niedrigen strahlungslosen Rekombination srate beitragen soll. Da die Perowskit Struktur aus negativen und positiven Ionen besteht, sind solche Polaronen wegen der Coulomb-Wechselwirkung zwischen Ladungsträger und Ionengitter bei mittlerer Stärke der Elektron-Phonon-Kopplung zu erwarten. Es wird angenommen, dass sie die Ladungsträger und Defekte abschirmen, was Rekombination und Trapping verhindert und zu langen Ladungsträgerlebensdauern führt. Die ARPES-Ergebnisse einer Gruppe stützen diese Annahme und zeigen, dass die effektive Masse von CsPbBr3 um 50% höher ist als die theoretische effektive Masse für die orthorhombische Phase. In der vorliegenden Arbeit wird diese Hypothese experimentell mit Hilfe von Photonenenergie-abhängigen ARPES-Spektren und theoretisch mit Hilfe von GW-Bandstrukturberechnungen an CsPbBr3-Perowskiten untersucht. Denn der Beitrag von Polaronen zum Ladungstransport lässt sich durch eine Zunahme der mit ARPES gemessenen effektiven-Masse nachweisen. Meine Experimente an kristallinem CsPbBr3 ergaben jedoch keine erhöhte effektive Masse, für die man große Polaronen postulieren könnte. Tatsächlich stimmen die aus ARPES ermittelten effektiven Massen gut mit den theoretischen Vorhersagen überein. Der zweite Teil meiner Dissertation untersucht die Möglichkeit, LHPs durch den Einbau von Mn2+ Ionen spontan zu magnetisieren. Es wurde berichtet, dass die Mn-Dotierung in einem der häufigsten LHPs - \MAPbI3 - Ferromagnetismus durch "Superaustausch" hervorruft. Außerdem berichteten zwei weitere Arbeiten über Ferromagnetismus sogar bei Raumtemperatur, hervorgerufen jedoch durch Doppalaustausch. In der vorliegenden Arbeit werden die magnetischen Eigenschaften eines weiten Konzentrationsbereichs von Mn-dotiertem MAPbI3 und Dreifachkation-Filmen mittels XAS, XMCD und SQUID-Messungen untersucht. Basierend auf der XAS-Linienform und der Summenregelanalyse der XMCD-Spektren wurde eine reine Mn2+ Konfiguration bestätigt. Negative Curie-Temperaturen werden aus einem Fit der Magnetisierung mit dem Curie-Weiss-Gesetz abgeleitet. Eine remanente Magnetisierung, die auf Ferromagnetismus hindeuten würde, wird jedoch bis hinunter zu 2K nicht beobachtet. Was den Doppelaustausch betrifft, so schließt die elementspezifische XAS eine ausreichende Menge an Mn3+ als Voraussetzung für diesen Mechanismus aus. Nach all diesen Erkenntnissen gibt es keinen Hinweis auf einen signifikanten Doppelaustausch oder Ferromagnetismus in Mn-dotierten LHP. Das magnetische Verhalten ist eher paramagnetisch als ferromagnetisch. Im letzten Kapitel der Dissertation werden orthorhombische ARPES-Strukturen bei CsPbBr3 beobachtet, einschließlich eines zusätzlichen VBM am Γ-Punkt. Das VBM von CsPbBr3 zeigt eine temperaturabhängige Aufspaltung, die zwischen 38K und 300 K um 190 meV abnimmt und von einer Verschiebung eines Sattelpunktes am kubischen M-Punkt stammt. A problem is modeled using a rule based language and then grounded and solved. Solving results in a set of stable models that correspond to solutions of the modeled problem. In this thesis, we present the design and implementation of the clingo system---perhaps, the most widely used ASP system. It features a rich modeling language originating from the field of knowledge representation and reasoning, efficient grounding algorithms based on database evaluation techniques, and high performance solving algorithms based on Boolean satisfiability (SAT) solving technology. The contributions of this thesis lie in the design of the modeling language, the design and implementation of the grounding algorithms, and the design and implementation of an Application Programmable Interface (API) facilitating the use of ASP in real world applications and the implementation of complex forms of reasoning beyond the traditional ASP workflow. We begin by providing nationally representative evidence that managers’ risk-aversion is negatively correlated with the likelihood that their firms engage in any worker training. Using a novel vignette study, we then demonstrate that risk-tolerant and risk-averse decision makers have significantly different training preferences. Risk aversion results in increased sensitivity to turnover risk. Managers who are risk-averse offer less general training and are more reluctant to train workers with a history of job mobility. Adopting a weighting approach to flexibly control for observed differences in the characteristics of risk-averse and risk-tolerant managers, we show that our findings cannot be explained by heterogeneity in either managers’ observed characteristics or the type of firms where they work. All managers, irrespective of their risk preferences, are sensitive to the investment risk associated with training, avoiding training that is more costly or that targets those with less occupational expertise or nearing retirement. This provides suggestive evidence that the risks of training are primarily due to the risk that trained workers will leave the firm (turnover risk) rather than the risk that the benefits of training do not outweigh the costs (investment risk). To increase the usability of this scale within a clinical and health services research context, we aimed to validate a German short version (12 items) of this scale in individuals with depression including the examination of the relations from need frustration and need satisfaction to ill-being and quality of life (QOL). This cross-sectional study involved 344 adults diagnosed with depression (Mage (SD) = 47.5 years (11.1); 71.8% females). Confirmatory factor analyses indicated that the short version of the BPNSFS was not only reliable, but also fitted a six-factor structure (i.e., satisfaction/frustration X type of need). Subsequent structural equation modeling showed that need frustration related positively to indicators of ill-being and negatively to QOL. Surprisingly, need satisfaction did not predict differences in ill-being or QOL. The short form of the BPNSFS represents a practical instrument to measure need satisfaction and frustration in people with depression. Further, the results support recent evidence on the importance of especially need frustration in the prediction of psychopathology. In a panel estimation for OECD countries between 1970 and 2016, although inequality is, on average countercyclical and significantly associated with the capital share, one-third of the countries display a pro- or noncyclical relationship. To analyze the observed pattern, we incorporate distributive shocks into an RBC model, where agents are ex ante heterogeneous with respect to wealth and ability. We find that whether wealth and income inequality behave countercyclically or not depends on the elasticity of intertemporal substitution and the persistence of shocks. We match the model to quarterly US data using Bayesian techniques. The parameter estimates point toward a non-monotonic relationship between productivity and inequality fluctuations. On impact, inequality increases in response to TFP shocks but subsequently declines. Furthermore, TFP shocks explain 17% of inequality fluctuations. We argue that this reasoning is problematic: It involves arbitrary thresholds, and the results are potentially confounded by the different surface orders of the compared structures and non-syntactic factors. We present three experiments with designs that do not require defining thresholds of 'low' or 'high' coreference values. Instead, we focus on grammatical contrasts (wh-movement vs. relativization, subject vs. object wh-movement) and aim to identify and reduce confounds. The results show that reconstruction for A'-movement of DPs is not very robust in German, contra previous findings. Our results are compatible with the view that the surface order and non-syntactic factors (e.g. plausibility, referential accessibility of an R-expression) heavily influence coreference possibilities. Thus, the data argue against a theory that includes both reconstruction and a hard Condition C constraint. There is a residual contrast between sentences with subject/object movement, which is compatible with an account without reconstruction (and an additional non-syntactic factor) or an account with reconstruction (and a soft Condition C constraint). We derive second-best policy rules for CDR subsidies and carbon taxes when no global carbon price exists but a national government implements a unilateral climate policy. We find that the optimal carbon tax differs from an optimal CDR subsidy because of carbon leakage and a balance of resource trade effect. First, the optimal removal subsidy tends to be larger than the carbon tax because of lower supply-side leakage on fossil resource markets. Second, net carbon exporters exacerbate this wedge to increase producer surplus of their carbon resource producers, implying even larger removal subsidies. Third, net carbon importers may set their removal subsidy even below their carbon tax when marginal environmental damages are small, to appropriate producer surplus from carbon exporters. Prior studies have shown that those who disclose health-related information across OSNs often tend to regret it and delete their publications afterwards. Hence, deleted posts containing sensitive data can be seen as manifestations of online regrets. In this work, we present an analysis of deleted content on Twitter during the outbreak of the COVID-19 pandemic. For this, we collected more than 3.67 million tweets describing COVID-19 symptoms (e.g., fever, cough, and fatigue) posted between January and April 2020. We observed that around 24% of the tweets containing personal pronouns were deleted either by their authors or by the platform after one year. As a practical application of the resulting dataset, we explored its suitability for the automatic classification of regrettable content on Twitter.
