TY - THES A1 - Winkler, Henning T1 - Synthese von thermoplastisch verarbeitbaren Fettsäure-Acylderivaten der Stärke und Proteine T1 - Synthesis of thermoplastic processable fatty acid acyl derivatives of starch and proteins N2 - In den vergangenen Jahren wurden stetig wachsende Produktionskapazitäten von Biokunststoffen aus nachwachsenden Rohstoffe nverzeichnet. Trotz großer Produktionskapazitäten und einem geeigneten Eigenschaftsprofil findet Stärke nur als hydrophile, mit Weichmachern verarbeitete thermoplastische Stärke (TPS) in Form von Blends mit z. B. Polyestern Anwendung. Gleiches gilt für Kunststoffe auf Proteinbasis. Die vorliegende Arbeit hat die Entwicklung von Biokunststoffen auf Stärkebasis zum Ziel, welche ohne externe Weichmacher thermoplastisch verarbeitbar und hydrophob sind sowie ein mechanisches Eigenschaftsprofil aufweisen, welches ein Potenzial zur Herstellung von Materialien für eine Anwendung als Verpackungsmittel bietet. Um die Rohstoffbasis für Biokunststoffe zu erweitern, soll das erarbeitete Konzept auf zwei industriell verfügbare Proteintypen, Zein und Molkenproteinisolat (WPI), übertragen werden. Als geeignete Materialklasse wurden Fettsäureester der Stärke herausgearbeitet. Zunächst fand ein Vergleich der Säurechlorid-Veresterung und der Umesterung von Fettsäurevinylestern statt, woraus letztere als geeignetere Methode hervorging. Durch Variation der Reaktionsparameter konnte diese optimiert und auf eine Serie der Fettsäurevinylester von Butanoat bis Stearat für DS-Werte bis zu 2,2-2,6 angewandt werden. Möglich war somit eine systematische Studie unter Variation der veresterten Fettsäure sowie des Substitutionsgrades (DS). Sämtliche Produkte mit einem DS ab 1,5 wiesen eine ausgprägte Löslichkeit in organischen Lösungsmitteln auf wodurch sowohl die Aufnahme von NMR-Spektren als auch Molmassenbestimmung mittels Größenausschlusschromatographie mit gekoppelter Mehrwinkel-Laserlichtstreuung (GPC-MALLS) möglich waren. Durch dynamische Lichtstreuung (DLS) wurde das Löslichkeitsverhalten veranschaulicht. Sämtliche Produkte konnten zu Filmen verarbeitet werden, wobei Materialien mit DS 1,5-1,7 hohe Zugfestigkeiten (bis zu 42 MPa) und Elastizitätsmodule (bis 1390 MPa) aufwiesen. Insbesondere Stärkehexanoat mit DS <2 sowie Stärkebutanoat mit DS >2 hatten ein mechanisches Eigenschaftsprofil, welches insbesondere in Bezug auf die Festigkeit/Steifigkeit vergleichbar mit Verpackungsmaterialien wie Polyethylen war (Zugfestigkeit: 15-32 MPa, E-Modul: 300-1300 MPa). Zugfestigkeit und Elastizitätsmodul nahmen mit steigender Kettenlänge der veresterten Fettsäure ab. Ester längerkettiger Fettsäuren (C16-C18) waren spröde. Über Weitwinkel-Röntgenstreuung (WAXS) und Infrarotspektroskopie (ATR-FTIR) konnte der Verlauf der Festigkeiten mit einer zunehmenden Distanz der Stärke im Material begründet werden. Es konnten von DS und Kettenlänge abhängige Glasübergänge detektiert werden, die kristallinen Strukturen der langkettigen Fettsäuren zeigten einen Schmelzpeak. Die Hydrophobie der Filme wurde anhand von Kontaktwinkeln >95° gegen Wasser dargestellt. Blends mit biobasierten Polyterpenen sowie den in der Arbeit hergestellten Zein-Acylderivaten ermöglichten eine weitere Verbesserung der Zugfestigkeit bzw. des Elastizitätsmoduls hochsubstituierter Produkte. Eine thermoplastische Verarbeitung mittels Spritzgießen war sowohl für Produkte mit hohem als auch mittlerem DS-Wert ohne jeglichen Zusatz von Weichmachern möglich. Es entstanden homogene, transparente Prüfstäbe. Untersuchungen der Härte ergaben auch hier für Stärkehexanoat und –butanoat mit Polyethylen vergleichbare Werte. Ausgewählte Produkte wurden zu Fasern nach dem Schmelzspinnverfahren verarbeitet. Hierbei wurden insbesondere für hochsubstituierte Derivate homogenen Fasern erstellt, welche im Vergleich zur Gießfolie signifikant höhere Zugfestigkeiten aufwiesen. Stärkeester mit mittlerem DS ließen sich ebenfalls verarbeiten. Zunächst wurden für eine Übertragung des Konzeptes auf die Proteine Zein und WPI verschiedene Synthesemethoden verglichen. Die Veresterung mit Säurechloriden ergab hierbei die höchsten Werte. Im Hinblick auf eine gute Löslichkeit in organischen Lösungsmitteln wurde für WPI die Veresterung mit carbonyldiimidazol (CDI)-aktivierten Fettsäuren in DMSO und für Zein die Veresterung mit Säu-rechloriden in Pyridin bevorzugt. Es stellte sich heraus, dass acyliertes WPI zwar hydrophob, jedoch ohne Weichmacher nicht thermoplastisch verarbeitet werden konnte. Die Erstellung von Gießfolien führte zu Sprödbruchverhalten. Unter Zugabe der biobasierten Ölsäure wurde die Anwendung von acyliertem WPI als thermoplastischer Filler z. B. in Blends mit Stärkeestern dargestellt. Im Gegensatz hierzu zeigte acyliertes Zein Glasübergänge <100 °C bei ausreichender Stabilität (150-200 °C). Zeinoleat konnte ohne Weichmacher zu einer transparenten Gießfolie verarbeitet werden. Sämtliche Derivate erwiesen sich als ausgeprägt hydrophob. Zeinoleat konnte über das Schmelzspinnverfahren zu thermoplastischen Fasern verarbeitet werden. N2 - In recent years, a steadily growing production capacity of bioplastic based on renewable resources was noticed. Despite its huge production capacities and an appropriate property profile (ubiquitous occurrence, easy extraction), starch is only applied in addition of plasticizers in a hydrophilic, thermoplastic form in blends with e. g. polyesters. The same applies to bioplastics based on proteins. The actual study has the aim to develop starch-based bioplastics, which are hydrophobic, thermoplastic without the addition of any plasticizer and have mechanical properties to be a suitable alternative material in the area of food packaging. To obtain a variation of the raw materials for bioplastics, the concept shall be applied to two types of industrial available proteins, whey protein isolate (WPI) and Zein. Fatty acid esters of starch came out to be a suitable class of materials. Initially, the methods of esterifying acid chlorides and the transesterification of fatty acid vinyl esters were compared with the latter being more appropriate. Reaction parameters of this method were optimized and it was applied to a complete series of vinyl ester reagents (butanoate to stearate), leading to degree of substitution (DS)-values up to 2.2-2.6. With that, a systematic study of the variation of the fatty acid ester chain as well as the DS became possible. It came out that all products with a DS >1.5 showed a well-marked solubility in organic solvents, whereby solution NMR-studies as well as measurements of the molecular weight distributions by using size exclusion chroma-tography with multi-angle laser light scattering (SEC-MALLS) were possible. The different solution behavior was studied by dynamic light scattering (DLS). All soluble products could be formed into films via casting, where materials with a DS of 1.5-1.7 showed the highest values concerning tensile strength (up to 42 MPa) and Youngs modulus (up to 1390 MPa). Especially starch hexanoate with DS <2 and starch butanoate with a DS >2 revealed mechanical properties which are comparable to usually applied polymers for food packaging, e. g. polyethylene (tensile strength: 15-20 MPa, E-Mod: 300-1300 MPa). Tensile strength and Youngs modulus were reduced with increasing length of the esterified fatty acid. Wide-angle X-Ray scattering (WAXS) and infrared spectroscopy (ATR-FTIR) explained this tendency by an increasing intermolecular distance of the starch in the material. Glassy transitions of the materials were detected and showed a dependency on the type of esterified fatty acid and the DS. The crystalline structures of the esterified long-chain fatty acids revealed a melting peak. All films came out to be hydrophobic with contact angles against water >95°. The tensile strength and the Youngs modulus of the highly substituted products could be further improved by blending them with biobased polyterpenes as well as the acylated Zein. A thermoplastic processing without the use of any plasticizer additives was possible for products with a medium and high DS. Homogeneous, transparent testing specimens were obtained. The specific mechanical values were comparable with the casted films, although the highest values for the tensile strength and the elongation were lower. Investigations of the hardness showed comparable values to polyethylene. Selected samples were further processed to fibers by melt spinning. Especially starch esters with high DS revealed homogeneous fibers with a significant increase in the tensile strength compared to the film or testing specimen. Even fatty acid starch esters with a medium DS were processed by the melt-spinning, but their higher glassy transition lead to a reduced softening behavior. To transfer this concept to the class of proteins, different methods of synthesis were studied in the first step, which differed in their amount of acylation. The acylation using fatty acid chlorides lead to highest values. With regard to a well-marked organic solvent solubility, in the case of WPI the acylation with carbonyldiimidazol (CDI)-activated fatty acid was established. For Zein, the acid chloride acylation in pyridine gave the desired results. It came out the fatty acid acylated soluble WPI could not be thermoplastic processed without additional plasticizers. By using biobased oleic acid as additive, the potential of acylated WPI as a thermoplastic filler in blends with e. g. fatty acid esters of starch was shown. In contrast, fatty acid acyl derivatives of Zein revealed well marked glassy transitions <100 °C with an adequate thermal stability. While Zeinoleate could be formed into transparent films via solvent casting without any plasticizer additives, low amounts of tall oil enabled film-forming in the case of acyl derivatives with shorter fatty acids as well. All derivatives revealed a well-marked hydrophobicity. Finally, Zeinoleate was thermoplastically processed into fibers by melt-spinning without any further additives. Recently, two non-catalytic starch-binding proteins, pivotal for normal starch turnover in Arabidopsis leaves, namely, EARLY STARVATION 1 (ESV1) and its homolog LIKE EARLY STARVATION 1 (LESV), have been identified. Both share nearly 38% sequence homology. As ESV1 has been found to influence glucan phosphorylation via two starch-related dikinases, alpha-glucan, water dikinase (GWD) and phosphoglucan, water dikinase (PWD), through modulating the surface glucan structures of the starch granules and thus affecting starch degradation, we assess the impact of its homolog LESV on starch metabolism. Thus, the 65-kDa recombinant protein LESV and the 50-kDa ESV1 were analyzed regarding their influence on the action of GWD and PWD on the surface of the starch granules. We included starches from various sources and additionally assessed the effect of these non-enzymatic proteins on other starch-related enzymes, such as starch synthases (SSI and SSIII), starch phosphorylases (PHS1), isoamylase and beta-amylase. The data obtained indicate that starch phosphorylation, hydrolyses and synthesis were affected by LESV and ESV1. Furthermore, incubation with LESV and ESV1 together exerted an additive effect on starch phosphorylation. In addition, a stable alteration of the glucan structures at the starch granule surface following treatment with LESV and ESV1 was observed. Here, we discuss all the observed changes that point to modifications in the glucan structures at the surface of the native starch granules and present a model to explain the existing processes. The water-soluble glycogen in animal cells and the amylopectin which is the major component of water-insoluble starch granules residing in plant plastids are chemically similar as they consist of α-1,6 branched α-1,4 glucan chains. Synthesis and degradation of transitory starch and of glycogen are accomplished by a set of enzymatic activities that to some extend are also similar in plants and animals. Chain elongation, branching, and debranching are achieved by synthases, branching enzymes, and debranching enzymes, respectively. Similarly, both types of polyglucans contain low amounts of phosphate esters whose abundance varies depending on species and organs. Starch is selectively phosphorylated by at least two dikinases (GWD and PWD) at the glucosyl carbons C6 and C3 and dephosphorylated by the phosphatase SEX4 and SEX4-like enzymes. In Arabidopsis insufficiency in starch phosphorylation or dephosphorylation results in largely impaired starch turnover, starch accumulation, and often in retardation of growth. In humans the progressive neurodegenerative epilepsy, Lafora disease, is the result of a defective enzyme (laforin) that is functional equivalent to the starch phosphatase SEX4 and capable of glycogen dephosphorylation. Patients lacking laforin progressively accumulate unphysiologically structured insoluble glycogen-derived particles (Lafora bodies) in many tissues including brain. Previous results concerning the carbon position of glycogen phosphate are contradictory. Currently it is believed that glycogen is esterified exclusively at the carbon positions C2 and C3 and that the monophosphate esters, being incorporated via a side reaction of glycogen synthase (GS), lack any specific function but are rather an enzymatic error that needs to be corrected. In this study a versatile and highly sensitive enzymatic cycling assay was established that enables quantification of very small G6P amounts in the presence of high concentrations of non-target compounds as present in hydrolysates of polysaccharides, such as starch, glycogen, or cytosolic heteroglycans in plants. Following validation of the G6P determination by analyzing previously characterized starches G6P was quantified in hydrolysates of various glycogen samples and in plant heteroglycans. Interestingly, glucosyl C6 phosphate is present in all glycogen preparations examined, the abundance varying between glycogens of different sources. Additionally, it was shown that carbon C6 is severely hyperphosphorylated in glycogen of Lafora disease mouse model and that laforin is capable of removing C6 phosphate from glycogen. After enrichment of phosphoglucans from amylolytically degraded glycogen, several techniques of two-dimensional NMR were applied that independently proved the existence of 6-phosphoglucosyl residues in glycogen and confirmed the recently described phosphorylation sites C2 and C3. C6 phosphate is neither Lafora disease- nor species-, or organ-specific as it was demonstrated in liver glycogen from laforin-deficient mice and in that of wild type rabbit skeletal muscle. The distribution of 6-phosphoglucosyl residues was analyzed in glycogen molecules and has been found to be uneven. Gradual degradation experiments revealed that C6 phosphate is more abundant in central parts of the glycogen molecules and in molecules possessing longer glucan chains. Glycogen of Lafora disease mice consistently contains a higher proportion of longer chains while most short chains were reduced as compared to wild type. Together with results recently published (Nitschke et al., 2013) the findings of this work completely unhinge the hypothesis of GS-mediated phosphate incorporation as the respective reaction mechanism excludes phosphorylation of this glucosyl carbon, and as it is difficult to explain an uneven distribution of C6 phosphate by a stochastic event. Indeed the results rather point to a specific function of 6-phosphoglucosyl residues in the metabolism of polysaccharides as they are present in starch, glycogen, and, as described in this study, in heteroglycans of Arabidopsis. In the latter the function of phosphate remains unclear but this study provides evidence that in starch and glycogen it is related to branching. Moreover a role of C6 phosphate in the early stages of glycogen synthesis is suggested. By rejecting the current view on glycogen phosphate to be a stochastic biochemical error the results permit a wider view on putative roles of glycogen phosphate and on alternative biochemical ways of glycogen phosphorylation which for many reasons are likely to be mediated by distinct phosphorylating enzymes as it is realized in starch metabolism of plants. Better understanding of the enzymology underlying glycogen phosphorylation implies new possibilities of Lafora disease treatment. N2 - Pflanzen und Tiere speichern Glukose in hochmolekularen Kohlenhydraten, um diese bei Bedarf unter anderem zur Gewinnung von Energie zu nutzen. Amylopectin, der größte Bestandteil des pflanzlichen Speicherkohlenhydrats Stärke, und das tierische Äquivalent Glykogen sind chemisch betrachtet ähnlich, denn sie bestehen aus verzweigten Ketten, deren Bausteine (Glukosylreste) auf identische Weise miteinander verbunden sind. Zudem kommen in beiden Kohlenhydraten kleine aber ähnliche Mengen von Phosphatgruppen vor, die offenbar eine tragende Rolle in Pflanzen und Tieren spielen. Ist in Pflanzen der Einbau oder die Entfernung von Phosphatgruppen in bzw. aus Stärke gestört, so ist oft der gesamte Stärkestoffwechsel beeinträchtigt. Dies zeigt sich unter anderem in der übermäßigen Akkumulation von Stärke und in Wachstumsverzögerungen der gesamten Pflanze. Beim Menschen und anderen Säugern beruht eine schwere Form der Epilepsie (Lafora disease) auf einer Störung des Glykogenstoffwechsels. Sie wird durch das erblich bedingte Fehlen eines Enzyms ausgelöst, das Phosphatgruppen aus dem Glykogen entfernt. Während die Enzyme, die für die Entfernung des Phosphats aus Stärke und Glykogen verantwortlich sind, hohe Ähnlichkeit aufweisen, ist momentan die Ansicht weit verbreitet, dass der Einbau von Phosphat in beide Speicherkohlenhydrate auf höchst unterschiedliche Weise erfolgt. In Pflanzen sind zwei Enzyme bekannt, die Phosphatgruppen an unterschiedlichen Stellen in Glukosylreste einbauen (Kohlenstoffatome 6 und 3). In Tieren soll eine seltene, unvermeidbare und zufällig auftretende Nebenreaktion eines Enzyms, das eigentlich die Ketten des Glykogens verlängert (Glykogen-Synthase), den Einbau von Phosphat bewirken, der somit als unwillkürlich gilt und weithin als „biochemischer Fehler“ (mit fatalen Konsequenzen bei ausbleibender Korrektur) betrachtet wird. In den Glukosylresten des Glykogens sollen ausschließlich die C-Atome 2 und 3 phosphoryliert sein. Die Ergebnisse dieser Arbeit zeigen mittels zweier unabhängiger Methoden, dass Glykogen auch am Glukosyl-Kohlenstoff 6 phosphoryliert ist, der Phosphatposition, die in der Stärke am häufigsten vorkommt. Die Tatsache, dass in dieser Arbeit Phosphat neben Stärke auch erstmals an Glukosylresten von anderen pflanzlichen Kohlenhydraten (wasserlösliche Heteroglykane) nachgewiesen werden konnte, lässt vermuten, dass Phosphorylierung ein generelles Phänomen bei Polysacchariden ist. Des Weiteren wiesen die Ergebnisse darauf hin, dass Phosphat im Glykogen, wie auch in der Stärke, einem bestimmten Zweck dient, der im Zusammenhang mit der Regulation von Kettenverzweigung steht, und dass kein zufälliges biochemisches Ereignis für den Einbau verantwortlich sein kann. Aufgrund der grundlegenden Ähnlichkeiten im Stärke- und Glykogenstoffwechsel, liegt es nahe, dass die Phosphorylierung von Glykogen, ähnlich der von Stärke, ebenfalls durch spezifische Enzyme bewirkt wird. Ein besseres Verständnis der Mechanismen, die der Glykogen-Phosphorylierung zugrunde liegen, kann neue Möglichkeiten der Behandlung von Lafora disease aufzeigen. This review covers the advances made in the study of these processes. The analyses presented herein depict a scenario in which starch synthase isoform 4 (SS4) provides the elongating activity necessary for the initiation of starch granule formation. However, this protein does not act alone; other polypeptides are required for the initiation of an appropriate number of starch granules per chloroplast. The functions of this group of polypeptides include providing suitable substrates (maltooligosaccharides) to SS4, the localization of the starch initiation machinery to the thylakoid membranes, and facilitating the correct folding of SS4. The number of starch granules per chloroplast is tightly regulated and depends on the developmental stage of the leaves and their metabolic status. Plastidial phosphorylase (PHS1) and other enzymes play an essential role in this process since they are necessary for the synthesis of the substrates used by the initiation machinery. The mechanism of starch granule formation initiation in Arabidopsis seems to be generalizable to other plants and also to the synthesis of long-term storage starch. The latter, however, shows specific features due to the presence of more isoforms, the absence of constantly recurring starch synthesis and degradation, and the metabolic characteristics of the storage sink organs. Allerdings basieren auch neue Lösungen meist nicht auf Biopolymeren und in einem noch geringeren Anteil auf wasserbasierten Beschichtungssystemen aus nachwachsenden Rohstoffen. Dies stellt den Anknüpfungspunkt dieser Arbeit dar, in der untersucht wurde, ob das Biopolymer Stärke das Potenzial zum wasserbasierten Filmbildner für Farben und Lacke besitzt. Dabei müssen angelehnt an etablierte synthetische Marktprodukte die folgenden Kriterien erfüllt werden: Die wässrige Dispersion muss mindestens einen 30%igen Feststoffgehalt haben, bei Raumtemperatur verarbeitet werden können und Viskositäten zwischen 10^2-10^3 mPa·s aufweisen. Die finale Beschichtung muss einen geschlossenen Film bilden und sehr gute Haftfestigkeiten zu einer spezifischen Oberfläche, in dieser Arbeit Glas, besitzen. Als Grundlage für die Modifizierung der Stärke wurde eine Kombination von molekularem Abbau und chemischer Funktionalisierung ausgewählt. Da nicht bekannt war, welchen Einfluss die Stärkeart, die gewählte Abbaureaktion als auch verschiedene Substituenten auf die Dispersionsherstellung und deren Eigenschaften sowie die Beschichtungseigenschaften ausüben könnten, wurden die strukturellen Parameter getrennt voneinander untersucht. Das erste Themengebiet beinhaltete den oxidativen Abbau von Kartoffel- und Palerbsenstärke mittels des Hypochlorit-Abbaus (OCl-) und des ManOx-Abbaus (H2O2, KMnO4). Mit beiden Abbaureaktionen konnten vergleichbare gewichtsmittlere Molmassen (Mw) von 2·10^5-10^6 g/mol (GPC-MALS) hergestellt werden. Allerdings führten die gewählten Reaktionsbedingungen beim ManOx-Abbau zur Bildung von Gelpartikeln. Diese lagen im µm-Bereich (DLS und Kryo-REM-Messungen) und hatten zur Folge, dass die ManOx-Proben deutlich erhöhte Viskositäten (c: 7,5 %; 9-260 mPa·s) im Vergleich zu den OCl--Proben (4-10 mPa·s) bei scherverdünnendem Verhalten besaßen und die Eigenschaften von viskoelastischen Gelen (G‘ > G‘‘) zeigten. Des Weiteren wiesen sie reduzierte Heißwasserlöslichkeiten (95 °C, vorrangig: 70-99 %) auf. Der OCl--Abbau führte zu hydrophileren (Carboxylgruppengehalt bis zu 6,1 %; ManOx: bis zu 3,1 %), nach 95 °C-Behandlung vollständig wasserlöslichen abgebauten Stärken, die ein Newtonsches Fließverhalten mit Eigenschaften einer viskoelastischen Flüssigkeit (G‘‘ > G‘) hatten. Die OCl--Proben konnten im Vergleich zu den ManOx-Produkten (10-20 %) zu konzentrierteren Dispersionen (20-40 %) verarbeitet werden, die gleichzeitig die Einschränkung von anwendungsrelevanten Mw auf < 7·10^5 g/mol zuließen (Konzentration sollte > 30 % sein). Außerdem führten nur die OCl--Proben der Kartoffelstärke zu transparenten (alle anderen waren opak) geschlossenen Beschichtungsfilmen. Somit hebt sich die Kombination von OCl--Abbau und Kartoffelstärke mit Hinblick auf die Endanwendung ab. Das zweite Themengebiet umfasste Untersuchungen zum Einfluss von Ester- und Hydroxyalkylether-Substituenten auf Basis einer industriell abgebauten Kartoffelstärke (Mw: 1,2·10^5 g/mol) vor allem auf die Dispersionsherstellung, die rheologischen Eigenschaften der Dispersionen und die Beschichtungseigenschaften in Kombination mit Glassubstraten. Dazu wurden Ester und Ether mit DS/MS-Werten von 0,07-0,91 synthetisiert. Die Derivate konnten zu wasserbasierten Dispersionen mit Konzentrationen von 30-45 % verarbeitet werden, wobei bei hydrophoberen Modifikaten ein Co-Lösemittel, Diethylenglycolmonobutylether (DEGBE), eingesetzt werden musste. Die Feststoffgehalte sanken dabei für beide Derivatklassen vor allem mit zunehmender Alkylkettenlänge. Die anwendungsrelevanten Viskositäten (323-1240 mPa·s) stiegen auf Grund von Wechselwirkungen tendenziell mit DS/MS und Alkylkettenlänge an. Hinsichtlich der Beschichtungseigenschaften erwiesen sich die Ester vergleichend zu den Ethern als die bevorzugte Substituentenklasse, da nur die Ester geschlossene, defektfreie und mehrheitlich transparente Beschichtungsfilme bildeten, die exzellente bis sehr gute Haftfestigkeiten (ISO Klasse: 0 und 1) auf Glas besaßen. Die Ether bildeten mehrheitlich brüchige Filme. Basierend auf der Kombination der Ergebnisse aus Lösemittelaustausch, den rheologischen Untersuchungen und zusätzlichen Oberflächenspannungsmessungen (30-61 mN/m) konnte geschlossen werden, dass wahrscheinlich fehlende oder schlechte Haftfestigkeiten vorrangig akkumuliertem Wasser in den Beschichtungsfilmen (visuell: trüb oder weiß) geschuldet sind, während die Brüchigkeit vermutlich auf Wechselwirkungen (H-Brücken Wechselwirkungen, hydrophobe Wechselwirkungen) zwischen den Polymeren zurückgeführt werden kann. Insgesamt scheint die Kombination aus Kartoffelstärke basierend auf dem OCl--Abbau mit Mw < 7·10^5 g/mol und einem Estersubstituenten eine gute Wahl für wasserbasierte Dispersionen mit hohen Feststoffkonzentrationen (> 30 %), guter Filmbildung und exzellenten Haftungen auf Glas zu sein. N2 - In the last few decades, the coating industry, like many other industries, has changed its thinking towards more environmentally friendly paints and varnishes. However, even new solutions are usually not based on biopolymers and, to an even lesser extent, on water-based coating systems made from renewable raw materials. This is the starting point of this work, which aimed to investigate whether the biopolymer starch has the potential to be a water-based film former for paints and varnishes. Based on established synthetic market products, the following criteria must be fulfilled: The aqueous dispersion must have a solid content of at least 30%, be processable at room temperature and have viscosities of 10^2-10^3 mPa·s. The final coating must form a continuous film and have very good adhesion to a specific substrate, in this thesis glass. A combination of molecular degradation and chemical functionalisation was selected as the basis for the modification of the starch. Since it was not known what influence the type of starch, the chosen degradation reaction as well as different substituents could have on the dispersion production and its properties as well as on the coating properties, the structural parameters were investigated separately. The first part dealt with the oxidative degradation of potato and smooth pea starch by hypochlorite degradation (OCl-) and ManOx degradation (H2O2, KMnO4). With both degradation reactions comparable weight average molar masses (Mw) of 2·10^5-10^6 g/mol (SEC-MALS) could be synthesized. However, the selected reaction conditions led to the formation of gel particles during ManOx degradation. These were in the µm-range (DLS and cryo-REM measurements) and resulted in ManOx samples having significantly higher viscosities (c: 7.5%; 9-260 mPa·s) compared to OCl- samples (4-10 mPa·s) with shear-thinning behaviour and showing the properties of viscoelastic gels (G' > G''). Furthermore, they exhibited reduced hot water solubilities (95 °C, primarily: 70-99%). OCl- degradation led to more hydrophilic (carboxyl group content up to 6.1%; ManOx: up to 3.1%), after 95 °C treatment completely water-soluble degraded starches, which had Newtonian flow behaviour with properties of a viscoelastic liquid (G'' > G'). Compared to the ManOx products (10-20%), the OCl- samples could be processed to more concentrated dispersions (20-40%), which at the same time allowed the restriction of application-relevant Mw to < 7·10^5 g/mol (concentration should be > 30%). In addition, only the OCl- samples of the potato starch led to transparent (all others were opaque) continuous coating films. Thus, the combination of OCl- degradation and potato starch stands out with regard to the final application. The second part included investigations on the influence of ester and hydroxyalkyl ether substituents on the basis of an industrially degraded potato starch (Mw: 1.2·10^5 g/mol), particularly on dispersion preparation, rheological properties of the dispersions and the coating properties in combination with glass substrates. For this purpose, esters and ethers with DS/MS values of 0.07-0.91 were synthesized. The derivatives could be prepared to water-based dispersions with concentrations of 30-45%, whereby a co-solvent, diethylene glycol monobutyl ether (DEGBE), had to be used for more hydrophobic derivatives. The solid content of both derivative classes decreased most of all with increasing alkyl chain length. The application-relevant viscosities (323-1240 mPa·s) tended to increase due to interactions with DS/MS and alkyl chain length. With regard to the coating properties, esters proved to be the preferred substituent class compared to ethers, since only the esters formed continuous, defect free and mostly transparent coating films which had excellent to very good adhesion (ISO class: 0 and 1) on glass. The majority of ethers formed brittle films. Based on the combination of the results from solvent exchange, rheological investigations and additional surface tension measurements (30-61 mN/m), it could be concluded that probably missing or poor adhesion was primarily due to accumulated water in the coating films (visually: turbid or white), while the brittleness can probably be attributed to interactions (H-bridge interactions, hydrophobic interactions) between the polymers. Overall, the combination of potato starch based on OCl- degradation with Mw < 7·10^5 g/mol and an ester substituent seems to be a good choice for water-based dispersions with high solid concentrations (> 30%), good film formation and excellent adhesion to glass. Several approaches and bioanalytical tools can be used to expand the knowledge on the different parts involved in the starch metabolism. In this sense, a comprehensive analysis targeting two of the main groups of molecules involved in this process: proteins, as effectors/regulators of the starch metabolism, and maltodextrins as starch components and degradation products, was conducted in this research work using potato plants (Solanum tuberosum L. cv. Desiree) as model of study. On one side, proteins physically interacting to potato starch were isolated and analyzed through mass spectrometry and western blot for their identification. Alternatively, starch interacting proteins were explored in potato tubers from transgenic plants having antisense inhibition of starch-related enzymes and on tubers stored under variable environmental conditions. Most of the proteins recovered from the starch granules corresponded to previously described proteins having a specific role in the starch metabolic pathway. Another set of proteins could be grouped as protease inhibitors, which were found weakly interacting to starch. Variations in the protein profile obtained after electrophoresis separation became clear when tubers were stored under different temperatures, indicating a differential expression of proteins in response to changing environmental conditions. On the other side, since maltodextrin metabolism is thought to be involved in both starch initiation and degradation, soluble maltooligosaccharide content in potato tubers was analyzed in this work under diverse experimental variables. For this, tuber disc samples from wild type and transgenic lines strongly repressing either the plastidial or cytosolic form of the -glucan phosphorylase and phosphoglucomutase were incubated with glucose, glucose-6-phosphate, and glucose-1-phosphate solutions to evaluate the influence of such enzymes on the conversion of the carbon sources into soluble maltodextrins, in comparison to wild-type samples. Relative maltodextrin amounts analyzed through capillary electrophoresis equipped with laser-induced fluorescence (CE-LIF) revealed that tuber discs could immediately uptake glucose-1-phosphate and use it to produce maltooligosaccharides with a degree of polymerization of up to 30 (DP30), in contrast to transgenic tubers with strong repression of the plastidial glucan phosphorylase. The results obtained from the maltodextrin analysis support previous indications that a specific transporter for glucose-1-phosphate may exist in both the plant cells and the plastidial membranes, thereby allowing a glucose-6-phosphate independent transport. Furthermore, it confirms that the plastidial glucan phosphorylase is responsible for producing longer maltooligosaccharides in the plastids by catalyzing a glucan polymerization reaction when glucose-1-phosphate is available. All these findings contribute to a better understanding of the role of the plastidial glucan phosphorylase as a key enzyme directly involved in the synthesis and degradation of glucans and their implication on starch metabolism. In diesem Sinne wurde in dieser Forschungsarbeit eine umfassende Analyse durchgeführt, die auf zwei der wichtigsten Molekülgruppen abzielt, die an diesem Prozess beteiligt sind: Proteine als Effektoren/Regulatoren des Stärkestoffwechsels und Maltodextrine als Stärkebestandteile und Abbauprodukte, wobei Kartoffelpflanzen (Solanum tuberosum L. cv. Desiree) als Untersuchungsmodell dienten. Einerseits wurden Proteine, die physisch mit Kartoffelstärke interagieren, isoliert und mittels Massenspektrometrie und Western Blot analysiert, um sie zu identifizieren. Andererseits wurden die mit Stärke interagierenden Proteine in Kartoffelknollen von transgenen Pflanzen mit Antisense-Hemmung von stärkeverwandten Enzymen und in Knollen, die unter variablen Umweltbedingungen gelagert wurden, untersucht. Variationen im Proteinprofil nach der Elektrophorese-Trennung wurden deutlich, wenn die Knollen bei unterschiedlichen Temperaturen gelagert wurden, was auf eine unterschiedliche Expression von Proteinen als Reaktion auf wechselnde Umweltbedingungen hindeutet. Da man davon ausgeht, dass der Maltodextrin-Stoffwechsel sowohl an der Entstehung als auch am Abbau von Stärke beteiligt ist, wurde in dieser Arbeit der Gehalt an löslichen Maltooligosacchariden in Kartoffelknollen unter verschiedenen experimentellen Variablen analysiert. Conversion of glucose 1-phosphate to starch is exceptionally efficient. In this communication, tuber slices were incubated with either of four solutions containing equimolar [U-C-14]glucose 1-phosphate, [U-C-14]sucrose, [U-C-14]glucose 1-phosphate plus unlabelled equimolar sucrose or [U-C-14]sucrose plus unlabelled equimolar glucose 1-phosphate. C-14-incorporation into starch was monitored. In slices from freshly harvested tubers each unlabelled compound strongly enhanced C-14 incorporation into starch indicating closely interacting paths of starch biosynthesis. However, enhancement disappeared when the tubers were stored. The two paths (and, consequently, the mutual enhancement effect) differ in temperature dependence. At lower temperatures, the glucose 1-phosphate-dependent path is functional, reaching maximal activity at approximately 20 degrees C but the flux of the sucrose-dependent route strongly increases above 20 degrees C. Results are confirmed by in vitro experiments using [U-C-14]glucose 1-phosphate or adenosine-[U-C-14]glucose and by quantitative zymograms of starch synthase or phosphorylase activity. In mutants almost completely lacking the plastidial phosphorylase isozyme(s), the glucose 1-phosphate-dependent path is largely impeded. Irrespective of the size of the granules, glucose 1-phosphate-dependent incorporation per granule surface area is essentially equal. Furthermore, within the granules no preference of distinct glucosyl acceptor sites was detectable. Thus, the path is integrated into the entire granule biosynthesis. In vitro C-14-incorporation into starch granules mediated by the recombinant plastidial phosphorylase isozyme clearly differed from the in situ results. Taken together, the data clearly demonstrate that two closely but flexibly interacting general paths of starch biosynthesis are functional in potato tuber cells. The two plastidial phosphorylases are shown to function as homodimers containing two 91-kDa (PhoA) subunits and two 110-kDa (PhoB) subunits. Both lack the typical 80-amino-acid insertion found in the higher plant plastidial forms. PhoB is exquisitely sensitive to inhibition by ADP-glucose and has a low affinity for malto-oligosaccharides. PhoA is more similar to the higher plant plastidial phosphorylases: it is moderately sensitive to ADP-glucose inhibition and has a high affinity for unbranched malto-oligosaccharides. Molecular analysis establishes that STA4 encodes PhoB. Chlamydomonas reinhardtii strains carrying mutations at the STA4 locus display a significant decrease in amounts of starch during storage that correlates with the accumulation of abnormally shaped granules containing a modified amylopectin structure and a high amylose content.
