This thesis investigates the modification of the microphysical properties of Arctic aerosols in the transition from Arctic haze to the summer season. Therefore, lidar measurements of Ny-{\AA}lesund from April 2021 to the end of July 2021 are evaluated based on the aerosols' optical properties. An overview of those properties will be provided. Furthermore, parallel radiosonde data is considered for indication of hygroscopic growth. The annual aerosol cycle in 2021 differs from expectations based on previous studies from Tunved et al. (2013) and Quinn et al. (2007). Developments of backscatter, extinction, aerosol depolarisation, lidar ratio and color ratio show a return of the Arctic haze in May. The haze had already reduced in April, but regrew afterwards. The average Arctic aerosol displays hygroscopic behaviour, meaning growth due to water uptake. To determine such a behaviour is generally laborious because various meteorological circumstances need to be considered. Two case studies provide further information on these possible events. Im Mittelpunkt steht ein Problem, das bereits in der fr{\"u}hesten f{\"a}cher{\"u}bergreifenden Methodik des modernen Fremdsprachenunterrichts aufgeworfen wurde. (Seidelmann, Christian Friedrich: Tractatus philosophico-philologicus de methodo recte tractandi linguas exoticas speciatim gallicam, italicam et anglicam (1724). Faksimiliert, {\"u}bersetzt und herausgegeben von Franz Josef Zapp und Konrad Schr{\"o}der, Augsburg 1984.) Seidelmann hat schon vor mehr als 250 Jahren darauf hingewiesen, daß f{\"u}r den Fremdsprachenunterricht ungeeignete Grammatiken zur Verf{\"u}gung stehen. Sie weisen einerseits L{\"u}cken auf und enthalten andererseits unn{\"o}tigen Ballast: 'Ne quisquam sibi persuadeat, Grammaticas, maximam partem hodiernas, esse compendia, in quibus Linguarum fundamenta sive prima principio contineantur .... ' ('Niemand darf der Meinung sein, daß die Grammatiken, und erst recht die derzeitigen, Zusammenfassungen sind, die die Grundlagen oder die wichtigsten Elemente der Sprachen enthalten.') Seidelmann 1724; zitiert nach der zweisprachigen Ausgabe von Zapp und Schr{\"o}der, Augsburg 1984, S. 81. Vor bald 100 Jahren betonte Vi{\"e}tor im Zusammenhang mit der Diskussion um die '{\"U}berb{\"u}rdung' der Sch{\"u}ler die Notwendigkeit, die Verbalflexion gewissermaßen 'auszumisten'. Wie meine Arbeit u.a. zeigt, reichen ganze 42 Verbparadigmen aus, um das Lernziel der kommunikativen Kompetenz zu realisieren. Das heißt, man kann auf {\"u}ber die H{\"a}lfte der bis zu 90 Verbalparadigmen, die in Lehrwerken f{\"u}r den Franz{\"o}sischunterricht vermittelt werden, verzichten. Die 42 Konjugationsmuster k{\"o}nnen als Teil eines nach koh{\"a}renten Prinzipien konstituierten und in diesem Sinne genormten sprachlichen Grundstocks f{\"u}r unterschiedliche Lehrgangstypen betrachtet werden. Der didaktische Nutzen der neuen Klassifikation l{\"a}ßt sich durch einen Vergleich mit der bisher in den Grammatischen Beiheften {\"u}blichen Einteilung belegen: 1) Die Lehrinhalte der Verbalflexion lassen sich reduzieren, wenn Verben mit identischen bzw. weitgehend identischen Flexionsmerkmalen einer Klasse zugeschlagen werden, 2) Meine Untersuchung hat ergeben, dass die Sch{\"u}ler nicht immer unterst{\"u}tzt werden, Neues mit bereits Bekanntem zu verkn{\"u}pfen und einen Zusammenhang zwischen Verben mit identischen Flexionsmerkmalen herzustellen. Von jeter wird nicht immer auf das bereits eingef{\"u}hrte Verb appeler verwiesen. Auch die Gemeinsamkeiten der Verben der Klassen atteindre, sentir und offrir werden im lektionsbegleitenden Teil bestimmter Grammatischer Beihefte dem Sch{\"u}ler nicht einsichtig gemacht und damit nicht als Lernhilfe genutzt. 3) Verben mit identischen Endungen im Infinitiv, aber ansonsten {\"u}berwiegend unterschiedlichen Flexionsmerkmalen, sollten verschiedenen Klassen zugeschlagen werden, um irrt{\"u}mliche Verallgemeinerungen zu vermeiden. F{\"u}r Verben der Klassen {\´e}lever und consid{\´e}rer wird vereinzelt im Gegensatz zu meiner Empfehlung nur eine Klasse angesetzt. Die Verben der Klasse employer und envoyer werden nicht immer deutlich genug von den Verben auf -AYER abgegrenzt. Die Pr{\"u}fungsaufgaben einer Zertifizierungs-organisation f{\"u}r das Englische konzentrieren sich auf rezeptive Kenntnisse und Fertigkeiten, bieten aber den in den Fremdsprachenkursen ausgebildeten produktiven Fertigkeiten wenig Raum. Sie untersucht, wie gesellschaftliches Wissen zur Impfung in meinungsf{\"u}hrenden Tages- und Wochenzeitungen sowie in ausgew{\"a}hlten Blogs w{\"a}hrend der Pandemie von Anfang 2020 bis Mitte 2022 (re)produziert wird. Ausgangspunkt sind - ankn{\"u}pfend an aktuelle Beitr{\"a}ge der kritischen Soziologie - die politisch-{\"o}konomischen Zusammenh{\"a}nge und Voraussetzungen, wie beispielsweise globale Lebens-, Wirtschafts- und Konsumweisen, durch die diese Pandemie erst m{\"o}glich wurde. Durch diese Perspektive kann die Pandemie als soziales Ereignis begriffen werden, anstatt als unvorhersehbare medizinische Katastrophe, wie es in den politischen und medialen Debatten den Anschein hatte. In der Analyse zeigt sich, dass diese Umst{\"a}nde in der medialen Auseinandersetzung keinerlei Widerhall fanden, was den Diskurs de-kontextualisiert und den herausgearbeiteten Radikalismen den Boden bereitet hat. Die Analyse erm{\"o}glicht es, zwei Zug{\"a}nge zum Diskurs zu unterscheiden: Ein Portal er{\"o}ffnet den Zugang {\"u}ber gesellschaftlich mehrheitlich anerkanntes, also orthodoxes Wissen, das zweite {\"u}ber gesellschaftlich mehrheitlich nicht anerkanntes, also heterodoxes Wissen. Entlang von benannten Themen, Problemen und L{\"o}sungen lassen sich auf einer Pro-Kontra-Achse sieben Wissenstypen rekonstruieren, die von radikaler Impfbef{\"u}rwortung bis radikaler Ablehnung reichen. Vier der Wissensbest{\"a}nde argumentieren f{\"u}r die Impfung, drei dagegen - der Raum des Indifferenten dazwischen bleibt medial unbesetzt. Anschließend werden diese Typen entlang von Werten, auf die rekurriert wird, und schließlich nach Ressourcen, Verantwortlichkeiten und Sprecher*innenrollen unterschieden, sodass eine klare und idealtypische Charakterisierung des jeweiligen Wissens entsteht. Durch die Analyse werden {\"u}berdies diskursive Verschiebungen im Zeitverlauf sichtbar. Es zeigen sich einerseits eine Polarisierung des Diskurses insgesamt, andererseits eine Ern{\"u}chterung innerhalb der Impfbef{\"u}rwortung sowie die interdependente Radikalisierung von Positionen an beiden Enden des Spektrums. Abschließend werden die Ergebnisse der Analyse komplement{\"a}r zum Ausgangspunkt gesellschaftstheoretisch eingebettet. Zum einen werden Eigendynamiken de-kontextualisierter und sich infolgedessen radikalisierender Diskurse reflektiert, innerhalb derer sich die Extreme so weit voneinander entfernen, dass sie sich schlussendlich wieder ber{\"u}hren. DNA methylation is a heritable variation in gene expression without changing the underlying genetic information. An understanding of this epigenetic mechanism in the context of non-native, predominantly sexually reproducing plant species might help to expand knowledge in biodiversity research on the interaction between plants and their habitats and, based on this, may enable more precise measures in conservation biology. For my studies, I combined chemical DNA demethylation of field-collected seed material from predominantly sexually reproducing species and rearing offsping under common climatic conditions to examine DNA methylation in an ecological-evolutionary context. The contrast of chemically treated (demethylated) plants, whose variation in DNA methylation was artificially reduced, and untreated control plants of the same species allowed me to study the impact of this mechanism on adaptive trait differentiation and local adaptation. With this experimental background, I conducted three studies examining the effect of DNA methylation in non-native species along a climatic gradient and also between climatically divergent regions. The first study focused on adaptive trait differentiation in two invasive perennial goldenrod species, Solidago canadensis sensu latu and S. gigantea AITON, along a climate gradient of more than 1000 km in length in Central Europe. I found population differences in flowering timing, plant height, and biomass in the temporally longer-established S. canadensis, but only in the number of regrowing shoots for S. gigantea. While S. canadensis did not show any population structure, I was able to identify three genetic groups along this climatic gradient in S. gigantea. Surprisingly, demethylated plants of both species showed no change in the majority of traits studied. In the subsequent second study, I focused on the longer-established goldenrod species S. canadensis and used molecular analyses to infer spatial epigenetic and genetic population differences in the same specimens from the previous study. I found weak genetic but no epigenetic spatial variation between populations. Additionally, I was able to identify one genetic marker and one epigenetic marker putatively susceptible to selection. However, the results of this study reconfirmed that the epigenetic mechanism of DNA methylation appears to be hardly involved in adaptive processes within the new range in S. canadensis. Finally, I conducted a third study in which I reciprocally transplanted short-lived plant species between two climatically divergent regions in Germany to investigate local adaptation at the plant family level. For this purpose, I used four plant families (Amaranthaceae, Asteraceae, Plantaginaceae, Solanaceae) and here I additionally compared between non-native and native plant species. Seeds were transplanted to regions with a distance of more than 600 kilometers and had either a temperate-oceanic or a temperate-continental climate. In this study, some species were found to be maladapted to their own local conditions, both in non-native and native plant species alike. In demethylated individuals of the plant species studied, DNA methylation had inconsistent but species-specific effects on survival and biomass production. The results of this study highlight that DNA methylation did not make a substantial contribution to local adaptation in the non-native as well as native species studied. In summary, my work showed that DNA methylation plays a negligible role in both adaptive trait variation along climatic gradients and local adaptation in non-native plant species that either exhibit a high degree of genetic variation or rely mainly on sexual reproduction with low clonal propagation. I was able to show that the adaptive success of these non-native plant species can hardly be explained by DNA methylation, but could be a possible consequence of multiple introductions, dispersal corridors and meta-population dynamics. However, some territorially breeding animals can have dramatic density fluctuations and little is known about the change from density-dependent processes to density-independence of breeding during a population increase or an outbreak. We suggest that territoriality, breeding suppression and its break-down can be understood with an incomplete-control model, developed for social breeders and social suppression. Results We studied density dependence in an arvicoline species, the bank vole, known as a territorial breeder with cyclic and non-cyclic density fluctuations and periodically high densities in different parts of its range. Our long-term data base from 38 experimental populations in large enclosures in boreal grassland confirms that breeding rates are density-regulated at moderate densities, probably by social suppression of subordinate potential breeders. We conducted an experiment, were we doubled and tripled this moderate density under otherwise the same conditions and measured space use, mortality, reproduction and faecal stress hormone levels (FGM) of adult females. We found that mortality did not differ among the densities, but the regulation of the breeding rate broke down: at double and triple densities all females were breeding, while at the low density the breeding rate was regulated as observed before. Spatial overlap among females increased with density, while a minimum territory size was maintained. Mean stress hormone levels were higher in double and triple densities than at moderate density. Conclusions At low and moderate densities, breeding suppression by the dominant breeders, But above a density-threshold (similar to a competition point), the dominance of breeders could not be sustained (incomplete control). In our experiment, this point was reached after territories could not shrink any further, while the number of intruders continued to increase with increasing density. Probably suppression becomes too costly for the dominants, and increasing number of other breeders reduces the effectiveness of threats. Risk-taking behavior might mediate the underlying life history trade-off, but empirical evidence supporting this hypothesis is still ambiguous. Using experimentally created populations of common voles (Microtus arvalis)—a species with distinct seasonal life history trajectories—we aimed to test whether individual differences in boldness behavior covary with risk taking, space use, and fitness. We quantified risk taking, space use (via automated tracking), survival, and reproductive success (via genetic parentage analysis) in 8 to 14 experimental, mixed-sex populations of 113 common voles of known boldness type in large grassland enclosures over a significant part of their adult life span and two reproductive events. Populations were assorted to contain extreme boldness types (bold or shy) of both sexes. Bolder individuals took more risks than shyer ones, which did not affect survival. Bolder males but not females produced more offspring than shy conspecifics. Daily home range and core area sizes, based on 95\% and 50\% Kernel density estimates (20 ± 10 per individual, n = 54 individuals), were highly repeatable over time. Individual space use unfolded differently for sex-boldness type combinations over the course of the experiment. While day ranges decreased for shy females, they increased for bold females and all males. Therefore, we expected that in winter and spring samples, when only OW can be sampled, the animals should be shyer than in summer and autumn, when both OW and YY animals can be sampled. We studied common vole (Microtus arvalis) populations, which express typical, intra-annual density fluctuation. We captured a total of 492 voles at different months over 3 years and examined boldness and activity level with two standardised behavioural experiments. Results: Behavioural variables of the two tests were correlated with each other. Boldness, measured as short latencies in both tests, was extremely high in spring compared to other seasons. Activity level was highest in spring and summer, and higher in males than in females. Conclusion: Being bold in laboratory tests may translate into higher risk-taking in nature by being more mobile while seeking out partners or valuable territories. Possible explanations include asset-protection, with OW animals being rather old with low residual reproductive value in spring. Therefore, OW may take higher risks during this season. Offspring born in spring encounter a lower population density and may have higher reproductive value than offspring of later cohorts. A constant connection between life history and animal personality, as suggested by the POL theory, however, was not found. Nevertheless, correlations of traits suggest the existence of animal personalities. We further compared vole behaviour using staged dyadic encounters in neutral arenas in both seasons. Survival of the non-breeding overwintering bank voles was not affected by competition. In the spring, the numbers of male bank voles, but not of females, were reduced significantly in the competition populations. Bank vole home ranges expanded with vole density in the presence of competitors, indicating food limitation. A comparison of behaviour between seasons based on an analysis of similarity revealed an avoidance of costly aggression against opponents, independent of species. Interactions were more aggressive during the summer than during the winter, and heterospecific encounters were more aggressive than conspecific encounters. Based on these results, we suggest that interaction types and their respective mechanisms are not either-or categories and may change over the seasons. During the winter, energy constraints and thermoregulatory needs decrease direct aggression, but food constraints increase indirect resource competition. Direct interference appears in the summer, probably triggered by each individual's reproductive and hormonal state and the defence of offspring against conspecific and heterospecific intruders. Taken together, our results suggest that the Bruce effect may be an adaptive breeding strategy for rodent females in cyclic populations specifically at low densities in the increase phase, when isolated, overwintered animals associate in MF pairs. During population lows infanticide risk and inbreeding risk may then be higher than during population highs, while also the fitness value of a litter in an increasing population is higher. In this paper, we present Flux-P as an approach to automate and standardize 13C-based metabolic flux analysis, using the Bio-jETI workflow framework. Exemplarily based on the FiatFlux software, it demonstrates how services can be created that carry out the different analysis steps autonomously and how these can subsequently be assembled into software workflows that perform automated, high-throughput intracellular flux analysis of high quality and reproducibility. We investigate whether the functional description reflects the typical environments of the corresponding organisms by dividing all species into disjoint groups based on whether they are aerotolerant and/or photosynthetic. Despite differences in the underlying concepts, both measures display some common features. Closely related organisms often display a similar functional behavior and in both cases the functional measures appear to correlate with the considered classes of environments. Carbon utilization spectra and nutrient profiles are complementary approaches toward a functional classification of organism-wide metabolic networks. Both approaches contain different information and thus yield different clusterings, which are both different from the classical taxonomy of organisms. The results indicate that, although there is a general length dependence of strand break cross sections, individual nucleotides do not contribute independently of the absolute strand break cross section of the whole DNA strand. The irradiation is made in the most likely energy range of 5 to 20 eV with an anionic resonance maximum around 10 eV independently of the DNA sequence. There is a clear difference between σSSB and σDSB of DNA single and double strands, where the strand break for ssDNA are always higher in all electron energies compared to dsDNA by the factor 3. A further part of this work deals with the characterization and analysis of new types of radiosensitizers used in chemoradiotherapy, which selectively increases the DNA damage upon radiation. Fluorinated DNA sequences with 2'-fluoro-2'-deoxycytidine (dFC) show an increased sensitivity at 7 and 10 eV compared to the unmodified DNA sequences by an enhancement factor between 2.1 and 2.5. However, due to the scarcity of hydrometeorological data, hydrological modeling for mountain regions in Central Asia involves large uncertainties. Addressing this problem, the first aim of this thesis was to develop hydrological modeling approaches that can increase the credibility of hydrological models in data sparse mountain regions. This was achieved by using additional data from remote sensing and atmospheric modeling. It was investigated whether spatial patterns from downscaled reanalysis data can be used for the interpolation of station-based precipitation data. This approach was compared to other precipitation estimates using a hydrologic evaluation based on hydrological modeling and a comparison of simulated and observed discharge, which demonstrated a generally good performance of this method. The study further investigated the value of satellite-derived snow cover data for model calibration. Trade-offs of good model performance in terms of discharge and snow cover were explicitly evaluated using a multiobjective optimization algorithm, and the results were contrasted with single-objective calibration and Monte Carlo simulations. The study clearly shows that the additional use of snow cover data improved the internal consistency of the hydrological model. In this context, it was further investigated for the first time how many snow cover scenes were required for hydrological model calibration. The second aim of this thesis was the application of the hydrological model in order to investigate the causes of observed streamflow increases in two headwater catchments of the Tarim River over the recent decades. This simulation-based approach for trend attribution was complemented by a data-based approach. The hydrological model was calibrated to discharge and glacier mass balance data and considered changes in glacier geometry over time. To account for the effects of sedimentation rate changes on the spectral expression of the orbital cycles we developed a new method: the Multi-band Wavelet Age modeling technique (MUBAWA). By using a Continuous Wavelet Transformation, we were able to track frequency shifts that resulted from changing sedimentation rates and thus calculated tuned age model encompassing the last 620 kyrs. The results show a good agreement with the directly dated age model that is available from the dating of volcanic ashes. Then we used the XRF data from CHB and developed a new and robust humid-arid index of east African climate during the last 620 kyrs. To disentangle the relationship of the selected elements we performed a principal component analysis (PCA). In a following step we applied a continuous wavelet transformation on the PC1, using the directly dated age model. The resulting wavelet power spectrum, unlike a normal power spectrum, displays the occurrence of cycles/frequencies in time. The results highlight that the precession cycles are most dominantly expressed under the 400 kyrs eccentricity maximum whereas weakly expressed during eccentricity minimum. This suggests that insolation is a key driver of the climatic variability observed at CHB throughout the last 620 kyrs. In addition, the prevalence of half-precession and obliquity signals was documented. The latter is attributed to the inter-tropical insolation gradient and not interpreted as an imprint of high latitudes forcing on climatic changes in the tropics. In addition, a windowed analysis of variability was used to detect changes in variance over time and showed that strong climate variability occurred especially along the transition from a dominant insolation-controlled humid climate background state towards a predominantly dry and less-insolation controlled climate. The last chapter dealt with non-linear aspects of climate changes represented by the sediments of the CHB. We use recurrence quantification analysis to detect non-linear changes within the potassium concentration of Chew Bahir sediment cores during the last 620 kyrs. The concentration of potassium in the sediments of the lake is subject to geochemical processes related to the evaporation rate of the lake water at the time of deposition. Based on recurrence analysis, two types of variabilities could be distinguished. Type 1 represents slow variations within the precession period bandwidth of 20 kyrs and a tendency towards extreme climatic events whereas type 2 represents fast, highly variable climatic transitions between wet and dry climate states. While type 1 variability is linked to eccentricity maxima, type 2 variability occurs during the 400 kyrs eccentricity minimum. The climate history presented here shows that during high eccentricity a strongly insolation-driven climate system prevailed, whereas during low eccentricity the climate was more strongly affected by short-term variability changes. The short-term environmental changes, reflected in the increased variability might have influenced the evolution, technological advances and expansion of early modern humans who lived in this region. In the Olorgesaille Basin the temporal changes in the occurrence of stone tools, which bracket the transition from Acheulean to Middle Stone Age (MSA) technologies at between 499-320 kyrs, could potentially correlate to the marked transition from a rather stable climate with less variability to a climate with increased variability in the CHB. Developers derive a set of test cases, and execute them frequently to ensure that their change did not adversely affect existing functionality. As the software product and its test suite grow, the time to feedback during regression test sessions increases, and impedes programmer productivity: developers wait longer for tests to complete, and delays in fault detection render fault removal increasingly difficult. Test case prioritization addresses the problem of long feedback loops by reordering test cases, such that test cases of high failure probability run first, and test case failures become actionable early in the testing process. We ask, given test execution schedules reconstructed from publicly available data, to which extent can their fault detection efficiency improved, and which technique yields the most efficient test schedules with respect to APFD? To this end, we recover regression 6200 test sessions from the build log files of Travis CI, a popular continuous integration service, and gather 62000 accompanying changelists. We evaluate the efficiency of current test schedules, and examine the prioritization results of state-of-the-art lightweight, history-based heuristics. We propose and evaluate a novel set of prioritization algorithms, which connect software changes and test failures in a matrix-like data structure. Our studies indicate that the optimization potential is substantial, because the existing test plans score only 30\% APFD. The predictive power of past test failures proves to be outstanding: simple heuristics, such as repeating tests with failures in recent sessions, result in efficiency scores of 95\% APFD. The best-performing matrix-based heuristic achieves a similar score of 92.5\% APFD. In contrast to prior approaches, we argue that matrix-based techniques are useful beyond the scope of effective prioritization, and enable a number of use cases involving software maintenance. We validate our findings from continuous integration processes by extending a continuous testing tool within development environments with means of test prioritization, and pose further research questions.
