A time-resolved luminescent competitive assay to detect L-selectin using aptamers as recognition elements
L-selectin is a protein with potential importance for numerous diseases and clinical disorders. In this paper, we present a new aptamer-based luminescent assay developed to detect L-selectin. The sensing system working principle is based on Forster Resonance Energy Transfer (FRET) from a donor terbium complex (TbC) to an acceptor cyanine dye (Cy5). In the present approach, the biotinylated aptamer is combined with Cy5-labelled streptavidin (Cy5-Strep) to yield an aptamer-based acceptor construct (Apta-Cy5-Strep), while L-selectin is conjugated using luminescent TbC. Upon aptamer binding to the TbC-labelled L-selectin (L-selectin-TbC), permanent donor-acceptor proximity is established which allows for radiationless energy transfer to occur. However, when unlabelled L-selectin is added, it competes with the L-selectin-TbC and the FRET signal decreases as the L-selectin concentration increases. FRET from the TbC to Cy5 was observed with time-gated time-resolved luminescence spectroscopy. A significant change in the corrected luminescence signal was observed in the dynamic range of 10 -500 ng/mL L-selectin, the concentration range relevant for accelerated cognitive decline of Alzheimer's disease, with a limit of detection (LOD) equal to 10 ng/mL. The aptasensor-based assay is homogeneous and can be realized within one hour. Therefore, this method has the potential to become an alternative to tedious heterogeneous analytical methods, e.g. based on enzyme-linked immunosorbent assay (ELISA). (C) 2015 Elsevier B.V. Cumarin- und DBD-Farbstoffe als Fluoreszenzsonden
Coumarin and dbd dyes as fluorescent probes
In dieser Arbeit werden drei Themen im Zusammenhang mit den spektroskopischen Eigenschaften von Cumarin- (Cou) und DBD-Farbstoffen ([1,3]Dioxolo[4,5-f][1,3]benzodioxol) behandelt. Der erste Teil zeigt die grundlegende spektroskopische Charakterisierung von 7-Aminocumarinen und ihre potentielle Anwendung als Fluoreszenzsonde für Fluoreszenzimmunassays. Im zweiten Teil werden mit die photophysikalischen Eigenschaften der Cumarine genutzt um Cou- und DBD-funktionalisierte Oligo-Spiro-Ketal-Stäbe (OSTK) und ihre Eigenschaften als Membransonden zu untersuchen. Der letzte Teil beschäftigt sich mit der Synthese und der Charakterisierung von Cou- und DBD-funktionalisierten Polyprolinen als Referenzsysteme für schwefelfunktionalisierte OSTK-Stäbe und ihrer Kopplung an Goldnanopartikel.
Immunochemische Analysemethoden sind in der klinischen Diagnostik sehr erfolgreich und werden heute auch für die Nahrungsmittelkontrolle und Überwachung von Umweltfragen mit einbezogen. Dadurch sind sie von großem Interesse für weitere Forschungen. Unter den verschiedenen Immunassays zeichnen sich lumineszenzbasierte Formate durch ihre herausragende Sensitivität aus, die dieses Format für zukünftige Anwendungen besonders attraktiv macht. Die Notwendigkeit von Multiparameterdetektionsmöglichkeiten erfordert einen Werkzeugkasten mit Farbstoffen, um die biochemische Reaktion in ein optisch detektierbares Signal umzuwandeln. Hier wird bei einem Multiparameteransatz jeder Analyt durch einen anderen Farbstoff mit einer einzigartigen Emissionsfarbe, die den blauen bis roten Spektralbereich abdecken, oder eine einzigartige Abklingzeit detektiert. Im Falle eines kompetitiven Immunassayformats wäre für jeden der verschiedenen Farbstoffe ein einzelner Antikörper erforderlich. In der vorliegenden Arbeit wird ein leicht modifizierter Ansatz unter Verwendung einer Cumarineinheit, gegen die hochspezifische monoklonale Antikörper (mAb) erzeugt wurden, als grundlegendes Antigen präsentiert. Durch eine Modifikation der Stammcumarineinheit an einer Position des Moleküls, die für die Erkennung durch den Antikörper nicht relevant ist, kann auf den vollen Spektralbereich von blau bis tiefrot zugegriffen werden. In dieser Arbeit wird die photophysikalische Charakterisierung der verschiedenen Cumarinderivate und ihrer entsprechenden Immunkomplexe mit zwei verschiedenen, aber dennoch hochspezifischen, Antikörpern präsentiert. Die Cumarinfarbstoffe und ihre Immunkomplexe wurden durch stationäre und zeitaufgelöste Absorptions- sowie Fluoreszenzemissionsspektroskopie charakterisiert. Darüber hinaus wurden Fluoreszenzdepolarisationsmessungen durchgeführt, um die Daten zu vervollständigen, die die verschiedenen Bindungsmodi der beiden Antikörper betonten. Im Gegensatz zu häufig eingesetzten Nachweissystemen wurde eine massive Fluoreszenzverstärkung bei der Bildung des Antikörper-Farbstoffkomplexes bis zu einem Faktor von 50 gefunden. Wegen der leichten Emissionsfarbenänderung durch das Anpassen der Cumarinsubstitution in der für die Antigenbindung nicht relevanten Position des Elternmoleküls, ist eine Farbstoff-Toolbox vorhanden, die bei der Konstruktion von kompetitiven Multiparameterfluoreszenzverstärkungsimmunassays verwendet werden kann.
Oligo-Spiro-Thio-Ketal-Stäbe werden aufgrund ihres hydrophoben Rückgrats leicht in Doppellipidschichten eingebaut und deshalb als optische Membransonde verwendet. Wegen ihres geringen Durchmessers wird nur eine minimale Störung der Doppellipidschicht verursacht. Durch die Markierung mit Fluoreszenzfarbstoffen sind neuartige Förster-Resonanz-Energietransfersonden mit hoch definierten relativen Orientierungen der Übergangsdipolmomente der Donor- und Akzeptorfarbstoffe zugänglich und macht die Klasse der OSTK-Sonden zu einem leistungsstarken, flexiblen Werkzeugkasten für optische Biosensoranwendungen. Mit Hilfe von stationären und zeitaufgelösten Fluoreszenzexperimenten wurde der Einbau von Cumarin- und DBD markierten OSTK-Stäben in großen unilamellaren Vesikeln untersucht und die Ergebnisse durch Fluoreszenzdepolarisationsmessungen untermauert.
Der letzte Teil dieser Arbeit beschäftigt sich mit der Synthese und Charakterisierung von Cou- und DBD-funktionalisierten Polyprolinen und ihrer Kopplung an Goldnanopartikel. Die farbstoffmarkierten Polyproline konnten erfolgreich hergestellt werden. Es zeigten sich deutlich Einflüsse auf die spektroskopischen Eigenschaften der Farbstoffe durch die Bindung an die Polyprolinhelix. Die Kopplung an die 5 nm großen AuNP konnte erfolgreich durchgeführt werden. Die Erfahrungen, die durch die Kopplung der Polyproline an die AuNP, gewonnen wurde, ist die Basis für eine Einzelmolekül-AFM-FRET-Nanoskopie mit OSTK-Stäben.
In this thesis, three topics are discussed in connection with the spectroscopic properties of coumarin (Cou) and DBD ([1,3]dioxolo[4,5-f][1,3]benzodioxole) dyes. The first part shows the basic spectroscopic characterization of 7-aminocumarins and their advantage as a fluorescence probe for fluorescence immunoassays. In the second part, the photophysical properties of the coumarins are used to investigate Cou- and DBD-functionalized oligo-spiro-ketal rods (OSTK) and their properties as membrane probes. The last part deals with the synthesis and characterization of Cou- and DBD-functionalized polyprolines as reference systems for sulfur-functionalized OSTK rods and their coupling to gold nanoparticles.
The fact that immunochemical analysis methods are very successful in clinical diagnostics and are now also included for food control and monitoring of environmental questions they are of great interest for further research. Among the various immunoassays, luminescence-based formats are distinguished by their outstanding sensitivity, which makes this format particularly attractive for future applications. The need for multiparameter detection capabilities requires a toolbox of dyes to convert the biochemical response to an optically detectable signal. Here, in the case of a multiparameter approach, each analyte is detected by another dye with a unique emission color which covers the blue to red spectral range or a unique decay time. In the case of a competitive immunoassay format, a single antibody would be required for each of the different dyes. In the present work, a slightly modified approach is presented as a basic antigen using a coumarin moiety against which highly specific antibodies have been produced. By modifying the parent coumarin moiety at a site of the molecule which is not relevant for recognition by the antibody, the full spectral range from blue to deep red can be accessed. This work presents the photophysical characterization of the different cumarine derivatives and their corresponding immuno complexes with two different but nevertheless highly specific monoclonal antibodies (mAb). The coumarin dyes and their immunocomplexes were characterized by steady-state time-resolved absorption and fluorescence emission spectroscopy. In addition, fluorescence depolarization measurements were performed to complete the data emphasizing the different binding modes of the two antibodies. In contrast to frequently used detection systems, a massive fluorescence enhancement was found in the formation of the antibody dye complex up to a factor of 50. Because of the slight change in the emission by adjusting the coumarin substitution in the position of the parent molecule which is not relevant for the antigen binding, a dye toolbox which can be used for the construction of competitive multiparameter fluorescence enhancement immunoassays has been created.
Due to their hydrophobic backbone, oligospirothioketal rods (OSTK) are easily incorporated into lipid bilayers and are therefore used as an optical membrane probe. Because of their narrow diameter, only a minimal disturbance of the lipid bilayer is caused. By labeling with fluorescent dyes, novel Förster resonance energy transfer probes are available with highly defined relative orientations of the transition dipole moments of the donor and acceptor dyes, making the class of the OSTK probes into a powerful, flexible toolbox for optical biosensor applications. The incorporation of cumarin and [1,3]-dioxolo[4,5-f][1,3]benzodioxol-labeled OSTK rods in large unilamellar vesicles was investigated using steady-state and time-resolved fluorescence experiments and the results were confirmed by fluorescence depolarization measurements.
The last part of this work deals with the synthesis and characterization of Cou- and DBD-functionalized polyprolines and their coupling to gold nanoparticles. The dye-labeled polyprolines were successfully synthesized. Influences on the spectroscopic properties of the dyes by binding to the polyproline helix were found. The coupling to 5 nm AuNP was successfully carried out. The experience gained by the coupling of the polyprolins to the AuNP can be used as a good basis for the investigation of a single molecule FRET AFM nanoscopy using OSTK rods.

DNA origami-based forster resonance energy-transfer Nanoarrays and their application as ratiometric sensors
DNA origami nanostructures provide a platform where dye molecules can be arranged with nanoscale accuracy allowing to assemble multiple fluorophores without dye-dye aggregation. Aiming to develop a bright and sensitive ratiometric sensor system, we systematically studied the optical properties of nanoarrays of dyes built on DNA origami platforms using a DNA template that provides a high versatility of label choice at minimum cost. The dyes are arranged at distances, at which they efficiently interact by Forster resonance energy transfer (FRET). To optimize array brightness, the FRET efficiencies between the donor fluorescein (FAM) and the acceptor cyanine 3 were determined for different sizes of the array and for different arrangements of the dye molecules within the array. By utilizing nanoarrays providing optimum FRET efficiency and brightness, we subsequently designed a ratiometric pH nanosensor using coumarin 343 as a pH-inert FRET donor and FAM as a pH responsive acceptor. Our results indicate that the sensitivity of a ratiometric sensor can be improved simply by arranging the dyes into a well-defined array. The dyes used here can be easily replaced by other analyte-responsive dyes, demonstrating the huge potential of DNA nanotechnology for light harvesting, signal enhancement, and sensing schemes in life sciences.

FRET Pairs with Fixed Relative Orientation of Chromophores
Synthetic routes to different oligospirothioketal (OSTK) Forster resonance energy transfer (FRET) constructs are described and the photophysics of these constructs were explored in different solvents. The FRET efficiencies were determined from the experimental data and compared with theoretical values. The influence of the outstanding rigidity of the novel OSTK compounds on the FRET is discussed.

Funktionalisierte OS(T)K-Stäbe und Sensorfluorophore zur optischen Sensorik
Functionalized OS(T)K-rods and dyes for optical sensor systems
Im Rahmen dieser Arbeit wurden zum einen erste Oligospiro(thio)ketal (OS(T)K)-basierte Modellsysteme (molekulare Sonden) für abstandsabhängige Messungen mittels Förster-Resonanz-Energietransfer (FRET) und zum anderen Sensorfluorophore, basierend auf einem DBD-Fluorophor und BAPTA, zur Messung der intrazellulären Calcium-Konzentration dargestellt.
Für die Synthese von molekularen Sonden für abstandsabhängige Messungen wurden verschiedenste einfach- und doppelt-markierte OS(T)K-Stäbe entwickelt und spektroskopisch untersucht. Die OS(T)K-Stäbe, sogenannte molekulare Stäbe, dienten als starre Abstandshalter zwischen den Fluorophoren. Als Fluorophore wurden Derivate von 6,7-Dihydroxy-4-methylcoumarin (Donor) und Acyl-DBD (Akzeptor) verwendet, die zusammen ein FRET-Paar bilden. Die Fluorophore wurden so funktionalisiert, dass sie sowohl unbeweglich bzw. „starr“, als auch beweglich bzw. „flexibel“ an den OS(T)K-Stab gebunden werden konnten. Für die Synthese der OS(T)K-Stäbe wurden ebenfalls eine Reihe an unterschiedlich langen und kurzen Stabbausteinen synthetisiert. Auf diese Weise wurden eine Vielzahl an verschiedensten einfach- und doppelt-markierten OS(T)K-Stäben dargestellt, deren Fluorophore sowohl „starr“ als auch „flexibel“ gebunden sind. Die dargestellten Stäbe wurden in verschiedensten Lösungsmitteln spektroskopisch untersucht, um anschließend das Verhalten in Vesikel, die eine biomimetische Umgebung darstellen, zu beurteilen. Es wurde festgestellt, dass sich die Stäbe erfolgreich in die Vesikelmembran einlagerten und hohe FRET-Effizienzen aufweisen.
Des Weiteren wurde ein FRET-Paar dargestellt, das sich durch 2-Photonenabsorpion im NIR-Bereich anregen lässt. Es wurde in den lebenden Zellen mittels Fluoreszenzlebenszeitmikroskopie (FLIM) untersucht.
Zur Untersuchung von intrazellulärem Calcium wurden zwei verschiedene DBD-Fluorophore über einen kurzen Linker mit dem Calcium-Chelator BAPTA verknüpft. Die dargestellten Fluorophore wurden sowohl in vitro als auch in vivo auf ihre Calcium-Sensitivität überprüft. Mittels FLIM wurden in lebenden Zellen die Fluoreszenzlebenszeitverteilungen der Fluorophore nach Calcium-Konzentrationsänderungen detektiert.
In this work, Oligospiro(thio)ketal (OS(T)K)-based model systems (molecular probes) for distance-dependent measurements by Förster resonance energy transfer (FRET) and sensor dyes, based on [1,3]dioxolo[4,5-f][1,3]benzodioxole (DBD) dyes and BAPTA, for measuring the intracellular calcium concentration, were presented. For synthesis of molecular probes, for distance-dependent measurements, various single and double-labeled OS(T)K rods were developed and examined spectroscopically. The OS(T)K rods, called molecular rods, served as rigid spacers between the fluorophores. The FRET pairs consist of a Coumarin (Cou) dye as donor and a DBD dye as acceptor. Cou and DBD were functionalized in such a way that they could be connected both "rigid" as well as "flexible" to the OS(T)K rod. For synthesis of the OS(T)K rods, a number of different long and short rod units were synthesized. In this way, a variety of different single- and double-labeled OS(T)K rods were presented. The rods were spectroscopically analyzed in various solvents to subsequently evaluate the behavior in large unilamellar vesicles (LUVs) that represent a biomimetic system for cell membranes. It was found that the rods successfully incorporated in the vesicle membrane. Furthermore, a FRET pair was shown that can be excited by 2-photon absorption in the NIR region. It was examined in living cells by fluorescence lifetime microscopy (FLIM). To study intracellular calcium, two different DBD dyes were linked via a short linker to the calcium chelator BAPTA. The dyes were examined for their calcium sensitivity both in vitro and in vivo. Using FLIM, the fluorescence lifetime distributions of the dyes were detected in living cells after calcium concentration changes.

Förster resonance energy transfer (FRET)-based nanophotonics using DNA origami structures
Förster-Resonanzenergietransfer (FRET) basierende Nanophotonik auf DNA Origami Strukturen
The field of nanophotonics focuses on the interaction between electromagnetic radiation and matter on the nanometer scale. The elements of nanoscale photonic devices can transfer excitation energy non-radiatively from an excited donor molecule to an acceptor molecule by Förster resonance energy transfer (FRET). The efficiency of this energy transfer is highly dependent on the donor-acceptor distance. Hence, in these nanoscale photonic devices it is of high importance to have a good control over the spatial assembly of used fluorophores. Based on molecular self-assembly processes, various nanostructures can be produced. Here, DNA nanotechnology and especially the DNA origami technique are auspicious self-assembling methods. By using DNA origami nanostructures different fluorophores can be introduced with a high local control to create a variety of nanoscale photonic objects. The applications of such nanostructures range from photonic wires and logic gates for molecular computing to artificial light harvesting systems for artificial photosynthesis.
In the present cumulative doctoral thesis, different FRET systems on DNA origami structures have been designed and thoroughly analyzed. Firstly, the formation of guanine (G) quadruplex structures from G rich DNA sequences has been studied based on a two-color FRET system (Fluorescein (FAM)/Cyanine3 (Cy3)). Here, the influences of different cations (Na+ and K+), of the DNA origami structure and of the DNA sequence on the G-quadruplex formation have been analyzed. In this study, an ion-selective K+ sensing scheme based on the G-quadruplex formation on DNA origami structures has been developed. Subsequently, the reversibility of the G-quadruplex formation on DNA origami structures has been evaluated. This has been done for the simple two-color FRET system which has then been advanced to a switchable photonic wire by introducing additional fluorophores (FAM/Cy3/Cyanine5 (Cy5)/IRDye®700). In the last part, the emission intensity of the acceptor molecule (Cy5) in a three-color FRET cascade has been tuned by arranging multiple donor (FAM) and transmitter (Cy3) molecules around the central acceptor molecule. In such artificial light harvesting systems, the excitation energy is absorbed by several donor and transmitter molecules followed by an energy transfer to the acceptor leading to a brighter Cy5 emission. Furthermore, the range of possible excitation wavelengths is extended by using several different fluorophores (FAM/Cy3/Cy5). In this part of the thesis, the light harvesting efficiency (antenna effect) and the FRET efficiency of different donor/transmitter/acceptor assemblies have been analyzed and the artificial light harvesting complex has been optimized in this respect.
Nanotechnologie hat in den letzten Jahrzehnten durch die Herstellung von Materialien mit außergewöhnlichen Eigenschaften für Anwendungen im Bereich der Medizin und Materialwissenschaften immer mehr an Popularität gewonnen. Die Herstellungsmethoden von Nanostrukturen sind weit gefächert. Auch Desoxyribonukleinsäure (DNS bzw. engl. DNA, deoxyribonucleic acid) kann für die Herstellung von Strukturen im Nanometerbereich genutzt werden. Diese sogenannte DNA-Nanotechnologie wurde in den frühen 1980er Jahren von Nadrian C. Seeman begründet. Ungefähr 30 Jahre später wurde eine neue Methodik für die Herstellung von DNA-Nanostrukturen von Paul W. K. Rothemund entwickelt, die er „scaffold DNA origami“ (Gerüst-DNA-Origami) nannte. DNA-Origami-Nanostrukturen können relativ einfach hergestellt werden und eignen sich perfekt für die Anordnung unterschiedlicher Moleküle (zum Beispiel Fluorophore) mit hoher räumlicher Kontrolle und Präzision. Daher können sie als Substrate genutzt werden, um verschiedene Förster-Resonanzenergietransfer (FRET) Systeme zu entwerfen und zu untersuchen. FRET ist ein strahlungsloser Energietransfer, bei dem die Anregungsenergie von einem Donor- auf ein Akzeptor-Molekül übertragen wird.
In dieser kumulativen Doktorarbeit wurden verschiedene FRET-Systeme auf DNA-Origami-Nanostrukturen entwickelt und mithilfe der Fluoreszenzspektroskopie untersucht. Hierbei wurde zuerst die durch einwertige Kationen (Kalium oder Natrium) induzierte Guanin-Quadruplex-Faltung von freier Telomer-DNA und Telomer-DNA auf DNA-Origami-Strukturen mittels FRET analysiert. Diese Untersuchungen haben gezeigt, dass die freie umgedrehte menschliche Telomer-Sequenz (RevHumTel, 5'-(GGG ATT)4) generell sensitiver auf K+ als auf Na+ reagiert. Durch die Immobilisierung der Telomer-DNA auf DNA-Origami-Strukturen kann eine vollständige Selektivität für K+ erreicht werden. Interessanterweise wird die Ionenselektivität aufgehoben, wenn die menschliche Telomer-Sequenz (HumTel, 5'-(TTA GGG)4) verwendet wird. Basierend auf der G-Quadruplex-Faltung konnten schaltbare FRET-Systeme entwickelt werden, da sich die G-Quadruplexe wieder entfalten, wenn die Kationen mithilfe von zum Beispiel Kryptanden entfernt werden. In den hier untersuchten FRET-Systemen konnte zwischen hoher FRET-Effizienz (gefalteter G-Quadruplex) und niedriger FRET-Effizienz (entfalteter DNA Einzelstrang) durch Zugabe KCl bzw. cryptand gewechselt werden. Da sich DNA-Origami-Strukturen recht einfach modifizieren lassen, wurde das ursprüngliche zwei-Farben-FRET-System durch Hinzufügen eines weiteren etwas rotverschobenen Farbstoffes erweitert (drei-Farben-FRET-Kaskade). Schließlich konnte ein schaltbarer photonischer Draht durch Einfügen eines vierten Farbstoffes entwickelt werden. Die Emissionsintensität des finalen Akzeptors ist in einer einfachen drei-Farben-FRET-Kaskade (ein Donor, ein Transmitter und ein Akzeptor) verhältnismäßig gering und kann durch das Anordnen von mehreren Donor- und Transmitter-Molekülen um ein zentrales Akzeptor-Molekül herum stark erhöht werden. In diesen sogenannten künstlichen Lichtsammelkomplexen absorbieren die Donor-Moleküle das Anregungslicht und übertragen dieses über mehrere FRET-Stufen zum Akzeptor-Molekül. Dadurch wird der Wellenlängenbereich der elektromagnetischen Strahlung, welcher vom Akzeptor absorbiert werden kann, vergrößert und die Emissionsintensität des Akzeptors verstärkt. In diesem Teil der Arbeit wurde die Anzahl der Farbstoffe und die Anordnung dieser unterschiedlichen Farbstoffe variiert und die Lichtsammeleffizienz und FRET-Effizienz bestimmt. Hierbei wurden diese Parameter optimiert und aufgrund der gefundenen Ergebnisse konnten Design-Regeln für solche künstlichen Lichtsammelkomplexe aufgestellt werden.

Ion-Selective formation of a guanine quadruplex on DNA origami structures
DNA origami nanostructures are a versatile tool that can be used to arrange functionalities with high local control to study molecular processes at a single-molecule level. Here, we demonstrate that DNA origami substrates can be used to suppress the formation of specific guanine (G) quadruplex structures from telomeric DNA. The folding of telomeres into G-quadruplex structures in the presence of monovalent cations (e.g. Na+ and K+) is currently used for the detection of K+ ions, however, with insufficient selectivity towards Na+. By means of FRET between two suitable dyes attached to the 3- and 5-ends of telomeric DNA we demonstrate that the formation of G-quadruplexes on DNA origami templates in the presence of sodium ions is suppressed due to steric hindrance. Hence, telomeric DNA attached to DNA origami structures represents a highly sensitive and selective detection tool for potassium ions even in the presence of high concentrations of sodium ions.

Lanthanide und Quantenpunkte : zeitaufgelöste Laserspektroskopie an biochemischen Förster-Resonanzenergietransfer (FRET) Systemen
Förster Resonance Energy Transfer (FRET) plays an important role for biochemical applications such as DNA sequencing, intracellular protein-protein interactions, molecular binding studies, in vitro diagnostics and many others. For qualitative and quantitative analysis, FRET systems are usually assembled through molecular recognition of biomolecules conjugated with donor and acceptor luminophores. Lanthanide (Ln) complexes, as well as semiconductor quantum dot nanocrystals (QD), possess unique photophysical properties that make them especially suitable for applied FRET. In this work the possibility of using QD as very efficient FRET acceptors in combination with Ln complexes as donors in biochemical systems is demonstrated. The necessary theoretical and practical background of FRET, Ln complexes, QD and the applied biochemical models is outlined. In addition, scientific as well as commercial applications are presented. FRET can be used to measure structural changes or dynamics at distances ranging from approximately 1 to 10 nm. The very strong and well characterized binding process between streptavidin (Strep) and biotin (Biot) is used as a biomolecular model system. A FRET system is established by Strep conjugation with the Ln complexes and QD biotinylation. Three Ln complexes (one with Tb3+ and two with Eu3+ as central ion) are used as FRET donors. Besides the QD two further acceptors, the luminescent crosslinked protein allophycocyanin (APC) and a commercial fluorescence dye (DY633), are investigated for direct comparison. FRET is demonstrated for all donor-acceptor pairs by acceptor emission sensitization and a more than 1000-fold increase of the luminescence decay time in the case of QD reaching the hundred microsecond regime. Detailed photophysical characterization of donors and acceptors permits analysis of the bioconjugates and calculation of the FRET parameters. Extremely large Förster radii of more than 100 Å are achieved for QD as acceptors, considerably larger than for APC and DY633 (ca. 80 and 60 Å). Special attention is paid to interactions with different additives in aqueous solutions, namely borate buffer, bovine serum albumin (BSA), sodium azide and potassium fluoride (KF). A more than 10-fold limit of detection (LOD) decrease compared to the extensively characterized and frequently used donor-acceptor pair of Europium tris(bipyridine) (Eu-TBP) and APC is demonstrated for the FRET system, consisting of the Tb complex and QD. A sub-picomolar LOD for QD is achieved with this system in azide free borate buffer (pH 8.3) containing 2 % BSA and 0.5 M KF. In order to transfer the Strep-Biot model system to a real-life in vitro diagnostic application, two kinds of imunnoassays are investigated using human chorionic gonadotropin (HCG) as analyte. HCG itself, as well as two monoclonal anti-HCG mouse-IgG (immunoglobulin G) antibodies are labeled with the Tb complex and QD, respectively. Although no sufficient evidence for FRET can be found for a sandwich assay, FRET becomes obvious in a direct HCG-IgG assay showing the feasibility of using the Ln-QD donor-acceptor pair as highly sensitive analytical tool for in vitro diagnostics.
Förster Resonanzenergietransfer (FRET) spielt eine wichtige Rolle in biochemischen Anwendungen, wie z.B. DNA-Sequenzierung, intrazellulären Protein-Protein-Wechselwirkungen, molekularen Bindungsstudien, in-vitro-Diagnostik und vielen anderen. Zur quantitativen und qualitativen Analyse werden FRET Systeme normalerweise durch molekulare Erkennung von Biomolekülen, die mit Donator- und Acceptorluminophoren markiert sind, ermöglicht. Durch die besonderen photophysikalischen Eigenschaften sowohl von Lanthanidkomplexen (Ln-Komplexen), als auch Halbleiternanokristallen (sog. Quantenpunkten oder Quantumdots - QD), sind diese besonders für FRET Anwendungen geeignet. In der vorliegenden Arbeit wird effizienter FRET zwischen Ln-Komplexen und QD in biochemischen Systemen demonstriert. Die notwendigen theoretischen und praktischen Grundlagen über FRET, Ln-Komplexe, QD und die verwendeten biochemischen Modelle werden dargestellt, und wissenschaftliche als auch kommerzielle Anwendungen werden präsentiert. FRET kann zur Messung von strukturellen Veränderungen und Dynamiken im Bereich von ca. 1 bis 10 nm verwendet werden. Der sehr starke und gut charakterisierte Bindungsprozess zwischen Streptavidin (Strep) und Biotin (Biot) wird als biomolekulares Modellsystem eingesetzt. Ein FRET System wird durch Streptavidinkonjugation mit Ln-Komplexen und QD-Biotinylierung etabliert. Drei Ln-Komplexe (einer mit Tb3+ und zwei mit Eu3+ als Zentralion) werden als Donatoren verwendet, und neben QD werden zwei weitere Acceptoren, das lumineszierende, quervernetzte Protein Allophycocyanin (APC) und ein kommerzieller Fluoreszenzfarbstoff (DY633), untersucht. FRET kann für alle Donator-Acceptor Paare nachgewiesen werden, zum einen durch sensibilisierte Acceptorlumineszenz und zum anderen durch eine über 1000-fach erhöhte Lumineszenzabklingzeit der QD mit über 100 Mikrosekunden. Mittels detailierter photophysikalischer Charakterisierung der Donatoren und Acceptoren können die Biokonjugate analysiert und die FRET Parameter berechnet werden. Für die QD FRET Systeme ergeben sich extrem große Försterradien von über 100 Å, die wesentlich größer sind als für APC und DY633 (ca. 80 bzw. 60 Å). Besondere Aufmerksamkeit gilt der Wechselwirkung mit den Zusatzreagenzien Boratpuffer, Bovines Serumalbumin (BSA), Natriumazid und Kaliumfluorid (KF) in den wässrigen Lösungen. Im Vergleich zum ausgiebig charakterisierten und vielfach verwendeten Donator-Acceptor Paar aus Europium-tris(Bipyridin) (Eu-TBP) und APC wird eine mehr als 10-fache Senkung der Nachweisgrenze für das FRET-System, bestehend aus Tb-Komplex und QD, erreicht. In azidfreiem Boratpuffer (pH 8,3) mit 2 % BSA und 0,5 M KF wird eine subpicomolare QD-Nachweisgrenze für dieses System aufgezeigt. Um den Transfer des Strep-Biot Modellsystems in eine echte in-vitro-diagnostische Anwendung zu demonstrieren, werden zwei Immuntests zum HCG-(Humanes Choriongonadotropin)-Nachweis untersucht. Sowohl HCG als auch monoklonale anti-HCG Maus-IgG-(Immunoglobulin G)-Antikörper werden mit dem Tb-Komplex bzw. mit QD markiert. Obwohl kein ausreichender Nachweis für FRET in einem immunometrischen Assay (oder Sandwichassay) erbracht werden kann, wird FRET in einem direkten HCG-IgG Assay erzielt, wodurch die Realisierbarkeit von Ln-QD Donator-Acceptor Paaren zur hochsensitiven Anwendung in der in-vitro-Diagnostik gezeigt werden kann.

Polyproline and the "spectroscopic ruler" revisited with single-molecule fluorescence
To determine whether Förster resonance energy transfer (FRET) measurements can provide quantitative distance information in single-molecule fluorescence experiments on polypeptides, we measured FRET efficiency distributions for donor and acceptor dyes attached to the ends of freely diffusing polyproline molecules of various lengths. The observed mean FRET efficiencies agree with those determined from ensemble lifetime measurements but differ considerably from the values expected from Förster theory, with polyproline treated as a rigid rod. At donor–acceptor distances much less than the Förster radius R0, the observed efficiencies are lower than predicted, whereas at distances comparable to and greater than R0, they are much higher. Two possible contributions to the former are incomplete orientational averaging during the donor lifetime and, because of the large size of the dyes, breakdown of the point-dipole approximation assumed in Förster theory. End-to-end distance distributions and correlation times obtained from Langevin molecular dynamics simulations suggest that the differences for the longer polyproline peptides can be explained by chain bending, which considerably shortens the donor–acceptor distances.

Quantenpunktbasiertes spektroskopisches Lineal mit Terbium-Komplexen als Donoren für optische FRET-Multiplexmessungen
Quantum-dot based spectroscopic ruler with terbium-complexes as donors for multiplexed optical FRET measurements
Der Förster-Resonanzenergietransfer (FRET) liefert einen wichtigen Beitrag bei der Untersuchung kleinskaliger biologischer Systeme und Prozesse. Möglich wird dies durch die r-6-Abhängigkeit des FRET, die es erlaubt Abstände und strukturelle Änderungen weit unterhalb der Beugungsgrenze des Lichts mit hoher Sensitivität und geringem Aufwand zu bestimmen. Die besonderen photophysikalischen Eigenschaften von Terbiumkomplexen (LTC) und Quantenpunkten (QD) machen sie zu geeigneten Kandidaten für hochsensitive und störungsarme Multiplex-Abstandsmessungen in biologischen Systemen und Prozessen. Die Abstandsbestimmungen setzen jedoch eine genaueste Kenntnis des Mechanismus des Energietransfers von LTC auf QD ebenso voraus, wie das Wissen um Größe und Gestalt letzterer. Quantenpunkte haben im Vergleich zu biologischen Strukturen ähnliche Dimensionen und können nicht als punktförmig betrachtet werden, wie es bei einfacheren Farbstoffen möglich ist. Durch ihre Form kommt es zu einer Abstandsverteilung innerhalb des Donor-Akzeptorsystems. Dies beeinflusst den Energietransfer und damit die experimentellen Ergebnisse. In dieser Arbeit wurde der Energietransfer von LTC auf QD untersucht, um zu einer Aussage hinsichtlich des Mechanismus der Energieübertragung und der dabei zu berücksichtigenden photophysikalischen und strukturellen Parameter von LTC und QD zu gelangen. Mit der Annahme einer Abstandsverteilung sollten die Größen der Quantenpunkte bestimmt und der Einfluss von Form und Gestalt auf den Energietransfer betrachtet werden. Die notwendigen theoretischen und praktischen Grundlagen wurden eingangs dargestellt. Daran schlossen sich Messungen zur photophysikalischen Charakterisierung der Donoren und Akzeptoren an, die Grundlage der Berechnung der FRET-Parameter waren. Die Förster-Radien zeigten die für den FRET von LTC auf QD typischen extrem hohen Werte von bis zu 11 nm. Zeitaufgelöste Messungen der FRET-induzierten Lumineszenz der Donoren und Akzeptoren in den beiden biomolekularen Modellsystemen Zink-Histidin und Biotin-Streptavidin beschlossen den praktischen Teil. Als Donor wurde Lumi4Tb gebunden an ein Peptid bzw. Streptavidin genutzt, Akzeptoren waren fünf verschiedene, kommerziell erhältliche Quantenpunkte mit Carboxyl- bzw. Biotinfunktionalisierung. Bei allen Donor-Akzeptor-Paarungen konnte FRET beobachtet und ausgewertet werden. Es konnte gezeigt werden, dass die gesamte Emission des Terbiums zum Energietransfer beiträgt und der Orientierungsfaktor ² den Wert 2/3 annimmt. Die Charakterisierung der Bindungsverhältnisse innerhalb der FRET-Paare von LTC und QD über Verteilungsfunktionen bietet über die Form der Verteilungskurve die Möglichkeit Aussagen über die Gestalt der FRET-Partner zu treffen. So war es möglich, die mittlere Form der Quantenpunkte als Sphäre zu bestimmen. Dies entsprach, insbesondere bei den in z-Richtung des Kristallgitters elongierten Quantenpunkten, nicht den Erwartungen. Dieser Befund ermöglicht daher bei zukünftigen Messungen eine Verbesserung der Genauigkeit bei Abstandsbestimmungen mit Quantenpunkten. Neben der Ermittlung der die FRET-Verteilung bestimmenden Gestalt der Quantenpunkte konnte im Rahmen dieser Arbeit anhand vergleichender Messungen die Dicke der Polymerhülle der QD bestimmt und so gezeigt werden, dass FRET-Paare aus lumineszenten Terbiumkomplexen und Quantenpunkten in der Lage sind, Abstände im Nano- bis Sub-Nanometerbereich aufzulösen.
Förster resonance energy transfer (FRET) plays an important role in the study of small-scale biological systems and processes. This is made possible by the r-6-dependence of FRET, which allows for determination of distances and structural changes far below the diffraction limit of light with high sensitivity and low costs. The unique photophysical properties of terbium complexes (LTC) and quantum dots (QDs) make them suitable candidates for high-sensitivity, low-noise multiplex distance measurements in biological systems and processes. Estimating distances with these FRET-pairs requires a precise knowledge of the mechanism of energy transfer from LTC to QD as well as the knowledge of size and shape of the latter. Quantum dots have, compared to biological structures, similar dimensions and therefore can not be considered as point-like, as it is possible with smaller dyes. Due to their shape, there is a distance distribution within the donor-acceptor system. This influences the energy transfer and hence the experimental results. In this work, the energy transfer from LTC to QD was examined to come to a conclusion regarding the mechanism of energy transfer and the photophysical and structural parameters of LTC and QD to be considered. The adoption of a FRET-distance distribution due to a size distribution of quantum dots should yield to a size estimation of the nanoparticles as well as a conclusion of the influence of shape and form on energy transfer. The necessary theoretical and practical principles were described at the outset of this work. This description of the basic concepts was followed by the photophysical characterization of the donors and acceptors and the calculation of FRET parameters. The calculated Förster radii were typical for the FRET from LTC to QD and showed extremely high values of up to 11 nm. Time-resolved measurements of the FRET-induced luminescence of donors and acceptors in two biomolecular model binding systems namely zinc-histidine and biotin-streptavidin binding rounded the practical part. FRET-donors used were commercially available Lumi4Tb complexes bound to streptavidin or a peptide, respectively. As FRET-acceptors five different commercially available quantum dots with carboxyl- or biotin-functionalisation were used. For all donor-acceptor pairs FRET could be observed and evaluated. It could be shown that the whole emission of terbium contributes to energy transfer. Furthermore the orientation factor ² was estimated to have a value of 2/3 when using LTC as FRET-donors and QD as FRET-acceptors. The characterization of the bonding within the FRET pairs of LTC and QD with distribution functions allows for statements about the shape of the FRET partners via shape of the distribution curves. It was possible to determine the average shape of the quantum dots as a sphere. This outcome was, especially for (in z-direction of the crystal lattice) elongated quantum dots, in the contrary to the expectations. This finding therefore allows for improving the accuracy of distance determinations with quantum dots. Based on comparative measurements it was also possible to determine the thickness of the polymer shell of the QD demonstrating that FRET pairs of luminescent terbium complexes and quantum dots are capable of determining distances in the nanometer to sub-nanometer range.
