Hier will die Arbeit Einblicke geben, wie die Geographielehrkr{\"a}fte diese Transformation mitgetragen und umgesetzt haben. Welche Wesensz{\"u}ge aus der Sozialisierung in der DDR haben sich bei der Gestaltung des Unterrichtes und dessen Ausrichtung auf die neuen Erziehungsziele erhalten? Hierzu wurden Geographielehrkr{\"a}fte befragt, die sowohl in der DDR als auch im geeinten Deutschland unterrichtet haben. Die Fragen bezogen sich in erster Linie auf die Art und Weise des Unterrichtens vor, w{\"a}hrend und nach der Wende und der daraus entstandenen Systemtransformation. Die Befragungen kommen zu dem Ergebnis, dass sich der Geographieunterricht in der DDR thematisch von dem in der BRD nicht sonderlich unterschied. Von daher bedurfte es keiner umfangreichen inhaltlichen Ver{\"a}nderung des Geographieunterrichts. Schon zu DDR-Zeiten wurden durch die Lehrkr{\"a}fte offenbar eigenm{\"a}chtig ideologiefreie physisch-geographische Themen oft ausgedehnt, um die Ideologie des Faches zu reduzieren. So fiel den meisten eine Anpassung ihres Unterrichts an das westdeutsche System relativ leicht. Die humanistisch gepr{\"a}gte Werteerziehung des DDR-Bildungssystems wurde unter Ausklammerung des sozialistischen Aspektes ebenso fortgef{\"u}hrt, da es auch hier viele Parallelen zum westdeutschen System gegeben hat. Deutlich wird eine Charakterisierung des Faches als Naturwissenschaft von Seiten der ostdeutschen Lehrkr{\"a}fte, obwohl das Fach an den Schulen den Gesellschaftswissenschaften zugeordnet wird und auch in der DDR eine starke wirtschaftsgeographische Ausrichtung hatte. Auf Druck der Europ{\"a}ischen Union, welche bereits Ende der 90er Jahre aus sicherheitstechnischen Gr{\"u}nden die Schließung des Atomkraftwerkes forderte, beschloss die Regierung Litauens, das Atomkraftwerk stillzulegen. Am 1. Januar 2005 wird der Betrieb des ersten Reaktors eingestellt. Die endg{\"u}ltige Abschaltung ist bis zum Januar 2009 vorgesehen. Auf diese Weise wird der Stadt Visaginas, und damit verbunden vielen ihrer Einwohner, die wirtschaftliche Existenzgrundlage entzogen. Bedingt durch den Wegfall ihres Arbeitsplatzes wird ein Großteil der Einwohner gezwungen sein, sich nach Besch{\"a}ftigungsalternativen umzuschauen. Da jedoch auch im Umland der Stadt keine ausreichenden M{\"o}glichkeiten bestehen, die freigesetzten Arbeitskr{\"a}fte aufzufangen, wird f{\"u}r viele Betroffene die einzige Alternative darin liegen, Visaginas auf der Suche nach einem neuen Arbeitsplatz zu verlassen. Despite several modular assembly methods developed in recent years, the plant biotechnology community has not widely adopted them yet, probably due to the lack of appropriate vectors and software tools. Here we present Plant X-tender, an extension of the highly efficient, scarfree and sequence-independent multigene assembly strategy AssemblX,based on overlapdepended cloning methods and rare-cutting restriction enzymes. Plant X-tender consists of a set of plant expression vectors and the protocols for most efficient cloning into the novel vector set needed for plant expression and thus introduces advantages of AssemblX into plant synthetic biology. The novel vector set covers different backbones and selection markers to allow full design flexibility. We have included ccdB counterselection, thereby allowing the transfer of multigene constructs into the novel vector set in a straightforward and highly efficient way. Vectors are available as empty backbones and are fully flexible regarding the orientation of expression cassettes and addition of linkers between them, if required. We optimised the assembly and subcloning protocol by testing different scar-less assembly approaches: the noncommercial SLiCE and TAR methods and the commercial Gibson assembly and NEBuilder HiFi DNA assembly kits. Plant X-tender was applicable even in combination with low efficient homemade chemically competent or electrocompetent Escherichia coli. We have further validated the developed procedure for plant protein expression by cloning two cassettes into the newly developed vectors and subsequently transferred them to Nicotiana benthamiana in a transient expression setup. Thereby we show that multigene constructs can be delivered into plant cells in a streamlined and highly efficient way. The connection between SFON and arithmetic was fully mediated by magnitude estimation and calculation skills at pre-school. Keeping such models in sync is a challenge within an ever changing business environment: once a process is changed, all its models have to be updated. Due to a large number of models and their complex relations, model maintenance becomes error-prone and expensive. Against this background, business process model abstraction emerged as an operation reducing the number of stored process models and facilitating model management. Business process model abstraction is an operation preserving essential process properties and leaving out insignificant details in order to retain information relevant for a particular purpose. Process model abstraction has been addressed by several researchers. The focus of their studies has been on particular use cases and model transformations supporting these use cases. This thesis systematically approaches the problem of business process model abstraction shaping the outcome into a framework. We investigate the current industry demand in abstraction summarizing it in a catalog of business process model abstraction use cases. The thesis focuses on one prominent use case where the user demands a model with coarse-grained activities and overall process ordering constraints. We develop model transformations that support this use case starting with the transformations based on process model structure analysis. Further, abstraction methods considering the semantics of process model elements are investigated. First, we suggest how semantically related activities can be discovered in process models-a barely researched challenge. The thesis validates the designed abstraction methods against sets of industrial process models and discusses the method implementation aspects. Second, we develop a novel model transformation, which combined with the related activity discovery allows flexible non-hierarchical abstraction. In plants, root hair growth requires polar nuclear migration into the outgrowing hair. However, knowledge about the dynamics and the regulatory mechanisms underlying nuclear movements in root epidermal cells remains limited. Here, we show that both auxin and Rho-of-Plant (ROP) signaling modulate polar nuclear position at the inner epidermal plasma membrane domain oriented to the cortical cells during cell elongation as well as subsequent polar nuclear movement to the outer domain into the emerging hair bulge in Arabidopsis (Arabidopsis thaliana). Auxin signaling via the nuclear AUXIN RESPONSE FACTOR7 (ARF7)/ARF19 and INDOLE ACETIC ACID7 pathway ensures correct nuclear placement toward the inner membrane domain. Moreover, precise inner nuclear placement relies on SPIKE1 Rho-GEF, SUPERCENTIPEDE1 Rho-GDI, and ACTIN7 (ACT7) function and to a lesser extent on VTI11 vacuolar SNARE activity. Strikingly, the directionality and/or velocity of outer polar nuclear migration into the hair outgrowth along actin strands also are ACT7 dependent, auxin sensitive, and regulated by ROP signaling. This polyimine network is converted in the second step into polyimidazolium chloride and finally to a polyNHC network. Furthermore a porous Cu(II)-coordinated polyNHC network can be generated. Supercritical drying generates polymer networks with high permanent surface areas and porosities which can be applied for different catalytic reactions.
