TY - THES A1 - Höllerer, Reinhard T1 - Modellierung und Optimierung von Bürgerdiensten am Beispiel der Stadt Landshut T1 - Modeling and optimization of civil services on the example of the city of Landshut N2 - Die Projektierung und Abwicklung sowie die statische und dynamische Analyse von Geschäftsprozessen im Bereich des Verwaltens und Regierens auf kommunaler, Länder- wie auch Bundesebene mit Hilfe von Informations- und Kommunikationstechniken beschäftigen Politiker und Strategen für Informationstechnologie ebenso wie die Öffentlichkeit seit Langem. Der hieraus entstandene Begriff E-Government wurde in der Folge aus den unterschiedlichsten technischen, politischen und semantischen Blickrichtungen beleuchtet. Die vorliegende Arbeit konzentriert sich dabei auf zwei Schwerpunktthemen: • Das erste Schwerpunktthema behandelt den Entwurf eines hierarchischen Architekturmodells, für welches sieben hierarchische Schichten identifiziert werden können. Diese erscheinen notwendig, aber auch hinreichend, um den allgemeinen Fall zu beschreiben. Den Hintergrund hierfür liefert die langjährige Prozess- und Verwaltungserfahrung als Leiter der EDV-Abteilung der Stadtverwaltung Landshut, eine kreisfreie Stadt mit rund 69.000 Einwohnern im Nordosten von München. Sie steht als Repräsentant für viele Verwaltungsvorgänge in der Bundesrepublik Deutschland und ist dennoch als Analyseobjekt in der Gesamtkomplexität und Prozessquantität überschaubar. Somit können aus der Analyse sämtlicher Kernabläufe statische und dynamische Strukturen extrahiert und abstrakt modelliert werden. Die Schwerpunkte liegen in der Darstellung der vorhandenen Bedienabläufe in einer Kommune. Die Transformation der Bedienanforderung in einem hierarchischen System, die Darstellung der Kontroll- und der Operationszustände in allen Schichten wie auch die Strategie der Fehlererkennung und Fehlerbehebung schaffen eine transparente Basis für umfassende Restrukturierungen und Optimierungen. Für die Modellierung wurde FMC-eCS eingesetzt, eine am Hasso-Plattner-Institut für Softwaresystemtechnik GmbH (HPI) im Fachgebiet Kommunikationssysteme entwickelte Methodik zur Modellierung zustandsdiskreter Systeme unter Berücksichtigung möglicher Inkonsistenzen (Betreuer: Prof. Dr.-Ing. Werner Zorn [ZW07a, ZW07b]). • Das zweite Schwerpunktthema widmet sich der quantitativen Modellierung und Optimierung von E-Government-Bediensystemen, welche am Beispiel des Bürgerbüros der Stadt Landshut im Zeitraum 2008 bis 2015 durchgeführt wurden. Dies erfolgt auf Basis einer kontinuierlichen Betriebsdatenerfassung mit aufwendiger Vorverarbeitung zur Extrahierung mathematisch beschreibbarer Wahrscheinlichkeitsverteilungen. Der hieraus entwickelte Dienstplan wurde hinsichtlich der erzielbaren Optimierungen im dauerhaften Echteinsatz verifiziert. N2 - The project development and implementation as well as the static and dynamic analysis of business processes in the context of administration and governance on a municipal, federal state or national level by information and communication technology, has concerned the media along with politicians and strategists for information technology as well as the general public for a long time. The here-from defined term of E-Government has been examined as the focal point of discussion from most diverse technical, political and semantic perspectives. The present work focuses on two main topics: • The first main topic approaches the development of a hierarchical architecture model for which seven hierarchical layers can be identified. These seem to be necessary as well as sufficient to describe the general case. The background is provided by the long-term processual and administrative experience as head of the IT department at the municipality of Landshut, an independent city with 69.000 inhabitants located in the north-east of Munich. It is representative of many administrative processes in the Federal Republic of Germany, but nonetheless still manageable concerning its complexity and quantity of processes. Therefore, static and dynamic structures can be extracted from the analysis of all core workflows and modelled abstractly. The emphases lie on the description of the existing operating procedures in a municipality. The transformation of the operating requirements in a hierarchical system, the modeling of the control and operational states within all layers, as well as the strategy of error recognition and troubleshooting create a transparent basis for extensive restructuring and optimisation. For modeling was used FMC-eCS, a methodology for the description of state discrete systems, developed at the Hasso-Plattner-Institute for Software Systems Engineering (HPI) in the subject area of communication systems. Furthermore inconsistent system statuses are taken into consideration (advisor: Prof. Dr.-Ing. Werner Zorn [ZW02, …, ZW10]). • The second main topic focuses on the quantitative modeling and optimisation of the E-Government process chain at Landshut´s Citizens Advice Bureau (2008 up to 2015). This is based on a continuous data acquisition with sophisticated pre-processing to extract mathematically describable probability distributions. Während ihrer Einsatzdauer unterliegen POFs unterschiedlichen Arten von Umweltbeanspruchungen, hauptsächlich durch hohe Temperatur, hohe Feuchtigkeit und mechanischen Belastungen. Zahlreiche experimentelle Forschungen beschäftigten sich mit der standardisierten Prüfung der Zuverlässigkeit von im Handel erhältlichen Fasern. Jedoch gab es bisher wenig Erfolg bei der Bemühung, zwei grundlegende optische Erscheinungen, Absorption und Streuung, die die Lichtausbreitung in Fasern stark beeinflussen, zu verstehen und praktisch zu modellieren: Diese beiden Effekte beschreiben nicht nur die Qualität neuer Fasern, sondern sie werden auch stark durch die Alterungsprozess beeinflusst. Der Hauptzweck dieser Doktorarbeit war es, ein praktisch verwendbares und theoretisch gut fundiertes Modell der Lichtausbreitung in nicht gealterten und gealterten POFs zu entwickeln und es durch optische Experimente zu verifizieren. Dabei wurden anwendungsorientierte Aspekte mit theoretischer POF-Modellierung kombiniert. Die Arbeit enthält die erste bekannte Anwendung der Wellenanalyse zur Untersuchung der winkelabhängigen Eigenschaften der Streuung in Lichtwellenleitern. Für die praktischen Experimente wurden mehrere POF-Proben unterschiedlicher Hersteller künstlich gealtert, indem sie bis 4500 Stunden bei 100 °C gelagert wurden (ohne Feuchtekontrolle). Die Parameter der jeweiligen Simulationen wurden mittels einer systematischen Optimierung an die gemessen optischen Eigenschaften der gealterten Proben angeglichen. Die Resultate deuten an, dass der Übertragungsverlust der gealterten Fasern in den ersten Tagen und Wochen der Alterung am stärksten durch eine wesentliche physikalische Verschlechterung der Kern-Mantel-Grenzfläche verursacht wird. Chemische Effekte des Alterungsprozesses scheinen im Faserkernmaterial zuerst nach einigen Monaten aufzutreten. N2 - This thesis discusses theoretical and practical aspects of modelling of light propagation in non-aged and aged step-index polymer optical fibres (POFs). Special attention has been paid in describing optical characteristics of non-ideal fibres, scattering and attenuation, and in combining application-oriented and theoretical approaches. The precedence has been given to practical issues, but much effort has been also spent on the theoretical analysis of basic mechanisms governing light propagation in cylindrical waveguides. As a result a practically usable general POF model based on the raytracing approach has been developed and implemented. A systematic numerical optimisation of its parameters has been performed to obtain the best fit between simulated and measured optical characteristics of numerous non-aged and aged fibre samples. The model was verified by providing good agreement, especially for the non-aged fibres. The relations found between aging time and optimal values of model parameters contribute to a better understanding of the aging mechanisms of POFs. Die Untersuchung dieser Prozesse in Raum und Zeit kann mit Hilfe geeigneter Modelle erfolgen. Ziele dieser Arbeit sind: i) die Entwicklung eines geeigneten Modellansatzes durch Kombination von Teilmodellen zur Simulation des N-Transportes im Boden und Grundwasser von Tieflandeinzugsgebieten und ii) die Untersuchung von Wechselwirkungen zwischen Gebietseigenschaften und N-Transport unter besonderer Berücksichtigung der potentiellen N-Zufuhr in die Oberflächengewässer. Der Modellansatz basiert auf der Kombination verschiedener Teilmodelle: das Bodenwasser- und -stickstoffmodell mRISK-N, das Grundwassermodell MODFLOW und das Stofftransportmodell RT3D. Zur Untersuchung der Wechselwirkungen mit den Gebietseigenschaften muss die Verteilung und Verfügbarkeit von Reaktionspartnern berücksichtigt werden. Dazu wurde ein Reaktionsmodul entwickelt, welches chemische Prozesse im Grundwasser simuliert. Hierzu gehören die Mineralisation organischer Substanz durch Sauerstoff, Nitrat und Sulfat sowie die Pyritoxidation durch Sauerstoff und Nitrat. Der Modellansatz wurde in verschiedenen Einzelstudien angewandt, wobei jeweils bestimmte Teilmodelle im Vordergrund stehen. Alle Modellstudien basieren auf Daten aus dem Schaugrabeneinzugsgebiet (ca. 25 km²), in der Nähe von Osterburg(Altmark) im Norden Sachsen-Anhalts. Die folgenden Einzelstudien wurden durchgeführt: i) Evaluation des Bodenmodells anhand von Lysimeterdaten, ii) Modellierung eines Tracerexperimentes im Feldmaßstab als eine erste Anwendung des Reaktionsmoduls, iii) Untersuchung hydraulisch-chemischer Wechselwirkungen an einem 2D-Grundwassertransekt, iv) Flächenverteilte Modellierung von Grundwasserneubildung und Bodenstickstoffaustrag im Untersuchungsgebiet als Eingangsdaten für nachfolgende Grundwassersimulationen, und v) Untersuchung der Ausbreitung von Nitrat im Grundwasser und des Durchbruchs in die Oberflächengewässer im Untersuchungsgebiet auf Basis einer 3D-Modellierung von Grundwasserströmung und reaktivem Stofftransport. Die Modellstudien zeigen, dass der Modellansatz geeignet ist, die Wechselwirkungen zwischen Stofftransport und –umsatz und den hydraulisch-chemischen Gebietseigenschaften zu modellieren. Die Ausbreitung von Nitrat im Sediment wird wesentlich von der Verfügbarkeit reaktiver Substanzen sowie der Verweilzeit im Grundwasserleiter bestimmt. Bei der Simulation des Untersuchungsgebietes wurde erst nach 70 Jahren eine der gegebenen Eintragssitutation entsprechende Nitratkonzentration im Grundwasserzustrom zum Grabensystem erreicht (konservativer Transport). Die Berücksichtigung von reaktivem Stofftransport führt zu einer deutlichen Reduktion der Nitratkonzentrationen. Die Modellergebnisse zeigen, dass der Grundwasserzustrom die beobachtete Nitratbelastung im Grabensystem nicht erklären kann, da der Großteil des Nitrates durch Denitrifikation verloren geht. Andere Quellen, wie direkte Einträge oder Dränagenzuflüsse müssen ebenfalls in Betracht gezogen werden. Die Prognosefähigkeit des Modells für das Untersuchungsgebiet wird durch die Datenunsicherheiten und die Schätzung der Modellparameter eingeschränkt. Dennoch ist der Modellansatz eine wertvolle Hilfe bei der Identifizierung von belastungsrelevanten Teilflächen (Stoffquellen und -senken) sowie bei der Modellierung der Auswirkungen von Managementmaßnahmen oder Landnutzungsveränderungen auf Grundlage von Szenario-Simulationen. Der Modellansatz unterstützt auch die Interpretation von Beobachtungsdaten, da so die lokalen Informationen in einen räumlichen und zeitlichen Zusammenhang gestellt werden können. N2 - Chemical transformations and hydraulic processes in soil and groundwater often lead to an apparent retention of nitrate in lowland catchments. Models are needed to evaluate the interaction of these processes in space and time. The objectives of this study are i) to develop a specific modelling approach by combining selected modelling tools simulating N-transport and turnover in soils and groundwater of lowland catchments, ii) to study interactions between catchment properties and nitrogen transport. Special attention was paid to potential N-loads to surface waters. The modelling approach combines various submodels for water flow and solute transport in soil and groundwater: The soil-water- and nitrogen-model mRISK-N, the groundwater flow model MODFLOW and the solute transport model RT3D. In order to investigate interactions of N-transport and catchment characteristics, the distribution and availability of reaction partners have to be taken into account. Therefore, a special reaction-module is developed, which simulates various chemical processes in groundwater, such as the degradation of organic matter by oxygen, nitrate, sulphate or pyrite oxidation by oxygen and nitrate. The model approach is applied to different simulation, focussing on specific submodels. All simulation studies are based on field data from the Schaugraben catchment, a pleistocene catchment of approximately 25 km², close to Osterburg(Altmark) in the North of Saxony-Anhalt. The following modelling studies have been carried out: i) evaluation of the soil-water- and nitrogen-model based on lysimeter data, ii) modelling of a field scale tracer experiment on nitrate transport and turnover in the groundwater as a first application of the reaction module, iii) evaluation of interactions between hydraulic and chemical aquifer properties in a two-dimensional groundwater transect, iv) modelling of distributed groundwater recharge and soil nitrogen leaching in the study area, to be used as input data for subsequent groundwater simulations, v) study of groundwater nitrate distribution and nitrate breakthrough to the surface water system in the Schaugraben catchment area and a subcatchment, using three-dimensional modelling of reactive groundwater transport. The various model applications prove the model to be capable of simulating interactions between transport, turnover and hydraulic and chemical catchment properties. The distribution of nitrate in the sediment and the resulting loads to surface waters are strongly affected by the amount of reactive substances and by the residence time within the aquifer. In the Schaugraben catchment simulations, it is found that a period of 70 years is needed to raise the average seepage concentrations of nitrate to a level corresponding to the given input situation, if no reactions are considered. Under reactive transport conditions, nitrate concentrations are reduced effectively. Simulation results show that groundwater exfiltration does not contribute considerably to the nitrate pollution of surface waters, as most nitrate entering soils and groundwater is lost by denitrification. Additional sources, such as direct inputs or tile drains have to be taken into account to explain surface water loads. The prognostic value of the models for the study site is limited by uncertainties of input data and estimation of model parameters. Nevertheless, the modelling approach is a useful aid for the identification of source and sink areas of nitrate pollution as well as the investigation of system response to management measures or landuse changes with scenario simulations. The modelling approach assists in the interpretation of observed data, as it allows to integrate local observations into a spatial and temporal framework. Both SECs involve diffusion as approach for hillslope processes and the stream power law to reflect riverbed incision. The 1D SEC settles sediment that was produced by fluvial incision in the appropriate minimum, while the supply-limited 2D SEC DANSER uses a fast filling algorithm to model sedimantation. It is based on a cellular automaton. A slope-dependent factor in the sediment flux extends the diffusion equation to nonlinear diffusion. The discharge accumulation is achieved with the D8-algorithm and an improved drainage accumulation routine. Lateral incision enhances the incision's modelling. Following empirical laws, it incises channels of several cells width. The coupling method enables different temporal and spatial resolutions of the SEC and the thermo-mechanical code. It transfers vertical as well as horizontal displacements to the surface model. A weighted smoothing of the 3D surface displacements is implemented. The smoothed displacement vectors transmit the deformation by bilinear interpolation to the surface model. These interpolation methods ensure mass conservation in both directions and prevent the two surfaces from drifting apart. The presented applications refer to the evolution of the Pamir orogen. A calibration of DANSER's parameters with geomorphological data and a DEM as initial topography highlights the advantage of lateral incision. Preserving the channel width and reflecting incision peaks in narrow channels, this closes the huge gap between current orogen-scale incision models and observed topographies. River capturing models in a system of fault-bounded block rotations reaffirm the importance of the lateral incision routine for capturing events with channel initiation. The models show a low probability of river capturings with large deflection angles. While the probability of river capturing is directly depending on the uplift rate, the erodibility inside of a dip-slip fault speeds up headward erosion along the fault: The model's capturing speed increases within a fault. Coupling DANSER with the thermo-mechanical code SLIM 3D emphasizes the versatility of the SEC. While DANSER has minor influence on the lithospheric evolution of an indenter model, the brittle surface deformation is strongly affected by its sedimentation, widening a basin in between two forming orogens and also the southern part of the southern orogen to south, east and west. N2 - Im Rahmen dieser Studie werden 1D und 2D Erosionsmodelle im Gebirgsmaßstab implementiert und mit Modellen für tektonische Deformation gekoppelt. Die Kopplungsmethode erlaubt unterschiedlich räumliche und zeitliche Auflösungen im tektonischen und im Erosionsmodell. Es werden sowohl vertikale als auch horizontale Bewegungen zwischen den Modellen transferiert. Darüber hinaus enthält die Kopplungsmethode ein Glättungsverfahren, um eventuelle Instabilitäten des tektonischen Modelles zu kompensieren. Beide Erosionsmodelle beziehen Hangerosion, Flusseinschneidung und Sedimentation ein. Der 1D Code nutzt Hack's Law, um die Wassermengen zu berechnen. Er garantiert Massenerhaltung, indem er Sedimente in Senken speichert. Das 2D Erosionsmodell DANSER basiert auf einem zellulären Automaten. Ein zusätzlicher steigungsabhängiger Faktor erweitert lineare zu nichtlinearer Diffusion. Wassermengen werden mit Hilfe des D8-Algorithmus und einer veränderten Form von O'Callaghans (1984) Algorithmus akkumuliert. Laterale Einschneidung, berechnet durch einen neuen Verteilungs-Algorithmus, verbessert die Modellierung von Flusssystemen. Flüsse sind dabei repräsentiert durch eine unterschiedliche Anzahl an Zellen orthogonal zur Fließrichtung. Ihre Breite wird nach empirischen Gesetzen ermittelt. Die präsentierten Anwendungen dienen der Studie des Pamirgebirges. Zunächst werden die Modellparameter anhand von Einschneidungs- und Erosionsraten sowie Sedimentdurchflüssen kalibriert. Ein digitales Höhenmodell dient als Anfangstopographie und zur Extraktion von Flussprofilen. Laterale Einschneidung zeigt eine deutliche Verbesserung zu bisher vorhandenen Modellen. Sie ermöglicht die Erhaltung der Flussbreite und zeigt hohe Einschneidungsraten in engen Flusspassagen. Modelle von Flussanzapfungen in einem System paralleler Verwerfungen bestätigen die Wichtigkeit von lateraler Einschneidung für Flussanzapfungsmodelle, die Hangerosion einbeziehen. Während die Modelle eine geringe Wahrscheinlichkeit von Flussanzapfungen mit hohem Ablenkungswinkel zeigen, belegen sie auch, dass deren (allgemeine) Wahrscheinlichkeit direkt von der Hebungsrate der Verwerfungen abhängt. Die Erodibilität beschleunigt lediglich die Geschwindigkeit von Flussanzapfungen. Ein Modell, das die Codes SLIM 3D und DANSER koppelt, dokumentiert die vielseitige Verwendbarkeit des neuen Codes: Es zeigt einen geringen Einfluss von Oberflächenprozessen auf die Lithosphärendeformation, während die Sedimentationsroutine erheblich auf spröde Oberflächendeformationen einwirkt. Das Modell legt nahe, dass Sedimentation ein zwischen zwei entstehenden Gebirgen gelegenes Becken weitet. Außerdem weitet sich der südlich von der interkontinentalen Kollisionszone gelegene Teil des Gebirge-Models ebenfalls durch Sedimentation. Während das forschende Lernen als normative Theorie das sollen beschreibt, geben die digitalen Werkzeuge, alte wie neue, das können in vielen Bereichen vor. In der vorliegenden Arbeit wird ein Prozessmodell aufgestellt, was den Versuch unternimmt, das forschende Lernen hinsichtlich interaktiver, gruppenbasierter Prozesse zu systematisieren. Basierend auf dem entwickelten Modell wurde ein Softwareprototyp implementiert, der den gesamten Forschungsprozess begleiten kann. Dabei werden Gruppenformation, Feedback- und Reflexionsprozesse und das Peer Assessment mit Bildungstechnologien unterstützt. Die Entwicklungen wurden in einem qualitativen Experiment eingesetzt, um Systemwissen über die Möglichkeiten und Grenzen der digitalen Unterstützung von forschendem Lernen zu gewinnen. N2 - Research-based learning and digital transformation are two of the most important influences on the development of pedagogies of universities in German-speaking countries. While research-based learning as a normative theory explains the should, the digital tools, both old and new, provide the can in many areas. In the present work, a process model was developed, which captures research-based learning with regard to interactive, group-based processes. Based on this model a software prototype was implemented that can accompany the entire research process. Thereby group formation, feedback and reflection processes and peer assessment are supported with educational technology. The developed software was used in a qualitative experiment to gain knowledge about the possibilities and limits of digital support for research-based learning. Klimaänderungen, ob natürlich entstanden oder anthropogen verursacht, äußern sich in erster Linie durch Änderungen der Auftrittswahrscheinlichkeiten der natürlichen Regime. In der vorliegenden Arbeit wurden dynamische Mechanismen des Regimeverhaltens und der dekadischen Klimavariabilität der Atmosphäre bei Abwesenheit zeitlich veränderlicher externer Einflussfaktoren untersucht. Das Hauptwerkzeug dafür war ein quasi-geostrophisches Dreischichtenmodell der winterlichen atmosphärischen Zirkulation auf der Nordhemisphäre, das eine spektrale T21-Auflösung, einen orographischen und einen zeitlich konstanten thermischen Antrieb mit nicht-zonalen Anteilen besitzt. Ein solches Modell vermag großskalige atmosphärische Strömungsvorgänge außerhalb der Tropen mit einiger Genauigkeit zu simulieren. Nicht berücksichtigt werden Feuchteprozesse, die Wechselwirkung der Atmosphäre mit anderen Teilen des Klimasystems sowie anthropogene Einflüsse. Für das Dreischichtenmodell wurde ein automatisiertes, iteratives Verfahren zur Anpassung des thermischen Modellantriebs neu entwickelt. Jede Iteration des Verfahrens besteht aus einer Testintegration des Modells, ihrer Auswertung, dem Vergleich der Ergebnisse mit den NCEP-NCAR-Reanalysedaten aus den Wintermonaten Dezember, Januar und Februar sowie einer auf diesem Vergleich basierenden Antriebskorrektur. Nach Konvergenz des Verfahrens stimmt das Modell sowohl bezüglich des zonal gemittelten Klimazustandes als auch bezüglich der zeitgemittelten nicht-zonalen außertropischen diabatischen Erwärmung nahezu perfekt mit den wintergemittelten Reanalysedaten überein. In einer 1000-jährigen Simulation wurden die beobachtete mittlere Zirkulation im Winter sowie ihre Variabilität realitätsnah reproduziert, insbesondere die Arktische Oszillation (AO) und ihre vertikale Ausdehnung. Der AO-Index des Modells weist deutliche dekadische Schwankungen auf, die allein durch die interne Modelldynamik bedingt sind. Darüber hinaus zeigt das Modell ein Regimeverhalten, das gut mit den Beobachtungsdaten übereintimmt. Es besitzt ein Regime, das in etwa der negativen Phase der Nordatlantischen Oszillation (NAO) entspricht und eines, das der positiven Phase der AO ähnelt. Eine weit verbreitete Hypothese ist die näherungsweise Übereinstimmung zwischen Regimen und stationären Lösungen der Bewegungsgleichungen. In der vorliegenden Arbeit wurde diese Hypothese für das Dreischichtenmodell überprüft, mit negativem Resultat. Es wurden mittels eines Funktionalminimierungsverfahrens sechs verschiedene stationäre Zustände gefunden. Diese sind allesamt durch eine äußerst unrealistische Zirkulation gekennzeichnet und sind daher weit vom Modellattraktor entfernt. Fünf der sechs Zustände zeichnen sich durch einen extrem starken subtropischen Jet in der mittleren und obereren Modellschicht aus. Da die Ursache des Regimeverhaltens des Dreischichtenmodells nach wie vor unklar war, wurde auf ein einfacheres Modell, nämlich ein barotropes Modell mit T21-Auflösung zurückgegriffen. Für die Anpassung des Oberflächenantriebs wurde eine modifizierte Form der iterativen Prozedur verwendet. Die zeitgemittelte Zirkulation des barotropen Modells stimmt sehr gut mit der zeitlich und vertikal gemittelten Zirkulation des Dreischichtenmodells überein. Das dominierende räumliche Muster der Variabilität besitzt eine AO-ähnliche Struktur. Zudem besitzt das barotrope Modell zwei Regime, die näherungsweise der positiven und negativen Phase der AO entsprechen und somit auch den Regimen des Dreischichtenmodells ähneln. Im Verlauf der Justierung des Oberflächenantriebs konnte beobachtet werden, dass die zwei Regime des barotropen Modells durch die Vereinigung zweier koexistierender Attraktoren entstanden. Der wahrscheinliche Mechanismus der Attraktorvereinigung ist eine Randkrise eines der beiden Attraktoren, gefolgt von einer explosiven Bifurkation des anderen Attraktors. Es wird die Hypothese aufgestellt, dass der beim barotropen Modell vorgefundene Mechanismus der Regimeentstehung für atmosphärische Zirkulationsmodelle mit realitätsnahem Regimeverhalten Allgemeingültigkeit besitzt. Gestützt wird die Hypothese durch vier Experimente mit dem Dreischichtenmodell, bei denen jeweils der Parameter der Bodenreibung verringert und die Antriebsanpassung wiederholt wurde. Bei diesen Experimenten erhöhte sich die Persistenz und die Separiertheit der Regime bei abnehmender Reibung drastisch und damit auch der Anteil dekadischer Zeitskalen an der Variabilität. Die Zunahme der Persistenz der Regime ist charakteristisch für die Annäherung an eine inverse innere Krise, deren Existenz aber nicht nachgewiesen werden konnte. N2 - Preferred quasi-stationary states of the planetary-scale atmospheric circulation, which may persist for one or several weeks, are referred to as atmospheric circulation regimes. Climate variations, either natural or anthropogenic, manifest themselves mainly in changes of the frequencies of occurrence of the natural regimes. In the presented work, dynamical mechanisms of regime behavior and decadal climate variability of the atmosphere in absence of time-varying external forcing factors have been examined using a quasi-geostrophic three-level model of the wintertime atmospheric circulation over the northern hemisphere. This model has spectral T21 resolution, an orographic and a time-constant thermal forcing including non-zonal components. Such kind of a model is able to simulate large-scale extratropical atmospheric processes with reasonable accuracy. However, moisture processes, the interaction between the atmosphere and other parts of the climate system, and anthropogenic influences are not accounted for. For the three-level model, a novel, automated, iterative procedure for the tuning of the thermal forcing has been developed. Every iteration of the procedure consists of a model test run, its evaluation, the comparison of the results with NCEP-NCAR reanalysis data for the winter months December, January, and February, and a forcing correction based on this comparison. After convergence of the procedure, the model matches the reanalysis data almost perfectly, as far as it concerns the zonal mean climate state and the time-mean non-zonal extratropical diabatic heating. In a 1000-year simulation, the observed time-mean circulation in winter as well as its variability have been reproduced with considerable realism, in particular the Arctic Oscillation (AO) and its deep vertical extent. The modeled AO index exhibits pronounced decadal variations, exclusively caused by internal model dynamics. Furthermore, the model's regime behavior is in good agreement with observations. It possesses one regime resembling the negative phase of the North Atlantic Oscillation (NAO) and another resembling the positive phase of the AO. A well-known hypothesis is the approximate correspondence between regimes and stationary solutions of the equations of motion. In the presented work, this hypothesis has been checked for the three-level model, but with negative result. Using a functional minimization method, six steady states have been found. All of them correspond to an extremely unrealistic circulation, and thus they are far away from the model's attractor. Five of the six steady states are characterized by a strongly exaggerated subtropical jet in the middle and upper model level. As the origin of regime behavior was still unclear, a simpler model, namely a T21 barotropic model, has been reverted to. For the adaptation of the surface forcing, a modified version of the tuning procedure has been applied. The time-mean circulation of the barotropic model matches the temporally and vertically averaged circulation of the three-level model very well. The dominant spatial pattern of variability has an AO-like structure. Furthermore, the barotropic model possesses two regimes which approximately correspond to the positive and negative AO phase and therefore resemble the regimes of the three-level model. During the tuning of the surface forcing it has been observed that the two regimes of the barotropic model have emerged from the unification of two coexisting attractors. The mechanism responsible for this attractor merging is probably a boundary crisis of one of these attractors, followed by an explosive bifurcation of the other attractor. It is hypothesized that the mechanism of regime genesis found in the barotropic model is universally valid for atmospheric circulation models with realistic regime behavior. This hypothesis is supported by four experiments with the three-level model, where the surface friction parameter has been decreased and the tuning procedure has been repeated, respectively. In these experiments, the persistence and separation of the regimes increases dramatically with decreasing friction, and thereby the fraction of decadal-scale variability. The increase of regime persistence is characteristic of approaching an inverse interior crisis, the existence of which, however, could not be proven. There are, however, numerous challenges associated with the development of mAbs as therapeutics. Mechanistic understanding of factors that govern the pharmacokinetics (PK) of mAbs is critical for drug development and the optimisation of effective therapies; in particular, adequate dosing strategies can improve patient quality life and lower drug cost. Physiologically-based PK (PBPK) models offer a physiological and mechanistic framework, which is of advantage in the context of animal to human extrapolation. Unlike for small molecule drugs, however, there is no consensus on how to model mAb disposition in a PBPK context. Current PBPK models for mAb PK hugely vary in their representation of physiology and parameterisation. Their complexity poses a challenge for their applications, e.g., translating knowledge from animal species to humans. In this thesis, we developed and validated a consensus PBPK model for mAb disposition taking into account recent insights into mAb distribution (antibody biodistribution coefficients and interstitial immunoglobulin G (IgG) pharmacokinetics) to predict tissue PK across several pre-clinical species and humans based on plasma data only. The model allows to a priori predict target-independent (unspecific) mAb disposition processes as well as mAb disposition in concentration ranges, for which the unspecific clearance (CL) dominates target-mediated CL processes. This is often the case for mAb therapies at steady state dosing. The consensus PBPK model was then used and refined to address two important problems: 1) Immunodeficient mice are crucial models to evaluate mAb efficacy in cancer therapy. Protection from elimination by binding to the neonatal Fc receptor is known to be a major pathway influencing the unspecific CL of both, endogenous and therapeutic IgG. The concentration of endogenous IgG, however, is reduced in immunodeficient mouse models, and this effect on unspecific mAb CL is unknown, yet of great importance for the extrapolation to human in the context of mAb cancer therapy. 2) The distribution of mAbs into solid tumours is of great interest. To comprehensively investigate mAb distribution within tumour tissue and its implications for therapeutic efficacy, we extended the consensus PBPK model by a detailed tumour distribution model incorporating a cell-level model for mAb-target interaction. We studied the impact of variations in tumour microenvironment on therapeutic efficacy and explored the plausibility of different mechanisms of action in mAb cancer therapy. The mathematical findings and observed phenomena shed new light on therapeutic utility and dosing regimens in mAb cancer treatment. Mechanistisches Verständnis der Pharmakokinetik (PK) von mAK ist wichtig für die Arzneimittelentwicklung sowie für die Therapieoptimierung. Adäquate Dosierungsstrategien können die Lebensqualität der Patienten erhöhen und die Gesundheitskosten senken. Physiologie-basierte PK (PBPK) Modelle bieten einen physiologischen und mechanistischen Rahmen für die Extrapolation von Tiermodellen auf den Menschen. Im Gegensatz zu kleinen chemischen Molekülen besteht für die PBPK Modellierung von mAK kein Konsens: Aktuelle Modelle unterscheiden sich stark hinsichtlich Physiologie und deren Parameterisierung. Die Komplexität dieser Modelle stellt eine große Herausforderung für ihre Anwendung dar. In der vorliegenden Arbeit entwickelten und validierten wir ein Konsens-PBPK-Modell für die mAK Disposition. Dabei wurden aktuelle Erkenntnisse zur mAK Verteilung berücksichtigt, um basierend auf Plasmadaten Vorhersagen für die PK im Gewebe verschiedener präklinischer sowie klinischer Spezies zu treffen. Das Modell erlaubt a priori Vorhersagen für die unspezifische (target-unabhängige) mAK Disposition als auch für die mAK Disposition in einem Konzentrationsbereich, für den die unspezifische Clearance (CL) die target-abhängige CL dominiert. Dies ist oft der Fall für mAK Therapien bei Steady-state-Dosierung. Changes in climate and land use affect terrestrial biosphere structure and functioning at present, and will likely impact on the terrestrial carbon balance during the coming decades - potentially providing a positive feedback to the climate system due to soil carbon releases under a warmer climate. Quantifying changes, and the associated uncertainties, in regional terrestrial carbon budgets resulting from these effects is relevant for the scientific understanding of the Earth system and for long-term climate mitigation strategies. A model describing the relevant processes that govern the terrestrial carbon cycle is a necessary tool to project regional carbon budgets into the future. This study (1) provides an extensive evaluation of the parameter-based uncertainty in model results of a leading terrestrial biosphere model, the Lund-Potsdam-Jena Dynamic Global Vegetation Model (LPJ-DGVM), against a range of observations and under climate change, thereby complementing existing studies on other aspects of model uncertainty; (2) evaluates different hypotheses to explain the age-related decline in forest growth, both from theoretical and experimental evidence, and introduces the most promising hypothesis into the model; (3) demonstrates how forest statistics can be successfully integrated with process-based modelling to provide long-term constraints on regional-scale forest carbon budget estimates for a European forest case-study; and (4) elucidates the combined effects of land-use and climate changes on the present-day and future terrestrial carbon balance over Europe for four illustrative scenarios - implemented by four general circulation models - using a comprehensive description of different land-use types within the framework of LPJ-DGVM. This study presents a way to assess and reduce uncertainty in process-based terrestrial carbon estimates on a regional scale. The results of this study demonstrate that simulated present-day land-atmosphere carbon fluxes are relatively well constrained, despite considerable uncertainty in modelled net primary production. Process-based terrestrial modelling and forest statistics are successfully combined to improve model-based estimates of vegetation carbon stocks and their change over time. Application of the advanced model for 77 European provinces shows that model-based estimates of biomass development with stand age compare favourably with forest inventory-based estimates for different tree species. Driven by historic changes in climate, atmospheric CO2 concentration, forest area and wood demand between 1948 and 2000, the model predicts European-scale, present-day age structure of forests, ratio of biomass removals to increment, and vegetation carbon sequestration rates that are consistent with inventory-based estimates. Alternative scenarios of climate and land-use change in the 21st century suggest carbon sequestration in the European terrestrial biosphere during the coming decades will likely be on magnitudes relevant to climate mitigation strategies. However, the uptake rates are small in comparison to the European emissions from fossil fuel combustion, and will likely decline towards the end of the century. Uncertainty in climate change projections is a key driver for uncertainty in simulated land-atmosphere carbon fluxes and needs to be accounted for in mitigation studies of the terrestrial biosphere. Quantitative Abschätzungen der Wirkung dieser Einflussfaktoren - sowie der mit ihnen verbundenen Unsicherheit - auf die terrestrische Kohlenstoffbilanz sind daher sowohl für das Verständnis des Erdsystems, als auch für eine langfristig angelegte Klimaschutzpolitik relevant. Um regionale Kohlenstoffbilanzen in die Zukunft zu projizieren, sind Modelle erforderlich, die die wesentlichen Prozesse des terrestrischen Kohlenstoffkreislaufes beschreiben. Die vorliegende Arbeit (1) analysiert die parameterbasierte Unsicherheit in Modellergebnissen eines der führenden globalen terrestrischen Ökosystemmodelle (LPJ-DGVM) im Vergleich mit unterschiedlichen ökosystemaren Messgrößen, sowie unter Klimawandelprojektionen, und erweitert damit bereits vorliegende Studien zu anderen Aspekten der Modelunsicherheit; (2) diskutiert unter theoretischen und experimentellen Aspekten verschiedene Hypothesen über die altersbedingte Abnahme des Waldwachstums, und implementiert die vielversprechenste Hypothese in das Model; (3) zeigt für eine europäische Fallstudie, wie Waldbestandsstatistiken erfolgreich für eine verbesserte Abschätzung von regionalen Kohlenstoffbilanzen in Wäldern durch prozessbasierten Modelle angewandt werden können; (4) untersucht die Auswirkung möglicher zukünftiger Klima- und Landnutzungsänderungen auf die europäische Kohlenstoffbilanz anhand von vier verschiedenen illustrativen Szenarien, jeweils unter Berücksichtigung von Klimawandelprojektionen vier verschiedener Klimamodelle. Eine erweiterte Version von LPJ-DGVM findet hierfür Anwendung, die eine umfassende Beschreibung der Hauptlandnutzungstypen beinhaltet. Die vorliegende Arbeit stellt einen Ansatz vor, um Unsicherheiten in der prozessbasierten Abschätzung von terrestrischen Kohlenstoffbilanzen auf regionaler Skala zu untersuchen und zu reduzieren. Die Ergebnisse dieser Arbeit zeigen, dass der Nettokohlenstoffaustausch zwischen terrestrischer Biosphäre und Atmosphäre unter heutigen klimatischen Bedingungen relativ sicher abgeschätzt werden kann, obwohl erhebliche Unsicherheit über die modelbasierte terrestrische Nettoprimärproduktion existiert. Prozessbasierte Modellierung und Waldbestandsstatistiken wurden erfolgreich kombiniert, um verbesserte Abschätzungen von regionalen Kohlenstoffvorräten und ihrer Änderung mit der Zeit zu ermöglichen. Die Anwendung des angepassten Modells in 77 europäischen Regionen zeigt, dass modellbasierte Abschätzungen des Biomasseaufwuchses in Wäldern weitgehend mit inventarbasierten Abschätzungen für verschiede Baumarten übereinstimmen. Unter Berücksichtigung von historischen Änderungen in Klima, atmosphärischem CO2-Gehalt, Waldfläche und Holzernte (1948-2000) reproduziert das Model auf europäischer Ebene die heutigen, auf Bestandsstatistiken beruhenden, Abschätzungen von Waldaltersstruktur, das Verhältnis von Zuwachs und Entnahme von Biomasse, sowie die Speicherungsraten im Kohlenstoffspeicher der Vegetation. These interaction are directly or indirectly impacted by a number of factors. These factors range from physiological characteristics of individual organisms, over specifics of their interaction to impacts of the environment. They may generate the potential for the application of different strategies by predators and prey. Within this thesis, I modelled predator-prey interactions and investigated a broad range of different factors driving the application of certain strategies, that affect the individuals or their populations. In doing so, I focused on phytoplankton-zooplankton systems as established model systems of predator-prey interactions. At the level of predator physiology I proposed, and partly confirmed, adaptations to fluctuating availability of co-limiting nutrients as beneficial strategies. These may allow to store ingested nutrients or to regulate the effort put into nutrient assimilation. We found that these two strategies are beneficial at different fluctuation frequencies of the nutrients, but may positively interact at intermediate frequencies. The corresponding experiments supported our model results. We found that the temporal structure of nutrient fluctuations indeed has strong effects on the juvenile somatic growth rate of {\itshape Daphnia}. Predator colimitation by energy and essential biochemical nutrients gave rise to another physiological strategy. High-quality prey species may render themselves indispensable in a scenario of predator-mediated coexistence by being the only source of essential biochemical nutrients, such as cholesterol. Thereby, the high-quality prey may even compensate for a lacking defense and ensure its persistence in competition with other more defended prey species. We found a similar effect in a model where algae and bacteria compete for nutrients. Now, being the only source of a compound that is required by the competitor (bacteria) prevented the competitive exclusion of the algae. In this case, the essential compounds were the organic carbon provided by the algae. Here again, being indispensable served as a prey strategy that ensured its coexistence. The latter scenario also gave rise to the application of the two metabolic strategies of autotrophy and heterotrophy by algae and bacteria, respectively. We found that their coexistence allowed the recycling of resources in a microbial loop that would otherwise be lost. Instead, these resources were made available to higher trophic levels, increasing the trophic transfer efficiency in food webs. The predation process comprises the next higher level of factors shaping the predator-prey interaction, besides these factors that originated from the functioning or composition of individuals. Here, I focused on defensive mechanisms and investigated multiple scenarios of static or adaptive combinations of prey defense and predator offense. I confirmed and extended earlier reports on the coexistence-promoting effects of partially lower palatability of the prey community. When bacteria and algae are coexisting, a higher palatability of bacteria may increase the average predator biomass, with the side effect of making the population dynamics more regular. This may facilitate experimental investigations and interpretations. If defense and offense are adaptive, this allows organisms to maximize their growth rate. Besides this fitness-enhancing effect, I found that co-adaptation may provide the predator-prey system with the flexibility to buffer external perturbations. On top of these rather internal factors, environmental drivers also affect predator-prey interactions. I showed that environmental nutrient fluctuations may create a spatio-temporal resource heterogeneity that selects for different predator strategies. I hypothesized that this might favour either storage or acclimation specialists, depending on the frequency of the environmental fluctuations. We found that many of these factors promote the coexistence of different strategies and may therefore support and sustain biodiversity. Thus, they might be relevant for the maintenance of crucial ecosystem functions that also affect us humans. Besides this, the richness of factors that impact predator-prey interactions might explain why so many species, especially in the planktonic regime, are able to coexist. N2 - Organismen interagieren miteinander und mit ihrer Umwelt. Innerhalb dieses Netzwerks von Interaktionen sind Fraßbeziehungen zwischen Räubern und ihrer Beute von zentraler Bedeutung. Sie werden auf verschiedenen Ebenen von unterschiedlichen Faktoren beeinflusst, was zur Ausprägung von diversen Strategien von Räuber oder Beute führen kann. Diese Faktoren und die Strategien die sie hervor bringen sind Gegenstand dieser Doktorarbeit. In mehreren Modellierungsstudien habe ich vielseitige Faktoren untersucht, die sich dem Aufbau einzelner Organismen, Verteidigungs- und Angriffsmechanismen sowie Umwelteinflüssen zuordnen lassen. Dabei konzentrierte ich mich auf ein etabliertes Modellsystem zur Erforschung von Räuber-Beute-Dynamiken und untersuchte die Fraßbeziehung zwischen Phytoplankton als Beute und Zooplankton als Räuber. Ich fand heraus, dass die Bereitstellung von essentiellen Ressourcen für Konkurrenten oder Räuber eine Strategie sein kann, mit der Beutearten sich vor dem Aussterben schützen können. Auch die direkte Verteidigung gegen den Räuber ist eine häufige Strategie zur Verringerung des Fraßdrucks und kann ebenfalls Koexistenz fördern. Für anpassungsfähige Verteidigung der Beute und Angriffsstärke des Räubers konnte ich zeigen, dass dies sowohl die Fitness erhöhen, als auch die Robustheit der Räuber-Beute-Dynamiken gegen äußere Störungen erhöhen kann. Weiterhin fand ich heraus, dass physiologische Anpassungsmechanismen wie Speicherung oder anpassungsfähige Aufnahme von Nährstoffen die Wachstumsrate des Räubers verbessern können, wenn die Qualität der verfügbaren Beute in der Umwelt des Räubers fluktuiert. Viele der Strategien, die ich in dieser Arbeit herausgestellt habe, können die Koexistenz von verschiedenen Arten fördern und damit zu erhöhter Biodiversität beitragen, welche wiederum entscheidend ist für die Stabilität von Ökosystemen und deren Nutzbarkeit. The Mesozoic Salta Rift extensional phase has created a dominant structural and lithological framework that controls Cenozoic deformation and exhumation patterns in the Central Andes. Studying the nature of these pre-existing anisotropies is a key to understanding the spatiotemporal distribution of exhumation and its controlling factors. The Eastern Cordillera in particular, has a structural grain that is in part controlled by Salta Rift structures and their orientation relative to Andean shortening. As a result, there are areas in which Andean deformation prevails and areas where the influence of the Salta Rift is the main control on deformation patterns. Between 23 and 24°S, lithological and structural heterogeneities imposed by the Lomas de Olmedo sub-basin (Salta Rift basin) affect the development of the Eastern Cordillera fold-and-thrust belt. The inverted northern margin of the sub-basin now forms the southern boundary of the intermontane Cianzo basin. The former western margin of the sub-basin is located at the confluence of the Subandean Zone, the Santa Barbara System and the Eastern Cordillera. Here, the Salta Rift basin architecture is responsible for the distribution of these morphotectonic provinces. In this study we use a multi-method approach consisting of low-temperature (U-Th-Sm)/He and apatite fission track thermochronology, detrital geochronology, structural and sedimentological analyses to investigate the Mesozoic structural inheritance of the Lomas de Olmedo sub-basin and Cenozoic exhumation patterns. Characterization of the extension-related Tacurú Group as an intermediate succession between Paleozoic basement and the syn-rift infill of the Lomas de Olmedo sub-basin reveals a Jurassic maximum depositional age. Zircon (U-Th-Sm)/He cooling ages record a pre-Cretaceous onset of exhumation for the rift shoulders in the northern part of the sub-basin, whereas the western shoulder shows a more recent onset (140–115 Ma). Variations in the sedimentary thickness of syn- and post-rift strata document the evolution of accommodation space in the sub-basin. While the thickness of syn-rift strata increases rapidly toward the northern basin margin, the post-rift strata thickness decreases toward the margin and forms a condensed section on the rift shoulder. Inversion of Salta Rift structures commenced between the late Oligocene and Miocene (24–15 Ma) in the ranges surrounding the Cianzo basin. The eastern and western limbs of the Cianzo syncline, located in the hanging wall of the basin-bounding Hornocal fault, show diachronous exhumation. At the same time, western fault blocks of Tilcara Range, south of the Cianzo basin, began exhuming in the late Oligocene to early Miocene (26–16 Ma). Eastward propagation to the frontal thrust and to the Paleozoic strata east of the Tilcara Range occurred in the middle Miocene (22–10 Ma) and the late Miocene–early Pliocene (10–4 Ma), respectively. Die mesozoische Salta Rift Extensionsphase hat einen strukturellen und lithologischen Rahmen geschaffen, der die känozoischen Heraushebungsmuster in den Zentralanden kontrolliert. Die Charakterisierung dieser Anisotropien ist daher entscheidend, um die räumlich-zeitliche Verteilung der Heraushebung und ihrer kontrollierenden Faktoren zu verstehen. Insbesondere die Östliche Kordillere weist einen strukturellen Rahmen auf, der teilweise von Salta Rift-Strukturen und ihrer Orientierung in Bezug auf die Verkürzung im Zuge der Gebirgsbildung der Anden kontrolliert wird. Dadurch wurden Gebiete geschaffen, in denen die jüngere Anden-Deformation überwiegt, und Gebiete, in denen der Einfluss des Salta Rifts die Deformationsmuster prägt. Zwischen 23 und 24°S beeinflussen lithologische und strukturelle Heterogenitäten des Lomas de Olmedo Beckens (Teil des Salta Rift Beckens) die Entwicklung des Faltengürtels der Östlichen Kordillere. Der invertierte nördliche Rand des Beckens bildet dabei die südliche Grenze des Cianzo Beckens, welches während der andinen Orogenese angelegt wurde. Der ehemalige westliche Rand des Lomas de Olmedo Beckens befindet sich am Übergang der Subandinen Zone, des Santa Barbara Systems und der Östlichen Kordillere. Hier ist die Architektur des Salta Rift-Beckens für die räumliche Verteilung dieser morphotektonischen Provinzen verantwortlich. In dieser Studie verwenden wir einen multi-methodischen Ansatz, bestehend aus Niedertemperatur (U-Th-Sm)/He und Apatit Spaltspur Thermochronologie, detritische Geochronologie sowie strukturelle und sedimentologische Analyse, um das mesozoische strukturelle Erbe des Lomas de Olmedo Beckens und die känozoischen Heraushebungsmuster zu untersuchen. Die mit Extension verbundene Tacurú-Gruppe bildet eine Einheit, die dem paläozoischen Grundgebirge und der syn-rift Auffüllung des Lomas de Olmedo Beckens zwischengeschaltet ist. Sie hat ein Jurassisches maximales Ablagerungsalter. Zirkon (U-Th-Sm)/He Abkühlungsalter zeigen einen präkretazischen Beginn der Heraushebung für die Riftschulter im nördlichen Teil des Beckens, während die westliche Schulter einen jüngeren Beginn aufweist (140–115 Ma). Variationen in der stratigraphischen Mächtigkeit von Syn- und Postrift-Gesteinen dokumentieren die Entwicklung des Akommodationsraums. Während die Mächtigkeit der Synrift-Gesteine zum nördlichen Beckenrand hin zunimmt, schwindet die Mächtigkeit der Postrift-Gesteine in Richtung des Beckenrandes und bildet dort eine kondensierte Abfolge. Die Inversion der Salta Rift Strukturen begann im Cianzo Becken zwischen dem späten Oligozän und Miozän (24–15 Ma) mit einer diachronen Heraushebung des östlichen und westlichen Schenkels der Cianzo Synklinale, welche sich im Hangenden der Hornocal Störung befindet. Gleichzeitig begann im Tilcara Gebirge, südlich des Cianzo Beckens, im späten Oligozän bis frühen Miozän (26–16 Ma) die Heraushebung westlicher Störungsblöcke.
