In the present study, we have assessed whether oestradiol and progesterone directly influence the uterine expression of Raldh1 and Raldh2 in ovariectomised mice. We investigated the effect of gestagen (promegestone 0.3 mg kg(-1) bodyweight), oestrogen (oestradiol 3 mu g kg(-1) bodyweight) and their combination on the uterine expression of Raldh2. Expression was analysed using in situ hybridisation and quantified using real-time detection reverse transcription-polymerase chain reaction. The results show that the expression of Raldh2 is rapidly (within 1-4 h) induced in stromal cells by oestrogen, but not by gestagen, treatment, whereas combined oestrogen + gestagen treatment leads to a more prolonged (48 h) response. In contrast, oestrogen, but not progesterone, treatment downregulates (within 4 - 24 h) Raldh1 expression in the uterine glandular epithelium. We conclude that the uterine RA concentrations are regulated by oestrogens via an effect on the expression of the Raldh synthesising enzymes. In many tissues, collagen molecules assemble to form a hierarchical structure. In the smallest supramolecular unit, named fibril, each molecule is displaced in the axial direction with respect to its neighbors. This staggering creates a periodic gap and overlap regions, where the gap regions exhibit 20\% less density. These fibril-forming collagens play an essential role in the strength of connective tissues. Despite much effort, directed at understanding collagen function and regulation, the influence of the chemical environment on the local structural and mechanical properties remains poorly understood. Recent studies, aimed at elucidating the effect of osmotic pressure, showed that collagen contracts upon water removal. This observation highlights the importance of water for the stabilization and mechanics of the collagen molecule. Using collagen mimetic peptides (CMPs), which fold into triple helical structures reminiscent of natural collagen, the primary goal of this work was to investigate the effect of the osmotic pressure on specific collagen-mimetic sequences. CMPs were used as the model system as they provide sequence control, which is essential for discriminating local from global structural changes and for relating the observed effects to existing knowledge about the full-length collagen molecule. Of specific interest was the structure of individual collagen triple helices as well as their organization into self-assembled higher order structures. These key structural features were monitored with infrared spectroscopy (IR) and synchrotron X-ray scattering, while varying the osmotic pressure. For controlling the osmotic pressure, CMP powder samples were incubated in air of defined relative humidity, ranging from dry conditions to highly "humid". In addition, to obtain more biologically relevant conditions, the CMPs were measured in ultrapure water and in solutions containing small molecule osmolytes. Using the sequences (Pro-Pro-Gly)10, (Pro-Hyp-Gly)10 and (Hyp-Hyp-Gly)10, it was shown that CMPs with different degrees of proline hydroxylation (Hyp = hydroxyproline) exhibit a sequence-specific response to osmotic pressure. IR spectroscopy revealed that osmotic pressure changes affect the strength of the triple helix stabilizing, interchain hydrogen bond and that the extent of this change depends on the degree of hydroxylation. X-ray scattering experiments further showed that changes in osmotic pressure affect both the molecular length as well as the higher order organization of CMPs. Starting from a pseudo-hexagonal packing in the dry state, all three CMPs showed isotropic swelling when increasing the water content to approximately 1.2 water molecules per amino acid, again to different extents depending on the degree of hydroxylation. When increasing the water content further, this pseudo-hexagonal arrangement breaks down. In the fully hydrated state, each CMP is characterized by its own specific and more complex packing geometry. While these changes in the lateral packing arrangement suggest swelling upon hydration, an overall decrease of the molecular length (i.e. contraction) was observed in the axial direction. Also for this structural feature, a strong dependency on the specific amino acid sequence was found. Interestingly, the observed contraction is the opposite of what has been reported for natural collagen. As (Pro-Pro-Gly)n, (Pro-Hyp-Gly)n and (Hyp-Hyp-Gly)n repeat units are found in collagen with a relatively high abundance, this suggests that other collagen sequence fragments need to respond to hydration in the opposite way to obtain a net elongation of the full-length collagen molecule. To test this hypothesis, sequences predicted to be sensitive to osmotic pressure were considered. One such sequence, consisting of two repeat units (Ala-Arg-Gly-Ser-Asp-Gly), was inserted as a guest into a (Pro-Pro-Gly) host. When compared to the canonical CMP sequences investigated earlier, the lateral helix packing follows a similar trend with increasing hydration; however, the host-guest CMP axially elongates with increasing water content. This behavior is more similar to what has been found for natural collagen and suggests that different sequences do determine the molecular length of collagen sequences differently. Interestingly, the canonical sequences are more abundant in the overlap region while the guest sequence is found in the gap region. This allows to speculate that sequences in the gap and overlap regions possess a specifically fine-tuned local response to osmotic pressure changes. Clearly, more experiments with additional sequences are needed to confirm this. In conclusion, the results obtained in this work indicate a highly sequence specific interaction between collagen and water. Osmotic pressure-induced conformational changes mostly originate from local geometries and bonding patterns and affect both the structure of individual triple helices as well as higher order assemblies. One key remaining question is how these conformational changes affect the local mechanical properties of the collagen molecule. As a first step, the stiffness (persistence length) of full-length collagen was determined using atomic force microscopy. Here, the measured absorption coefficients reflect temperature as well as fat content of milk and the reduced scattering coefficients probe physical changes of the light scattering fat droplets and casein micelles. Thermal processing reveals breakpoints within the temperature trend of the reduced scattering coefficient of fat containing milk. Found at 16 degrees C and 24 degrees C while cooling and heating, respectively, they are associated to the phase transitions of milk fat. Continuous isothermal measurement of the optical coefficients showed that the crystallization process requires several hours. This group has set itself the goal of investigating geological processes that take place on different temporal and spatial scales and have shaped the southern central Andes. This study focuses on claystones and carbonates of the Yacoraite Fm. that were deposited between Maastricht and Dan in the Cretaceous Salta Rift Basin. The former rift basin is located in northwest Argentina and is divided into the sub-basins Tres Cruces, Met{\´a}n-Aleman{\´i}a and Lomas de Olmedo. The overall motivation for this study was to gain new knowledge about the evolution of marine and lacustrine conditions during the Yacoraite Fm. Deposit in the Tres Cruces and Met{\´a}n-Aleman{\´i}a sub-basins. Other important aspects that were examined within the scope of this dissertation are the conversion of organic matter from Yacoraite Fm. into oil and its genetic relationship to selected oils produced and natural oil spills. The results of my study show that the Yacoraite Fm. began to be deposited under marine conditions and that a lacustrine environment developed by the end of the deposition in the Tres Cruces and Met{\´a}n-Aleman{\´i}a Basins. In general, the kerogen of Yacoraite Fm. consists mainly of the kerogen types II, III and II / III mixtures. Kerogen type III is mainly found in samples from the Yacoraite Fm., whose TOC values are low. Due to the adsorption of hydrocarbons on the mineral surfaces (mineral matrix effect), the content of type III kerogen with Rock-Eval pyrolysis in these samples could be overestimated. Investigations using organic petrography show that the organic particles of Yacoraite Fm. mainly consist of alginites and some vitrinite-like particles. The pyrolysis GC of the rock samples showed that the Yacoraite Fm. generates low-sulfur oils with a predominantly low-wax, paraffinic-naphthenic-aromatic composition and paraffinic wax-rich oils. Small proportions of paraffinic, low-wax oils and a gas condensate-generating facies are also predicted. Here, too, mineral matrix effects were taken into account, which can lead to a quantitative overestimation of the gas-forming character. The results of an additional 1D tank modeling carried out show that the beginning (10\% TR) of the oil genesis took place between ≈10 Ma and ≈4 Ma. Most of the oil (from ≈50\% to 65\%) was generated prior to the development of structural traps formed during the Plio-Pleistocene Diaguita deformation phase. Only ≈10\% of the total oil generated was formed and potentially trapped after the formation of structural traps. Important factors in the risk assessment of this petroleum system, which can determine the small amounts of generated and migrated oil, are the generally low TOC contents and the variable thickness of the Yacoraite Fm. The presence of a complexing amine group in addition to the charged betaine moieties in the polymer permits stabilization of more than stoichiometric amounts of the metal species in the blends. XRD measurements suggest that the homogeneity in the solid state can be kept up to about 1.5 mol of each metal that is incorporated ( anionic as well as cationic) per mol of repeat units of the copolymer. The blends were calcined under air at 600 degrees C to produce the simple as well as mixed nickel, cobalt and manganese molybdates. Characterization of the final phases by XRD and Raman spectroscopy indicates that the alpha- as well as the beta-molybdate phases can be prepared, and that the mixed structures are solid solutions of the simple NiMoO4, MnMoO4 and CoMoO4. In order to quantify the geological variations at a creeping hillslope in the Austrian Alps, we have collected six 2D refraction seismic profiles. We propose using a layer-based inversion strategy to reconstruct P-wave velocity models from first arrival times. Considering the geological complexity at such sites, the selected inversion approach eases the interpretability of geological structures given intrinsic optimization for only a discrete, user-defined, number of layers. As the applied layer-based inversion approach fits our travel time data equally well as traditional smooth inversion approaches, it represents a feasible mean to summarize the structural complexity often present at such sites. Analysis of the inversion results illustrates that bedrock topography clearly deviates from a previously assumed planar surface and exhibits distinct variations across the slope extension. To illustrate our workflow, we present field data collected to characterize a near-surface sedimentary environment Our field data base includes crosshole ground penetrating radar (GPR), seismic P-, and S-wave data sets collected between 25 m deep boreholes penetrating sand- and gravel dominated sediments. Furthermore, different typical borehole and direct-push logs are available. We perform a global joint inversion of traveltimes extracted from the crosshole geophysical data using a recently proposed approach based on particle swarm optimization. Our inversion strategy allows for generating consistent models of GPR, P-wave, and S-wave velocities including an appraisal of uncertainties. We analyze the observed complex relationships between geophysical velocities and target parameter logs using the alternating conditional expectation (ACE) algorithm. This nonparametric statistical tool allows us to perform multivariate regression analysis without assuming a specific functional relation between the variables. We are able to explain selected target parameters such as characteristic grain size values or natural gamma activity by our inverted geophysical data and to extrapolate these parameters to the inter-borehole plane covered by our crosshole experiments. We test and evaluate our approach using synthetic seismic travel times and field data collected across a creeping hillslope in the Austrian Alps. Our synthetic study mimics a layered near-surface environment, including a sharp velocity increase with depth and complex refractor topography. Analysing the generated ensemble of acceptable solutions using different statistical measures demonstrates that our inversion strategy is able to reconstruct the input velocity model, including reasonable, quantitative estimates of uncertainty. Our field data set is inverted, employing the same strategy, and we further compare our results with the velocity model obtained by a standard local optimization approach and the information from a nearby borehole. This comparison shows that both inversion strategies result in geologically reasonable models (in agreement with the borehole information). However, analysing the model variability of the ensemble generated using our global approach indicates that the result of the local optimization approach is part of this model ensemble. It has been shown that microgels effectively absorb surfactant ions. At low surfactant concentration this absorption is realized due to an ion exchange between microgel counterions and surfactant ions. The ion exchange is significantly affected by the amount of the microgel counterions initially trapped within the microgel particles which depends on the size of the microgel, its ionization degree, cross-linking density as well as polymer concentration in the solution. Increase of the surfactant concentration causes contraction of the microgels, which can be realized as either a continuous shrinking or a jump-like collapse transition depending on the system parameters. In the collapsed state additional absorption of surfactants by microgels takes place due to an energy gain from micellization or hydrophobic interactions. This leads to microgel precipitation and successive microgel overcharging at an excess of the surfactant in the solution. Five oxidative stress models were investigated including incubations of three human cell lines as well as the in vivo model Caenorhabditis elegans with tert-butyl hydroperoxide (t-BOOH) and analysis of murine kidney tissue after renal ischemia reperfusion injury (IRI). A comprehensive set of IsoP and epoxy-PUFA derived from biologically relevant PUFA (ARA, EPA and DHA) was simultaneously quantified by LC-ESI(-)-MS/MS. Following renal IRI only a moderate increase in the kidney levels of IsoP and no relevant change in the trans/cis-epoxy-PUFA ratio was observed. In all investigated cell lines (HCT-116, HepG2 and Caki-2) as well as C. elegans a dose dependent increase of both, IsoP and the trans/cis-epoxy-PUFA ratio in response to the applied t-BOOH was observed. This decreases image availability and thus constrains the application of automated algorithms for time series disturbance detection for large-scale abrupt permafrost disturbances at high temporal resolution. Since models and observations suggest that abrupt permafrost disturbances will intensify, we require disturbance products at continental-scale, which allow for meaningful integration into Earth system models. The main aim of this dissertation therefore, is to enhance our knowledge on the spatial extent and temporal dynamics of abrupt permafrost disturbances in a large-scale assessment. To address this, three research objectives were posed: 1. Assess the comparability and compatibility of Landsat-8 and Sentinel-2 data for a combined use in multi-spectral analysis in northern high latitudes. 2. Adapt an image mosaicking method for Landsat and Sentinel-2 data to create combined mosaics of high quality as input for high temporal disturbance assessments in northern high latitudes. 3. Automatically map retrogressive thaw slumps on the landscape-scale and assess their high temporal thaw dynamics. We assessed the comparability of Landsat-8 and Sentinel-2 imagery by spectral comparison of corresponding bands. Based on overlapping same-day acquisitions of Landsat-8 and Sentinel-2 we derived spectral bandpass adjustment coefficients for North Siberia to adjust Sentinel-2 reflectance values to resemble Landsat-8 and harmonise the two data sets. Furthermore, we adapted a workflow to combine Landsat and Sentinel-2 images to create homogeneous and gap-free annual mosaics. We determined the number of images and cloud-free pixels, the spatial coverage and the quality of the mosaic with spectral comparisons to demonstrate the relevance of the Landsat+Sentinel-2 mosaics. Lastly, we adapted the automatic disturbance detection algorithm LandTrendr for large-scale RTS identification and mapping at high temporal resolution. For this, we modified the temporal segmentation algorithm for annual gradual and abrupt disturbance detection to incorporate the annual Landsat+Sentinel-2 mosaics. We further parametrised the temporal segmentation and spectral filtering for optimised RTS detection, conducted further spatial masking and filtering, and implemented a binary object classification algorithm with machine-learning to derive RTS from the LandTrendr disturbance output. We applied the algorithm to North Siberia, covering an area of 8.1 x 106 km2. The spectral band comparison between same-day Landsat-8 and Sentinel-2 acquisitions already showed an overall good fit between both satellite products. However, applying the acquired spectral bandpass coefficients for adjustment of Sentinel-2 reflectance values, resulted in a near-perfect alignment between the same-day images. It can therefore be concluded that the spectral band adjustment succeeds in adjusting Sentinel-2 spectral values to those of Landsat-8 in North Siberia. The number of available cloud-free images increased steadily between 1999 and 2019, especially intensified after 2016 with the addition of Sentinel-2 images. This signifies a highly improved input database for the mosaicking workflow. In a comparison of annual mosaics, the Landsat+Sentinel-2 mosaics always fully covered the study areas, while Landsat-only mosaics contained data-gaps for the same years. The spectral comparison of input images and Landsat+Sentinel-2 mosaic showed a high correlation between the input images and the mosaic bands, testifying mosaicking results of high quality. Our results show that especially the mosaic coverage for northern, coastal areas was substantially improved with the Landsat+Sentinel-2 mosaics. By combining data from both Landsat and Sentinel-2 sensors we reliably created input mosaics at high spatial resolution for comprehensive time series analyses. This research presents the first automatically derived assessment of RTS distribution and temporal dynamics at continental-scale. In total, we identified 50,895 RTS, primarily located in ice-rich permafrost regions, as well as a steady increase in RTS-affected areas between 2001 and 2019 across North Siberia. From 2016 onward the RTS area increased more abruptly, indicating heightened thaw slump dynamics in this period. Overall, the RTS-affected area increased by 331 \% within the observation period. Contrary to this, five focus sites show spatiotemporal variability in their annual RTS dynamics, alternating between periods of increased and decreased RTS development. This suggests a close relationship to varying thaw drivers. The majority of identified RTS was active from 2000 onward and only a small proportion initiated during the assessment period. This highlights that the increase in RTS-affected area was mainly caused by enlarging existing RTS and not by newly initiated RTS. Overall, this research showed the advantages of combining Landsat and Sentinel-2 data in northern high latitudes and the improvements in spatial and temporal coverage of combined annual mosaics. The mosaics build the database for automated disturbance detection to reliably map RTS and other abrupt permafrost disturbances at continental-scale. The assessment at high temporal resolution further testifies the increasing impact of abrupt permafrost disturbances and likewise emphasises the spatio-temporal variability of thaw dynamics across landscapes. In this study we apply the disturbance detection algorithm LandTrendr for automated large-scale RTS mapping and high temporal thaw dynamic assessment to North Siberia (8.1 x 106km2). We adapted and parametrised the temporal segmentation algorithm for abrupt disturbance detection to incorporate Landsat+Sentinel-2 mosaics, conducted spectral filtering, spatial masking and filtering, and a binary machine-learning object classification of the disturbance output to separate between RTS and false positives (F1 score: 0.609). Ground truth data for calibration and validation of the workflow was collected from 9 known RTS cluster sites using very highresolution RapidEye and PlanetScope imagery. Our study presents the first automated detection and assessment of RTS and their temporal dynamics at largescale for 2001-2019. We identified 50,895 RTS and a steady increase in RTS-affected area from 2001 to 2019 across North Siberia, with a more abrupt increase from 2016 onward. Overall the RTS-affected area increased by 331\% compared to 2000 (2000: 20,158 ha, 2001-2019: 66,699 ha). Contrary to this, 5 focus sites show spatiotemporal variability in their annual RTS dynamics, with alternating periods of increased and decreased RTS development, indicating a close relationship to thaw drivers. We introduce a platform-independent, interactive program enabling us to quantify, elicit, and transfer expert knowledge into a set of subjective probabilities by applying experimental design theory, following the approach of Curtis and Wood (2004). Instead of determining the set of probabilities for all models in a single step, the computer-driven elicitation process is performed as a sequence of evaluations of relative weights for small subsets of models. From these, the probabilities for the whole model set are determined as a solution of an optimization problem. The result of this process is a set of logically consistent probabilities together with a measure of confidence determined from the amount of conflicting information which is provided by the expert during the relative weighting process. We experiment with different scenarios simulating likely expert behaviors in the context of knowledge elicitation and show the impact this has on the results. The obtained results found using much improved numerical methods show a magnetic field evolution with x-line formation and plasma acceleration.
