The construct consists of 1)an aliphatic chain to enable anchoring within membranes, 2)a maleimide moiety to react with the sulfhydryl group of a soluble (bio)molecule, and 3)a fluorescence moiety to allow the construct to be followed by fluorescence spectroscopy and microscopy. It is shown that the construct can be incorporated into preformed membranes, thus allowing application of the approach with biological membranes. The close proximity between the fluorophore and the maleimide moiety within the construct causes fluorescence quenching. This allows monitoring of the reaction with SH-containing molecules by measurement of increases in fluorescence intensity and lifetime. Notably, the construct distributes into laterally ordered membrane domains of lipid vesicles, which is probably triggered by the length of its membrane anchor. The advantages of the new construct can be employed for several biological, biotechnological, and medicinal applications. To enhance understanding of regional climate patterns, we present molecular biomarker records at subannual to annual resolution by mass spectrometry imaging (MSI) of sediments from the Lake Meerfelder Maar covering the Allerod-Younger Dryas transition. These analyses are supported by conventional extraction-based molecular-isotopic analyses, which both validate the imaging results and constrain the sources of the target compounds. The targeted fatty acid biomarkers serve as a gauge of the response of the local aquatic and terrestrial ecosystem to climate change. Based on the comparison of our data with existing data from Meerfelder Maar, we analyse the short-term environmental evolution in Western Europe during the studied time interval and confirm the previously reported delayed hydrological response to Greenland cooling. However, despite a detected delay of Western European environmental change of similar to 135 years, our biomarker data show statistically significant correlation with deuterium excess in Greenland ice core at - annual resolution during this time-transgressive cooling. This suggests a coherent atmospheric forcing across the North Atlantic realm during this transition. We propose that Western European cooling was postponed due to major reorganization of the westerlies that were intermittently forcing warmer and wetter air masses from lower latitudes to Western Europe and thus resulted in delayed cooling relative to Greenland. We investigated the main and interactive effects of two variables on algal FA concentrations (i.e., light intensity and P supply or temperature and P supply). Interactive effects of light intensity and P supply were most pronounced in C. meneghiniana, but were also found in S. quadricauda and C. ovata. Changes in several saturated and unsaturated FA concentrations with light were more distinct in the low-P treatments than in the high-P treatments. Interactive effects of temperature and P supply on various FA concentrations were observed in all four species, but there was no consistent pattern. In lake ecosystems, P limitation often coincides with high light intensities and temperatures in summer. Therefore, it is important to examine how combinations of these environmental conditions affect FA concentrations of primary producers that are important sources of FAs for higher trophic levels. Some FAs are not synthesised endogenously and, therefore, these circulating FAs reflect dietary intake, for example, the trans fatty acids (TFAs), saturated odd chain fatty acids (OCFAs), and linoleic acid, an n-6 polyunsaturated fatty acids (PUFA). It remains unclear if intake of TFA influence T2D risk and whether industrial TFAs (iTFAs) and ruminant TFAs (rTFAs) exert the same effect. Unlike even chain saturated FAs, the OCFAs have been inversely associated with T2D risk, but this association is poorly understood. Furthermore, the associations of n-6 PUFAs intake with T2D risk are still debated, while delta-5 desaturase (D5D), a key enzyme in the metabolism of PUFAs, has been consistently related to T2D risk. To better understand these relationships, the FA composition in circulating lipid fractions can be used as biomarkers of dietary intake and metabolism. The exploration of TFAs subtypes in plasma phospholipids and OCFAs and n-6 PUFAs within a wide range of lipid classes may give insights into the pathophysiology of T2D. Aim: This thesis aimed mainly to analyse the association of TFAs, OCFAs and n-6 PUFAs with self-reported dietary intake and prospective T2D risk, using seven types of TFAs in plasma phospholipids and deep lipidomics profiling data from fifteen lipid classes. Methods: A prospective case-cohort study was designed within the European Prospective Investigation into Cancer and Nutrition (EPIC)-Potsdam study, including all the participants who developed T2D (median follow-up 6.5 years) and a random subsample of the full cohort (subcohort: n=1248; T2D cases: n=820). The main analyses included two lipid profiles. The first was an assessment of seven TFA in plasma phospholipids, with a modified method for analysis of FA with very low abundances. The second lipid profile was derived from a high-throughout lipid profiling technology, which identified 940 distinct molecular species and allowed to quantify OCFAs and PUFAs composition across 15 lipid classes. Delta-5 desaturase (D5D) activity was estimated as 20:4/20:3-ratio. Using multivariable Cox regression models, we examined the associations of TFA subtypes with incident T2D and class-specific associations of OCFA and n-6 PUFAs with T2D risk. Results: 16:1n-7t, 18:1n-7t, and c9t11-CLA were positively correlated with the intake of fat-rich dairy foods. iTFA 18:1 isomers were positively correlated with margarine. After adjustment for confounders and other TFAs, higher plasma phospholipid concentrations of two rTFAs were associated with a lower incidence of T2D: 18:1n-7t and t10c12-CLA. In contrast, the rTFA c9t11-CLA was associated with a higher incidence of T2D. rTFA 16:1n-7t and iTFAs (18:1n-6t, 18:1n-9t, 18:2n-6,9t) were not statistically significantly associated with T2D risk. We observed heterogeneous integration of OCFA in different lipid classes, and the contribution of 15:0 versus 17:0 to the total OCFA abundance differed across lipid classes. Consumption of fat-rich dairy and fiber-rich foods were positively and red meat inversely correlated to OCFA abundance in plasma phospholipid classes. In women only, higher abundances of 15:0 in phosphatidylcholines (PC) and diacylglycerols (DG), and 17:0 in PC, lysophosphatidylcholines (LPC), and cholesterol esters (CE) were inversely associated with T2D risk. In men and women, a higher abundance of 15:0 in monoacylglycerols (MG) was also inversely associated with T2D. Conversely, a higher 15:0 concentration in LPC and triacylglycerols (TG) was associated with higher T2D risk in men. Women with a higher concentration of 17:0 as free fatty acids (FFA) also had higher T2D incidence. The integration of n-6 PUFAs in lipid classes was also heterogeneous. 18:2 was highly abundant in phospholipids (particularly PC), CE, and TG; 20:3 represented a small fraction of FA in most lipid classes, and 20:4 accounted for a large proportion of circulating phosphatidylinositol (PI) and phosphatidylethanolamines (PE). Higher concentrations of 18:2 were inversely associated with T2D risk, especially within DG, TG, and LPC. However, 18:2 as part of MG was positively associated with T2D risk. Higher concentrations of 20:3 in phospholipids (PC, PE, PI), FFA, CE, and MG were linked to higher T2D incidence. 20:4 was unrelated to risk in most lipid classes, except positive associations were observed for 20:4 enriched in FFA and PE. The estimated D5D activities in PC, PE, PI, LPC, and CE were inversely associated with T2D and explained variance of estimated D5D activity by genomic variation in the FADS locus was only substantial in those lipid classes. Conclusion: The TFAs' conformation is essential in their relationship to diabetes risk, as indicated by plasma rTFA subtypes concentrations having opposite directions of associations with diabetes risk. Plasma OCFA concentration is linked to T2D risk in a lipid class and sex-specific manner. Plasma n-6 PUFA concentrations are associated differently with T2D incidence depending on the specific FA and the lipid class. Overall, these results highlight the complexity of circulating FAs and their heterogeneous association with T2D risk depending on the specific FA structure, lipid class, and sex. My results extend the evidence of the relationship between diet, lipid metabolism, and subsequent T2D risk. In addition, my work generated several potential new biomarkers of dietary intake and prospective T2D risk. Die Fettsäure (FS)-Zusammensetzung in den Blutfettfraktionen kann als Biomarker für die Nahrungsaufnahme und den Stoffwechsel verwendet werden, um die Beziehung zwischen den FS und dem T2D-Risiko besser zu verstehen. Das Hauptziel dieser Arbeit war es, den Zusammenhang zwischen zirkulierenden trans-FS (TFS), ungeradkettigen gesättigten FS (UGFS) und n-6 poly ungesättigte FS (PUFS), die in verschiedenen Lipidklassen angereichert sind, mit dem T2D-Risiko zu untersuchen. Mit einer eingebetten Fall-Kohorten-Studie, die im Rahmen der prospektiven EPIC-Potsdam-Studie konzipiert wurde, untersuchte diese Arbeit zwei Lipidprofile im Hinblick auf das T2D-Risiko: (1) Sieben TFS-Subtypen in Plasma-Phospholipiden und (2) die Zusammensetzung von UGFS und PUFA in 15 Plasma-Lipidklassen. Die Aktivität der Delta-5-Desaturase (D5D) wurde als 20:4/20:3-Verhältnis geschätzt. Assoziationen mit dem Auftreten von T2D wurden mit multivariablen Cox-Regressionsmodellen untersucht. Von den üblicherweise aus Milchprodukten stammenden TFS waren 18:1n-7t und t10c12-CLA mit einer geringeren T2D-Inzidenz, c9t11-CLA mit einer höheren Inzidenz und 16:1n-7t nicht mit dem T2D-Risiko assoziiert. TFS aus industriellen Quellen (18:1n-6t, 18:1n-9t, 18:2n-6t) zeigten keinen statistisch signifikanten Zusammenhang mit dem T2D-Risiko. Die UGFS-Konzentration im Plasma war mit dem T2D-Risiko auf lipidklassen- und geschlechtsspezifische Weise assoziiert, wobei bei Frauen stärkere inverse Zusammenhänge für 15:0 in Monoacylglycerinen (MG), Phosphatidylcholinen (PC) und Diacylglycerinen (DG) sowie für 17:0 in PC, Lysophosphatidylcholinen (LPC) und Cholesterinestern (CE) beobachtet wurden. Höhere Konzentrationen von 18:2 waren in DG, TG und LPC invers mit dem T2D-Risiko assoziiert, während MG(18:2) positiv mit dem T2D-Risiko assoziiert war. Höhere Konzentrationen von 20:3 in Phospholipiden (PC, PE, Phosphatidylinositole (PI)), Fettsäuren (FFS), CE und MG waren mit einer höheren T2D-Inzidenz verbunden. 20:4 stand in den meisten Lipidklassen in keinem statistisch singifikanten Zusammenhang mit dem Risiko, außer bei in FFS und PE angereichertem 20:4, das positiv assoziiert war. Zusammenfassend ist die Konformation der TFS für ihren Zusammenhang mit dem Diabetesrisiko entscheidend. Die Assoziationen der UGFS-Plasma-Konzentrationen mit dem T2D-Risiko zeigten lipidklassen- und geschlechtsspezifische Unterschiede. Die Plasma-Konzentrationen der n-6-PUFA waren je nach spezifischer FA und Lipidklasse unterschiedlich mit der T2D-Inzidenz assoziiert. In addition, CLU183 neurons were stimulated with combinations of palmitate, linoleic acid and cholesterol to assess their effects on metabolic stress, mitochondrial function and insulin action. The dietary interventions resulted in a molecular signature of metabolic stress in the hypothalamus with decreased expression of occludin and subunits of mitochondrial electron chain complexes, elevated protein carbonylation, as well as c-Jun N-terminal kinase (JNK) activation. The rotifers grazed for 5 days on eitherNannochloropsis limneticaorMonoraphidium minutum, two food algae with distinct polyunsaturated fatty acid (PUFA) profiles, before the diets were switched to PUFA-freeSynechococcus elongatus, which was provided for three more days. We found between- and within-species differences in rotifer fatty acid compositions on the respective food sources and, in particular, highly specific acclimation reactions to the PUFA-free diet. The different reactions indicate inter- but also intraspecific differences in physiological traits, such as PUFA retention, allocation and bioconversion capacities, within the genusBrachionusthat are most likely accompanied by differences in their nutritional demands. Fettreduzierte Lebensmittel erreichen jedoch, trotz ihrer Weiterentwicklung, nicht die Schmackhaftigkeit ihrer Originale. Die traditionelle Sichtweise, dass die Attraktivität von Fetten allein durch Textur, Geruch, Aussehen und postingestive Effekte bestimmt wird, wird nun durch das Konzept einer gustatorischen Wahrnehmung ergänzt. Bei Nagetieren zeigte sich, dass Lipide unabhängig von den vorgenannten Eigenschaften erkannt werden, sowie, dass Fettsäuren, freigesetzt durch linguale Lipasen, als gustatorische Stimuli fungieren und Fettsäuresensoren in Geschmackszellen exprimiert sind. Die Datenlage für den Menschen erwies sich jedoch als sehr begrenzt, daher war es Ziel der vorliegenden Arbeit molekulare und histologische Voraussetzungen für eine gustatorische Fettwahrnehmung beim Menschen zu untersuchen. Zunächst wurde humanes Geschmacksgewebe mittels RT-PCR und immunhistochemischen Methoden auf die Expression von Fettsäuresensoren untersucht, sowie exprimierende Zellen in Kofärbeexperimenten charakterisiert und quantifiziert. Es wurde die Expression fettsäuresensitiver Rezeptoren nachgewiesen, deren Agonisten das gesamte Spektrum an kurz- bis langkettigen Fettsäuren abdecken (GPR43, GPR84, GPR120, CD36, KCNA5). Ein zweifelsfreier Nachweis des Proteins konnte für den auf langkettige Fettsäuren spezialisierten Rezeptor GPR120 in Typ-I- und Typ-III-Geschmackszellen der Wallpapillen erbracht werden. Etwa 85 % dieser GPR120-exprimierenden Zellen enthielten keine der ausgewählten Rezeptoren der Geschmacksqualitäten süß (TAS1R2/3), umami (TAS1R1/3) oder bitter (TAS2R38). Somit findet sich in humanen Geschmackspapillen nicht nur mindestens ein Sensor, sondern möglicherweise auch eine spezifische, fettsäuresensitive Zellpopulation. Weitere RT-PCR-Experimente und Untersuchungen mittels In-situ-Hybridisierung wurden zur Klärung der Frage durchgeführt, ob Lipasen in den Von-Ebner-Speicheldrüsen (VED) existieren, die freie Fettsäuren aus Triglyceriden als gustatorischen Stimulus freisetzen können. Es zeigte sich zwar keine Expression der bei Nagetieren gefundenen Lipase F (LIPF), jedoch der eng verwandten Lipasen K, M und N in den serösen Zellen der VED. In-silico-Untersuchungen der Sekundär- und Tertiärstrukturen zeigten die hohe Ähnlichkeit zu LIPF, erwiesen aber auch Unterschiede in den Bindungstaschen der Enzyme, welche auf ein differenziertes Substratspektrum hinweisen. Die Anwesenheit eines spezifischen Signalpeptids macht eine Sekretion der Lipasen in den die Geschmacksporen umspülenden Speichel wahrscheinlich und damit auch eine Bereitstellung von Fettsäuren als Stimuli für Fettsäuresensoren. Die Übertragung des durch diese Stimuli hervorgerufenen Signals von Geschmackszellen auf gustatorische Nervenfasern über P2X-Rezeptormultimere wurde mit Hilfe einer vorherigen Intervention mit einem P2X3 /P2X2/3-spezifischen Antagonisten an der Maus als Modellorganismus im Kurzzeit-Präferenztest untersucht. Es zeigte sich weder eine Beeinträchtigung der Wahrnehmung einer Fettsäurelösung, noch einer zuckerhaltigen Kontrolllösung, wohingegen die Wahrnehmung einer Bitterstofflösung reduziert wurde. Somit ist anhand der Ergebnisse dieser Arbeit eine Beteiligung des P2X3-Homomers bzw. des P2X2/3-Heteromers unwahrscheinlich, jedoch die des P2X2-Homomers und damit der gustatorischen Nervenfasern nicht ausgeschlossen. Die Ergebnisse dieser Arbeit weisen auf die Erfüllung grundlegender Voraussetzungen für die gustatorische Fett(säure)wahrnehmung hin und tragen zum Verständnis der sensorischen Fettwahrnehmung und der Regulation der Fettaufnahme bei. Das Wissen um die Regulation dieser Mechanismen stellt eine Grundlage zur Aufklärung der Ursachen und damit der Bekämpfung von Adipositas und assoziierten Krankheiten dar. N2 - High consumption of energy from fat is considered one of the main factors that evoke obesity which led to a worldwide effort to reduce dietary fat intake. However, despite their continuous improvement, fat-reduced foods do not yet reach the palatability of their originals. The traditional view that the attraction to fats is only determined by texture, odor, appearance as well as postingestive effects is now challenged by the concept of gustatory sensation of fats. After excluding or masking the aforementioned features, rodents showed continuous attraction towards lipid solutions. Also long-chain fatty acids liberated from triglycerides by lingual lipases as the main stimulus and the expression of fatty acid-sensitive receptors in taste buds was shown. In contrast, only little data exists for humans. Therefore, this thesis aimed at elucidating the molecular and cellular prerequisites for a gustatory detection of fat. First, human taste tissue was examined by RT-PCR and immunohistochemical methods for the expression of fatty acid sensors and the expressing cells were characterized and quantified by co-staining procedures. The expression of fatty acid-sensitive receptors was shown whose agonists cover the whole range of short- to long-chain fatty acids (GPR43, GPR84, GPR120, CD36, and KCNA5). Protein expression of the long-chain fatty acid receptor GPR120 in type I and type III taste cells was unambiguously demonstrated. About 85 % of the GPR120-expressing cells did not co-express receptors for sweet (TAS1R2/3), umami (TAS1R1/3) or bitter (TAS2R38) taste. Hence, not only does at least one fatty acid sensor exist in human taste papillae, but possibly also a special fatty acid-sensitive cell population. Additional RT-PCR experiments and in situ hybridization analyses were used to address the question, if lipases in the Von-Ebner-salivary glands (VEG) produce free fatty acids from triglycerides as gustatory stimuli. Unlike mice which express lipase F, lipases K, M and N, which are highly related to lipase F, were found to be expressed in the serous cells of the VEG. In silico approaches confirmed high similarities of the secondary and tertiary structures of these lipases to lipase F. Also a marked difference in the binding pocket of the enzymes was observed, which suggests differential substrate specificity. The presence of a signal peptide sequence proposes that the lipases K, M, and N are secreted into the trenches of gustatory papillae. This would result in lipolysis of dietary triglycerides and generation of stimuli for the fatty acid sensor GPR120. Next the hypothesis was tested whether GPR120-generated signals are conveyed to gustatory nerves via P2X-receptor-multimers. To this end short term preference tests were performed in mice after intervention with a P2X3-/P2X2/3-specific antagonist. However, this treatment did not affect the preference of mice for the fatty acid or for a sweet control solution, whereas recognition of a bitter solution was impaired. Thus, an involvement of the P2X3-homomer or the P2X3-/P2X2/3-heteromer seems unlikely. However, these results do not exclude a contribution of P2X2-receptor-heteromers and in turn gustatory nerves for the preference of fatty acid solutions. In summary, the results of this thesis give new insights into the molecular and cellular prerequisites for a gustatory component for fat detection in humans. They also help understanding fat sensation and the regulation of fat intake which in turn may eventually promote novel concepts for the treatment of obesity and related diseases. Der Fokus dieser Arbeit bestand in der gezielten Substitution der internalen Doppelbindungen der Fettsäureester durch reaktivere Gruppen, wie Acrylate oder Methacrylate, welche für Alkydharze in dieser Form so in der Literatur nicht beschrieben sind. Untersuchungen des Polymerisationsverhaltens dieser funktionalisierten Harze wurden mit der Photo DSC durchgeführt, wobei Bis – (4 – methoxybenzoyl) diethylgermanium als Photoinitiator diente. Die Ergebnisse haben gezeigt, dass die Harze radikalisch polymerisiert werden können und eine geringere Abhängigkeit von der Umgebungsatmosphäre (Luftsauerstoff bzw. Stickstoff) vorliegt. Dies ist so in der Literatur für funktionalisierte Alkydharze nicht bekannt. Abmischungen von unterschiedlichen Monomeren und funktionalisierten Harzen bewirkten eine Steigerung der Viskosität sowie eine Verringerung der Sauerstoffinhibierung im Zuge der photoinduzierten Polymerisation unter Luftsauerstoff für die Quecksilberdampflampe und der UV LED. Zur Untersuchung der sauerstoffinhibierenden Wirkung der Harze sind Synthesen unterschiedlicher, funktionalisierter Ölsäuremethylester als Modellsubstanzen durchgeführt worden. Ein verbessertes Polymerisationsverhalten und eine geringe Abhängigkeit von der Umgebungsatmosphäre konnte für die Modelle nachgewiesen werden. Zur Aufklärung des verbesserten Polymerisationsverhaltens sind gezielt Substituenten (Imidazol, Brom, Alkohol, Acetat) in den funktionalisierten Ölsäuremethylester eingebaut worden, um den Einfluss dieser aufzuzeigen. Im Rahmen dieser Synthesen sind neuartige Strukturen synthetisiert worden, welche so in der Literatur nicht beschrieben sind. Die Gegenüberstellung der Polymerisationszeit, der Umsatz der (Meth-)Acrylatgruppen sowie die Zeit zum Erreichen der maximalen Polymerisationsgeschwindigkeit unter Verwendung von unterschiedlichen UV Lichtquellen hat einen Einfluss der Substituenten auf das Polymerisationsverhalten gezeigt. N2 - The present work deals with the synthesis and characterization of functionalized alkyl resins and the photoinduced polymerization of them with different UV light sources. The focus of this work was the targeted substitution of the internal double bonds of fatty acid esters by more reactive groups such as acrylates or methacrylates, which are not described in the literature for alkyd resins in this form. Differences in the basic polymerizability of these functionalized resins were carried out with the Photo DSC, with Bis – (4 – methoxybenzoyl) diethylgermane serving as the photoinitiator. The results showed that the resins could be radically polymerized and also had a lower dependence on the ambient atmosphere. This is not described in the literature for functionalized alkyd resins. Blends of different monomers and the functionalized resins also showed that in addition to the increase in viscosity and the polymerizability of the monomers was improved under atmospheric oxygen. The reference used was methyl oleate, which had been functionalized via the same routes of synthesis and polymerized photochemically. In the context of these syntheses, novel monomers have been synthesized which are thus unknown in the literature.
The reference substances confirmed the behavior of the functionalized resins and showed improved polymerization behavior under atmospheric oxygen. To elucidate these properties, different functionalized methyl oleate have been synthesized to investigate the influence of the substituents on the polymerizability under atmospheric oxygen. In particular, the polymerization time, the conversion of the (meth) acrylate groups and the time to reach the maximum polymerization rate played a decisive role.
