The different methodologies used for assessing plant-plant interactions under changing environmental conditions may affect the outcome but they are not equally represented in the literature. Mechanistic experimental manipulations are rare compared with correlative approaches that infer future patterns from current observations along spatial climatic gradients. 3. Here, we utilize a unique climatic gradient in combination with a large-scale, long-term experiment to test whether predictions about plant-plant interactions yield similar results when using experimental manipulations, spatial gradients or temporal variation. We assessed shrub-annual interactions in three different sites along a natural rainfall gradient (spatial) during 9 years of varying rainfall (temporal) and 8 years of dry and wet manipulations of ambient rainfall (experimental) that closely mimicked regional climate scenarios. 4. The results were fundamentally different among all three approaches. Experimental water manipulations hardly altered shrub effects on annual plant communities for the assessed fitness parameters biomass and survival. Along the spatial gradient, shrub effects shifted from clearly negative to mildly facilitative towards drier sites, whereas temporal variation showed the opposite trend: more negative shrub effects in drier years. 5. Based on our experimental approach, we conclude that shrub-annual interaction will remain similar under climate change. In contrast, the commonly applied space-for-time approach based on spatial gradients would have suggested increasing facilitative effects with climate change. We discuss potential mechanisms governing the differences among the three approaches. 6. Our study highlights the critical importance of long-term experimental manipulations for evaluating climate change impacts. In a nine generation study in Israel, we manipulated rainfall (Drought -30\%; Irrigation +30\%; Control natural rainfall) at two sites which differ chiefly in rainfall quantity and variability. We analysed community responses to the manipulations by grouping species based on their climatic rainfall niche. These responses were compared to analyses based on single species, total densities, and commonly used taxonomic groupings. Climate Niche Groups yielded clear and consistent results: within communities, those species distributed in drier regions performed relatively better in the drought treatment, and those from wetter climates performed better when irrigated. In contrast, analyses based on other principles revealed little insight into community dynamics. Notably, most relationships were weaker at the drier, more variable site, suggesting that enhanced adaptation to variability may buffer climate change impacts. We provide robust experimental evidence that using climatic niches commonly applied in BEMs is a valid approach for eliciting community changes in response to climate change. However, we also argue that additional empirical information needs to be gathered using experiments in situ to correctly assess community vulnerability. We calculated Kr IV-VII oscillator strengths to consider radiative and collisional bound-bound transitions in detail in our NLTE stellar-atmosphere models for the analysis of Kr lines that are exhibited in high-resolution and high S/N ultraviolet (UV) observations of the hot white dwarf RE 0503-289. Results. We reanalyzed the effective temperature and surface gravity and determined T-eff = 70 000 +/- 2000 K and log (g/cm s(-2)) = 7.5 +/- 0.1. We newly identified ten Kr V lines and one Kr vi line in the spectrum of RE 0503-289. We measured a Kr abundance of 3.3 +/- 0.3 (logarithmic mass fraction). We discovered that the interstellar absorption toward RE 0503-289 has a multi-velocity structure within a radial-velocity interval of -40 km s(-1) < upsilon(rad) < +18 km s(-1). Conclusions. Reliable measurements and calculations of atomic data are a prerequisite for state-of-the-art NLTE stellar-atmosphere modeling. Also, they scale their aggressive behaviors according to the relatedness to other females: they are more aggressive to non-related clones. Our results demonstrate that even in species with very small genetic differences between individuals, kin recognition can be adaptive. As a general tendency, the levels of gene expression were lowest in gill and highest in ovarian tissues, while expression levels in the brain were intermediate in most cases. Overall, mean heterozygosity scored for all loci and populations was lower than that reported for the few other diaptomid species for which microsatellite loci have been to date described; this is possibly due to the intrinsically fragmented and isolated peculiar habitat inhabited by the species. Furthermore, the presence of indels within the flanking regions of selected loci was scored. Three hundred eighty-two participants took part in one of the 17 speed-dating sessions. After each speed-dating interaction, participants indicated how interesting they found the respective person as a long-term and short-term partner. Using social relations analyses, we computed perceiver effects (being more or less choosy) and target effects (being rated as more or less interesting) of long-term and short-term partner ratings. Self-enhancement was operationalized as the discrepancy between self-rated attractiveness and four components of actual attractiveness (observer-rated facial and vocal attractiveness, height and body mass index). Results indicated that self-enhancers were less choosy with respect to their interest for short-term partners, which was especially true for men, but more choosy with respect to long-term partners. With regard to popularity as a mate, potential partners indicated that they found self-enhancers more interesting as short-term partners but not as long-term partners. Aus diesem Grund ist die einfache Darstellung dieser Struktureinheit von großen Interesse in der organischen Chemie. Das erste Ziel der vorliegenden Arbeit war daher die Weiterentwicklung der Ringschlussmetathese-/ baseninduzierten Ring{\"o}ffnungs-/ Veresterungssequenz (RBRV-Sequenz) zur Synthese von (2Z,4E)-Diencarbons{\"a}ureethylestern ausgehend von Butenoaten. Dazu wurde zun{\"a}chst die RBRV-Sequenz optimiert. Diese aus drei Schritten bestehende Sequenz konnte in einem Eintopf-Verfahren angewendet werden. Die Ringschlussmetathese gelang mit einer Katalysatorbeladung von 1 mol\% des GRUBBS-Katalysators der zweiten Generation in Dichlormethan. F{\"u}r die baseninduzierte Ring{\"o}ffnung des β,γ-unges{\"a}ttigten δ Valerolactons wurde NaHMDS verwendet. Die Alkylierung der Carboxylatspezies gelang mit dem MEERWEIN-Reagenz. Die Anwendbarkeit der Sequenz wurde f{\"u}r verschiedene Substrate demonstriert. Die Erweiterung der Methode auf α-substituierte Butenoate unterlag starken Einschr{\"a}nkungen. So konnte der Zugang f{\"u}r α Hydroxyderivate realisiert werden. Bei der Anwendung der RBRV-Sequenz auf die α-substituierten Butenoate wurde festgestellt, dass diese sich nur in moderaten Ausbeuten umsetzen ließen und zudem nicht selektiv zu den (2E,4E)-konfigurierten α-substituierten-Dienestern reagierten. Der Einsatz von Eninen unter den Standardbedingungen der RBRV-Sequenz gelang nicht. Erst nach Modifizierung der Sequenz (h{\"o}here Katalysatorbeladung, Wechsel des L{\"o}sungsmittels) konnten die [3]Dendralen-Produkte in geringen Ausbeuten erhalten werden. Im zweiten Teil der Arbeit wurde der Einsatz von (2Z,4E)-Diencarbons{\"a}ureethylestern in der Totalsynthese von Naturstoffen untersucht. Dazu wurden zun{\"a}chst die Transformationsm{\"o}glichkeiten der Ester gepr{\"u}ft. Es konnte gezeigt werden, dass sich (2Z,4E)-Diencarbons{\"a}ureethylester insbesondere zur Synthese von (2Z,4E)-Aldehyden sowie zum Aufbau der (3Z,5E)-Dien-1-in-Struktur eignen. Anhand dieser Ergebnisse wurde im Anschluss die RBRV-Sequenz in der Totalsynthese eingesetzt. Dazu wurde zun{\"a}chst der (2Z,4E)-Dienester Microsphaerodiolin in seiner ersten Totalsynthese auf drei verschiedene Routen hergestellt. Im Anschluss wurden sechs verschiedene Polyacetylene mit einer (3Z,5E)-Dien-1-in-Einheit hergestellt. Schl{\"u}sselschritte in ihrer Synthese waren immer die RBRV-Sequenz zum Aufbau der Z,E-Dien-Einheit, die Transformation des Esters in ein terminales Alkin sowie die CADIOT-CHODKIEWICZ-Kupplung zum Aufbau unsymmetrischer Polyine. Alle sechs Polyacetylene wurden zum ersten Mal in einer Totalsynthese synthetisiert. Drei Polyacetylene wurden ausgehend von (S)-Butantriol enantiomerenrein dargestellt. Die initiale Detektion der zahlreich vorkommenden Bitterstoffe erfolgt bei der Maus durch 35 Bitterrezeptoren (Tas2rs), die sich im Zungengewebe befinden. Die Geschmacksinformation wird anschließend von der Zunge {\"u}ber das periphere (PNS) ins zentrale Nervensystem (ZNS) geleitet, wo deren Verarbeitung stattfindet. Die Verarbeitung der Geschmacksinformation konnte bislang nicht g{\"a}nzlich aufgekl{\"a}rt werden. Neue Studien deuten auf eine Expression von Tas2rs auch im PNS und ZNS entlang der Geschmacksbahn hin. {\"U}ber Vorkommen und Aufgaben dieser Rezeptoren bzw. Rezeptorzellen im Nervensystem ist bislang wenig bekannt. Im Rahmen dieser Arbeit wurde die Tas2r-Expression in verschiedenen Mausmodellen untersucht, Tas2r-exprimierende Zellen identifiziert und deren Funktionen bei der {\"U}bertragung der Geschmacksinformationen analysiert. Im Zuge der Expressionsanalysen mittels qRT-PCR konnte die Expression von 25 der 35 bekannten Bittergeschmacksrezeptoren im zentralen Nervensystem der Maus nachgewiesen werden. Die Expressionsmuster im PNS sowie im ZNS lassen dar{\"u}ber hinaus Vermutungen zu Funktionen in verschiedenen Bereichen des Nervensystems zu. Basierend auf den Ergebnissen der Expressionsanalysen war es m{\"o}glich, stark exprimierte Tas2rs mittels In-situ-Hybridisierung in verschiedenen Zelltypen zu visualisieren. Des Weiteren konnten immunhistochemische F{\"a}rbungen unter Verwendung eines genetisch modifizierten Mausmodells die Ergebnisse der Expressionsanalysen best{\"a}tigen. Sie zeigten eine Expression von Tas2rs, am Beispiel des Tas2r131-Rezeptors, in cholinergen, dopaminergen, GABAergen, noradrenergen und glycinerg-angesteuerten Projektionsneuronen sowie in Interneuronen. Die Ergebnisse der vorliegenden Arbeit zeigen daher erstmals das Vorkommen von Tas2rs in verschiedenen neuronalen Zelltypen in weiten Teilen des ZNS. Dies l{\"a}sst den Schluss zu, dass Tas2r-exprimierende Zellen potentiell multiple Funktionen innehaben. Anhand von Verhaltensexperimenten in genetisch modifizierten M{\"a}usen wurde die m{\"o}gliche Funktion von Tas2r131-exprimierenden Neuronen (Tas2r131-Neurone) bei der Geschmackswahrnehmung untersucht. Die Ergebnisse weisen auf eine Beteiligung von Tas2r131-Neuronen an der Signalweiterleitung bzw. -verarbeitung der Geschmacksinformation f{\"u}r eine Auswahl von Bittersubstanzen hin. Die Analysen zeigen dar{\"u}ber hinaus, dass Tas2r131-Neuronen nicht an der Geschmackswahrnehmung anderer Bitterstoffe sowie Geschmacksstimuli anderer Qualit{\"a}ten (s{\"u}ß, umami, sauer, salzig), beteiligt sind. It is composed of different types of protein filaments that form complex, dynamically rearranging, and interconnected networks. The cytoskeleton serves a multitude of cellular functions which further depend on the cell context. In animal cells, the cytoskeleton prominently shapes the cell's mechanical properties and movement. In plant cells, in contrast, the presence of a rigid cell wall as well as their larger sizes highlight the role of the cytoskeleton in long-distance intracellular transport. As it provides the basis for cell growth and biomass production, cytoskeletal transport in plant cells is of direct environmental and economical relevance. However, while knowledge about the molecular details of the cytoskeletal transport is growing rapidly, the organizational principles that shape these processes on a whole-cell level remain elusive. This thesis is devoted to the following question: How does the complex architecture of the plant cytoskeleton relate to its transport functionality? The answer requires a systems level perspective of plant cytoskeletal structure and transport. To this end, I combined state-of-the-art confocal microscopy, quantitative digital image analysis, and mathematically powerful, intuitively accessible graph-theoretical approaches. This thesis summarizes five of my publications that shed light on the plant cytoskeleton as a transportation network: (1) I developed network-based frameworks for accurate, automated quantification of cytoskeletal structures, applicable in, e.g., genetic or chemical screens; (2) I showed that the actin cytoskeleton displays properties of efficient transport networks, hinting at its biological design principles; (3) Using multi-objective optimization, I demonstrated that different plant cell types sustain cytoskeletal networks with cell-type specific and near-optimal organization; (4) By investigating actual transport of organelles through the cell, I showed that properties of the actin cytoskeleton are predictive of organelle flow and provided quantitative evidence for a coordination of transport at a cellular level; (5) I devised a robust, optimization-based method to identify individual cytoskeletal filaments from a given network representation, allowing the investigation of single filament properties in the network context. The developed methods were made publicly available as open-source software tools. Altogether, my findings and proposed frameworks provide quantitative, system-level insights into intracellular transport in living cells. Despite my focus on the plant cytoskeleton, the established combination of experimental and theoretical approaches is readily applicable to different organisms. Die vorliegende Arbeit untersucht die Zug{\"a}nglichkeit virtueller Klassenzimmer f{\"u}r blinde Nutzende, um eine m{\"o}glichst gleichberechtigte Teilhabe an synchronen, kollaborativen Lernszenarien zu erm{\"o}glichen. Im Rahmen einer Produktanalyse werden dazu virtuelle Klassenzimmer auf ihre Zug{\"a}nglichkeit und bestehende Barrieren untersucht und Richtlinien f{\"u}r die zug{\"a}ngliche Gestaltung von virtuellen Klassenzimmern definiert. Anschließend wird ein alternatives Benutzungskonzept zur Darstellung und Bedienung virtueller Klassenzimmer auf einem zweidimensionalen taktilen Braille-Display entwickelt, um eine m{\"o}glichst gleichberechtigte Teilhabe blinder Lernender an synchronen Lehrveranstaltungen zu erm{\"o}glichen. Nach einer ersten Evaluation mit blinden Probanden erfolgt die prototypische Umsetzung des Benutzungskonzepts f{\"u}r ein Open-Source-Klassenzimmer.
