Alterations in the maternal or fetal metabolism might impact on fetal growth and influence birth outcomes. Discerning associations between the maternal and fetal metabolome and surrogate parameters of fetal growth could give new insight into the complex relationship between intrauterine conditions, birth outcomes, and later life disease susceptibility. Methods: Using flow injection tandem mass spectrometry, targeted metabolomics was performed in serum samples obtained from 226 mother/child pairs at delivery. Associations between neonatal birth weight and concentrations of 163 maternal and fetal metabolites were analyzed. Results: After FDR adjustment using the Benjamini-Hochberg procedure lysophosphatidylcholines (LPC) 14:0, 16:1, and 18:1 were strongly positively correlated with birth weight. In a stepwise linear regression model corrected for established confounding factors of birth weight, LPC 16: 1 showed the strongest independent association with birth weight (CI: 93.63 - 168.94; P = 6.94x10(-11)). The association with birth weight was stronger than classical confounding factors such as offspring sex (CI: - 258.81- -61.32; P = 0.002) and maternal smoking during pregnancy (CI: -298.74 - -29.51; P = 0.017). Conclusions: After correction for multiple testing and adjustment for potential confounders, LPC 16:1 showed a very strong and independent association with birth weight. We used a system of 46 small wetlands (kettle holes)—natural small-scale freshwater habitats rarely considered in nature conservation policies—embedded in an intensively managed agricultural matrix in northern Germany. We compared two types of kettle holes with distinct topographies (flatsloped, ephemeral, frequently plowed kettle holes vs. steep-sloped, more permanent ones) and determined 254 vascular plant species within these ecosystems, as well as plant functional traits and nearest neighbor distances to other kettle holes. Differences in alpha and beta diversity between steep permanent compared with ephemeral flat kettle holes were mainly explained by species sorting and niche processes and mass effect processes in ephemeral flat kettle holes. The plant-community composition as well as the community trait distribution in terms of life span, breeding system, dispersal ability, and longevity of seed banks significantly differed between the two habitat types. Flat ephemeral kettle holes held a higher percentage of non-perennial plants with a more persistent seed bank, less obligate outbreeders and more species with seed dispersal abilities via animal vectors compared with steep-sloped, more permanent kettle holes that had a higher percentage of wind-dispersed species. In the flat kettle holes, plant-species richness was negatively correlated with the degree of isolation, whereas no such pattern was found for the permanent kettle holes. Synthesis: Environment acts as filter shaping plant diversity (alpha and beta) and plant-community trait distribution between steep permanent compared with ephemeral flat kettle holes supporting species sorting and niche mechanisms as expected, but we identified a mass effect in ephemeral kettle holes only. Our results, within a perfect-model experiment setting, indicate that the "online DA" approach did not outperform the "off-line" one, despite its considerable additional implementation complexity. On the other hand, it was observed that the nonlinear response of tree growth to surface temperature and soil moisture does deteriorate the operation of the time-averaged EnKF methodology. Moreover, for the first time we show that this skill loss appears significantly sensitive to the structure of the growth rate function, used to represent the principle of limiting factors (PLF) within the forward model. Seasonal drought early warning is thus vital for the whole region. In this study, the predictability of hydrological droughts during the main runoff period from December to May is assessed using statistical approaches. Three methods (multiple linear models, artificial neural networks, random forest regression trees) are compared in terms of their ability to forecast streamflow with up to 12 months of lead time. The following four main findings result from the study. 1. There are stations in the basin at which standardised streamflow is predictable with lead times up to 12 months. The results show high inter-station differences of forecast skill but reach a coefficient of determination as high as 0.73 (cross validated). 2. A large range of potential predictors is considered in this study, comprising well-established climate indices, customised teleconnection indices derived from sea surface temperatures and antecedent streamflow as a proxy of catchment conditions. El Nino and customised indices, representing sea surface temperature in the Atlantic and Indian oceans, prove to be important teleconnection predictors for the region. Antecedent streamflow is a strong predictor in small catchments (with median 42\% explained variance), whereas teleconnections exert a stronger influence in large catchments. 3. Multiple linear models show the best forecast skill in this study and the greatest robustness compared to artificial neural networks and random forest regression trees, despite their capabilities to represent nonlinear relationships. 4. Employed in early warning, the models can be used to forecast a specific drought level. Even if the coefficient of determination is low, the forecast models have a skill better than a climatological forecast, which is shown by analysis of receiver operating characteristics (ROCs). In particular, the polar jet is located between cold arctic air to the north and warmer subtropical air to the south. Strongly meandering states therefore often lead to extreme surface weather. Some algorithms exist which can detect the 2-D (latitude and longitude) jets' core around the hemisphere, but all of them use a minimal threshold to determine the subtropical and polar jet stream. This is particularly problematic for the polar jet stream, whose wind velocities can change rapidly from very weak to very high values and vice versa. We develop a network-based scheme using Dijkstra's shortest-path algorithm to detect the polar and subtropical jet stream core. This algorithm not only considers the commonly used wind strength for core detection but also takes wind direction and climatological latitudinal position into account. Furthermore, it distinguishes between polar and subtropical jet, and between separate and merged jet states. The parameter values of the detection scheme are optimized using simulated annealing and a skill function that accounts for the zonal-mean jet stream position (Rikus, 2015). After the successful optimization process, we apply our scheme to reanalysis data covering 1979-2015 and calculate seasonal-mean probabilistic maps and trends in wind strength and position of jet streams. We present longitudinally defined probability distributions of the positions for both jets for all on the Northern Hemisphere seasons. This shows that winter is characterized by two well-separated jets over Europe and Asia (ca. 20 degrees W to 140 degrees E). In contrast, summer normally has a single merged jet over the western hemisphere but can have both merged and separated jet states in the eastern hemisphere. We find that globally available DEMs commonly overestimate river elevations in steep topography. The distributions of elevation errors become increasingly wider and right skewed if adjacent hillslope gradients are steep. Our analysis indicates that the AW3D DEM has the highest precision and lowest bias for the analysis of river profiles in mountainous topography. The new 12m resolution TanDEM-X DEM has a very low precision, most likely due to the combined effect of steep valley walls and the presence of water surfaces in valley bottoms. These models are Earth-like in terms of convective vigour, thermal structure, surface heat-flux and the geographic distribution of heterogeneity. The models generate a degree-2-dominated spectrum of dynamic topography with negative amplitudes above subducted slabs (i.e. circum-Pacific regions and southern Eurasia) and positive amplitudes elsewhere (i.e. Africa, north-western Eurasia and the central Pacific). Model predictions are compared with published observations and subsidence patterns from well data, both globally and for the Australian and southern African regions. We find that our models reproduce the long-wavelength component of these observations, although observed smaller-scale variations are not reproduced. We subsequently define "geodynamic rules" for how different surface tectonic settings are affected by mantle processes: (i) locations in the vicinity of a subduction zone show large negative dynamic topography amplitudes; (ii) regions far away from convergent margins feature long-term positive dynamic topography; and (iii) rapid variations in dynamic support occur along the margins of overriding plates (e.g. the western US) and at points located on a plate that rapidly approaches a subduction zone (e.g. India and the Arabia Peninsula). These 3-D models differentiate various sedimentary, crustal, and mantle units and integrate different geophysi- cal data such as seismic observations and the gravity field. We extracted the temperature-depth distributions in 1 km intervals down to 6 km below the upper thermal boundary condition. The geothermal gradient was then calculated for these intervals between the upper thermal boundary condi- tion and the respective depth levels (1, 2, 3, 4, 5, and 6 km below the upper thermal boundary condition). According to our results, the geothermal gradient decreases with increas- ing depth and shows varying lateral trends and values for these two different margins. We compare the 3-D geologi- cal structural models and the geothermal gradient variations for both thermal models and show how radiogenic heat pro- duction, sediment insulating effect, and thermal lithosphere- asthenosphere boundary (LAB) depth influence the shallow thermal field pattern. Aphids are among the most detrimental agricultural pests worldwide, and parasitoid wasps are frequently employed for their control. The use of asexual parasitoids may improve the effectiveness of biological control because only females kill hosts and because asexual populations have a higher growth rate than sexuals. However, asexuals may have a reduced capacity to track evolutionary change in their host populations. We used a factorial experiment to compare the ability of sexual and asexual populations of the parasitoid Lysiphlebus fabarum to control caged populations of black bean aphids (Aphis fabae) of high and low clonal diversity. The aphids came from a natural population, and one-­third of the aphid clones harbored Hamiltonella defensa, a heritable bacterial endosymbiont that increases resistance to parasitoids. We followed aphid and parasitoid population dynamics for 3 months but found no evidence that the reproductive mode of parasitoids affected their effectiveness as biocontrol agents, independent of host clonal diversity. Parasitoids failed to control aphids in most cases, because their introduction resulted in strong selection for clones protected by H. defensa. The increasingly resistant aphid populations escaped control by parasitoids, and we even observed parasitoid extinctions in many cages. The rapid evolution of symbiont-­conferred resistance in turn imposed selection on parasitoids. In cages where asexual parasitoids persisted until the end of the experiment, they became dominated by a single genotype able to overcome the protection provided by H. defensa. Thus, there was evidence for parasitoid counteradaptation, but it was generally too slow for parasitoids to regain control over aphid populations. Intolerant rice varieties generally lack the SUB1A gene but some intermediate tolerant varieties, such as IR64, carry the allelic variant SUB1A-2. Differential effects of the two alleles have so far not been addressed. As a first step, we have therefore quantified and compared the expression of nearly 2500 rice TF genes between IR64 and its derived tolerant near isogenic line IR64-Sub1, which carries the SUB1A-1 allele. Gene expression was studied in internodes, where the main difference in expression between the two alleles was previously shown. Results: Nineteen and twenty-six TF genes were identified that responded to submergence in IR64 and IR64-Sub1, respectively. Only one gene was found to be submergence-responsive in both, suggesting different regulatory pathways under submergence in the two genotypes. These differentially expressed genes (DEGs) mainly included MYB, NAC, TIFY and Zn-finger TFs, and most genes were downregulated upon submergence. In IR64, but not in IR64-Sub1, SUB1B and SUB1C, which are also present in the Sub1 locus, were identified as submergence responsive. Four TFs were not submergence responsive but exhibited constitutive, genotype-specific differential expression. Most of the identified submergence responsive DEGs are associated with regulatory hormonal pathways, i.e. gibberellins (GA), abscisic acid (ABA), and jasmonic acid (JA), apart from ethylene. An in-silico promoter analysis of the two genotypes revealed the presence of allele-specific single nucleotide polymorphisms, giving rise to ABRE, DRE/CRT, CARE and Site II cis-elements, which can partly explain the observed differential TF gene expression. We measured certain fitness-related aboveground traits and used them along literature-derived traits to determine the influence of morphological similarities on the strength and direction of the feedback. We found that the average strength of negative feedback increased from the intraspecific over the interspecific to the functional group level. However, individual accessions and species differed in the direction and strength of the feedback. None of our results could be explained by morphological dissimilarities or individual traits. Synthesis. Our results indicate that negative plant-soil feedback is stronger if the involved plants belong to more distantly related species. Our results indicated that the split between the two species took place during the Pleistocene, similar to 1.8Mya, possibly driven by adaptations of R. timorensis to the drier climate found on Java compared to the other islands of Sundaland. Although both markers identified two well-differentiated clades, there was a largely discrepant pattern between mitochondrial and nuclear markers. While nDNA separated the individuals into the two species, largely in agreement with their museum label, mtDNA revealed that all R. timorensis sampled to the east of the Sunda shelf carried haplotypes from R. unicolor and one Rusa unicolor from South Sumatra carried a R. timorensis haplotype. This process was completely blocked by the treatment of a combination of the ETA receptor blockers-BQ123 and the ETB receptor blocker BQ788. The results highlight the presence of smoke layers of 1-2 km thickness, located at about 5 km a.s.l. altitude over Granada and Leipzig and around 2.5 km a.s.l. at Warsaw. These layers were intense, as they accounted for more than 30\% of the total AOD (aerosol optical depth) in all cases, and presented optical and microphysical features typical for different aging degrees: color ratio of lidar ratios (LR532/LR355) around 2, alpha-related angstrom exponents of less than 1, effective radii of 0.3 mu m and large values of single scattering albedos (SSA), nearly spectrally independent. The Hominin Sites and Paleolakes Drilling Project (HSPDP) was developed to address these issues. The project has focused its efforts on the eastern African Rift Valley, where much of the evidence for early hominins has been recovered. We have collected about 2 km of sediment drill core from six basins in Kenya and Ethiopia, in lake deposits immediately adjacent to important fossil hominin and archaeological sites. Collectively these cores cover in time many of the key transitions and critical intervals in human evolutionary history over the last 4 Ma, such as the earliest stone tools, the origin of our own genus Homo, and the earliest anatomically modern Homo sapiens. The Kathmandu Valley is a bowl-shaped basin with a severe air pollution problem. This paper reports measurements of two major greenhouse gases (GHGs), methane (CH4) and carbon dioxide (CO2), along with the pollutant CO, that began during the campaign and were extended for 1 year at the SusKat-ABC supersite in Bode, a semi-urban location in the Kathmandu Valley. Simultaneous measurements were also made during 2015 in Bode and a nearby rural site (Chanban) similar to 25 km (aerial distance) to the southwest of Bode on the other side of a tall ridge. The ambient mixing ratios of methane (CH4), carbon dioxide (CO2), water vapor, and carbon monoxide (CO) were measured with a cavity ring-down spectrometer (G2401; Picarro, USA) along with meteorological parameters for 1 year (March 2013-March 2014). These measurements are the first of their kind in the central Himalayan foothills. At Bode, the annual average mixing ratios of CO2 and CH4 were 419.3 (+/- 6.0) ppm and 2.192 (+/- 0.066) ppm, respectively. These values are higher than the levels observed at background sites such as Mauna Loa, USA (CO2: 396.8 +/- 2.0 ppm, CH4: 1.831 +/- 0.110 ppm) and Waliguan, China (CO2: 397.7 +/- 3.6 ppm, CH4: 1.879 +/- 0.009 ppm) during the same period and at other urban and semi-urban sites in the region, such as Ahmedabad and Shadnagar (India). They varied slightly across the seasons at Bode, with seasonal average CH4 mixing ratios of 2.157 (+/- 0.230) ppm in the pre-monsoon season, 2.199 (+/- 0.241) ppm in the monsoon, 2.210 (+/- 0.200) ppm in the post-monsoon, and 2.214 (+/- 0.209) ppm in the winter season. The average CO2 mixing ratios were 426.2 (+/- 25.5) ppm in the pre-monsoon, 413.5 (+/- 24.2) ppm in the monsoon, 417.3 (+/- 23.1) ppm in the postmonsoon, and 421.9 (+/- 20.3) ppm in the winter season. The maximum seasonal mean mixing ratio of CH4 in winter was only 0.057 ppm or 2.6\% higher than the seasonal minimum during the pre-monsoon period, while CO2 was 12.8 ppm or 3.1\% higher during the pre-monsoon period (seasonal maximum) than during the monsoon (seasonal minimum). On the other hand, the CO mixing ratio at Bode was 191\% higher during the winter than during the monsoon season. The enhancement in CO2 mixing ratios during the pre-monsoon season is associated with additional CO2 emissions from forest fires and agro-residue burning in northern South Asia in addition to local emissions in the Kathmandu Valley. Published CO = CO2 ratios of different emission sources in Nepal and India were compared with the observed CO = CO2 ratios in this study. This comparison suggested that the major sources in the Kathmandu Valley were residential cooking and vehicle exhaust in all seasons except winter. In winter, brick kiln emissions were a major source. Simultaneous measurements in Bode and Chanban (15 July-3 October 2015) revealed that the mixing ratios of CO2, CH4, and CO were 3.8, 12, and 64\% higher in Bode than Chanban. The Kathmandu Valley thus has significant emissions from local sources, which can also be attributed to its bowl-shaped geography that is conducive to pollution build-up. At Bode, all three gas species (CO2, CH4, and CO) showed strong diurnal patterns in their mixing ratios with a pronounced morning peak (ca. 08:00), a dip in the afternoon, and a gradual increase again through the night until the next morning. CH4 and CO at Chanban, however, did not show any noticeable diurnal variations. This scenario is supported by data-constrained numerical simulations of the eruption. Observations show that dense cool filament plasma was re-directed and heated in situ, producing coronal-temperature emission around the reconnection region. We present a simple calculation algorithm to separate open-water CH4 fluxes measured with automatic chambers into diffusion- and ebullition-derived components. This helps to reveal underlying dynamics, to identify potential environmental drivers and, thus, to calculate reliable CH4 emission estimates. The flux separation is based on identification of ebullition-related sudden concentration changes during single measurements. Therefore, a variable ebullition filter is applied, using the lower and upper quartile and the interquartile range (IQR). Automation of data processing is achieved by using an established R script, adjusted for the purpose of CH4 flux calculation. The algorithm was validated by performing a laboratory experiment and tested using flux measurement data (July to September 2013) from a former fen grassland site, which converted into a shallow lake as a result of rewetting. Ebullition and diffusion contributed equally (46 and 55 \%) to total CH4 emissions, which is comparable to ratios given in the literature. Moreover, the separation algorithm revealed a concealed shift in the diurnal trend of diffusive fluxes throughout the measurement period. The proposed strategy processes a time-resolved sequence of flow snapshots in two steps. First, the snapshot data are clustered into a small number of representative states, called centroids, in the state space. These centroids partition the state space in complementary non-overlapping regions (centroidal Voronoi cells). Departing from the standard algorithm, the probabilities of the clusters are determined, and the states are sorted by analysis of the transition matrix. Second, the transitions between the states are dynamically modelled using a Markov process. Physical mechanisms are then distilled by a refined analysis of the Markov process, e. g. using finite-time Lyapunov exponent (FTLE) and entropic methods. This CROM framework is applied to the Lorenz attractor (as illustrative example), to velocity fields of the spatially evolving incompressible mixing layer and the three-dimensional turbulent wake of a bluff body. For these examples, CROM is shown to identify non-trivial quasi-attractors and transition processes in an unsupervised manner. A total of four different diets with increasing vitamin A content between 5.24 and 104.80 mu mol retinol (5000-100 000 IU vitamin A)/4184 kJ (1000 kcal) metabolisable energy were fed from the age of 8 weeks up to 52 (MS) and 78 weeks (LAB). The daily energy intake was recorded throughout the experimental period. The body condition score was evaluated weekly using a seven-category system, and food allowances were adjusted to maintain optimal body condition. Body composition was assessed at the age of 26 and 52 weeks for both breeds and at the age of 78 weeks for the LAB breed only using dual-energy X-ray absorptiometry. The growth curves of the dogs followed a breed-specific pattern. However, data on energy intake showed considerable variability between the two breeds as well as when compared with predicted energy intake. The human intestinal microbiome is enriched in genes involved in the degradation of indigestible polysaccharides. The extent to which dietary fibres are fermented and in which molar ratio SCFA are formed depends on their physicochemical properties and on the individual microbiome. Acetate and propionate play an important role in lipid and glucose metabolism. Acetate serves as a substrate for de novo lipogenesis in liver, whereas propionate can be utilised for gluconeogenesis. The conversion of fermentable dietary fibre to SCFA provides additional energy to the host which could promote obesity. However, epidemiologic studies indicate that diets rich in fibre rather prevent than promote obesity development. This may be due to the fact that SCFA are also ligands of free fatty acid receptors (FFAR). Activation of FFAR leads to an increased expression and secretion of enteroendocrine hormones such as glucagon-like-peptide 1 or peptide YY which cause satiety. In the CMC, Ar-Ar spot ages locally date white-mica formation at peak P-T conditions and during early exhumation at 279-242 Ma. In a local garnet mica-schist intercalation (570-585 degrees C, 11-13 kbar) Ar-Ar spot ages refer to the ascent from the subduction channel at 307-274 Ma. Portions of the CMC were isobarically heated to 510-580 degrees C at 6.6-8.5 kbar. The age of peak P-T conditions in the AF can only vaguely be approximated at >= 310 Ma by relict fission-track ages consistent with the observation that frontal accretion occurred prior to basal accretion. Zircon fission-track dating indicates cooling below similar to 280 degrees C at similar to 248 Ma in the CMC and the AF, when a regional unconformity also formed. Ar-Ar white-mica spot ages in parts of the CMC and within the entire AF and HF point to heterogeneous resetting during Mesozoic extensional and shortening events at similar to 245-240 Ma, similar to 210-200 Ma, similar to 174-159 Ma and similar to 142-127 Ma. The zircon fission-track ages are locally reset at 109-96 Ma. All resetting of Ar-Ar white-mica ages is proposed to have occurred by in situ dissolution/precipitation at low temperature in the presence of locally penetrating hydrous fluids. In contrast to quantitative methods that permit fine-grained (kinetic) analysis, qualitative approaches allow for addressing large-scale networks. Sixty-five athletes (22.83 years +/- 2.45; 25 women) were randomly assigned to an incentive-to-fake condition or a control condition. Participants in the incentive-to-fake condition were manipulated to believe that athletes with lenient doping attitudes would be referred to a tedious 45-minute anti-doping program. Attitudes were measured with the pictorial doping brief IAT (BIAT) and with the Performance Enhancement Attitude Scale (PEAS). A one-way MANOVA revealed significant differences between conditions after the manipulation in PEAS scores, but not in the doping BIAT. In the light of our hypothesis this suggests that participants successfully faked an exceedingly negative attitude to doping when completing the PEAS, but were unsuccessful in doing so on the reaction time-based test. This study assessed BIAT faking in a setting that aimed to resemble a situation in which participants want to hide their attempts to cheat. The two measures of attitude were differentially affected by the implicit incentive. Our findings provide evidence that the pictorial doping BIAT is relatively robust against spontaneous and naive faking attempts. Methods: Data from 12,910 outpatients with dementia (mean age 79.2 years; SD = 7.6 years) treated between January 2003 and December 2013 in Germany were included. Continuous treatment was analysed using Kaplan-Meier curves and log-rank tests. In addition, multivariate Cox regression models were fitted with continuous treatment as dependent variable and the predictors antidementia agent, age, gender, medical comorbidities, physician specialty, and health insurance status. Results: After one year of follow-up, nearly 60\% of patients continued drug treatment. Donezepil (HR: 0.88; 95\% CI: 0.82-0.95) and memantine (HR: 0.85; 0.79-0.91) patients were less likely to be discontinued treatment as compared to rivastigmine users. Patients were less likely to be discontinued if they were treated by specialist physicians as compared to general practitioners (HR: 0.44; 0.41-0.48). Younger male patients and patients who had private health insurance had a lower discontinuation risk. Regarding comorbidity, patients were more likely to be continuously treated with the index substance if a diagnosis of heart failure or hypertension had been diagnosed at baseline. To investigate the interconnection of different monsoon systems during the pre-industrial Holocene, five transient global climate model simulations have been analysed with respect to the rainfall trend and variability in different sub-domains of the Afro-Asian monsoon region. Our analysis suggests that on millennial timescales with varying orbital forcing, the monsoons do not behave as a tightly connected global system. According to the models, the Indian and North African monsoons are coupled, showing similar rainfall trend and moderate correlation in centennial rainfall variability in all models. The East Asian monsoon changes independently during the Holocene. The dissimilarities in the seasonality of the monsoon sub-systems lead to a stronger response of the North African and Indian monsoon systems to the Holocene insolation forcing than of the East Asian monsoon and affect the seasonal distribution of Holocene rainfall variations. Within the Indian and North African monsoon domain, precipitation solely changes during the summer months, showing a decreasing Holocene precipitation trend. In the East Asian monsoon region, the precipitation signal is determined by an increasing precipitation trend during spring and a decreasing precipitation change during summer, partly balancing each other. A synthesis of reconstructions and the model results do not reveal an impact of the different seasonality on the timing of the Holocene rainfall optimum in the different sub-monsoon systems. This has led the species to be classified as globally Vulnerable. By the beginning of the last century, populations were partially migratory and moved south during the breeding season. Currently, the main breeding population inhabits the Ibera wetlands in the province of Corrientes, north-east Argentina, where it is resident all year round. There are two remaining small populations in the province of Formosa, north-east Argentina, and in southern Paraguay, which are separated from the main population by the Parana-Paraguay River and its continuous riverine forest habitat. The populations of Corrientes and Formosa are separated by 300 km and the grasslands between populations are non-continuous due to habitat transformation. We used mtDNA sequences and eight microsatellite loci to test if there were evidences of genetic isolation between Argentinean populations. We found no evidence of genetic structure between populations (Phi(ST) = 0.004, P = 0.32; Fst = 0.01, P = 0.06), which can be explained by either retained ancestral polymorphism or by dispersal between populations. We found no evidence for a recent demographic bottleneck in nuclear loci. Our results indicate that these populations could be managed as a single conservation unit on a regional scale. Using a yeast-two-hybrid (Y2H) screen with prototype FV (PFV) Gag protein as bait we identified human polo-like kinase 2 (hPLK2), a member of cell cycle regulatory kinases, as a new interactor of PFV capsids. Further Y2H studies confirmed interaction of PFV Gag with several PLKs of both human and rat origin. A consensus Ser-Thr/Ser-Pro (S-T/S-P) motif in Gag, which is conserved among primate FVs and phosphorylated in PFV virions, was essential for recognition by PLKs. In the case of rat PLK2, functional kinase and polo-box domains were required for interaction with PFV Gag. Fluorescently-tagged PFV Gag, through its chromatin tethering function, selectively relocalized ectopically expressed eGFP-tagged PLK proteins to mitotic chromosomes in a Gag STP motif-dependent manner, confirming a specific and dominant nature of the Gag-PLK interaction in mammalian cells. The functional relevance of the Gag-PLK interaction was examined in the context of replication-competent FVs and single-round PFV vectors. Although STP motif mutated viruses displayed wild type (wt) particle release, RNA packaging and intra-particle reverse transcription, their replication capacity was decreased 3-fold in single-cycle infections, and up to 20-fold in spreading infections over an extended time period. Strikingly similar defects were observed when cells infected with single-round wt Gag PFV vectors were treated with a pan PLK inhibitor. Analysis of entry kinetics of the mutant viruses indicated a post-fusion defect resulting in delayed and reduced integration, which was accompanied with an enhanced preference to integrate into heterochromatin. Blood was sampled at baseline (fasted), 1, 2, 3, 4, 5, 6, 8 and 12 h after supplementation. Plasma and chylomicron concentrations of TCT and a-TCP were measured at each time point. Plasma TAG were measured enzymatically, and plasma antioxidant capacity was assessed by the Trolox equivalent antioxidant capacity assay. In fasted dogs, levels of TCT were 0·07 ( SD 0·03) mmol/l. Following the administration of the TRF, total plasma TCT peaked at 2 h (7·16 ( SD 3·88) mmol/l; P, 0·01) and remained above baseline levels (0·67 ( SD 0·44) mmol/l; P, 0·01) at 12 h. The TCT response in chylomicrons paralleled the increase in TCT in plasma with a maximum peak (3·49 ( SD 2·06) mmol/l; P, 0·01) at 2 h post-dosage. a-TCP was the major vitamin E detected in plasma and unaffected by TRF supplementation. The Trolox equivalent values increased from 2 h (776 ( SD 51·2) mmol/l) to a maximum at 12 h (1130 ( SD 7·72) mmol/l; P,0·01). The results show that TCT are detected in postprandial plasma of dogs. The granitoid magmatic activity occurred in relation to the subduction of the Neo-Tethys oceanic crust beneath the Iranian crust during Tertiary times. The granitoids are mainly granodiorite, quartz monzodiorite, monzonite and quartz diorite. Chemically, the magmatic rocks are characterized by ASI < 1.04, AI < 0.87 and high contents of CaO (up to ∼ 14.5 wt \%), which are consistent with the I-type magmatic series. Low FeO t /(FeO t +MgO) values (< 0.75) as well as low Nb, Y and K 2 O contents of the investigated rocks resemble the calc-alkaline series. Low SiO 2 , K 2 O/Na 2 O and Al 2 O 3 accompanied by high CaO and FeO contents indicate melting of metabasites as an appropriate source for the intrusions. Negative Ti and Nb anomalies verify a metaluminous crustal origin for the protoliths of the investigated igneous rocks. These are comparable with compositions of the associated mafic migmatites, in the Takab metamorphic complex, which originated from the partial melting of amphibolites. Therefore, crustal melting and a collision-related origin for the Takab calc-alkaline intrusions are proposed here on the basis of mineralogy and geochemical characteristics. The P-T evolution during magmatic crystallization and subsolidus cooling stages is determined by the study of mineral chemistry of the granodiorite and the quartz diorite. Magmatic crystallization pressure and temperature for the quartz-diorite and the granodiorite are estimated to be P ∼ 7.8 ± 2.5 kbar, T ∼ 760 ± 75 ◦C and P ∼ 5 ± 1 kbar, T ∼ 700 ◦C, respectively. Pursuing previous work, we employed temporal diagnostics for syllable structure, consisting of static correspondences between any given phonological organisation and its presumed phonetic indices. We show that such correspondences offer only a partial understanding of the relation between syllabic organisation and continuous indices of that organisation. We analyse the failure of the diagnostics and put forth a new approach in which different phonological organisations prescribe different ways in which phonetic indices change as phonetic parameters are scaled. However, high-dose supplements or the use of multiple supplements may exceed the recommended daily allowance (RDA) of particular nutrients or even result in a daily intake above tolerable upper limits (UL). The present case report presents nutritional intake data and supplement use of a highly trained male swimmer competing at international level. Habitual energy and micronutrient intake were analysed by 3 d dietary reports. Supplement use and dosage were assessed, and total amount of nutrient supply was calculated. Micronutrient intake was evaluated based on RDA and UL as presented by the European Scientific Committee on Food, and maximum permitted levels in supplements (MPL) are given. The athlete's diet provided adequate micronutrient content well above RDA except for vitamin D. Simultaneous use of ten different supplements was reported, resulting in excess intake above tolerable UL for folate, vitamin E and Zn. Additionally, daily supplement dosage was considerably above MPL for nine micronutrients consumed as artificial products. Risks and possible side effects of exceeding UL by the athlete are discussed. Athletes with high energy intake may be at risk of exceeding UL of particular nutrients if multiple supplements are added. Therefore, dietary counselling of athletes should include assessment of habitual diet and nutritional supplement intake. Drought triggers diverse cellular processes including the inhibition of photosynthesis, the accumulation of cell-damaging reactive oxygen species and gene expression reprogramming, besides others. Transcription factors (TF) are central regulators of transcriptional reprogramming and expression of many TF genes is affected by drought, including members of the NAC family. Here, we identify the NAC factor JUNGBRUNNEN1 (JUB1) as a regulator of drought tolerance in tomato (Solanum lycopersicum). Expression of tomato JUB1 (SlJUB1) is enhanced by various abiotic stresses, including drought. Inhibiting SlJUB1 by virus-induced gene silencing drastically lowers drought tolerance concomitant with an increase in ion leakage, an elevation of hydrogen peroxide (H2O2) levels and a decrease in the expression of various drought-responsive genes. In contrast, overexpression of AtJUB1 from Arabidopsis thaliana increases drought tolerance in tomato, alongside with a higher relative leaf water content during drought and reduced H2O2 levels. AtJUB1 was previously shown to stimulate expression of DREB2A, a TF involved in drought responses, and of the DELLA genes GAI and RGL1. We show here that SlJUB1 similarly controls the expression of the tomato orthologs SlDREB1, SlDREB2 and SlDELLA. Furthermore, AtJUB1 directly binds to the promoters of SlDREB1, SlDREB2 and SlDELLA in tomato. To reduce uncertainties and inconsistencies in the estimation of the landslide distribution area, we propose an objective definition based on the shortest distance from the seismic wave emission line containing 95\% of the total landslide area. Without any empirical calibration the model explains 56\% of the variance in our dataset, and predicts 35 to 49 out of 83 cases within a factor of 2, depending on how we account for uncertainties on the seismic source depth. For most cases with comprehensive landslide inventories we show that our prediction compares well with the smallest region around the fault containing 95\% of the total landslide area. Aspects ignored by the model that could explain the residuals include local variations of the threshold of acceleration and processes modulating the surface ground shaking, such as the distribution of seismic energy release on the fault plane, the dynamic stress drop, and rupture directivity. The algorithms are based on a manually controlled inversion of optical data which allows for detailed sensitivity studies. The algorithms allow us to derive particle effective radius as well as volume and surface area concentration with comparably high confidence. The retrieval of the real and imaginary parts of the complex refractive index still is a challenge in view of the accuracy required for these parameters in climate change studies in which light absorption needs to be known with high accuracy. It is an extreme challenge to retrieve the real part with an accuracy better than 0.05 and the imaginary part with accuracy better than 0.005-0.1 or +/- 50 \%. Single-scattering albedo can be computed from the retrieved microphysical parameters and allows us to categorize aerosols into high-and low-absorbing aerosols. On the basis of a few exemplary simulations with synthetic optical data we discuss the current status of these manually operated algorithms, the potentially achievable accuracy of data products, and the goals for future work. One algorithm was used with the purpose of testing how well microphysical parameters can be derived if the real part of the complex refractive index is known to at least 0.05 or 0.1. The other algorithm was used to find out how well microphysical parameters can be derived if this constraint for the real part is not applied. The optical data used in our study cover a range of Angstrom exponents and extinction-to-backscatter (lidar) ratios that are found from lidar measurements of various aerosol types. We also tested aerosol scenarios that are considered highly unlikely, e.g. the lidar ratios fall outside the commonly accepted range of values measured with Raman lidar, even though the underlying microphysical particle properties are not uncommon. The goal of this part of the study is to test the robustness of the algorithms towards their ability to identify aerosol types that have not been measured so far, but cannot be ruled out based on our current knowledge of aerosol physics. We computed the optical data from monomodal logarithmic particle size distributions, i.e. we explicitly excluded the more complicated case of bimodal particle size distributions which is a topic of ongoing research work. Another constraint is that we only considered particles of spherical shape in our simulations. We considered particle radii as large as 7-10 mu m in our simulations where the Potsdam algorithm is limited to the lower value. We considered optical-data errors of 15\% in the simulation studies. Carrying out a dimensional analysis of the stress balance we obtain dimensionless numbers that characterize the flow. Requiring that these numbers remain the same under scaling we obtain conditions that relate the geometric scaling factors, the parameters for the ice softness, surface mass balance and basal friction as well as the ice-sheet intrinsic response time to each other. We demonstrate that these scaling laws are the same for both the (two-dimensional) flow-line case and the three-dimensional case. The theoretically predicted ice-sheet scaling behavior agrees with results from numerical simulations that we conduct in flow-line and three-dimensional conceptual setups. We further investigate analytically the implications of geometric scaling of ice sheets for their response time. With this study we provide a framework which, under several assumptions, allows for a fundamental comparison of the ice-dynamic behavior across different scales. Here, pollen (palynological) richness and diversity (Hill N (1)) are compared with vegetation richness and diversity from forest and savannah ecosystems in the New World and Old World tropics (Neotropics and Palaeotropics). Modern pollen data were obtained from artificial pollen traps deployed in 1-ha vegetation study plots from which vegetation inventories had been completed in Bolivia and Ghana. Pollen counts were obtained from 15 to 22 traps per plot, and aggregated pollen sums for each plot were > 2,500. The palynological richness/diversity values from the Neotropics were moist evergreen forest = 86/6.8, semi-deciduous dry forest = 111/21.9, wooded savannah = 138/31.5, and from the Palaeotropics wet evergreen forest = 144/28.3, semi-deciduous moist forest = 104/4.4, forest-savannah transition = 121/14.1; the corresponding vegetation richness/diversity was 100/36.7, 80/38.7 and 71/39.4 (Neotropics), and 101/54.8, 87/45.5 and 71/34.5 (Palaeotropics). No consistent relationship was found between palynological richness/diversity, and plot vegetation richness/diversity, due to the differential influence of other factors such as landscape diversity, pollination strategy, and pollen source area. Palynological richness exceeded vegetation richness, while pollen diversity was lower than vegetation diversity. For this purpose, we follow a Peak-over-Threshold approach employing a Poisson point process and the Generalised Pareto Distribution. By considering the effect of sea level rise as well as potential adaptation scenarios on the involved parameters, we are able to study the development of the annual damage. An application to the city of Copenhagen shows that a doubling of losses can be expected from a mean sea level increase of only 11 cm. In general, we find that for varying parameters the expected losses can be well approximated by one of three analytical expressions depending on the extreme value parameters. These findings reveal the complex interplay of the involved parameters and allow conclusions of fundamental relevance. For instance, we show that the damage typically increases faster than the sea level rise itself. This in turn can be of great importance for the assessment of sea level rise impacts on the global scale. Our results are accompanied by an assessment of uncertainty, which reflects the stochastic nature of extreme events. Method We included 314 patients with diabetes mellitus or mild renal impairment undergoing coronary angiography and collected blood and urine before and 24 hours after the CM application. Patients were followed for 90 days for the composite endpoint major adverse renal events (MARE: need for dialysis, doubling of serum creatinine after 90 days, unplanned emergency rehospitalization or death). Correction of urine VDBP concentrations for creatinine excretion confirmed its predictive value and was consistent with increased levels of urinary Kidney Injury Molecule1 (KIM-1) and baseline plasma creatinine in patients with above mentioned complications. The impact of urinary VDBP and KIM-1 on MARE was independent of known CIN risk factors such as anemia, preexisting renal failure, preexisting heart failure, and diabetes. The main challenge in this approach is to bridge the gap in scale between the widely scattered data used for calibration of the model and the high resolution in the target area required for the application. We present a multi-stage 3-D geomechanical-numerical approach which provides a state-of-the-art model of the stress field for a reservoir-scale area from widely scattered data records. Therefore, we first use a large-scale regional model which is calibrated by available stress data and provides the full 3-D stress tensor at discrete points in the entire model volume. The modelled stress state is used subsequently for the calibration of a smaller-scale model located within the large-scale model in an area without any observed stress data records. We exemplify this approach with two-stages for the area around Munich in the German Molasse Basin. As an example of application, we estimate the scalar values for slip tendency and fracture potential from the model results as measures for the criticality of fault reactivation in the reservoir-scale model. The modelling results show that variations due to uncertainties in the input data are mainly introduced by the uncertain material properties and missing S-Hmax magnitude estimates needed for a more reliable model calibration. This leads to the conclusion that at this stage the model's reliability depends only on the amount and quality of available stress information rather than on the modelling technique itself or on local details of the model geometry. The essential variable in the models is syllable structure. When mapped to discrete coordination topologies, syllabic organization imposes systematic patterns of variability on the temporal dynamics of speech articulation. We simulated these dynamics under different syllabic parses and evaluated simulations against experimental data from Arabic and English, two languages claimed to parse similar strings of segments into different syllabic structures. Model simulations replicated several key experimental results, including the fallibility of past phonetic heuristics for syllable structure, and exposed the range of conditions under which such heuristics remain valid. More importantly, the modelling approach consistently diagnosed syllable structure proving resilient to multiple sources of variability in experimental data including measurement variability, speaker variability, and contextual variability.
