Wie bei allen Proteinen dieser Enzymfamilie ist auch die Polypeptidkette von Pel-15 zu einer einstr{\"a}ngigen, rechtsg{\"a}ngigen, parallelen β-Helix aufgewunden. In diesem Strukturmotiv enth{\"a}lt jede Windung drei β-Str{\"a}nge, die jeweils durch flexible Schleifenbereiche miteinander verbunden sind. Insgesamt acht Windungen stapeln sich in Pel-15 {\"u}bereinander und bilden entlang der Helixachse fl{\"a}chige, parallele β-Faltbl{\"a}tter aus. Im Bereich dieser β-Faltbl{\"a}tter existiert ein ausgedehntes Netzwerk von Wasserstoffbr{\"u}ckenbindungen, durch das der hydrophobe Kern, der sich im Inneren der β-Helix befindet, vom umgebenden L{\"o}sungsmittel abgeschirmt wird. Besondere Abschlussstrukturen an beiden Enden der β-Helix, wie sie typischerweise bei anderen Ver-tretern dieser Strukturklasse ausgepr{\"a}gt werden, sind in Pel-15 nicht zu beobachten. Stattdessen sind die terminalen Bereiche der β-Helix {\"u}ber Salzbr{\"u}cken und hydrophobe Seitenkettenkontakte stabilisiert. In der vorliegenden Dissertation wurde die Pektatlyase Pel-15 hinsichtlich ihres Faltungsgleichgewichtes, ihrer enzymatischen Aktivit{\"a}t und der Kinetik ihrer Strukturbildung charakterisiert. In eine evolution{\"a}r konservierte Helixwindung wurden destabilisierende Mutationen eingef{\"u}hrt, und deren Auswirkungen mittels spektroskopischer Methoden analysiert. Die Ergebnisse zeigen, dass Pel-15 in Gegenwart des Denaturierungsmittels Guanidiniumhydrochlorid einen hyperfluoreszenten Gleichgewichtsustand (HF) populiert, der nach Messungen von Faltungs- und Entfaltungskinetiken ein konformationelles Ensemble aus den Zust{\"a}nden HFslow und HFfast darstellt. Diese HF-Zust{\"a}nde sind durch eine hohe Aktivierungsbarriere voneinander getrennt. In R{\"u}ckfaltungsexperimenten populieren nur etwa 80 \% der faltenden Molek{\"u}le den Zwischenzustand HFslow, der mit einer Zeitkonstante von ca. 100 s zu HFfast weiterreagiert. Die Denaturierungsmittelabh{\"a}ngigkeit dieser Reaktion ist sehr gering, was eine trans-/cis-Prolylisomerisierung als geschwindigkeitslimitierenden Schritt nahelegt. Die Existenz eines cis-Peptides in der nativen Struktur macht es erforderlich, den denaturierten Zustand als ein Ensemble kinetisch separierter Konformationen, kurz: DSE, zu betrachten, das durch die Spezies Ufast und Uslow populiert wird. Nach dem in dieser Arbeit aufgestellten „Minimalmodell der Pel-15 Faltung" stehen die HF-Spezies (HFslow, HFfast) mit den Konformationen des DSE in einem thermodynamischen Kreisprozess. Das Modell positioniert HFfast und die native Konformation N auf die „native Seite" der Aktivierungsbarriere und tr{\"a}gt damit der Tatsache Rechnung, dass die Gleichgewichtseinstellung zwischen diesen Spezies zu schnell ist, um mit manuellen Techniken erfasst zu werden. Die hochaffine Bindung von Ca2+ (Kd = 10 μM) verschiebt sich das Faltungsgleichgewicht bereits in Gegenwart von 1 mM CaCl2 soweit auf die Seite des nativen Zustandes, das HFfast nicht l{\"a}nger nachweisbar ist. Entgegen anf{\"a}nglicher Vermutungen kommt einer lokalen, evolution{\"a}r konservierten Disulfidbr{\"u}cke im Zentrum der β-Helix eine wichtige Stabilisierungsfunktion zu. Die Disulfidbr{\"u}cke befindet sich in einem kurzen Schleifenbereich der β-Helix nahe dem aktiven Zentrum. Obwohl ihr Austausch gegen die Reste Val und Ala die freie Stabilisierungsenthalpie des Proteins um ca. 10 kJ/mol reduziert, l{\"a}sst die Struktur im Bereich der Mutationsstelle keine gravierende Ver{\"a}nderung erkennen. Auch die katalytisch relevante Ca2+-Bindungsaffinit{\"a}t bleibt unbeeinflusst; dennoch zeigen Enzymaktivit{\"a}tstests f{\"u}r VA-Mutanten eine Reduktion der enzymatischen Aktivit{\"a}t um fast 50 \% an. Die evolution{\"a}r konservierte Helixwindung im Allgemeinen und die in ihr enthaltene Disulfidbr{\"u}cke im Besonderen m{\"u}ssen nach den vorliegenden Ergebnissen also eine zentrale Funktion sowohl f{\"u}r die Struktur des katalytischen Zentrums als auch f{\"u}r die Strukturbildung der β-Helix w{\"a}hrend der Faltungsreaktion besitzen. Die Ergebnisse dieser Arbeit finden in mehreren Punkten Anklang an Faltungseigenschaften, die f{\"u}r andere β -Helixproteine beschrieben wurden. Vor allem aber pr{\"a}destinieren sie Pel-15 als ein neues, β-helikales Modellprotein. In Arabidopsis (Arabidopsis thaliana), TREHALOSE-6-PHOSPHATE SYNTHASE1 (TPS1) catalyzes the synthesis of the sucrose-signaling metabolite trehalose 6-phosphate (Tre6P) and is essential for embryogenesis and normal postembryonic growth and development. To understand its molecular functions, we transformed the embryo-lethal tps1-1 null mutant with various forms of TPS1 and with a heterologous TPS (OtsA) from Escherichia coli, under the control of the TPS1 promoter, and tested for complementation. TPS1 protein localized predominantly in the phloem-loading zone and guard cells in leaves, root vasculature, and shoot apical meristem, implicating it in both local and systemic signaling of Suc status. The protein is targeted mainly to the nucleus. Restoring Tre6P synthesis was both necessary and sufficient to rescue the tps1-1 mutant through embryogenesis. However, postembryonic growth and the sucrose-Tre6P relationship were disrupted in some complementation lines. A point mutation (A119W) in the catalytic domain or truncating the C-terminal domain of TPS1 severely compromised growth. Despite having high Tre6P levels, these plants never flowered, possibly because Tre6P signaling was disrupted by two unidentified disaccharide-monophosphates that appeared in these plants. We expressed tagged forms of TREHALOSE-6-PHOSPHATE SYNTHASE1 (TPS1) to determine where Tre6P is synthesized in arabidopsis (Arabidopsis thaliana), and investigated the impact of localized changes in Tre6P levels, in axillary buds or vascular tissues, on shoot branching in wild-type and branching mutant backgrounds. TPS1 is expressed in axillary buds and the subtending vasculature, as well as in the leaf and stem vasculature. Expression of a heterologous Tre6P phosphatase (TPP) to lower Tre6P in axillary buds strongly delayed bud outgrowth in long days and inhibited branching in short days. TPP expression in the vasculature also delayed lateral bud outgrowth and decreased branching. Increased Tre6P in the vasculature enhanced branching and was accompanied by higher expression of FLOWERING LOCUS T (FT) and upregulation of sucrose transporters. Increased vascular Tre6P levels enhanced branching in branched1 but not in ft mutant backgrounds. The chosen model taxon, the genus Campylomormyrus, exhibits a wide diversity of electric organ discharge (EOD) waveform types. Their EOD is age, sex, and species specific and is an important character for discriminating among species that are otherwise cryptic. After having established a complementary set of molecular markers, I examined the radiation of Campylomormyrus by a combined approach of molecular data (sequence data from the mitochondrial cytochrome b and the nuclear S7 ribosomal protein gene, as well as 18 microsatellite loci, especially developed for the genus Campylomormyrus), observation of ontogeny and diversification of EOD waveform, and morphometric analysis of relevant morphological traits. I built up the first convincing phylogenetic hypothesis for the genus Campylomormyrus. Taking advantage of microsatellite data, the identified phylogenetic clades proved to be reproductively isolated biological species. This way I detected at least six species occurring in sympatry near Brazzaville/Kinshasa (Congo Basin). By combining molecular data and EOD analyses, I could show that there are three cryptic species, characterised by their own adult EOD types, hidden under a common juvenile EOD form. In addition, I confirmed that adult male EOD is species-specific and is more different among closely related species than among more distantly related ones. This result and the observation that the EOD changes with maturity suggest its function as a reproductive isolation mechanism. As a result of my morphometric shape analysis, I could assign species types to the identified reproductively isolated groups to produce a sound taxonomy of the group. Besides this, I could also identify morphological traits relevant for the divergences between the identified species. Among them, the variations I found in the shape of the trunk-like snout, suggest the presence of different trophic specializations; therefore, this trait might have been involved in the ecological radiation of the group. Mit dem Ziel die Anzahl der Tierversuche zu verringern, die notwendig sind um das toxikologische Profil einer Pr{\"u}fsubstanz zu bestimmen, wurde der Embryonale Stammzelltest (EST) entwickelt. Als Grundlage des EST dienen embryonale Stammzellen (ES-Zellen) einer Zelllinie. ES-Zellen sind Zellen, die sich in der fr{\"u}hen embryonalen Entwicklung in die Zellen der Keimbl{\"a}tter entwickeln k{\"o}nnen. Daraus wiederum differenzieren die vielen verschiedenen, unterschiedlich spezialisierten Zelltypen des komplexen Organismus. Im EST wird die Konzentration einer Pr{\"u}fsubstanz bestimmt, bei der die Differenzierung von ES-Zellen zu Herzmuskelzellen zu 50 \% inhibiert wird. Zus{\"a}tzlich wird die Konzentration der Pr{\"u}fsubstanz bestimm\&\#176;t, bei der 50 \% der ES-Zellen (IC50D3) bzw. Fibroblastenzellen (IC503T3) absterben. Die allgemeine Toxizit{\"a}t ist damit von der spezifischen Toxizit{\"a}t der Pr{\"u}fsubstanz auf die ES-Zellen und deren Differenzierung unterscheidbar. Die Parameter fliessen in ein biostatistisches Modell zur Pr{\"a}diktion des embryotoxischen Potentials der Pr{\"u}fsubstanzen ein. Es wurde ein Versuchsprotokoll entwickelt, wonach die ES-Zellen sich verst{\"a}rkt zu Endothelzellen differenzieren. Die Endothelzellen, die im lebenden Organismus die Wand der sp{\"a}teren Blutgef{\"a}sse, wie Venen und Arterien bilden, wurden mittels molekularbiologischer Methoden auf der RNA- und der Protein-Ebene nachgewiesen und quantifiziert. Verschiedene Zellkulturmethoden, Wachstumsfaktoren, als auch Wachstumsfaktorkonzentrationen wurden auf deren Verm{\"o}gen die Differenzierung der ES-Zellen zu Endothelzellen zu induzieren, untersucht. Nach der Etablierung des Differenzierungsprotokolls wurden sieben Substanzen auf deren Verm{\"o}gen gepr{\"u}ft, die Differenzierung von ES-Zellen zu Endothelzellen zu inhibieren. Die Endothelzellen wurden dabei {\"u}ber die Expression der RNA von zwei endothelzellspezifischen Genen quantifiziert. Im Vergleich dazu wurden die IC50D3 und die IC503T3 der Pr{\"u}fsubstanz bestimmt, um eine Absch{\"a}tzung des embryotoxischen Potentials der Pr{\"u}fsubstanz zu erm{\"o}glichen. Die Ergebnisse zeigten, dass eine Absch{\"a}tzung des embryotoxischen Potentials der sieben Pr{\"u}fsubstanzen in nicht-, schwach- oder stark embryotoxisch vorgenommen werden konnte. Es ist zu schlussfolgern, dass der weiterentwickelte in vitro Embryotoxizit{\"a}tsassay sensitiv und reproduzierbar ist. Mit der Verwendung von verschiedenen Differenzierungsendpunkten kann die Pr{\"a}diktionskraft des Assays deutlich verbessert, und die Anzahl von Tierversuchen verringert werden. This study aims (a) to develop a comprehensive multi-objective calibration framework using remote sensing vegetation data and hydrological signatures (flow duration curve - FDC, and baseflow index) in addition to discharge, and (b) to apply this framework for calibration of the Soil and Water Assessment Tool (SWAT) in a typical Andean catchment. Overall, our calibration approach outperformed traditional discharge-based and FDC signature-based calibration strategies in terms of vegetation, streamflow, and flow partitioning simulation. New hydrological insights for the region are the following: baseflow is the main component of the streamflow sustaining the long dry-season flow, and pasture areas offer higher water yield and baseflow than other land-cover types. Methods One-hundred and nineteen young male handball players (under (U)-13 (U13) [n = 85]) and U15 [n = 34]) volunteered to participate in this study. The participating athletes had a mean background of sytematic handball training of 5.5 ± 2.8 years and they exercised on average 540 ± 10.1 min per week including sport-specific team handball training and strength and conditioning programs. Players were tested for passive shoulder range-of-motion (ROM) for both internal (IR) and external rotation (ER) and isometric strength (i.e., IR and ER) of the dominant/non-dominant shoulders, overhead medicine ball throw (OMB), hip isometric abductor (ABD) and adductor (ADD) strength, hip ROM, jumps (countermovement jump [CMJ] and triple leg-hop [3H] for distance), linear sprint test, modified 505 change-of-direction (COD) test and handball throwing speed (7 m [HT7] and 9 m [HT9]). Results U15 players outperformed U13 in upper (i.e., HT7 and HT9 speed, OMB, absolute IR and ER strength of the dominant and non-dominant sides; Cohen's d: 0.76-2.13) and lower body (i.e., CMJ, 3H, 20-m sprint and COD, hip ABD and ADD; d: 0.70-2.33) performance measures. Regarding shoulder ROM outcomes, a lower IR ROM was found of the dominant side in the U15 group compared to the U13 and a higher ER ROM on both sides in U15 (d: 0.76-1.04). It seems that primarily anthropometric characteristics (i.e., body height, body mass) and upper body strength/power (OMB distance) are the most important factors that explain the throw speed variance in male handball players, particularly in U13. To differentiate the vegetation variability due to ENSO, we removed the ENSO signal from the climate data using empirical orthogonal teleconnection (EOT) analysis. Then, we simulated the ecosystem carbon and water fluxes under the historical climate without components related to ENSO teleconnections. We found ENSO-driven patterns in vegetation response and confirmed that EOT analysis can successfully produce coupled tropical Pacific sea surface temperature-eastern African rainfall teleconnection from observed datasets. We further simulated eastern African vegetation response under future climate change as it is projected by climate models and under future climate change combined with a predicted increased ENSO intensity. Our EOT analysis highlights that climate simulations are still not good at capturing rainfall variability due to ENSO, and as we show here the future vegetation would be different from what is simulated under these climate model outputs lacking accurate ENSO contribution. Als n{\"a}chstes wurde die fr{\"u}here Vegetation simuliert, f{\"u}r die fossile Pollen-Daten zum Abgleich vorliegen. {\"U}ber Computermodelle lassen sich Wissensl{\"u}cken zwischen Standorten {\"u}berbr{\"u}cken, bei denen wir {\"u}ber fossile Pollen-Daten verf{\"u}gen, sodass ein vollst{\"a}ndigeres Bild der Vergangenheit entsteht. Zus{\"a}tzlich validiert wurde die Leistungsf{\"a}higkeit des Modells durch die hohe {\"U}bereinstimmung zwischen Modell und Pollen-Daten, wo sie im Raum {\"u}berlappen. Nachdem das Modell getestet und f{\"u}r die Region validiert war, konnte eine der seit langem offenen Fragen {\"u}ber die ostafrikanische Vegetation angegangen werden, n{\"a}mlich wie Ostafrika seines Tropenwaldes verlustig gehen konnte. In den Tropen wird die heutige Vegetation weltweit haupts{\"a}chlich von W{\"a}ldern dominiert, mit Ausnahme der Tropengebiete Ostafrikas, wo W{\"a}lder nur noch stellenweise an der K{\"u}ste und im Hochland vorkommen. Durch eine Reihe von Simulationsexperimenten konnte aufgezeigt werden, unter welchen Bedingungen jene Waldgebiete fr{\"u}her zusammenhingen und schließlich fragmentiert wurden. Die Studie hat erwiesen, wie empfindlich die ostafrikanische Vegetation f{\"u}r die Klimaschwankungen ist, die durch den k{\"u}nftigen Klimawandel zu erwarten sind. Weitere Auswirkungen auf das ostafrikanische Klima ergeben sich aus dem El Ni{\~n}o/Southern Oscillation-Ph{\"a}nomen (ENSO), das aus Temperaturfluktuationen zwischen dem Ozean und der Atmosph{\"a}re herr{\"u}hrt und k{\"u}nftig an Intensit{\"a}t zunehmen d{\"u}rfte. Although the conjoint occurrence between brain-structural and brain-functional changes, the decrease of plasticity, and changes in cognitive abilities suggests a certain causality between these processes, concrete evidence for the relation between brain development, language processing, and language performance is rare. Therefore, the current dissertation investigates the tripartite relationship between behavior (in the form of language performance and cognitive maturation as prerequisite for language processing), brain structure (in the form of gray matter maturation), and brain function (in the form of brain activation evoked by complex sentence processing). Previous developmental studies indicate a missing increase of activation in accordance to sentence complexity (functional selectivity) in language-relevant brain areas in children. To determine the factors contributing to the functional development of language-relevant brain areas, different methodologies and data acquisition techniques were used to investigate the processing of center-embedded sentences in 5- and 6-year-old children, 7- and 8-year-old children, and adults. Behavioral results indicate that children between 5 and 8 years show difficulties in processing double embedded sentences and that their performance for these type of sentences is positively correlated with digit span. In 7- and 8-year-old children, it was found that especially the processing of long-distance relations between the initial phrase and its corresponding verb appears to be associated with the subject's verbal working memory capacity. In contrast, children's performance for double embedded sentences in the younger age group positively correlated with their performance in a standardized sentence comprehension test. This finding supports the hypothesis that processing difficulties in this age group may be mainly attributed to difficulties in processing case marking information. These findings are discussed with respect to current accounts of language and working memory development. A second study aimed at investigating the structural maturation of brain areas involved in sentence comprehension. To do this, whole-brain magnetic resonance images from 59 children between 5 and 8 years were collected and children's gray matter was analyzed by using voxel-based morphometry. Children's grammatical proficiency was assessed by a standardized sentence comprehension test. A confirmatory factory analysis corroborated a grammar-relevant and a verbal working memory-relevant factor underlying the measured performance. While children's ability to assign thematic roles is positively correlated with gray matter probability (GMP) in the left inferior temporal gyrus and the left inferior frontal gyrus, verbal working memory-related performance is positively correlated with GMP in the left parietal operculum extending into the posterior superior temporal gyrus. These areas have been previously shown to be differentially engaged in adults' complex sentence processing. Thus, the findings of the second study suggest a specific correspondence between children's GMP in language-relevant brain regions and differential cognitive abilities which underlie complex sentence comprehension. In a third study, functional brain activity during the processing of center-embedded sentences was investigated in three different age groups (5-6 years, 7-8 years, and adults). Although all age groups engage a qualitatively comparable network of the left pars opercularis (PO), the left inferior parietal lobe extending into the posterior superior temporal gyrus (IPL/pSTG), the supplementary motor area (SMA) and the cerebellum, functional selectivity of these regions was only observable in adults. However, functional activation of the language-related regions (PO and IPL/pSTG) predicted sentence comprehension performance for all age groups. To solve the question of the complex interplay between different maturational factors, a fourth study analyzed the predictive power of gray matter probability, verbal working memory capacity, and behavioral differences in performance for simple and complex sentence for the functional selectivity of each activated region. These analyses revealed that the establishment of the adult-like functional selectivity for complex sentences is predicted by a reduction of the left PO's gray matter probability across age groups while that of the IPL/pSTG is additionally predicted by verbal working memory capacity. So unterschiedlich die Jetquellen von ihrer Erscheinung sein m{\"o}gen, zwei Tatsachen scheinen sie zu vereinen: Alle Jetquellen zeigen ebenfalls Hinweise auf die Existenz einer Akkretionsscheibe und von Magnetfeldern. Damit sind die wichtigsten Punkte einer Theorie der Jetentstehung schon umrissen. Sie muss sowohl die komplexe Struktur der Jetquelle ber{\"u}cksichtigen - ein System bestehend aus einem Zentralobjekt, der es umgebenden Scheibe, und dem Jet - als auch die magnetohydrodynamische Wechselwirkung zwischen diesen Komponenten. Subject pronouns are always technically ambiguous, and the application of syntactic constraints on their interpretation depends on properties of the antecedent that is to be retrieved. While pronouns can freely corefer with non-quantified referential antecedents, linking a pronoun to a quantified antecedent is only possible in certain syntactic configurations via variable binding. We report the results from a judgment task and three online reading comprehension experiments investigating pronoun resolution with quantified and non-quantified antecedents. Results from both the judgment task and participants' eye movements during reading indicate that comprehenders freely allow pronouns to corefer with non-quantified antecedents, but that retrieval of quantified antecedents is restricted to specific syntactic environments. On base of a thorough review of the literature on hydrothermolysis and hydrothermal carbonisation of sugars in addition to the chemistry of hydroxymethylfurfural, mechanistic considerations of the formation of hydrothermal carbon are proposed. On the base of these reaction schemes, the mineral borax, is introduced as an additive for the hydrothermal carbonisation of glucose. It was found to be a highly active catalyst, resulting in decreased reaction times and increased carbon yields. The chemical impact of borax, in the following is exploited for the modification of the micro- and nanostructure of hydrothermal carbon. From the borax - mediated aggregation of those primary species, widely applicable, low density, pure hydrothermal carbon aerogels with high porosities and specific surface areas are produced. To conclude the first section of the thesis, a short series of experiments is carried out, for the purpose of demonstrating the applicability of the HC model to "real" biowaste i.e. watermelon waste as feedstock for the production of useful materials. In part two cyano - containing ionic liquids are employed as precursors for the synthesis of high - performance, heteroatom - containing carbon materials. By varying the ionic liquid precursor and the carbonisation conditions, it was possible to design highly active non - metal electrocatalyst for the reduction of oxygen. In the direct reduction of oxygen to water (like used in polymer electrolyte fuel cells), compared to commercial platinum catalysts, astonishing activities are observed. In another example the selective and very cost efficient electrochemical synthesis of hydrogen peroxide is presented. In a last example the synthesis of graphitic boron carbon nitrides from the ionic liquid 1 - Ethyl - 3 - methylimidazolium - tetracyanoborate is investigated in detail. Importantly, the human brain inherits from evolution a predisposition to map cardinality onto space, as revealed by the presence of spatial-numerical associations (SNAs) in humans and animals. Here, the mapping of cardinal information onto physical space is addressed as a hallmark signature characterizing numerical cognition. According to traditional approaches, cognition is defined as complex forms of internal information processing, taking place in the brain (cognitive processor). On the contrary, embodied cognition approaches define cognition as functionally linked to perception and action, in the continuous interaction between a biological body and its physical and sociocultural environment. Embracing the principles of the embodied cognition perspective, I conducted four novel studies designed to unveil how SNAs originate, develop, and adapt, depending on characteristics of the organism, the context, and their interaction. I structured my doctoral thesis in three levels. At the grounded level (Study 1), I unfold the biological foundations underlying the tendency to map cardinal information across space; at the embodied level (Study 2), I reveal the impact of atypical motor development on the construction of SNAs; at the situated level (Study 3), I document the joint influence of visuospatial attention and task properties on SNAs. Furthermore, I experimentally investigate the presence of associations between physical and numerical distance, another numerical property fundamental for the development of efficient mathematical minds (Study 4). In Study 1, I present the Brain's Asymmetric Frequency Tuning hypothesis that relies on hemispheric asymmetries for processing spatial frequencies, a low-level visual feature that the (in)vertebrate brain extracts from any visual scene to create a coherent percept of the world. Computational analyses of the power spectra of the original stimuli used to document the presence of SNAs in human newborns and animals, support the brain's asymmetric frequency tuning as a theoretical account and as an evolutionarily inherited mechanism scaffolding the universal and innate tendency to represent cardinality across horizontal space. In Study 2, I explore SNAs in children with rare genetic neuromuscular diseases: spinal muscular atrophy (SMA) and Duchenne muscular dystrophy (DMD). SMA children never accomplish independent motoric exploration of their environment; in contrast, DMD children do explore but later lose this ability. The different SNAs reported by the two groups support the critical role of early sensorimotor experiences in the spatial representation of cardinality. In Study 3, I directly compare the effects of overt attentional orientation during explicit and implicit processing of numerical magnitude. First, the different effects of attentional orienting based on the type of assessment support different mechanisms underlying SNAs during explicit and implicit assessment of numerical magnitude. Secondly, the impact of vertical shifts of attention on the processing of numerical distance sheds light on the correspondence between numerical distance and peri-personal distance. In Study 4, I document the presence of different SNAs, driven by numerical magnitude and numerical distance, by employing different response mappings (left vs. right and near vs. distant). In the field of numerical cognition, the four studies included in the present thesis contribute to unveiling how the characteristics of the organism and the environment influence the emergence, the development, and the flexibility of our attitude to represent cardinal information across space, thus supporting the predictions of the embodied cognition approach. Furthermore, they inform a taxonomy of body-centred factors (biological properties of the brain and sensorimotor system) modulating the spatial representation of cardinality throughout the course of life, at the grounded, embodied, and situated levels. If the awareness for different variables influencing SNAs over the course of life is important, it is equally important to consider the organism as a whole in its sensorimotor interaction with the world. Inspired by my doctoral research, here I propose a holistic perspective that considers the role of evolution, embodiment, and environment in the association of cardinal information with directional space. The new perspective advances the current approaches to SNAs, both at the conceptual and at the methodological levels. We present a language - Babelsberg - that unifies the constructs for en- capsulation and abstraction by using only object-oriented method definitions for both declarative and imperative code. Our prototype - Babelsberg/R - is an extension to Ruby, and continues to support Ruby's object-oriented se- mantics. It allows programmers to add constraints to existing Ruby programs in incremental steps by placing them on the results of normal object-oriented message sends. It is implemented by modifying a state-of-the-art Ruby virtual machine. The performance of standard object-oriented code without con- straints is only modestly impacted, with typically less than 10\% overhead compared with the unmodified virtual machine. A central question, then, is how to overcome such obstacles to cooperation. We study this question in the context of Germany, by asking whether the propensity of immigrant youth to cooperate with native peers was affected by a major integration reform: the introduction of birthright citizenship. Our unique setup exploits data from a large-scale lab-in-the-field experiment in a quasi-experimental evaluation framework. We find that the policy caused male, but not female, immigrants to significantly increase their cooperativeness toward natives. We show that the increase in out-group cooperation among immigrant boys is an outcome of more trust rather than a reflection of stronger other- regarding preferences towards natives. In exploring factors that may explain these behavioral effects, we present evidence that the policy also led to a near-closure of the educational achievement gap between young immigrant men and their native peers. Windinstabilit{\"a}t durch Dopplerde-shadowing des Gases. Die Instabilit{\"a}t bewirkt, dass Windgas in dichte Schalen komprimiert wird, die von starken Stoßfronten begrenzt sind. Schnelle Wolken entstehen im Raum zwischen den Schalen und stoßen mit diesen zusammen. Dies erzeugt R{\"o}ntgenflashes, die die beobachtete R{\"o}ntgenstrahlung heißer Sterne erkl{\"a}ren k{\"o}nnen. 2. Wind runway durch radiative Wellen. Der runaway zeigt, warum beobachtete liniengetriebene Winde schnelle, kritische L{\"o}sungen anstelle von Brisenl{\"o}sungen (oder shallow solutions) annehmen. Here we apply this method to the problem of ice flow to examine the dynamic similitude of isothermal ice sheets in shallow-shelf approximation against the scaling of their geometry and physical parameters. Carrying out a dimensional analysis of the stress balance we obtain dimensionless numbers that characterize the flow. Requiring that these numbers remain the same under scaling we obtain conditions that relate the geometric scaling factors, the parameters for the ice softness, surface mass balance and basal friction as well as the ice-sheet intrinsic response time to each other. We demonstrate that these scaling laws are the same for both the (two-dimensional) flow-line case and the three-dimensional case. The theoretically predicted ice-sheet scaling behavior agrees with results from numerical simulations that we conduct in flow-line and three-dimensional conceptual setups. We further investigate analytically the implications of geometric scaling of ice sheets for their response time. With this study we provide a framework which, under several assumptions, allows for a fundamental comparison of the ice-dynamic behavior across different scales. It proves to be useful in the design of conceptual numerical model setups and could also be helpful for designing laboratory glacier experiments. We find that the initiation of a MISI in one basin can induce a destabilization in the other. The underlying mechanism of basin interaction is based on dynamic thinning and a consecutive motion of the ice divide which induces a thinning in the adjacent basin and a successive initiation of the instability. Our simplified and symmetric topographic setup allows scaling both the geometry and the transition time between both instabilities. We find that the ice profile follows a universal shape that is scaled with the horizontal extent of the ice sheet and that the same exponent of 1/2 applies for the scaling relation between central surface elevation and horizontal extent as in the pure shallow ice approximation (Vialov profile). Altering the central bed elevation, we find that the extent of grounding-line retreat in one basin determines the degree of interaction with the other. Different scenarios of basin interaction are discussed based on our modeling results as well as on a conceptual flux-balance analysis. With point-like laser irradiation, radial inward and outward hydrodynamic surface flows are remotely switched on and off. In this way, ensembles of microparticles can be moved toward or away from the irradiation center. Particle motion is analyzed according to varying parameters, such as surfactant and salt concentration, illumination condition, surface hydrophobicity, and surface structure. The physical origin of this process is the so-called light-driven diffusioosmosis (LDDO), a phenomenon that was discovered in the framework of this thesis and is described experimentally and theoretically in this work. To compensate for the change in osmotic pressure near the surface, a hydrodynamic flow along the surface is generated. Surface-surfactant interaction largely governs LDDO. It is shown that surfactant adsorption depends on the isomerization state of the surfactant. Photoisomerization, therefore, triggers a surfactant attachment or detachment from the surface. This change is considered to be one of the reasons for the formation of LDDO flow. These flows are introduced not only by a focused laser source but also by global irradiation. Porous particles show reversible repulsive and attractive interactions when dispersed in the solution of photosensitive surfactant. Repulsion and attraction is controlled by the irradiation wavelength. Illumination with red light leads to formation of aggregates, while illumination with blue light leads to the formation of a well-separated grid with equal interparticle distances, between 2µm and 80µm, depending on the particle surface density. These long-range interactions are considered to be a result of an increase or decrease of surfactant concentration around each particle, depending on the irradiation wavelength. Surfactant molecules adsorb inside the pores of the particles. A light-induced photoisomerization changes adsorption to the pores and drives surfactant molecules to the outside. The concentration gradients generate symmetric flows around each single particle resulting in local LDDO. Die Ionen und Isotopenverh{\"a}ltnisse im Formationswasser gel{\"o}ster Komponenten in GrSk belegen einen gemeinsamen Genesepfad mit W{\"a}ssern anderer Rotliegend-Reservoire des Nordostdeutschen Beckens (NEGB). Die Isotopenverh{\"a}ltnisse von ⁸⁷Sr/⁸⁶Sr ≈ 0,7158 und von δ³⁴SV CDT ≈ 4,1 per mille des Sulfats weisen auf die Anreicherung des Fluids mit schweren Isotopen durch die Fluid Gestein-Wechselwirkung mit Vulkaniten und Rotliegend Sandsteinen des Unteren Perms hin. Das im Formationswasser bei Reservoirbedingungen gel{\"o}ste Gas (Gas/Wasser ≤ 2 bei STP) enth{\"a}lt Stickstoff (δ¹⁵NAir ≈ 0,6 per mille) und thermogenes Methan (δ¹³CV-PDB ≈ - 18 per mille) aus organischen Karbonablagerungen (Kerogen Typ - III Kohlen) hoher Reife. Die Isotopenverh{\"a}ltnisse der Edelgase belegen eine krustale Herkunft des Gasgemisches. Die berechnete Verweilzeit τ (⁴He) der Gase im Reservoir liegt zwischen 275 und 317 Ma und {\"u}berschreitet damit bei gegebener Konzentration von Mutternukliden im Reservoirgestein das allgemein angenommene Zeitalter der Sedimentgruppe. Das l{\"a}sst sich durch eine Zuwanderung von Gasen aus {\"a}lteren Sedimentfolgen erkl{\"a}ren. Die Ver{\"a}nderungen der physikochemischen Fluidparameter w{\"a}hrend des Anlagenbetriebs sind haupts{\"a}chlich temperaturbedingt. Bei stabilen Produktionsbedingungen und einer Temperatur von ca. 100 °C stabilisieren sich auch die Fluideigenschaften. Bei In situ Bedingungen {\"u}bertage betr{\"a}gt die Dichte ρ = 1,1325 ± 0,0002 g ∙ mL⁻¹, das Redoxpotential Eh = -105,5 ± 1,3 mV und der pH = 6,61 ± 0,002. Die relative Zusammensetzung der Gasphase bei stabilen Produktionsbedingungen zeigt dagegen eine geringe Erh{\"o}hung des Stickstoffanteils sowie des Anteils der Kohlenwasserstoffe (Ethan, Propan, usw.) und Abnahme des relativen Methananteils im Laufe des Betriebs. Die quantitative Untersuchung der sekund{\"a}ren mineralischen Ausf{\"a}llungen im Fluid mittels sequentieller Extraktion zeigte, dass Schwermetalle als eine Hauptkomponente der Fluidfestphase gr{\"o}ßtenteils in Verbindung mit organischen Molek{\"u}len vorliegen. Experimente zum Einfluss organischer Verbindungen unterschiedlicher Substanzklassen auf eine Mobilisierung der Schwermetalle aus dem Reservoirgestein ergaben, dass die Verbindungen wie Fetts{\"a}uren und PAK (polyzyklische aromatische Kohlenwasserstoffe) die Freisetzung von Kupfer, Nickel, Chrom und Blei verhindern bzw. zu derer Immobilisierung beitragen. Im Gegensatz dazu wird die Mobilit{\"a}t von Zink in Anwesenheit von diesen Verbindungen erh{\"o}ht. Niedermolekulare Monocarbons{\"a}uren und stickstoffhaltige Heteroaromaten tragen, mit Ausnahme von Blei, zur Freisetzung bzw. Mobilisierung von Schwermetallen aus dem Reservoirgestein bei. Die gewonnenen Erkenntnisse dieser Arbeit best{\"a}tigen das Risiko massiver Ausf{\"a}llungen auf der kalten Seite der Geothermieanlage bei Inbetriebnahme des Kraftwerks, wenn keine an den Fluidchemismus angepassten Pr{\"a}ventionsmethoden eingesetzt werden. The aim of the project is to reveal the impact of the transition from subduction to continent-continent collision of the African plate with the Cyprus-Anatolian plate. A visual quality check, frequency analysis and filtering were applied to the seismic data and reveal a good data quality. Subsequent first break picking, finite-differences ray tracing and inversion of the offshore wide-angle data leads to a first-arrival tomographic model. This model reveals (1) P-wave velocities lower than 6.5 km/s in the crust, (2) a variable crustal thickness of about 28 - 37 km and (3) an upper crustal reflection at 5 km depth beneath the ESM. Two land shots on Turkey, also recorded on Cyprus, airgun shots south of Cyprus and geological and previous seismic investigations provide the information to derive a layered velocity model beneath the Anatolian plateau and for the ophiolite complex on Cyprus. The analysis of the reflections provides evidence for a north-dipping plate subducting beneath Cyprus. The main features of this layered velocity model are (1) an upper and lower crust with large lateral changes of the velocity structure and thickness, (2) a Moho depth of about 38 - 45 km beneath the Anatolian plateau, (3) a shallow north-dipping subducting plate below Cyprus with an increasing dip and (4) a typical ophiolite sequence on Cyprus with a total thickness of about 12 km. The offshore-onshore seismic data complete and improve the information about the velocity structure beneath Cyprus and the deeper part of the offshore tomographic model. Thus, the wide-angle seismic data provide detailed insights into the 2-D geometry and velocity structures of the uplifted and overriding Cyprus-Anatolian plate. Subsequent gravity modelling confirms and extends the crustal P-wave velocity model. The deeper part of the subducting plate is constrained by the gravity data and has a dip angle of ~ 28°. Dazu wurden sie insgesamt drei Mal im Abstand von jeweils sechs Monaten im ersten Jahr nach ihrem Studiumsabschluss befragt (T1: in den ersten zwei Wochen nach dem Staatsexamen, T2: kurzzeitig nach dem Berufseinstieg, T3: im Schnitt 9.5 Monate nach dem Berufseinstieg). Die Ergebnisse zeigten zun{\"a}chst, dass unl{\"a}ngst examinierte Jung-{\"A}rztInnen, die sich vergleichsweise schlechter auf den Beruf durch das Studium vorbereitet f{\"u}hlten, ihren bevorstehenden Berufseinstieg negativer bewerteten und schon vor diesem beanspruchter waren. Die Bewertung des Berufseinstiegs vermittelte dabei den Zusammenhang zwischen einer schlechten Vorbereitung und der Beanspruchung. Arbeitsspezifische Copingfunktionalit{\"a}t wiederum pufferte den Zusammenhang zwischen einer schlechten Vorbereitung und der Bewertung des Berufseinstiegs. Das Problem einer als schlecht empfundenen Vorbereitung verdeutlichte sich in der L{\"a}ngsschnittanalyse - sie sagte eine h{\"o}here Beanspruchung zum zweiten Messzeitpunkt, d.h. nach dem Berufseinstieg, vorher. In der Untersuchung der Beanspruchungsentwicklung {\"u}ber die drei Messzeitpunkte hinweg fanden sich nur wenige Ver{\"a}nderungen. Es ließ sich zwar eine deutliche Zunahme der mittleren Depressivit{\"a}ts-Auspr{\"a}gungen {\"u}ber den Berufseinstieg hinweg herausstellen (T1-T2); auf anderen Beanspruchungsindikatoren zeigte sich jedoch kein direkter Effekt des Arbeitsbeginns bzw. fand sich auch keine Adaptation der Jung-{\"A}rztInnen an ihre neue Situation im Sinne einer sich verringernden Beanspruchung im weiteren Verlauf (T2-T3). In der Erkl{\"a}rung interindividueller Unterschiede in der Beanspruchung im Untersuchungszeitraum zeigte sich, dass die sich mit dem Berufseinstieg einstellende Arbeitsbelastung zum zweiten und dritten Messzeitpunkt erwartungsgem{\"a}ß positiv mit Beanspruchung assoziiert war. Die Arbeitsbelastungs-Beanspruchungs-Beziehung bestand jedoch nur im Querschnitt; in der L{\"a}ngsschnittanalyse fand sich kein Effekt der T2-Arbeitsbelastung auf die T3-Beanspruchung. Ausgangsunterschiede in psychischen Ressourcen wirkten einerseits direkt auf die Beanspruchung zu T2, zum Teil moderierten sie aber auch den Zusammenhang zwischen der Arbeitsbelastung und Beanspruchung: Eine h{\"o}here Resilienz und die Wahrnehmung sozialer Unterst{\"u}tzung sagten eine geringere Beanspruchung nach dem Berufseinstieg vorher. Jung-{\"A}rztInnen, die sich durch eine st{\"a}rkere Arbeitsbelastung auszeichneten, aber {\"u}ber ein funktionaleres Bew{\"a}ltigungsverhalten im Arbeitskontext verf{\"u}gten, waren kurzzeitig nach dem Berufseinstieg weniger beansprucht als stark arbeitsbelastete Jung-{\"A}rztInnen mit weniger funktionalem Coping. Verringerungen in den psychischen Ressourcen {\"u}ber den Berufseinstieg hinweg wirkten sich direkt, d.h. per se ung{\"u}nstig auf die Beanspruchung zum dritten Messzeitpunkt aus. Zudem interagierten sie mit der zu diesem Zeitpunkt bestehenden Arbeitsbelastung in Vorhersage der Beanspruchung. St{\"a}rker arbeitsbelastete Jung-{\"A}rztInnen, deren Copingfunktionalit{\"a}t und Wahrnehmung sozialer Unterst{\"u}tzung vom ersten zum dritten Messzeitpunkt abgenommen hatte, waren am Ende des Untersuchungszeitraums am st{\"a}rksten beansprucht. Hinsichtlich der Auswirkungen des Berufseinstiegs auf die Pers{\"o}nlichkeit der Jung-{\"A}rztInnen fanden sich ung{\"u}nstige Ver{\"a}nderungen: Sowohl die Auspr{\"a}gungen psychischer Ressourcen (Widerstandsf{\"a}higkeit, Wahrnehmung sozialer Unterst{\"u}tzung hinsichtlich der Arbeitst{\"a}tigkeit) als auch die der Big Five-Faktoren nahmen im Mittel ab. Interindividuelle Unterschiede in den Ver{\"a}nderungen ließen sich auf die Beanspruchung kurzzeitig nach dem Berufseinstieg (T2) bzw. auf deren Entwicklung in den Folgemonaten (T2-T3) zur{\"u}ckf{\"u}hren: Jene Jung-{\"A}rztInnen, die vergleichsweise stark beansprucht auf den Berufseinstieg reagiert hatten bzw. deren Beanspruchung im weiteren Verlauf zunahm, zeigten entsprechend ung{\"u}nstige Ver{\"a}nderungen. Die Ergebnisse zusammengefasst verdeutlicht sich folgende Problematik: Jung-{\"A}rztInnen, die weniger gut, d.h. pers{\"o}nlichkeitsbasiert gesch{\"u}tzt den Berufseinstieg absolvieren, reagieren st{\"a}rker beansprucht und sind dann auch diejenigen, deren Pers{\"o}nlichkeit sich in den ersten Arbeitsmonaten ung{\"u}nstig ver{\"a}ndert. Jung-{\"A}rztInnen mit geringen psychischen Ressourcen sind folglich nicht nur besonders vulnerabel f{\"u}r die Entwicklung von Beanspruchung angesichts belastender Arbeitsbedingungen, sondern ihre vergleichsweise hohe Beanspruchung bedingt eine weitere Verringerung des Schutz- und Pufferpotenzials ihrer Pers{\"o}nlichkeit. The article tests this claim by assessing whether World Bank projects perform worse in implementation environments with a higher corruption level. The article focuses specifically on bribery between public officials and firms during the procurement of needed goods and services. Approach and Methods: I use data from the World Bank's Enterprise Surveys to avoid the often-criticized corruption perception indices and to allow for an assessment of effects at the subnational level. The analysis builds on an assessment of the performance ratings of 1,228 World Bank projects and covers 87 different countries. Finding: Overall, the article finds a small but statistically significant correlation between the corruption level and project performance. This result indicates that the corruption level of recipient countries should be considered during the design and implementation of projects. Policy Implications: Nonetheless, the relatively small correlation and the low pseudo R-squareds advise not overestimating the relevance of corruption for project performance. Es ist allerdings unklar, auf welche Weise angehende Physiklehrkr{\"a}fte Aspekte des kompetenzorientierten Experimentierens in ihrem Unterricht in schulpraktischen Ausbildungsphasen einsetzen, auf welche Weise sie solche Unterrichtsversuche mit Experimentierbezug reflektieren und wie strukturiert (im Sinne der Ablaufmodelle) sie dabei vorgehen. In der vorliegenden Studie wurde deshalb untersucht, auf welche Weise Praxissemesterstudierende Experimentierprozesse in ihren Unterrichtsversuchen reflektieren. Hierf{\"u}r wurde betrachtet, zu welchen Anteilen die Experimentierphasen in den Reflexionen adressiert werden. Um weiterhin herauszufinden, mit welcher Qualit{\"a}t die Experimentplanung reflektiert wird und inwiefern sich Vorstrukturierung f{\"u}r die Planungsphase zeigt, wurde diese differenzierter betrachtet. Auf Basis empirischer Vorarbeiten wurde vermutet, dass Fragenentwicklung, Hypothesenbildung und Experimentplanung seltener thematisiert werden als die anderen Teilkompetenzen und dass die Planungsphase haupts{\"a}chlich stark vorstrukturierte Elemente enth{\"a}lt, statt den Lernenden Freir{\"a}ume f{\"u}r selbstst{\"a}ndige Planungen zu lassen. Zur Untersuchung der Fragestellung wurden Kodiermanuale zur Erfassung experimentierbezogener Kompetenzen in schriftlichen Reflexionen entwickelt und validiert. Analysiert wurden 40 Reflexionstexte von 14 Studierenden des Physik-Lehramts im Praxissemester an der Universit{\"a}t Potsdam. Als Untersuchungsmethode wurde die qualitative Inhaltsanalyse genutzt. Die Texte wurden bez{\"u}glich der Umsetzung eines Reflexionsmodells und auf das Vorkommen der Teilkompetenzen des Experimentierzyklus untersucht. Die Ergebnisse best{\"a}tigten das geringe Vorkommen der Fragenentwicklung und Hypothesenbildung sowie die tendenziell geschlossenen Planungsinhalte. Zudem konnte festgestellt werden, dass die Planungsphase eher oberfl{\"a}chlich reflektiert und vor allem Arbeitsauftr{\"a}ge wiedergegeben wurden. Allgemein zeigten sich haupts{\"a}chlich beschreibende Tendenzen in den Reflexionen und eher wenige Alternativen und Konsequenzen. The aim of this thesis was to increase knowledge of uncultivated methanogenic microorganisms in submarine and terrestrial permafrost deposits, their community composition, the role they play with regard to methane emissions, and their phylogeny. It is assumed that methanogenic communities in warmer submarine permafrost may serve as a model to anticipate the response of methanogenic communities in colder terrestrial permafrost to rising temperatures. The compositions of methanogenic communities were examined in terrestrial and submarine permafrost sediment samples. The submarine permafrost studied in this research was 10°C warmer than the terrestrial permafrost. By polymerase chain reaction (PCR), DNA was extracted from each of the samples and analyzed by molecular microbiological methods such as PCR-DGGE, RT-PCR, and cloning. Furthermore, these samples were used for in vitro experiment and FISH. The submarine permafrost analysis of the isotope composition of CH4 suggested a relationship between methane content and in situ active methanogenesis. Furthermore, active methanogenesis was proven using 13C-isotope measurements of methane in submarine permafrost sediment with a high TOC value and a high methane concentration. In the molecular-microbiological studies uncultivated lines of Methanosarcina, Methanomicrobiales, Methanobacteriacea and the Groups 1.3 and Marine Benthic from Crenarchaeota were found in all submarine and terrestrial permafrost samples. Methanosarcina was the dominant group of the Archaea in all submarine and terrestrial permafrost samples. The archaeal community composition, in particular, the methanogenic community composition showed diversity with changes in temperatures. Furthermore, cell count of methanogens in submarine permafrost was 10 times higher than in terrestrial permafrost. In vitro experiments showed that methanogens adapt quickly and well to higher temperatures. Ethical guidelines stated transparency about model development and validation as a requirement for trustworthy AI. Abundant guidance exists to provide transparency through reporting, but poorly reported medical AI tools are common. To close this transparency gap, we developed and piloted a framework to quantify the transparency of medical AI tools with three use cases. Our framework comprises a survey to report on the intended use, training and validation data and processes, ethical considerations, and deployment recommendations. The transparency of each response was scored with either 0, 0.5, or 1 to reflect if the requested information was not, partially, or fully provided. Additionally, we assessed on an analogous three-point scale if the provided responses fulfilled the transparency requirement for a set of trustworthiness criteria from ethical guidelines. The degree of transparency and trustworthiness was calculated on a scale from 0\% to 100\%. Our assessment of three medical AI use cases pin-pointed reporting gaps and resulted in transparency scores of 67\% for two use cases and one with 59\%. We report anecdotal evidence that business constraints and limited information from external datasets were major obstacles to providing transparency for the three use cases. The observed transparency gaps also lowered the degree of trustworthiness, indicating compliance gaps with ethical guidelines. All three pilot use cases faced challenges to provide transparency about medical AI tools, but more studies are needed to investigate those in the wider medical AI sector. Applying this framework for an external assessment of transparency may be infeasible if business constraints prevent the disclosure of information.
