The studied samples are characterised by different chemical compositions (low and high calcium content), variable firing temperatures and different amounts of soluble salts as damage products. In the low-Ca samples, the decrease of the phyllosilicate content (from 23.4 to 6.9 wt\%) is associated to the increase of the amorphous fraction (from 24 to 48\%). This clear negative correlation between the phyllosilicate content and the amorphous fraction indicates that in low-Ca systems vitrification processes are overwhelming with respect to nucleation and recrystallisation processes. By contrast, high-Ca samples present newly formed Ca(Mg) silicates (diopside from 5.7 to 27.2\%; anorthite from 1.4 to 8.7\%) and aluminium silicates (gehlenite only in two samples, 6.2 and 7.7\%) associated to the decrease of quartz (from 27.7 to 11.5\%), phyllosilicate (from 6.5\% until complete break down) and amorphous (from 30 to 14\%) phase fractions. These findings support the role played by the CaO(MgO) content deriving from carbonates decomposition which reacts with Al2O3 and SiO2 oxides from dehydroxylated clay minerals and quartz grains. The above results have been obtained by X-ray powder diffraction data using the combined Rietveld refinement - internal standard method in order to estimate both the crystalline and the amorphous phase fractions. In addition, the coexistence of two distinct plagioclases in high-Ca samples was modelled as follows: a primary albite, which tends to incorporate Ca during the firing process as demonstrated by the increasing of gamma crystallographic angle, and a newly formed anorthite. Finally, by difference between the X-ray fluorescence data and the chemical compositions inferred by QPA, it proved possible to roughly estimate the residual chemical composition attributable to the amorphous fraction. The functional significance of a putative ARE in the GI-GPx promoter was validated by transcriptional activation of reporter gene constructs upon exposure to electrophiles (tBHQ, SFN, and curcumin) or overexpression of Nrf2 and by reversal of these effects by mutation of the ARE in the promoter and by overexpressed Keap1. Binding of Nrf2 to the ARE sequence in authentic gpx2 was corroborated by chromatin immunoprecipitation. The recent completion of several algal genome sequences and expressed sequence tag collections and the establishment of nuclear and organellar transformation methods has opened the way for functional genomics approaches using algal model systems. The thermo-acidophilic unicellular red alga Galdieria sulphuraria represents a particularly interesting species for a genomics approach owing to its extraordinary metabolic versatility such as heterotrophic and mixotrophic growth on more than 50 different carbon sources and its adaptation to hot acidic environments. However, the ab initio prediction of genes required for unknown metabolic pathways from genome sequences is not trivial. A compelling strategy for gene identification is the comparison of similarly sized genomes of related organisms with different physiologies. Using this approach, candidate genes were identified that are critical to the metabolic versatility of Galdieria. Expressed sequence tags and high-throughput genomic sequence reads covering >70\% of the G. sulphuraria genome were compared to the genome of the unicellular, obligate photoautotrophic red alga Cyanidioschyzon merolae. The sample is composed of all GEMS galaxies with Sersic indices n < 2.5, derived from fits to the galaxy images. We account fully for selection effects through careful analysis of image simulations; we are limited by the depth of the redshift and HST data to the study of galaxies with M-V less than or similar to -20, or equivalently, log (M/M-circle dot) greater than or similar to 10. We find strong evolution in the magnitude-size scaling relation for galaxies with M-V less than or similar to -20, corresponding to a brightening of similar to 1 mag arcsec(-2) in rest-frame V band by z similar to 1. Yet disks at a given absolute magnitude are bluer and have lower stellar mass-to-light ratios at z similar to 1 than at the present day. As a result, our findings indicate weak or no evolution in the relation between stellar mass and effective disk size for galaxies with log (M/M-circle dot) greater than or similar to 10 over the same time interval. This is strongly inconsistent with the most naive theoretical expectation, in which disk size scales in proportion to the halo virial radius, which would predict that disks are a factor of 2 denser at fixed mass at z similar to 1. The lack of evolution in the stellar mass-size relation is consistent with an "inside-out'' growth of galaxy disks on average (galaxies increasing in size as they grow more massive), although we cannot rule out more complex evolutionary scenarios}, language = {en} } @phdthesis{Bari2005, author = {Bari, Rajendra P.}, title = {Molecular characterization of phosphate signaling in arabidopsis thaliana}, address = {Potsdam}, pages = {X, 141 S. : graph. In plants, this channel has been shown by green fluorescent protein fusion to localize to the vacuolar membrane, which led to speculations that the TPK1 gene product would be a component of the nonselective, Ca2+ and voltage- dependent slow-vacuolar (SV) cation channel found in many plants species. Using yeast ( Saccharomyces cerevisiae) as an expression system for TPK1, we show functional expression of the channel in the vacuolar membrane. In isolated vacuoles of yeast yvc1 disruption mutants, the TPK1 gene product shows ion channel activity with some characteristics very similar to the SV-type channel. The open channel conductance of TPK1 in symmetrically 100mM KCl is slightly asymmetric with roughly 40 pS at positive membrane voltages and 75 pS at negative voltages. Similar to the SV-type channel, TPK1 is activated by cytosolic Ca2+, requiring micromolar concentration for activation. However, in contrast to the SV- type channel, TPK1 exhibits strong selectivity for K+ over Na+, and its activity turned out to be independent of the membrane voltage over the range of +/- 80mV. These substances regulate and modulate many physiological and behavioral processes. Various inactivation mechanisms for these substances exist to terminate biogenic amine-mediated signal transduction. In vertebrates, the enzymes monoamine oxidase and/or catechol-O-methyl-transferase are involved in these processes. In insects, however, in which both enzymes are low in abundance or absent, biogenic amines are inactivated mainly by N- acetylation or O-sulphation. In Droso-philo, beta-alanyl conjugation mediated by the Ebony protein has recently been shown to be a novel and alternative pathway for biogenic amine inactivation. Here, we report the cloning of ebony cDNA (Peaebony) from a brain-specific cDNA library of the cockroach Periplaneta americana. The open reading frame encodes a protein of 860 amino acid residues (PeaEbony). The PeaEbony polypeptide shares homology to Ebony sequences from Anopheles gambiae, Apis mellifera, and Drosophila melonogaster. In addition, PeaEbony exhibits sequence similarity to a family of microbial non-ribosomal peptide synthetases. The mRNA encoding PeaEbony is highly expressed in the cockroach brain and to a lesser extent in the salivary glands. PeaEbony is, therefore, probably involved in the inactivation of various biogenic amines through beta-alanyl conjugation in the cockroach CNS. Since the salivary glands in Periplaneta are innervated by dopaminergic and serotonergic neurons, PeaEbony probably also biochemically modifies dopamine and serotonin in these acinar glands. Arch. Geologic data imply that this climatic pattern has been established or significantly enhanced during Miocene surface uplift of this Andean segment. We report evidence for important climatic and ecologic changes in the eastern foreland of the Patagonian Andes that appear to be the result of this uplift. To provide constraints on Miocene plant ecosystems and precipitation in the eastern (leeward) foreland of the Patagonian Andes, we determined carbon and oxygen isotope values of pedogenic carbonate nodules from a similar to500 m thick section of the continental Santa Cruz Formation. The age of these deposits was constrained by Ar/Ar dating of intercalated tuffs, which range from similar to22 to 14 Ma. At similar to16.5 Ma, the delta(13)C values increase by similar to3parts per thousand, the delta(18)O values decrease by >2parts per thousand, and the scatter in the oxygen isotope data increases significantly. We interpret these changes as the consequence of >1 km surface uplift in this Andean segment (from the delta(18)O values), and increased aridity to its east (from the delta(13)C values and the increased scatter in the delta(18)O values). Sediments overlying the Santa Cruz Formation are very limited in extent and volume, and dominated by coarse conglomerates related to Pleistocene and older glaciations. It thus seems that, by similar to14 Ma, deposition in the eastern foreland of the Southern Patagonian Andes had essentially ceased as the result of rain shadow formation. (C) 2004 Elsevier B.V. In this paper, an attempt was made to determine the CSC with both batch and column experiments. Two types of sediments were tested: (a) homogeneous quartz sand and (b) mineralogically heterogeneous sediment, taken from the Hanford formation in southeast Washington. Stepwise decreasing concentrations of NaNO3 solution were applied until fine particles were released from the sediments and the CSC was determined. Two methods were used to minimize the interference of particle release due to physical forces (shear stress) in the batch experiments: (a) postexperimental correction for mechanical effects, and (b) minimization of shear stress on the sediments during the experiment. CSCs from batch experiments were compared to those obtained from column experiments. It was found that both the amount of particles released and the CSC were an order of magnitude higher for the Hanford sediment than for the Sand. Moreover, particle detachment above the CSC was observed for the Hanford sediment. This suggests that the concept of sharp CSCs could be problematic in natural heterogeneous sediments where fine particles may mobilize at salt concentrations significantly above the CSC, thus unexpectedly enhancing colloid-facilitated transport of contaminants.
