We propose a new methodology based on time series of synthetic aperture radar (SAR) interferometric coherence images to map sediment transport regimes across arid and semiarid landscapes. Using 4 years of Sentinel-1 data, we analyze sediment transport regimes for the south-central Andes in northwestern Argentina characterized by steep topographic and climatic gradients. We observe seasonally low coherence during the regional wet season, particularly on hillslopes and in alluvial channels. The spatial distribution of coherence is compared to drainage areas extracted from digital topography to identify two distinct transitions within watersheds: (a) a hillslope-to-fluvial and (b) a fluvial-to-alluvial transition. While transitions within a given basin can be well-constrained, the relative role of each sediment transport domain varies widely over the climatic and topographic gradients. In semiarid regions, we observe larger relative contributions from hillslopes compared to arid regions. Across regional gradients, the range of coherence within basins positively correlates to previously published millennial catchment-wide erosion rates and to topographic metrics used to indicate long-term uplift. We propose an alternative, fault-agnostic approach based on the conservation of energy: the Energy-Conserving Seismicity Framework (ENCOS). Working in energy space has the advantage that the radiated energy is a better predictor of the damage potential of earthquake waves than the seismic moment release. In a region, ENCOS balances the stationary power available to cause earthquakes with the long-term seismic energy release represented by the energy-frequency distribution's first moment. Accumulation and release are connected through the average seismic efficiency, by which we mean the fraction of released energy that is converted into seismic waves. Besides measuring earthquakes in energy, ENCOS differs from moment balance essentially in that the energy accumulation rate depends on the total stress in addition to the strain rate tensor. To validate ENCOS, we exemplarily model the energy-frequency distribution around Southern California. We estimate the energy accumulation rate due to tectonic loading assuming poroelasticity and hydrostasis. Using data from the World Stress Map and assuming the frictional limit to estimate the stress tensor, we obtain a power of 0.8 GW. The uncertainty range, 0.3-2.0GW, originates mainly from the thickness of the seismogenic crust, the friction coefficient on preexisting faults, and models of Global Positioning System (GPS) derived strain rates. Furthermore, students without SEN preferred peers without SEN as a friend. This homophily-effect was shown for students with SEN, too. However, students with and without SEN rated the quality of their friendships similarly and no interactions between the SEN status of oneself or of the friend was found for the quality of the friendship. While these findings provide evidence for the pre-activation proposal, recent replication attempts together with inconsistencies in the results from the literature cast doubt on the robustness of this phenomenon. Our study presents the first attempt to use the effect of gender on predictability in German to study the pre-activation hypothesis, capitalizing on the fact that all German nouns have a gender and that their preceding determiners can show an unambiguous gender marking when the noun phrase has accusative case. Despite having a relatively large sample size (of 120 subjects), both our preregistered and exploratory analyses failed to yield conclusive evidence for or against an effect of pre-activation. The sign of the effect is, however, in the expected direction: the more unexpected the gender of the determiner, the larger the negativity. The recent, inconclusive replication attempts by Nieuwland et al. (2018) and others also show effects with signs in the expected direction. We conducted a Bayesian random-ef-fects meta-analysis using our data and the publicly available data from these recent replication attempts. Previous model studies and coarse resolution (0.25° × 0.25°, ∼25 km × 25 km) passive microwave assessments of trends in the volume and timing of snowfall, snowmelt, and glacier melt in HMA have identified key spatial and seasonal heterogeneities in the response of snow to changes in regional climate. Here we use recently developed, continuous, internally consistent, and high-resolution passive microwave data (3.125 km × 3.125 km, 1987-2016) from the special sensor microwave imager instrument family to refine and extend previous estimates of changes in the snow regime of HMA. We find an overall decline in snow volume across HMA; however, there exist spatially contiguous regions of increasing snow volume—particularly during the winter season in the Pamir, Karakoram, Hindu Kush, and Kunlun Shan. Detailed analysis of changes in snow-volume trends through time reveal a large step change from negative trends during the period 1987-1997, to much more positive trends across large regions of HMA during the periods 1997-2007 and 2007-2016. We also find that changes in high percentile monthly snow-water volume exhibit steeper trends than changes in low percentile snow-water volume, which suggests a reduction in the frequency of high snow-water volumes in much of HMA. After each element (except for the result), a fixation dot moved equally often to either the left or right side and participants pressed left or right buttons to discriminate its movement direction (four times per trial). First and second operand magnitude (small/large), operation (addition/subtraction), result correctness (right/wrong) and movement direction (left/right) were balanced across 128 trials. Manual reaction times of dot movement discriminations were considered in relation to previous arithmetic elements. We found no evidence of early attentional shifts after first operand and operator presentation. Discrimination performance was modulated consistent with SNAs after the second operand, suggesting that attentional shifts occur once there is access to all elements necessary to complete an arithmetic operation. This investigation is conducted by deploying an integrated modeling workflow that consists of a mesoscale hydrologic model (mHM) and a fully distributed groundwater model, OpenGeoSys (OGS). mHM is forced with climate simulations of five general circulation models under three representative concentration pathways. The diffuse recharges estimated by mHM are used as boundary forcings to the OGS groundwater model to compute changes in groundwater levels and travel time distributions. Simulation results indicate that groundwater recharges and levels are expected to increase slightly under future climate scenarios. Meanwhile, the mean travel time is expected to decrease compared to the historical average. However, the ensemble simulations do not all agree on the sign of relative change. Changes in mean travel time exhibit a larger variability than those in groundwater levels. The ensemble simulations do not show a systematic relationship between the projected change (in both groundwater levels and travel times) and the warming level, but they indicate an increased variability in projected changes with adjusting the enhanced warming level from 1.5 to 3 ∘C. The effect typically appears in monoculture production sites and leads to production decreases of up to 50\%, even though the cultivation practice remains the same. However, an indication of replant disease is challenged by the lack of specification of the particular microbial group responsible for ARD. The objective of this study was to establish an algorithm for estimating growth suppression in orchards irrespective of the unknowns in the complex causal relationship by assessing plant-soil interaction in the orchard several years after planting. Based on a comparison between no-replant and replant soils, the Alternaria group (Ag) was identified as a soil-fungal population responding to replant with abundance. The trunk cross-sectional area (CSA) was found to be a practical and robust parameter representing below-ground and above-ground tree performance. Suppression of tree vigour was therefore calculated by dividing the two inversely related parameters, Q = ln(Ag)/CSA, as a function of soil-fungal proportions and plant responses at the single-tree level. On this basis, five clusters of tree vigour suppression (Q) were defined: (1) no tree vigour suppression/vital (0\%), (2) escalating (- 38\%), (3) strong (- 53\%), (4) very strong (- 62\%), and (5) critical (- 74\%). By calculating Q at the level of the single tree, trees were clustered according to tree vigour suppression. The weighted frequency of clusters in the field allowed replant impact to be quantified at field level. Applied to a case study on sandy brown, dry diluvial soils in Brandenburg, Germany, the calculated tree vigour suppression was 46\% compared to the potential tree vigour on no-replant soil in the same field. The objective of this study was to establish an algorithm for estimating growth suppression in orchards irrespective of the unknowns in the complex causal relationship by assessing plant-soil interaction in the orchard several years after planting. Based on a comparison between no-replant and replant soils, the Alternaria group (Ag) was identified as a soil-fungal population responding to replant with abundance. The trunk cross-sectional area (CSA) was found to be a practical and robust parameter representing below-ground and above-ground tree performance. Suppression of tree vigour was therefore calculated by dividing the two inversely related parameters, Q = ln(Ag)/CSA, as a function of soil-fungal proportions and plant responses at the single-tree level. On this basis, five clusters of tree vigour suppression (Q) were defined: (1) no tree vigour suppression/vital (0\%), (2) escalating (- 38\%), (3) strong (- 53\%), (4) very strong (- 62\%), and (5) critical (- 74\%). By calculating Q at the level of the single tree, trees were clustered according to tree vigour suppression. The weighted frequency of clusters in the field allowed replant impact to be quantified at field level. Applied to a case study on sandy brown, dry diluvial soils in Brandenburg, Germany, the calculated tree vigour suppression was 46\% compared to the potential tree vigour on no-replant soil in the same field. Methods: Between early April and late-July 2020, a total of 3654 individuals in Lower Saxony, Germany, participated in an online symptom-tracking survey conducted through the app covid-nein-danke.de. The questionnaire comprised items on typical COVID-19 symptoms, age range, gender, employment in patient-facing healthcare, housing status, postal code, previous illnesses, permanent medication, vaccination status, results of reverse transcription polymerase chain reaction (RT-PCR) and antibody tests for COVID-19 diagnosis, and consequent COVID-19 treatment if applicable. Odds ratio estimates with corresponding 95\% CIs were computed for each medication and symptom by using logistic regression models. Results: Data analysis suggested a statistically significant inverse relationship between typical COVID-19 symptoms self-reported by the participants and self-reported statin therapy and, to a lesser extent, antihypertensive therapy. When COVID-19 diagnosis was based on restrictive symptom criteria (ie, presence of 4 out of 7 symptoms) or a positive RT-PCR test, a statistically significant association was found solely for statins (odds ratio 0.28, 95\% CI 0.1-0.78). Conclusions: Individuals taking statin medication are more likely to have asymptomatic COVID-19, in which case they may be at an increased risk of transmitting the disease unknowingly. We suggest that the results of this study be incorporated into symptoms-based surveillance and decision-making protocols in regard to COVID-19 management. In addition, two kinetic parameters (mechanical work [W], maximal force [F-max]) were analysed using the judo-specific measurement and information system JERGo (c). For this purpose, athletes were asked to do their judo-specific pulling movements while standing and with a dynamic change of position (i.e. Morote-seoi-nage). Results: Regarding pulling movements while standing, significant correlations (0.62 <= r(P) <= 0.72) were found between isokinetic tests (PITFlex, PITEx, PITRot) and mechanical work during judo-specific movement. Further, significant correlations (0.59 <= r(P) <= 0.65) were detected between isokinetic tests (PITEx, PITRot) and judo-specific pulling movements (Fmax). Regarding pulling movements with a change of position, significant correlations (0.47 <= r(P) <= 0.88) were observed between isokinetics (PITFlex, PITEx, PITRot) and the kinetic pulling parameters (W, Fmax), irrespective of the examined arm. No significant differences in magnitude of correlation coefficients were found between PIT of the trunk flexors, extensors, and rotators and judo-specific movements. Further, the regression analysis indicated that PIT of the trunk extensors is the single best predictor for mechanical work during pulling movements while standing (46.9 \%). Trunk rotator PIT is the single best predictor for mechanical work during pulling movements with a change of position (69.4 \%). We used eye-tracking to measure their fixation duration on facial expressions depicting different emotions, including negative (fear, anger), positive (happy), and neutral faces. Both Germans and the not equal Akhoe Hai parallel to om gazed longer at fearful faces, but shorter on angry faces, challenging the notion of a general bias toward negative emotions. For happy faces, fixation durations varied between the two groups, suggesting more flexibility in the response to positive emotions. One use case of ELISA is to differentiate between different states (e.g. healthy vs. diseased). Another goal is to quantitatively assess the biomarker in question, like autoantibodies. Thus, the ELISA technology is used for the discovery and verification of new autoantibodies, too. Of key interest, however, is the development of immunoassays for the sensitive and specific detection of such biomarkers at early disease stages. Therefore, users have to deal with many parameters, such as buffer systems or antigen-autoantibody interactions, to successfully establish an ELISA. Often, fine-tuning like testing of several blocking substances is performed to yield high signal-to-noise ratios.
Methods: We developed an ELISA to detect IgA and IgG autoantibodies against chitinase-3-like protein 1 (CHI3L1), a newly identified autoantigen in inflammatory bowel disease (IBD), in the serum of control and disease groups (n = 23, respectively). Microwell plates with different surface modifications (PolySorp and MaxiSorp coating) were tested to detect reproducibility problems.
Results: We found a significant impact of the surface properties of the microwell plates. IgA antibody reactivity was significantly lower, since it was in the range of background noise, when measured on MaxiSorp coated plates (p < 0.0001). The IgG antibody reactivity did not differ on the diverse plates, but the plate surface had a significant influence on the test result (p = 0.0005).
Conclusion: With this report, we want to draw readers' attention to the properties of solid phases and their effects on the detection of autoantibodies by ELISA. The affective-reflective theory of physical inactivity and exercise (ART) proposes a theoretical framework for explaining how the automatic affective process (type‑1 process) will influence exercise behavior, i.e., through the automatic activation of exercise-related associations and a subsequent affective valuation of exercise. This study aimed to empirically test this assumption of the ART with data from 69 study participants. A single-measurement study, including within-subject experimental variation, was conducted. Automatic associations with exercise were first measured with a single-target implicit association test. The somato-affective core of the participants' automatic valuation of exercise-related pictures was then assessed via heart rate variability (HRV) analysis, and the affective valence of the valuation was tested with a facial expression (FE; smile and frown) task. Exercise behavior was assessed via self-report. Multiple regression (path) analysis revealed that automatic associations predicted HRV reactivity (β = -0.24, p = .044); the signs of the correlation between automatic associations and the smile FE score was in the expected direction but remained nonsignificant (β = -0.21, p = .078). HRV reactivity predicted self-reported exercise behavior (β = -0.28, p = .013) (the same pattern of results was achieved for the frown FE score). The HRV-related results illustrate the potential role of automatic negative affective reactions to the thought of exercise as a restraining force in exercise motivation. For better empirical distinction between the two ART type‑1 process components, automatic associations and the affective valuation should perhaps be measured separately in the future. The power spectrum was analyzed for theta (4-7 Hz) and alpha-2 (10-12 Hz) frequency bands in pre-defined frontal, central, and parietal clusters of electrocortical sources. Repeated measures analysis of variance (rmANOVA) showed a significant main effect of task difficulty for postural sway (p < 0.001; d = 6.36). Concomitantly, the power spectrum changed in frontal, bilateral central, and bilateral parietal clusters. RmANOVAs revealed significant main effects of task difficulty for theta band power in the frontal (p < 0.001, d = 1.80) and both central clusters (left: p < 0.001, d = 1.49; right: p < 0.001, d = 1.42) as well as for alpha-2 band power in both parietal clusters (left: p < 0.001, d = 1.39; right: p < 0.001, d = 1.05) and in the central right cluster (p = 0.005, d = 0.92). Increases in theta band power (frontal, central) and decreases in alpha-2 power (central, parietal) with increasing balance task difficulty may reflect increased attentional processes and/or error monitoring as well as increased sensory information processing due to increasing postural demands. Despite the countering nature of these alternative narratives, they strengthen the societal expectation of professional success as a key pathway to inclusion, thereby reinforcing the rigidity of this narrative. Small microplastics enter the water either directly or accumulate through disintegration of larger plastic particles. These particles might then be ingested by filter-feeding zooplankton, such as rotifers. Particles released into the water may also interact with the biota through the formation of aggregates, which might alter the uptake by zooplankton. In this study, we tested for size-specific aggregation of polystyrene microspheres and their ingestion by a common freshwater rotifer Brachionus calyciflorus. The ingestion of three sizes of polystyrene microspheres (MS) 1-, 3-, and 6-mu m was investigated. Each MS size was tested in combination with three different treatments: MS as the sole food intake, MS in association with food algae and MS aggregated with biogenic matter. After 72 h incubation in pre-filtered natural river water, the majority of the 1-mu m spheres occurred as aggregates. The larger the particles, the higher the relative number of single particles and the larger the aggregates. All particles were ingested by the rotifer following a Type-II functional response. The presence of algae did not influence the ingestion of the MS for all three sizes. The biogenic aggregation of microspheres led to a significant size-dependent alteration in their ingestion. Rotifers ingested more microspheres (MS) when exposed to aggregated 1- and 3-mu m MS as compared to single spheres, whereas fewer aggregated 6-mu m spheres were ingested. This indicates that the small particles when aggregated were in an effective size range for Brachionus, while the aggregated larger spheres became too large to be efficiently ingested. These observations provide the first evidence of a size- and aggregation-dependent feeding interaction between microplastics and rotifers. Microplastics when aggregated with biogenic particles in a natural environment can rapidly change their size-dependent availability. The dye was excited via RET from the UCNPs which was excited in the near-infrared (NIR). The change of the dye diffusion speed (free vs coupled) was investigated by MST. RET shows significant changes in the decay times of the dye as well as of the UCNPs. MST reveals significant changes in the diffusion speed. A unique amphiphilic coating polymer (customized mussel protein (CMP) polymer) for UCNP surface coating was used, which mimics blood protein adsorption and mussel food protein adhesion to transfer the UCNP into the aqueous phase and to allow surface functionalization. The CMP provides very good water dispersibility to the UCNPs and minimizes ligand exchange and subsequent UCNP aging reactions because of the interlinkage of the CMP on the UCNP surface. Moreover, CMP provides N-3-functional groups for dick chemistry-based functionalization demonstrated with the dye 5-carboxytetramethylrhodamine (TAMRA). This establishes the principle coupling scheme for suitable biomarkers such as antibodies. The CMP provides very stable aqueous UCNP dispersions that are storable up to 3 years in a fridge at 5 degrees C without dissolution or coagulation. The outstanding properties of CMP in shielding the UCNP from unwanted solvent effects is reflected in the distinct increase of the photoluminescence decay times after UCNP functionalization. The UCNP-to-TAMRA energy transfer is also spectroscopically investigated at low temperatures (4-200 K), revealing that one of the two green Er(III) emission bands contributes the major part to the energy transfer. The TAMRA fluorescence decay time increases by a factor of 9500 from 2.28 ns up to 22 mu s due to radiationless energy transfer from the UCNP after NIR excitation of the latter. However, we know little about whether key interlocutors in national governments perceive the International Financial Institutions as biased actors who do the bidding for powerful member states or as impartial executors of policy. In order to better understand these perceptions, we surveyed high-level civil servants who are chiefly responsible for four policy areas from more than 100 countries. We found substantial variations in impartiality perceptions. What explains these variations? By developing an argument of selective awareness, we extend rationalist and ideational perspectives on IO impartiality to explain domestic perceptions. Using novel survey data, we test whether staffing underrepresentation, voting underrepresentation, alignment to the major shareholders and overlapping economic policy paradigms are associated with impartiality perceptions. We find substantial evidence that shared economic policy paradigms influence impartiality perceptions. In contrast, the SSA over the IGP does not exhibit a pronounced spatial variation. BC dominates (>= 75\%) the aerosol absorption over the IGP and the Himalayan foothills throughout the year. Higher BC concentration at elevated locations in the Himalayas leads to lower SSA at elevated locations in the Himalayas. The contribution of dust to aerosol absorption is higher throughout the year over the IGP than over the Himalayan foothills. The aerosol absorption over South Asia is very high, exceeding available observations over East Asia, and also exceeds previous model estimates. The results revealed increased post-error slowing along with reduced P300 and error-related negativity (ERN) amplitudes in high (vs. low) procrastination participants-effects that indicate impaired attention and error-related processing in this group. This pattern of results did not differ as a function of task difficulty and motivation condition. However, when the task got more difficult executive attention deficits became even more apparent at the behavioral level in high procrastinators, as indexed by increased reaction time variability. We also investigated whether BDNF was associated with somatic comorbidity burden, acute coronary syndrome (ACS) or congestive heart failure (CHF). Methods: The following variables were assessed for 225 hospitalised patients with CHD: BDNF concentrations, depression [Patient Health Questionnaire-9 (PHQ-9)], somatic comorbidity (Charlson Comorbidity Index), CHF, ACS, platelet count, smoking status and antidepressant treatment. Results: Regression models revealed that BDNF was not associated with severity of depression. Although depressed patients (PHQ-9 score >7) had significantly lower BDNF concentrations compared to non-depressed patients (p = 0.04), this was not statistically significant after controlling for confounders (p = 0.15). Cognitive-affective symptoms and somatic comorbidity burden each closely missed a statistically significant association with BDNF concentrations (p = 0.08, p = 0.06, respectively). BDNF was reduced in patients with CHF (p = 0.02). There was no covariate-adjusted, significant association between BDNF and ACS. Conclusion: Serum BDNF concentrations are associated with cardiovascular dysfunction. We demonstrate how the shapes of spherical viruses can be understood from the perspective of elasticity theory of thin two-component shells. We develop a theory of shape transformations of an icosahedral shell upon addition of a softer, but still crystalline, material onto its surface. We show how the soft component "invades" the regions with the highest elastic energy and stress imposed by the 12 topological defects on the surface. We explore the phase diagram as a function of the surface fraction of the soft material, the shell size, and the incommensurability of the elastic moduli of the rigid and soft phases. We find that, as expected, progressive filling of the rigid shell by the soft phase starts from the most deformed regions of the icosahedron. With a progressively increasing soft-phase coverage, the spherical segments of domes are filled first (12 vertices of the shell), then the cylindrical segments connecting the domes (30 edges) are invaded, and, ultimately, the 20 flat faces of the icosahedral shell tend to be occupied by the soft material. We present a detailed theoretical investigation of the first two stages of this invasion process and develop a model of morphological changes of the cone structure that permits noncircular cross sections. Plain Language Summary Increasing air and sea surface temperatures at high latitudes leads to accelerated thaw, destabilization, and erosion of perennially frozen soils (i.e., permafrost), which are often rich in organic carbon. Coastal erosion leads to an increased mobilization of organic carbon into the Arctic Ocean, which there can be converted into greenhouse gases and may therefore contribute to further warming. Carbon decomposition can be limited if organic matter is efficiently deposited on the seafloor, buried in marine sediments, and thus removed from the short-term carbon cycle. Basins, canyons, and troughs near the coastline can serve as sediment traps and potentially accommodate large quantities of organic carbon along the Arctic coast. Here we use biomarkers (source-specific molecules), stable carbon isotopes, and radiocarbon to identify the sources of organic carbon in the nearshore zone of the southern Canadian Beaufort Sea near Herschel Island. Methods: Twenty-eight psychotherapytrainees (M= 28.54 years of age,SD= 3.19) participated as blind raters. They evaluated six video-recorded therapy segments of trained SPsand real patients using the Authenticity of Patient Demonstrations Scale. Results: The authenticity scores of real patients and SPs did notdiffer (p= .43). The descriptive results indicated that the highest score of authenticity was given to an SP. Further, the real patients did notdiffer significantly from the SPs concerning perceived impairment (p= .33) and the likelihood of being a real patient (p= .52). Conclusions: The current results suggest that psychotherapy trainees were unable to distinguish the SPs from real patients. We therefore stronglyrecommend incorporating training SPs before application. This work addresses the question of structural identification of both, the fluorescent and non-fluorescent reaction products obtained in the thermal reaction of citric acid and urea. The reaction products produced during autoclave-microwave reactions in the melt were thoroughly investigated as a function of the reaction temperature and the reaction products were subsequently separated by a series of solvent extractions and column chromatography. The evolution of a green molecular fluorophore, namely HPPT, was confirmed and a full characterization study on its structure and photophysical properties was conducted. The additional blue fluorescence is attributed to oligomeric ureas, which was confirmed by complementary optical and structural characterization. These two components form strong hydrogen-bond networks which eventually react to form solid, semi-crystalline particles with a size of similar to 7 nm and an elemental composition of 46\% C, 22\% N, and 29\% O. The change in availability and type of carcasses throughout ecosystems can involve negative effects for scavenging communities. To address this issue, there have been recent conservation management measures of carrion provision in natural systems. However, the optimal conditions under which exposing carcasses to optimize conservation outcomes are still limited. Here, we used camera traps throughout elevational and vegetational gradients to monitor the consumption of 48 deer carcasses over a study period of six years by evaluating 270,279 photographs resulting out of 15,373 trap nights. We detected 17 species visiting carcass deployments, including five endangered species. Our results show that large carcasses, the winter season, and a heterogeneous surrounding habitat enhanced the frequency of carcass visits and the species richness of scavenger assemblages. Contrary to our expectations, carcass species, condition (fresh/frozen), and provision schedule (continuous vs single exposure) did not influence scavenging frequency or diversity. The carcass visitation frequency increased with carcass mass and lower temperatures. The effect of large carcasses was especially pronounced for mesopredators and the Eurasian lynx (Lynx lynx ). Lynx were not too influenced in its carrion acquisition by the season, but exclusively preferred remote habitats containing higher forest cover. Birds of prey, mesopredators, and top predators were also positively influenced by the visiting rate of ravens (Corvus corax ), whereas no biotic or abiotic preferences were found for wild boars (Sus scrofa ). Age-related changes in cellular function were determined by senescence-associated beta-galactosidase activity, increasing presence of cell cycle regulators, such as p16, p53, and p21, accumulation of protein aggregates, and restricted proteolysis in terms of decreasing (macro-)autophagy. Furthermore, the culture system was functionally characterized for alterations in cell morphology and contractility. An increase in cellular size associated with induced expression of atrial natriuretic peptides demonstrated a stress-induced hypertrophic phenotype in neonatal cardiomyocytes. In this paper, we use a rich data set to compare participants of a German start-up subsidy program for unemployed individuals to a group of regular founders who started from non-unemployment and did not receive the subsidy. The data allows us to analyze their business performance up until 40 months after business formation. We find that formerly subsidized founders lag behind not only in survival and job creation, but especially also in innovation activities. The gaps in these business outcomes are relatively constant or even widening over time. Hence, we do not see any indication of catching up in the longer run. While the gap in survival can be entirely explained by initial differences in observable start-up characteristics, the gap in business development remains and seems to be the result of restricted access to capital as well as differential business strategies and dynamics. The role of Nf1 in muscle and the cause for the NF1-associated myopathy are mostly unknown. Methods To dissect the function ofNf1in muscle, we created muscle-specific knockout mouse models for NF1, inactivatingNf1in the prenatal myogenic lineage either under the Lbx1 promoter or under the Myf5 promoter. Mice were analysed during prenatal and postnatal myogenesis and muscle growth. Results Nf1(Lbx1)and Nf1(Myf5)animals showed only mild defects in prenatal myogenesis. Nf1(Lbx1)animals were perinatally lethal, while Nf1(Myf5)animals survived only up to approximately 25 weeks. A comprehensive phenotypic characterization of Nf1(Myf5)animals showed decreased postnatal growth, reduced muscle size, and fast fibre atrophy. Proteome and transcriptome analyses of muscle tissue indicated decreased protein synthesis and increased proteasomal degradation, and decreased glycolytic and increased oxidative activity in muscle tissue. High-resolution respirometry confirmed enhanced oxidative metabolism in Nf1(Myf5)muscles, which was concomitant to a fibre type shift from type 2B to type 2A and type 1. Moreover, Nf1(Myf5)muscles showed hallmarks of decreased activation of mTORC1 and increased expression of atrogenes. Remarkably, loss of Nf1 promoted a robust activation of AMPK with a gene expression profile indicative of increased fatty acid catabolism. Additionally, we observed a strong induction of genes encoding catabolic cytokines in muscle Nf1(Myf5)animals, in line with a drastic reduction of white, but not brown adipose tissue. Conclusions Our results demonstrate a cell autonomous role for Nf1 in myogenic cells during postnatal muscle growth required for metabolic and proteostatic homeostasis. Cell-free systems are emerging as an attractive alternative for the production of proteins due to their flexible nature without any cell membrane constraints. In a bioproduction context, open systems based on cell lysates derived from different sources, and with batch-to-batch consistency, have acted as a catalyst for cell-free synthesis of target proteins. Most importantly, proteins can be processed for downstream applications like purification and functional analysis without the necessity of transfection, selection, and expansion of clones. In the last 5 years, there has been an increased availability of new cell-free lysates derived from multiple organisms, and their use for the synthesis of a diverse range of proteins. Despite this progress, major challenges still exist in terms of scalability, cost effectiveness, protein folding, and functionality. In this review, we present an overview of different cell-free systems derived from diverse sources and their application in the production of a wide spectrum of proteins. Further, this article discusses some recent progress in cell-free systems derived from Chinese hamster ovary and Sf21 lysates containing endogenous translocationally active microsomes for the synthesis of membrane proteins. We particularly highlight the usage of internal ribosomal entry site sequences for more efficient protein production, and also the significance of site-specific incorporation of non-canonical amino acids for labeling applications and creation of antibody drug conjugates using cell-free systems. Our main findings are that (1) the ML models achieve higher prediction accuracy than linear regression, (2) the results reflect basic hydrological principles, but (3) further inference is hindered by the heterogeneity of results across algorithms. Thus, we conclude that the problem of equifinality as known from classical hydrological modeling also exists for ML and severely hampers its potential for inference.
