It is based on 1.5 years of weekly rainfall samples. In the first step, the isotopic composition of the samples is analyzed by local meteoric water lines (LMWLs) and single-factor linear correlations. Additionally, the contribution of several regional and local factors is quantified by multiple linear regression (MLR) of all possible factor combinations and by relative importance analysis. This approach is novel for the interpretation of isotopic records and enables an objective quantification of the explained variance in isotopic records for individual factors. In this study, the local factors are extracted from local climate records, while the regional factors are derived from atmospheric backward trajectories of water particles. The regional factors, i.e., precipitation, temperature, relative humidity and the length of backward trajectories, are combined with equivalent local climatic parameters to explain the response variables delta O-18, delta H-2, and d-excess of precipitation at the station of measurement. The results indicate that (i) MLR can better explain the isotopic variation in precipitation (R-2 = 0.8) compared to single-factor linear regression (R-2 = 0.3); (ii) the isotopic variation in precipitation is controlled dominantly by regional moisture regimes (similar to 70 \%) compared to local climatic conditions (similar to 30 \%); (iii) the most important climatic parameter during the rainy season is the precipitation amount along the trajectories of air mass movement; (iv) the influence of local precipitation amount and temperature is not sig-nificant during the rainy season, unlike the regional precipitation amount effect; (v) secondary fractionation processes (e.g., sub-cloud evaporation) can be identified through the d-excess and take place mainly in the dry season, either locally for delta O-18 and delta H-2, or along the air mass trajectories for d-excess. The analysis shows that regional and local factors vary in importance over the seasons and that the source regions and transport pathways, and particularly the climatic conditions along the pathways, have a large influence on the isotopic composition of rainfall. Although the general results have been reported qualitatively in previous studies (proving the validity of the approach), the proposed method provides quantitative estimates of the controlling factors, both for the whole data set and for distinct seasons. Therefore, it is argued that the approach constitutes an advancement in the statistical analysis of isotopic records in rainfall that can supplement or precede more complex studies utilizing atmospheric models. Due to its relative simplicity, the method can be easily transferred to other regions, or extended with other factors. The results illustrate that the interpretation of the isotopic composition of precipitation as a recorder of local climatic conditions, as for example performed for paleorecords of water isotopes, may not be adequate in the southern part of the Indochinese Peninsula, and likely neither in other regions affected by monsoon processes. These processes have not been fully addressed in the VMD so far. The thesis is based on statistical analyses of the isotopic data of Global Network of Isotopes in Precipitation (GNIP), of meteorological and hydrological data from Vietnamese agencies, and of the stable water isotopes and monitoring data collected as part of this work. First, the controlling processes of precipitation were quantified by the combination of trajectory analysis, multi-factor linear regression, and relative importance analysis (hereafter, a model-based statistical approach). The validity of this approach is confirmed by similar, but mainly qualitative results obtained in other studies. The total variation in precipitation isotopes (δ18O and δ2H) can be better explained by multiple linear regression (up to 80\%) than single-factor linear regression (30\%). The relative importance analysis indicates that atmospheric moisture regimes control precipitation isotopes rather than local climatic conditions. The most crucial factor is the upstream rainfall along the trajectories of air mass movement. However, the influences of regional and local climatic factors vary in importance over the seasons. The developed model-based statistical approach is a robust tool for the interpretation of precipitation isotopes and could also be applied to understand the controlling processes of precipitation in other regions. Second, the concept of the two-component lumped-parameter model (LPM) in conjunction with stable water isotopes was applied to examine the surface-groundwater interaction in the VMD. A calibration framework was also set up to evaluate the behaviour, parameter identifiability, and uncertainties of two-component LPMs. The modelling results provided insights on the subsurface flow conditions, the recharge contributions, and the spatial variation of groundwater transit time. The subsurface flow conditions at the study site can be best represented by the linear-piston flow distribution. The contributions of the recharge sources change with distance to the river. The mean transit time (mTT) of riverbank infiltration increases with the length of the horizontal flow path and the decreasing gradient between river and groundwater. River water infiltrates horizontally mainly via the highly permeable aquifer, resulting in short mTTs (<40 weeks) for locations close to the river (<200 m). The vertical infiltration from precipitation takes place primarily via a low-permeable overlying aquitard, resulting in considerably longer mTTs (>80 weeks). Notably, the transit time of precipitation infiltration is independent of the distance to the river. All these results are hydrologically plausible and could be quantified by the presented method for the first time. This study indicates that the highly complex mechanism of surface-groundwater interaction at riverbank infiltration systems can be conceptualized by exploiting two-component LPMs. It is illustrated that the model concept can be used as a tool to investigate the hydrological functioning of mixing processes and the flow path of multiple water components in riverbank infiltration systems. Lastly, a suite of time series analysis approaches was applied to examine the groundwater dynamics in the VMD. The assessment was focused on the time-variant trends of groundwater levels (GWLs), the groundwater memory effect (representing the time that an aquifer holds water), and the hydraulic response between surface water and multi-layer alluvial aquifers. The analysis indicates that the aquifers act as low-pass filters to reduce the high-frequency signals in the GWL variations, and limit the recharge to the deep groundwater. The groundwater abstraction has exceeded groundwater recharge between 1997 and 2017, leading to the decline of groundwater levels (0.01-0.55 m/year) in all considered aquifers in the VMD. The memory effect varies according to the geographical location, being shorter in shallow aquifers and flood-prone areas and longer in deep aquifers and coastal regions. Groundwater depth, season, and location primarily control the variation of the response time between the river and alluvial aquifers. These findings are important contributions to the hydrogeological literature of a little-known groundwater system in an alluvial setting. It is suggested that time series analysis can be used as an efficient tool to understand groundwater systems where resources are insufficient to develop a physical-based groundwater model. This doctoral thesis demonstrates that important aspects of hydrological processes can be understood by statistical analysis of stable water isotope and monitoring data. The approaches developed in this thesis can be easily transferred to regions in similar tropical environments, particularly those in alluvial settings. H. rhodopensis plants remained green and viable throughout the dark treatment. Transcriptomic analysis revealed that darkness regulated several transcription factor (TF) genes. Stress-and autophagy-related TFs such as ERF8, HSFA2b, RD26, TGA1, and WRKY33 were up-regulated, while chloroplast-and flowering-related TFs such as ATH1, COL2, COL4, RL1, and PTAC7 were repressed. PHYTOCHROME INTERACTING FACTOR4, a negative regulator of photomorphogenesis and promoter of senescence, also was down-regulated. In response to darkness, most of the photosynthesis-and photorespiratory-related genes were strongly down-regulated, while genes related to autophagy were up-regulated. We identified 750 non-redundant, experimentally verified sites with three-dimensional (3D) structural information available in the protein data bank (PDB) and grouped them according to their respective kinase family. We studied the spatial distribution of amino acids around phosphorserines, phosphothreonines, and phosphotyrosines to extract signature 3D-profiles. Characteristic spatial distributions of amino acid residue types around phosphorylation sites were indeed discernable, especially when kinase-family-specific target sites were analyzed. To test the added value of using spatial information for the computational prediction of phosphorylation sites, Support Vector Machines were applied using both sequence as well as structural information. When compared to sequence-only based prediction methods, a small but consistent performance improvement was obtained when the prediction was informed by 3D-context information. They include metabolic pathway maps, protein-protein interaction networks as well as kinase interaction networks. First, we present an integrated analysis of metabolic pathway maps and protein-protein interaction networks (PIN). It has long been established that successive enzymatic steps are often catalyzed by physically interacting proteins forming permanent or transient multi-enzyme complexes. Inspecting high-throughput PIN data, it has been shown recently that, indeed, enzymes involved in successive reactions are generally more likely to interact than other protein pairs. In this study, we expanded this line of research to include comparisons of the respective underlying network topologies as well as to investigate whether the spatial organization of enzyme interactions correlates with metabolic efficiency. Analyzing yeast data, we detected long-range correlations between shortest paths between proteins in both network types suggesting a mutual correspondence of both network architectures. We discovered that the organizing principles of physical interactions between metabolic enzymes differ from the general PIN of all proteins. While physical interactions between proteins are generally dissortative, enzyme interactions were observed to be assortative. Thus, enzymes frequently interact with other enzymes of similar rather than different degree. Enzymes carrying high flux loads are more likely to physically interact than enzymes with lower metabolic throughput. In particular, enzymes associated with catabolic pathways as well as enzymes involved in the biosynthesis of complex molecules were found to exhibit high degrees of physical clustering. Single proteins were identified that connect major components of the cellular metabolism and hence might be essential for the structural integrity of several biosynthetic systems. Besides metabolic aspects of PINs, we investigated the characteristic topological properties of protein interactions involved in signaling and regulatory functions mediated by kinase interactions. Characteristic topological differences between PINs associated with metabolism, and those describing phosphorylation networks were revealed and shown to reflect the different modes of biological operation of both network types. The construction of phosphorylation networks is based on the identification of specific kinase-target relations including the determination of the actual phosphorylation sites (P-sites). The computational prediction of P-sites as well as the identification of involved kinases still suffers from insufficient accuracies and specificities of the underlying prediction algorithms, and the experimental identification in a genome-scale manner is not (yet) doable. Computational prediction methods have focused primarily on extracting predictive features from the local, one-dimensional sequence information surrounding P-sites. However the recognition of such motifs by the respective kinases is a spatial event. A powerful toolbox to study these highly dynamic processes in the native environment is provided by fluorescence fluctuation spectroscopy (FFS) techniques. In more detail, FFS takes advantage of the inherent dynamics present in biological systems, such as diffusion, to infer molecular parameters from fluctuations of the signal emitted by an ensemble of fluorescently tagged molecules. In particular, two parameters are accessible: the concentration of molecules and their transit times through the observation volume. In addition, molecular interactions can be measured by analyzing the average signal emitted per molecule - the molecular brightness - and the cross-correlation of signals detected from differently tagged species. In the present work, several FFS techniques were implemented and applied in different biological contexts. In particular, scanning fluorescence correlation spectroscopy (sFCS) was performed to measure protein dynamics and interactions at the plasma membrane (PM) of cells, and number and brightness (N\&B) analysis to spatially map molecular aggregation. To account for technical limitations and sample related artifacts, e.g. detector noise, photobleaching, or background signal, several correction schemes were explored. In addition, sFCS was combined with spectral detection and higher moment analysis of the photon count distribution to resolve multiple species at the PM. Using scanning fluorescence cross-correlation spectroscopy and cross-correlation N\&B, the interactions of amyloid precursor-like protein 1 (APLP1), a synaptic membrane protein, were investigated. It is shown for the first time directly in living cells, that APLP1 undergoes specific interactions at cell-cell contacts. It is further demonstrated that zinc ions induce formation of large APLP1 clusters that enrich at contact sites and bind to clusters on the opposing cell. Altogether, these results provide direct evidence that APLP1 is a zinc ion dependent neuronal adhesion protein. In the context of APLP1, discrepancies of oligomeric state estimates were observed, which were attributed to non-fluorescent states of the chosen red fluorescent protein (FP) tag mCardinal (mCard). Therefore, multiple FPs and their performance in FFS based measurements of protein interactions were systematically evaluated. The study revealed superior properties of monomeric enhanced green fluorescent protein (mEGFP) and mCherry2. Furthermore, a simple correction scheme allowed unbiased in situ measurements of protein oligomerization by quantifying non-fluorescent state fractions of FP tags. The procedure was experimentally confirmed for biologically relevant protein complexes consisting of up to 12 monomers. In the last part of this work, fluorescence correlation spectroscopy (FCS) and single particle tracking (SPT) were used to characterize diffusive transport dynamics in a bacterial biofilm model. Biofilms are surface adherent bacterial communities, whose structural organization is provided by extracellular polymeric substances (EPS) that form a viscous polymer hydrogel. The presented study revealed a probe size and polymer concentration dependent (anomalous) diffusion hindrance in a reconstituted EPS matrix system caused by polymer chain entanglement at physiological concentrations. This result indicates a meshwork-like organization of the biofilm matrix that allows free diffusion of small particles, but strongly hinders diffusion of larger particles such as bacteriophages. Finally, it is shown that depolymerization of the matrix by phage derived enzymes rapidly facilitated free diffusion. In the context of phage infections, such enzymes may provide a key to evade trapping in the biofilm matrix and promote efficient infection of bacteria. Still, the knowledge about the mechanisms of the nutrient exchange between the symbiotic partners is very limited. Therefore, transport processes of both, the plant and the fungal partner, are investigated in this study. In order to enhance the understanding of the molecular basis underlying this tight interaction between the roots of Medicago truncatula and the AM fungus Rhizophagus irregularis, genes involved in transport processes of both symbiotic partners are analysed here. The AM-specific regulation and cell-specific expression of potential transporter genes of M. truncatula that were found to be specifically regulated in arbuscule-containing cells and in non-arbusculated cells of mycorrhizal roots was confirmed. A model for the carbon allocation in mycorrhizal roots is suggested, in which carbohydrates are mobilized in non-arbusculated cells and symplastically provided to the arbuscule-containing cells. New insights into the mechanisms of the carbohydrate allocation were gained by the analysis of hexose/H+ symporter MtHxt1 which is regulated in distinct cells of mycorrhizal roots. Metabolite profiling of leaves and roots of a knock-out mutant, hxt1, showed that it indeed does have an impact on the carbohydrate balance in the course of the symbiosis throughout the whole plant, and on the interaction with the fungal partner. The primary metabolite profile of M. truncatula was shown to be altered significantly in response to mycorrhizal colonization. Additionally, molecular mechanisms determining the progress of the interaction in the fungal partner of the AM symbiosis were investigated. The R. irregularis transcriptome in planta and in extraradical tissues gave new insight into genes that are differentially expressed in these two fungal tissues. Over 3200 fungal transcripts with a significantly altered expression level in laser capture microdissection-collected arbuscules compared to extraradical tissues were identified. Among them, six previously unknown specifically regulated potential transporter genes were found. These are likely to play a role in the nutrient exchange between plant and fungus. While the substrates of three potential MFS transporters are as yet unknown, two potential sugar transporters are might play a role in the carbohydrate flow towards the fungal partner. Mortality due to non-predation factors is often ignored even though anecdotal evidence of non-predation mass mortality of zooplankton has been reported repeatedly. One way to estimate non-predation mortality rate is to measure the removal rate of carcasses, for which sinking is the primary removal mechanism especially in quiescent shallow water bodies. Objectives and Results We used sediment traps to quantify in situ carcass sinking velocity and non-predation mortality rate on eight consecutive days in 2013 for the cladoceran Bosmina longirostris in the oligo-mesotrophic Lake Stechlin; the outcomes were compared against estimates derived from in vitro carcass sinking velocity measurements and an empirical model correcting in vitro sinking velocity for turbulence resuspension and microbial decomposition of carcasses. Our results show that the latter two approaches produced unrealistically high mortality rates of 0.58-1.04 d(-1), whereas the sediment trap approach, when used properly, yielded a mortality rate estimate of 0.015 d(-1), which is more consistent with concurrent population abundance data and comparable to physiological death rate from the literature. Ecological implications Zooplankton carcasses may be exposed to water column microbes for days before entering the benthos; therefore, non-predation mortality affects not only zooplankton population dynamics but also microbial and benthic food webs. While numerous studies have shown that AGNs can be variable at all wavelengths, the nature of the variability is still not well understood. Studying the properties of LLAGNs may help to understand better galaxy evolution, and how AGNs transit between active and inactive states. In this thesis, we develop a method to extract variability properties of AGNs. Using multi-epoch deep photometric observations, we subtract the contribution of the host galaxy at each epoch to extract variability and estimate AGN accretion rates. This pipeline will be a powerful tool in connection with future deep surveys such as PANSTARS. The second study in this thesis describes a survey of X-ray selected AGN hosts at redshifts z>1.5 and compares them to quiescent galaxies. These in-memory databases (IMDBs) profit from DRAM's low latency and high throughput as well as from the removal of costly abstractions used in disk-based systems, such as the buffer cache. However, as the DRAM technology approaches physical limits, scaling these databases becomes difficult. Non-volatile memory (NVM) addresses this challenge. This new type of memory is persistent, has more capacity than DRAM (4x), and does not suffer from its density-inhibiting limitations. Yet, as NVM has a higher latency (5-15x) and a lower throughput (0.35x), it cannot fully replace DRAM. IMDBs thus need to navigate the trade-off between the two memory tiers. We present a solution to this optimization problem. Leveraging information about access frequencies and patterns, our solution utilizes NVM's additional capacity while minimizing the associated access costs. Unlike buffer cache-based implementations, our tiering abstraction does not add any costs when reading data from DRAM. As such, it can act as a drop-in replacement for existing IMDBs. Our contributions are as follows: (1) As the foundation for our research, we present Hyrise, an open-source, columnar IMDB that we re-engineered and re-wrote from scratch. Hyrise enables realistic end-to-end benchmarks of SQL workloads and offers query performance which is competitive with other research and commercial systems. At the same time, Hyrise is easy to understand and modify as repeatedly demonstrated by its uses in research and teaching. (2) We present a novel memory management framework for different memory and storage tiers. By encapsulating the allocation and access methods of these tiers, we enable existing data structures to be stored on different tiers with no modifications to their implementation. Besides DRAM and NVM, we also support and evaluate SSDs and have made provisions for upcoming technologies such as disaggregated memory. (3) To identify the parts of the data that can be moved to (s)lower tiers with little performance impact, we present a tracking method that identifies access skew both in the row and column dimensions and that detects patterns within consecutive accesses. Unlike existing methods that have substantial associated costs, our access counters exhibit no identifiable overhead in standard benchmarks despite their increased accuracy. (4) Finally, we introduce a tiering algorithm that optimizes the data placement for a given memory budget. In the TPC-H benchmark, this allows us to move 90\% of the data to NVM while the throughput is reduced by only 10.8\% and the query latency is increased by 11.6\%. With this, we outperform approaches that ignore the workload's access skew and access patterns and increase the query latency by 20\% or more. Individually, our contributions provide novel approaches to current challenges in systems engineering and database research. Nachdem zun{\"a}chst er{\"o}rtert wird, was im Rahmen der Untersuchung unter dem Begriff der transnationalen Kriminalit{\"a}t verstanden wird, wird der internationale Rechtsrahmen zur Bek{\"a}mpfung anhand von f{\"u}nf beispielhaft ausgew{\"a}hlten transnationalen Kriminalit{\"a}tsph{\"a}nomenen beschrieben. Im darauffolgenden Hauptteil der Untersuchung wird der Frage nachgegangen, weshalb dieser durch die internationale Staatengemeinschaft geschaffene Rechtsrahmen, gerade in fragilen Staaten, kaum einen Beitrag dazu leistet solchen Kriminalit{\"a}tsph{\"a}nomenen effektiv zu begegnen. Dabei wird festgestellt, dass die Genese des internationalen Rechtsrahmens zu einem Legitimit{\"a}tsdefizit desselbigen f{\"u}hrt. Auch die mangelhafte Ber{\"u}cksichtigung der speziellen Lebensrealit{\"a}ten, die in vielen fragilen Staaten vorzufinden sind, wirkt sich negativ auf die Durchsetzbarkeit des internationalen Rechtsrahmens aus. Es wird dargelegt, dass unterschiedlich hohe menschenrechtliche Schutzstandards zu Normenkollisionen bei der internationalen Zusammenarbeit der Staaten f{\"u}hren, insbesondere im Rahmen der internationalen Rechtshilfe. Sowohl Adipositas als auch die Suszeptibilit{\"a}t gegen{\"u}ber Diabetes sind zu einem erheblichen Teil genetisch determiniert. Die relevanten Risikogene, deren Interaktion mit der Umwelt, insbesondere mit Bestandteilen der Nahrung, und die Pathomechanismen, die zur Insulinresistenz und Diabetes f{\"u}hren, sind nicht vollst{\"a}ndig aufgekl{\"a}rt. In der vorliegenden Arbeit sollte durch Genexpressionsanalysen des weißen Fettgewebes (WAT) und der Langerhansschen Inseln die Entstehung und Progression von Adipositas und Typ-2-Diabetes untersucht werden, um relevante Pathomechanismen und neue Kandidatengene zu identifizieren. Zu diesem Zweck wurden Di{\"a}t-Interventionsstudien mit NZO- und verwandten NZL-M{\"a}usen, zwei polygenen Mausmodellen f{\"u}r das humane metabolische Syndrom, durchgef{\"u}hrt. Eine kohlenhydrathaltige Hochfett-Di{\"a}t (HF: 14,6 \% Fettanteil) f{\"u}hrte in beiden Mausmodellen zu fr{\"u}her Adipositas, Insulinresistenz und Typ 2 Diabetes. Eine fettreduzierte Standarddi{\"a}t (SD: 3,3 \% Fettanteil), welche die Entstehung von Adipositas und Diabetes stark verz{\"o}gert, sowie eine diabetesprotektive kohlenhydratfreie Hochfett-Di{\"a}t (CHF: 30,2 \% Fettanteil) dienten als Kontrolldi{\"a}ten. Mit Hilfe der Microarray-Technologie wurden genomweite Expressionsprofile des WAT erstellt. Pankreatische Inseln wurden durch laserbasierte Mikropr{\"a}paration (Laser Capture Microdissection; LCM) isoliert und ebenfalls hinsichtlich ihres Expressionsprofils analysiert. Differenziell exprimierte Gene wurden durch Real-Time-PCR validiert. Im WAT der NZO-Maus bewirkte die HF-Di{\"a}t eine reduzierte Expression nukle{\"a}rer Gene der oxidativen Phosphorylierung und von lipogenen Enzymen. Dies deutet auf eine inad{\"a}quate Fettspeicherung und -verwertung in diesen Tieren hin. Die Reduktion in der Fettspeicherung und -oxidation ist spezifisch f{\"u}r das adip{\"o}se NZO-Modell und konnte bei der schlanken SJL Maus nicht beobachtet werden, was auf eine m{\"o}gliche Beteiligung an der Entstehung der Insulinresistenz hinweist. Zus{\"a}tzlich wurde best{\"a}tigt, dass die Expansion des Fettgewebes bei der adip{\"o}sen NZO-Maus eine zeitlich verz{\"o}gerte Infiltration von Makrophagen in das WAT und dort eine lokale Immunantwort ausl{\"o}st. Dar{\"u}ber hinaus wurde die Methode der LCM etabliert und zur Gewinnung hochangereicherter RNA aus den Langerhansschen Inseln eingesetzt. In erstmalig durchgef{\"u}hrten genomweiten Expressionsanalysen wurde zu einem fr{\"u}hen Zeitpunkt in der Diabetesentwicklung der Einfluss einer diabetogenen HF-Di{\"a}t und einer diabetesprotektiven CHF-Di{\"a}t auf das Expressionsprofil von pankreatischen Inselzellen verglichen. Im Gegensatz zum WAT bewirkt die diabetogene HF-Di{\"a}t in Inselzellen einerseits, eine erh{\"o}hte Expression von nukle{\"a}ren Genen f{\"u}r die oxidative Phosphorylierung und andererseits von Genen, die mit Zellproliferation assoziiert sind. Zudem wurden 37 bereits annotierte Gene identifiziert, deren differenzielle Expression mit der Diabetesentwicklung korreliert. Das Peptidhormon Cholecystokinin (Cck, 11,8-fach erh{\"o}ht durch die HF) stellt eines der am st{\"a}rksten herauf regulierten Gene dar. Die hohe Anreicherung der Cck-mRNA in Inselzellen deutet auf eine bisher unbekannte Funktion des Hormons in der Regulation der Inselzellproliferation hin. Der Transkriptionsfaktor Mlxipl (ChREBP; 3,8-fach erniedrigt durch die HF) stellt in Langerhansschen Inseln eines der am st{\"a}rksten herunter regulierten Gene dar. Ferner wurde ChREBP, dessen Funktion als glucoseregulierter Transkriptionsfaktor f{\"u}r lipogene Enzyme bislang in der Leber, aber nicht in Inselzellen nachgewiesen werden konnte, erstmals immunhistochemisch in Inselzellen detektiert. Dies deutet auf eine neue, bisher unbekannte regulatorische Funktion von ChREBP im Glucosesensor-Mechanismus der Inselzellen hin. Eine durchgef{\"u}hrte Korrelation der mit der Diabetesentwicklung assoziierten, differenziell exprimierten Inselzellgene mit Genvarianten aus humanen genomweiten Assoziationsstudien f{\"u}r Typ-2-Diabetes (WTCCC, Broad-DGI-T2D-Studie) erm{\"o}glichte die Identifizierung von 24 neuartigen Diabetes-Kandidatengenen. Auf welchen Organisationsebenen der Lebensgemeinschaften waren die Auswirkungen dieser Stressoren erkennbar? • Der zweite Aspekt behandelte die Entwicklung der Artenzahl, die Gleichverteilung der Dominanz von Arten (Eveness) und die Diversit{\"a}t von Planktongemeinschaften entlang des Produktivit{\"a}tsgradienten. Speziell wurde untersucht, ob die Artenzahl und die Diversit{\"a}t eine monoton positive oder eine unimodale Funktion der Produktivit{\"a}t waren und ob die Eveness eine monoton abnehmende Funktion der Produktivit{\"a}t war. Zur besseren Vorhersagbarkeit der Entwicklung dieser Indizes wurden in einem n{\"a}chsten Schritt zus{\"a}tzliche biotische und abiotische Faktoren (z.B. Konsumenteneffekte, physikalische St{\"o}rung, Immigration) ber{\"u}cksichtigt. Zuletzt wurde die Hypothese getestet, dass unter dem Einfluss von extremem physiologischem Stress keine Abh{\"a}ngigkeit zwischen den betrachteten Indizes und der Produktivit{\"a}t von {\"O}kosystemen besteht. Die Untersuchungen der vorliegenden Arbeit f{\"u}hrten zu folgenden Ergebnissen und Schlussfolgerungen: 1) Der Reifungsprozess der Planktongemeinschaft war unter neutralen Bedingungen nicht eindeutig an einzelnen Kriterien festzumachen. Vielmehr schienen idiosynkratische Effekte einzelner Arten auf die Zusammensetzung und Funktion der Organismengemeinschaft von Bedeutung. Coenobiumbildende Kieselalgen sowie gr{\"o}ßere Cladoceren und Copepoden dominierten sehr rasch die Planktongemeinschaft und vermittelten den Eindruck eines reifen {\"O}kosystems fast unmittelbar nach der Neutralisierung des Tagebausees. 2) Der Einfluss von physiologischem S{\"a}urestress und physikalischer St{\"o}rung der Schichtung durch das eintretende Flutungswasser war gegen{\"u}ber einem neutralen, ungest{\"o}rten Teilbecken des Tagebausees (Referenzzustand) eindeutig zu erkennen. Die isolierte Betrachtung der Wirkung der Stressoren lieferte hinsichtlich fast aller Kriterien Anzeichen einer Verj{\"u}ngung des Systems sensu Odum (1969, 1985). 3) Im betrachteten {\"O}kosystem existierte eine Hierarchie innerhalb der Stressoren. Der Einfluss des S{\"a}urestresses dominierte gegen{\"u}ber dem Einfluss der physikalischen St{\"o}rung, wahrscheinlich indem er die Reaktionsm{\"o}glichkeiten der Planktongemeinschaft einschr{\"a}nkte. 4) F{\"u}r Prim{\"a}rproduzenten war die Artenzahl eine monoton positive Funktion der realisierten Biomasse (einem Surrogatparameter f{\"u}r die Produktivit{\"a}t des Systems). Die Eveness war eine monoton negative Funktion der Produktivit{\"a}t. Die beobachtete unimodale Beziehung zwischen der Diversit{\"a}t und der Produktivit{\"a}t der Prim{\"a}rproduzenten muss als eine Folge der mathematischen Formulierung dieser Indizes betrachtet werden. 5) Die Ergebnisse multivariater Modelle zur Vorhersage der Artenzahl und der Eveness der Prim{\"a}rproduzenten in Abh{\"a}ngigkeit zus{\"a}tzlicher erfassbarer biotischer und abiotischer Faktoren erm{\"o}glichten eine differenziertere Betrachtung der Ergebnisse: • Im Tagebausee Goitsche war die Eveness haupts{\"a}chlich von dichteabh{\"a}ngigen Prozessen gesteuert (negative Abh{\"a}ngigkeit zum Quadrat der Biomassen und zu den Lichtverh{\"a}ltnissen). • Die Entwicklung der Artenzahl war neben dem prim{\"a}ren Einfluss der zunehmenden Biomasse auch durch qualitative und quantitative Aspekte der Konsumentengemeinschaft (Diversit{\"a}t und Biomasse des Zooplanktons) beeinflusst. Der Einfluss einer erh{\"o}hten Immigration auf die Artenzahl wurde nur zu Beginn der Flutung des Tagebausees beobachtet. 6) Auf Ebene der Konsumenten war die einzige eindeutig feststellbare Abh{\"a}ngigkeit ein Anstieg der Artenzahl mit steigender Biomasse. In Chloroplasten, den gr{\"u}nen Organellen der Pflanzenzellen, findet ein Großteil der Regulation der Genexpression auf Ebene der Initiation dieses Schrittes statt. Eine Vielzahl von Eigenschaften der RNA und von Faktoren, die an die RNA binden, entfalten einen Einfluss auf diesen Schritt. Bisher unvollst{\"a}ndig aufgekl{\"a}rt ist die Rolle einer konservierten Nukleotidsequenz in der untranslatierten Region der RNA -- der Shine-Dalgarno-Sequenz. Diese stellt in Bakterien, wie z.B. E. coli als Ribosomenbindestelle sicher, dass Ribosomen den Anfang der zu translatierenden Sequenz zuverl{\"a}ssig erkennen. Im Rahmen dieser Arbeit wurden diverse DNA-Konstrukte in Plastiden von Tabak eingebracht. Hierzu z{\"a}hlten Konstrukte, die sowohl eine erh{\"o}hte Anzahl von Ribosomenbindestellen enthielten als auch vermehrte Startpunkte der Translation. Zus{\"a}tzlich wurden Konstrukte hergestellt, die die Situation von mehreren zu translatierenden Regionen in der RNA nachstellten. Es konnte festgestellt werden, dass plastid{\"a}re Ribosomen die strangaufw{\"a}rts gelegenen Translationsstartpunkte bevorzugen -- im Gegensatz zu E. coli, wo alle Startpunkte gleichm{\"a}ßig genutzt wurden. Hierdurch zeigten die prokaryotischen Ribosomen aus Chloroplasten, die sich aus bakteriellen Systemen ableiten, Eigenschaften von eukaryotischen Ribosomen. Ein zweites Teilprojekt dieser Arbeit besch{\"a}ftigte sich mit der Inkompatibilit{\"a}t von Chloroplasten mit dem Kerngenom. In Kreuzungen von Arten der Gattung Pelargonium fielen Kombinationen auf, bei denen die Tochterpflanzen bleiche Blattbereiche bis hin zu vollst{\"a}ndig weißen Pflanzen zeigten. Dieses Ph{\"a}nomen wird als Bastardbleichheit bezeichnet. In der Gattung Pelargonium werden Chloroplasten von beiden Elternteilen an die Tochterpflanzen vererbt. Da das Ph{\"a}nomen der Bastardbleichheit nur in einem der Plastiden vorkommt, nicht jedoch im anderen in der gleichen Pflanze, muss von einem Effekt ausgegangen werden, der von Plastiden ausgeht. Die Interaktionen zwischen Zellkern und Chloroplasten sind offensichtlich stark gest{\"o}rt. Zur detaillierten Untersuchung dieses Effekts wurde die Nukleotidsequenz von drei Chloroplastengenomen aufgekl{\"a}rt. Es konnte eine Reihe von Sequenzunterschieden der Genome ermittelt werden. Aus diesen wurde eine Reihe von Unterschieden beobachtet, die einen solchen Effekt zur Folge haben k{\"o}nnen. Due to the increasing amount of data in recent years, the problem has become even more challenging on the one hand, but has led to a renaissance in duplicate detection research on the other hand. This thesis examines the effects and opportunities of transitive relationships on the duplicate detection process. Transitivity implies that if record pairs ⟨ri,rj⟩ and ⟨rj,rk⟩ are classified as duplicates, then also record pair ⟨ri,rk⟩ has to be a duplicate. However, this reasoning might contradict with the pairwise classification, which is usually based on the similarity of objects. An essential property of similarity, in contrast to equivalence, is that similarity is not necessarily transitive. First, we experimentally evaluate the effect of an increasing data volume on the threshold selection to classify whether a record pair is a duplicate or non-duplicate. Our experiments show that independently of the pair selection algorithm and the used similarity measure, selecting a suitable threshold becomes more difficult with an increasing number of records due to an increased probability of adding a false duplicate to an existing cluster. Thus, the best threshold changes with the dataset size, and a good threshold for a small (possibly sampled) dataset is not necessarily a good threshold for a larger (possibly complete) dataset. As data grows over time, earlier selected thresholds are no longer a suitable choice, and the problem becomes worse for datasets with larger clusters. Second, we present with the Duplicate Count Strategy (DCS) and its enhancement DCS++ two alternatives to the standard Sorted Neighborhood Method (SNM) for the selection of candidate record pairs. DCS adapts SNMs window size based on the number of detected duplicates and DCS++ uses transitive dependencies to save complex comparisons for finding duplicates in larger clusters. We prove that with a proper (domain- and data-independent!) threshold, DCS++ is more efficient than SNM without loss of effectiveness. Third, we tackle the problem of contradicting pairwise classifications. Usually, the transitive closure is used for pairwise classifications to obtain a transitively closed result set. However, the transitive closure disregards negative classifications. We present three new and several existing clustering algorithms and experimentally evaluate them on various datasets and under various algorithm configurations. The results show that the commonly used transitive closure is inferior to most other clustering algorithms, especially for the precision of results. In scenarios with larger clusters, our proposed EMCC algorithm is, together with Markov Clustering, the best performing clustering approach for duplicate detection, although its runtime is longer than Markov Clustering due to the subexponential time complexity. An interaction between the food quality and the MPs treatments was found in the reproduction of B. calyciflorus. PA and SiO2 beads had no effect on fitness response. The aim of my thesis was to investigate the ingestion and the effect of MPs in rotifers from a more natural scenario and to individuate processes such as the aggregation of MPs, the food dilution effect and the increasing concentrations of MPs that could influence the final outcome of MPs in the environment. In fact, in a near natural scenario MPs interaction with bacteria and algae, aggregations together with the size and concentration are considered drivers of ingestion and effect. The aggregation of MPs makes smaller MPs more available for rotifers and larger MPs less ingested. The negative effect caused by the ingestion of MPs was modulated by their size but also by the quantity and the quality of food that cause variable responses. In fact, rotifers in the environment are subjected to food limitation and the presence of MPs could exacerbate this condition and decrease the population and the reproduction input. Finally, in a scenario incorporating an entire zooplanktonic community, MPs were ingested by most individuals taking into account their feeding mode but also the concentration of MPs, which was found to be essential for the availability of MPs. Our study addresses this aim using bibliometric methods—bibliographic coupling and co-word analysis as they are particularly suitable for identifying current trends and future research priorities at the forefront of the research. To model learning strategies, we impose restrictions on the hypotheses made, for example requiring the conjectures to follow a monotonic behaviour. This way, we can study the impact a certain restriction has on learning. Recently, the literature shifted towards map charting. Here, various seemingly unrelated restrictions are contrasted, unveiling interesting relations between them. The results are then depicted in maps. For explanatory learning, the literature already provides maps of common restrictions for various forms of data presentation. In the case of behaviourally correct learning, where the learners are required to converge semantically instead of syntactically, the same restrictions as in explanatory learning have been investigated. However, a similarly complete picture regarding their interaction has not been presented yet. In this thesis, we transfer the map charting approach to behaviourally correct learning. In particular, we complete the partial results from the literature for many well-studied restrictions and provide full maps for behaviourally correct learning with different types of data presentation. We also study properties of learners assessed important in the literature. We are interested whether learners are consistent, that is, whether their conjectures include the data they are built on. While learners cannot be assumed consistent in explanatory learning, the opposite is the case in behaviourally correct learning. Even further, it is known that learners following different restrictions may be assumed consistent. We contribute to the literature by showing that this is the case for all studied restrictions. We also investigate mathematically interesting properties of learners. In particular, we are interested in whether learning under a given restriction may be done with strongly Bc-locking learners. CH vibrations were identified by their characteristic isotopic shifts in the spectra of deuterated triangular [4]phenylenes. Two CC stretching modes, at 100 cm-1 and 1176 cm-1, suitable as probes for bond strength changes in the excited state, were identified. The isotope effect on the internal conversion rates of triangular [4]phenylene was evaluated from measurements of temperature dependent lifetime. Isotope dependency and the magnitude of the internal conversion rates indicate that internal conversion in triangular [4]phenylene is most likely induced by CH vibrations. The results obtained by LESS and lifetime measurements were compared with PM3 PECI calculations of the excited state structure. While the BCI systems of almost all other groups require the user to be trained over several weeks or even months, the group of Prof. Dr. Klaus-Robert M{\"u}ller in Berlin and Potsdam, which I belong to, was one of the first research groups in this field which used machine learning techniques on a large scale. The adaptivity of the processing system to the individual brain patterns of the subject confers huge advantages for the user. Thus BCI research is considered a hot topic in machine learning and computer science. It requires interdisciplinary cooperation between disparate fields such as neuroscience, since only by combining machine learning and signal processing techniques based on neurophysiological knowledge will the largest progress be made. In this work I particularly deal with my part of this project, which lies mainly in the area of computer science. I have considered the following three main points: Establishing a performance measure based on information theory: I have critically illuminated the assumptions of Shannon's information transfer rate for application in a BCI context. By establishing suitable coding strategies I was able to show that this theoretical measure approximates quite well to what is practically achieveable. Transfer and development of suitable signal processing and machine learning techniques: One substantial component of my work was to develop several machine learning and signal processing algorithms to improve the efficiency of a BCI. Based on the neurophysiological knowledge that several independent EEG features can be observed for some mental states, I have developed a method for combining different and maybe independent features which improved performance. In some cases the performance of the combination algorithm outperforms the best single performance by more than 50 \%. Furthermore, I have theoretically and practically addressed via the development of suitable algorithms the question of the optimal number of classes which should be used for a BCI. It transpired that with BCI performances reported so far, three or four different mental states are optimal. For another extension I have combined ideas from signal processing with those of machine learning since a high gain can be achieved if the temporal filtering, i.e., the choice of frequency bands, is automatically adapted to each subject individually. Implementation of the Berlin brain computer interface and realization of suitable experiments: Finally a further substantial component of my work was to realize an online BCI system which includes the developed methods, but is also flexible enough to allow the simple realization of new algorithms and ideas. In the field of chemistry, this translates to practices such as waste prevention, use of renewable feedstocks, and catalysis; concepts based on the principles of green chemistry. Polymers are an important product in the chemical industry and are also in the focus of these changes. In this thesis, more sustainable approaches to make two classes of polymers, polypeptoids and polyesters, are described. Polypeptoids or poly(alkyl-N-glycines) are isomers of polypeptides and are biocompatible, as well as degradable under biologically relevant conditions. In addition to that, they can have interesting properties such as lower critical solution temperature (LCST) behavior. They are usually synthesized by the ring opening polymerization (ROP) of N-carboxy anhydrides (NCAs), which are produced with the use of toxic compounds (e.g. phosgene) and which are highly sensitive to humidity. In order to avoid the direct synthesis and isolation of the NCAs, N-phenoxycarbonyl-protected N-substituted glycines are prepared, which can yield the NCAs in situ. The conditions for the NCA synthesis and its direct polymerization are investigated and optimized for the simplest N-substituted glycine, sarcosine. The use of a tertiary amine in less than stoichiometric amounts compared to the N-phenoxycarbonyl--sarcosine seems to accelerate drastically the NCA formation and does not affect the efficiency of the polymerization. In fact, well defined polysarcosines that comply to the monomer to initiator ratio can be produced by this method. This approach was also applied to other N-substituted glycines. Dihydroxyacetone is a sustainable diol produced from glycerol, and has already been used for the synthesis of polycarbonates. Here, it was used as a comonomer for the synthesis of polyesters. However, the polymerization of dihydroxyacetone presented difficulties, probably due to the insolubility of the macromolecular chains. To circumvent the problem, the dimethyl acetal protected dihydroxyacetone was polymerized with terephthaloyl chloride to yield a soluble polymer. When the carbonyl was recovered after deprotection, the product was insoluble in all solvents, showing that the carbonyl in the main chain hinders the dissolution of the polymers. Therefore, in this study we investigated the role of the precuneus in egocentric spatial updating in a real environment setting in 20 healthy young participants who underwent two conditions in a cross-over design: (a) stimulation, achieved through applying continuous theta-burst stimulation (cTBS) to inhibit the precuneus and (b) sham condition (activated coil turned upside down). In both conditions, participants had to walk back with blindfolded eyes to objects they had previously memorized while walking with open eyes. Simplified trials (without spatial updating) were used as control condition, to make sure the participants were not affected by factors such as walking blindfolded, vestibular or working memory deficits. A significant interaction was found, with participants performing better in the sham condition compared to real stimulation, showing smaller errors both in distance and angle. Building on a consistent dataset and method, we find that for countries with per capita incomes of below USD 15,000 per year (at PPP-adjusted 2011 USD) carbon pricing has, on average, progressive distributional effects. We also develop a novel decomposition technique to show that distributional outcomes are primarily determined by differences among income groups in consumption patterns of energy, rather than of food, goods or services. We argue that an inverse U-shape relationship between energy expenditure shares and income explains why carbon pricing tends to be regressive in countries with relatively higher income. In order to be able to appropriately interpret such sources of palaeoclimatic information, suitable approaches of statistical modelling as well as methods of time series analysis are necessary, which are applicable to short, noisy, and non-stationary uni- and multivariate data sets. Correlations between different climatic proxy data within one or more climatological archives contain significant information about the climatic change on longer time scales. Based on an appropriate statistical decomposition of such multivariate time series, one may estimate dimensions in terms of the number of significant, linear independent components of the considered data set. In the presented work, a corresponding approach is introduced, critically discussed, and extended with respect to the analysis of palaeoclimatic time series. Temporal variations of the resulting measures allow to derive information about climatic changes. For an example of trace element abundances and grain-size distributions obtained near the Cape Roberts (Eastern Antarctica), it is shown that the variability of the dimensions of the investigated data sets clearly correlates with the Oligocene/Miocene transition about 24 million years before present as well as regional deglaciation events. Grain-size distributions in sediments give information about the predominance of different transportation as well as deposition mechanisms. Finite mixture models may be used to approximate the corresponding distribution functions appropriately. In order to give a complete description of the statistical uncertainty of the parameter estimates in such models, the concept of asymptotic uncertainty distributions is introduced. The relationship with the mutual component overlap as well as with the information missing due to grouping and truncation of the measured data is discussed for a particular geological example. An analysis of a sequence of grain-size distributions obtained in Lake Baikal reveals that there are certain problems accompanying the application of finite mixture models, which cause an extended climatological interpretation of the results to fail. As an appropriate alternative, a linear principal component analysis is used to decompose the data set into suitable fractions whose temporal variability correlates well with the variations of the average solar insolation on millenial to multi-millenial time scales. Within this graph theoretical framework, vertices are identified with grid points taken from the data set representing a region on the the Earth's surface, and edges correspond to strong statistical interrelationships between the dynamics on pairs of grid points. The resulting climate networks are neither perfectly regular nor completely random, but display the intriguing and nontrivial characteristics of complexity commonly found in real world networks such as the internet, citation and acquaintance networks, food webs and cortical networks in the mammalian brain. Among other interesting properties, climate networks exhibit the "small-world" effect and possess a broad degree distribution with dominating super-nodes as well as a pronounced community structure. We have performed an extensive and detailed graph theoretical analysis of climate networks on the global topological scale focussing on the flow and centrality measure betweenness which is locally defined at each vertex, but includes global topological information by relying on the distribution of shortest paths between all pairs of vertices in the network. The betweenness centrality field reveals a rich internal structure in complex climate networks constructed from reanalysis and atmosphere-ocean coupled general circulation model (AOGCM) surface air temperature data. Our novel approach uncovers an elaborately woven meta-network of highly localized channels of strong dynamical information flow, that we relate to global surface ocean currents and dub the backbone of the climate network in analogy to the homonymous data highways of the internet. This finding points to a major role of the oceanic surface circulation in coupling and stabilizing the global temperature field in the long term mean (140 years for the model run and 60 years for reanalysis data). Carefully comparing the backbone structures detected in climate networks constructed using linear Pearson correlation and nonlinear mutual information, we argue that the high sensitivity of betweenness with respect to small changes in network structure may allow to detect the footprints of strongly nonlinear physical interactions in the climate system. The results presented in this thesis are thoroughly founded and substantiated using a hierarchy of statistical significance tests on the level of time series and networks, i.e., by tests based on time series surrogates as well as network surrogates. This is particularly relevant when working with real world data. Specifically, we developed new types of network surrogates to include the additional constraints imposed by the spatial embedding of vertices in a climate network. Our methodology is of potential interest for a broad audience within the physics community and various applied fields, because it is universal in the sense of being valid for any spatially extended dynamical system. It can help to understand the localized flow of dynamical information in any such system by combining multivariate time series analysis, a complex network approach and the information flow measure betweenness centrality. Die SULT-vermittelte Bioaktivierung zu einem genotoxischen Schwefels{\"a}ureester wurde f{\"u}r den benzylischen Alkohol 1-Hydroxymethylpyren des Hepatokanzerogens 1-Methylpyren in fr{\"u}heren Arbeiten gezeigt. In der vorliegenden Arbeit wurde {\"u}berpr{\"u}ft, ob die benzylischen Alkohole weiterer alkylierter PAK {\"u}ber Sulfonierung zu genotoxischen Schwefels{\"a}ureestern umgesetzt werden. Hierzu wurde eine Gruppe von 17 Modellsubstanzen ausgew{\"a}hlt, um die Ableitung von Struktur-Aktivit{\"a}ts-Beziehungen zu erm{\"o}glichen. Das genotoxische Potenzial authentischer benzylischer Schwefels{\"a}ureester der Modellsubstanzen wurde zun{\"a}chst in vitro {\"u}ber DNA-Adduktbildung im zellfreien System und Mutagenit{\"a}t im Salmonella-R{\"u}ckmutationstest untersucht. Die Sulfate zeigten große Reaktivit{\"a}tsunterschiede in Abh{\"a}ngigkeit von der Struktur des aromatischen Systems und der Position der Alkylseitenkette, wobei die Endpunkte DNA-Adduktbildung und Mutagenit{\"a}t gut korrelierten. Des Weiteren wurde der Salmonella-Mutagenit{\"a}tstest mit den benzylischen Alkoholen der untersuchten alkylierten PAK und gentechnisch ver{\"a}nderten S. typhimurium-St{\"a}mmen, die SULT-Formen des Menschen heterolog exprimieren, durchgef{\"u}hrt. Bis auf die Alkohole 2- und 4-HMP zeigten alle untersuchten benzylischen Alkohole deutliche mutagene Effekte in einem oder mehreren humane SULT exprimierenden St{\"a}mmen. Die durchgef{\"u}hrten in vitro-Versuche zeigten das Potenzial der benzylischen Metabolite alkylierter PAK f{\"u}r genotoxische Wirkungen. Nachfolgend musste gekl{\"a}rt werden, welche Relevanz die beobachteten Effekte f{\"u}r die komplexere in vivo-Situation haben. Nach Verabreichung verschiedener benzylischer Schwefels{\"a}ureester und Alkohole an m{\"a}nnliche Ratten konnten DNA-Addukte in den untersuchten Organen detektiert werden, was im Fall der Schwefels{\"a}ureester deren systemische Bioverf{\"u}gbarkeit und im Fall der benzylischen Alkohole deren Umsatz durch SULT der m{\"a}nnlichen Ratte zeigte. Da im Gegensatz zum Menschen die SULT-Expression in der Ratte auf die Leber fokussiert ist, musste ein Großteil des Umsatzes zu genotoxischen Sulfaten in der Leber stattgefunden haben. DNA-Addukte wurden jedoch auch in extrahepatischen Organen gefunden, was {\"u}ber einen hepatischen Export der gebildeten reaktiven Sulfate und deren Transport {\"u}ber den Blutkreislauf zu diesen Geweben erkl{\"a}rt werden kann. F{\"u}r die weiterf{\"u}hrenden in vivo-Studien wurden die benzylischen Alkohole 1-HMP und 1-HM-8-MP ausgew{\"a}hlt, die trotz großer struktureller {\"A}hnlichkeit toxikodynamische Unterschiede zeigten. Zur Untersuchung der Bedeutung des SULT-vermittelten Toxifizierungsweges als auch konkurrierender detoxifizierender oxidativer Stoffwechselprozesse, wurden f{\"u}r 1-HMP und 1-HM-8-MP in vivo-Inhibitionsstudien mit SULT-Inhibitoren und f{\"u}r 1-HM-8-MP auch mit ADH/ALDH-Inhibitoren durchgef{\"u}hrt. Eine Vorbehandlung mit dem SULT-Hemmstoff Pentachlorphenol f{\"u}hrte zu einer Reduktion der DNA-Adduktniveaus in Organen 1-HMP- und 1-HM-8-MP-behandelter Tiere. Die Verabreichung von Quercetin hatte keine Auswirkung auf die DNA-Adduktniveaus. Die Hemmung der DNA-Adduktbildung bei Verabreichung von Pentachlorphenol verdeutlichte jedoch, dass benzylische Alkohole alkylierter PAK in vivo {\"u}ber Sulfonierung bioaktiviert werden. Eine Vorbehandlung mit dem ADH-Inhibitor 4-Methylpyrazol und dem ADH-Substrat Ethanol f{\"u}hrte zu erh{\"o}hten DNA-Adduktniveaus in Organen 1-HM-8-MP-behandelter Tiere. Den gleichen Effekt, jedoch in geringerem Ausmaß, hatte auch die Vorbehandlung mit dem ALDH-Inhibitor Disulfiram. Dies deutet darauf hin, dass oxidative Modifikationen an der Seitenkette des 1-HM-8-MP einen Detoxifizierungsmechanismus darstellen. Nach Verabreichung benzylischer Metabolite alkylierter PAK wurden oftmals hohe Adduktniveaus in der Niere detektiert. Moreover, the taphonomy of sedimentary DNA, especially in tropical environments, is poorly understood. This study elucidates challenges and opportunities of sedimentary ancient DNA approaches for reconstructing tropical biodiversity. We present shotgun-sequenced metagenomic profiles and DNA degradation patterns from multiple sediment cores from Mubwindi Swamp, located in Bwindi Impenetrable Forest (Uganda), one of the most diverse forests in Africa. We describe the taxonomic composition of the sediments covering the past 2200 years and compare the sedimentary DNA data with a comprehensive set of environmental and sedimentological parameters to unravel the conditions of DNA degradation. Consistent with the preservation of authentic ancient DNA in tropical swamp sediments, DNA concentration and mean fragment length declined exponentially with age and depth, while terminal deamination increased with age. DNA preservation patterns cannot be explained by any environmental parameter alone, but age seems to be the primary driver of DNA degradation in the swamp. Besides degradation, the presence of living microbial communities in the sediment also affects DNA quantity. Critically, 92.3\% of our metagenomic data of a total 81.8 million unique, merged reads cannot be taxonomically identified due to the absence of genomic references in public databases. Of the remaining 7.7\%, most of the data (93.0\%) derive from Bacteria and Archaea, whereas only 0-5.8\% are from Metazoa and 0-6.9\% from Viridiplantae, in part due to unbalanced taxa representation in the reference data. The plant DNA record at ordinal level agrees well with local pollen data but resolves less diversity. Our animal DNA record reveals the presence of 41 native taxa (16 orders) including Afrotheria, Carnivora, and Ruminantia at Bwindi during the past 2200 years. In principle, the fraction of binary stars at a certain separation range can be estimated based on the number of measured microlensing events. However, a binary system may produce a light curve which can be fitted well as a single lens light curve, in particullary if the data sampling is poor and the errorbars are large. We investigate what fraction of microlensing events produced by binary stars for different separations may be well fitted by and hence misinterpreted as single lens events for various observational conditions. We find that this fraction strongly depends on the separation of the binary components, reaching its minimum at between 0.6 and 1.0 Einstein radius, where it is still of the order of 5\% The Einstein radius is corresponding to few A.U. for typical Galactic microlensing scenarios. The rate for misinterpretation is higher for short microlensing events lasting up to few months and events with smaller maximum amplification. For fixed separation it increases for binaries with more extreme mass ratios. Problem of degeneracy in photometric light curve solution between binary lens and binary source microlensing events was studied on simulated data, and data observed by the PLANET collaboration. The fitting code BISCO using the PIKAIA genetic algorithm optimizing routine was written for optimizing binary-source microlensing light curves observed at different sites, in I, R and V photometric bands. Tests on simulated microlensing light curves show that BISCO is successful in finding the solution to a binary-source event in a very wide parameter space. Flux ratio method is suggested in this work for breaking degeneracy between binary-lens and binary-source photometric light curves. Models show that only a few additional data points in photometric V band, together with a full light curve in I band, will enable breaking the degeneracy. Very good data quality and dense data sampling, combined with accurate binary lens and binary source modeling, yielded the discovery of the lowest-mass planet discovered outside of the Solar System so far, OGLE-2005-BLG-390Lb, having only 5.5 Earth masses. This was the first observed microlensing event in which the degeneracy between a planetary binary-lens and an extreme flux ratio binary-source model has been successfully broken. For events OGLE-2003-BLG-222 and OGLE-2004-BLG-347, the degeneracy was encountered despite of very dense data sampling. From light curve modeling and stellar evolution theory, there was a slight preference to explain OGLE-2003-BLG-222 as a binary source event, and OGLE-2004-BLG-347 as a binary lens event. However, without spectra, this degeneracy cannot be fully broken. No planet was found so far around a white dwarf, though it is believed that Jovian planets should survive the late stages of stellar evolution, and that white dwarfs will retain planetary systems in wide orbits. We want to perform high precision astrometric observations of nearby white dwarfs in wide binary systems with red dwarfs in order to find planets around white dwarfs. We selected a sample of observing targets (WD-RD binary systems, not published yet), which can possibly have planets around the WD component, and modeled synthetic astrometric orbits which can be observed for these targets using existing and future astrometric facilities. Modeling was performed for the astrometric accuracy of 0.01, 0.1, and 1.0 mas, separation between WD and planet of 3 and 5 A.U., binary system separation of 30 A.U., planet masses of 10 Earth masses, 1 and 10 Jupiter masses, WD mass of 0.5M and 1.0 Solar masses, and distances to the system of 10, 20 and 30 pc. It was found that the PRIMA facility at the VLTI will be able to detect planets around white dwarfs once it is operating, by measuring the astrometric wobble of the WD due to a planet companion, down to 1 Jupiter mass.
